  19. Genomics:GTL Contractor-Grantee Workshop IV and Metabolic Engineering Working Group Inter-Agency Conference on Metabolic Engineering 2006

    SciTech Connect (OSTI)

Mansfield, Betty Kay; Martin, Sheryl A

    2006-02-01

Welcome to the 2006 joint meeting of the fourth Genomics:GTL Contractor-Grantee Workshop and the six Metabolic Engineering Working Group Inter-Agency Conference. The vision and scope of the Genomics:GTL program continue to expand and encompass research and technology issues from diverse scientific disciplines, attracting broad interest and support from researchers at universities, DOE national laboratories, and industry. Metabolic engineering's vision is the targeted and purposeful alteration of metabolic pathways to improve the understanding and use of cellular pathways for chemical transformation, energy transduction, and supramolecular assembly. These two programs have much complementarity in both vision and technological approaches, as reflected in this joint workshop. GLT's challenge to the scientific community remains the further development and use of a broad array of innovative technologies and computational tools to systematically leverage the knowledge and capabilities brought to us by DNA sequencing projects. The goal is to seek a broad and predictive understanding of the functioning and control of complex systems--individual microbes, microbial communities, and plants. GTL's prominent position at the interface of the physical, computational, and biological sciences is both a strength and challenge. Microbes remain GTL's principal biological focus. In the complex 'simplicity' of microbes, they find capabilities needed by DOE and the nation for clean and secure energy, cleanup of environmental contamination, and sequestration of atmospheric carbon dioxide that contributes to global warming. An ongoing challenge for the entire GTL community is to demonstrate that the fundamental science conducted in each of your research projects brings us a step closer to biology-based solutions for these important national energy and environmental needs.

The Idaho National Engineering Laboratory (INEL) Technology Logic Diagram (TLD) was developed to provide a decision support tool that relates Environmental Restoration (ER) and Waste Management (WM) problems at the INEL to potential technologies that can remediate these problems. The TLD identifies the research, development, demonstration, testing, and evaluation needed to develop these technologies to a state that allows technology transfer and application to an environmental restoration need. It is essential that follow-on engineering and system studies be conducted to build on the output of this project. These studies will begin by selecting the most promising technologies identified in this TLD and finding an optimum mix of technologies that will provide a socially acceptable balance between cost and risk to meet the site windows of opportunity. The TLD consists of three separate volumes: Volume I includes the purpose and scope of the TLD, a brief history of the INEL Waste Area Groups, and environmental problems they represent. A description of the TLD, definitions of terms, a description of the technology evaluation process, and a summary of each subelement, is presented. Volume II describes the overall layout and development of the TLD in logic diagram format. This section addresses the environmental restoration of contaminated INEL sites. Volume III (this volume) provides the Technology Evaluation Data Sheets (TEDS) for Environmental Restoration and Waste Management (EM) activities that are reference by a TEDS code number in Volume II. Each of these sheets represents a single logic trace across the TLD. These sheets contain more detail than provided for technologies in Volume II. Data sheets are arranged alphanumerically by the TEDS code number in the upper right corner of each sheet.

The lubricating oil system provides a means to introduce a lubricant in the form of a film to reduce friction and wear between surfaces that bear against each other as they move.1 The oil film which is established also cools the parts by carrying generated heat away from hot surfaces, cleans and carries dirt or metal wear particles to the filter media, and helps seal the piston to the cylinder during combustion. Most systems are pressure lubricated and distribute oil under pressure to bearings, gears, and power assemblies. Lubricating oil usually reaches main, connecting rod, and camshaft bearings through drilled passages in the cylinder block and crankshaft or through piping and common manifolds. Many parts rely on oil for cooling, so if the lube oil system fails to perform its function the engine will overheat. Metal to metal surfaces not separated by a thin film of oil rapidly build up frictional heat. As the metals reach their melting point, they tend to weld together in spots or streaks. Lube oil system failures can cause significant damage to an engine in a short period of time. Proper maintenance and operation of the lubricating oil system is essential if your engine is to accomplish its mission.

A historic return interval of 100 years for large fires in the U.S. southwestern deserts is being replaced by one where fires may reoccur as frequently as every 20 to 30 years. The shortened return interval, which translates to an increase in fires, has implications for management of Soil Corrective Action Units (CAUs) and Corrective Action Sites (CASs) for which the Department of Energy, National Nuclear Security Administration Nevada Field Office has responsibility. A series of studies was initiated at uncontaminated analog sites to better understand the possible impacts of erosion and transport by wind and water should contaminated soil sites burn. The first of these studies was undertaken at the Jacob Fire site approximately 12 kilometers (7.5 miles) north of Hiko, Nevada. A lightning-caused fire burned approximately 200 hectares during August 6-8, 2008. The site is representative of a transition between Mojave and Great Basin desert ecoregions on the Nevada National Security Site (NNSS), where the largest number of Soil CAUs/CASs are located. The area that burned at the Jacob Fire site was primarily a Coleogyne ramosissima (blackbrush) and Ephedra nevadensis (Mormon tea) community, also an abundant shrub assemblage in the similar transition zone on the NNSS. This report summarizes three years of measurements after the fire. Seven measurement campaigns at the Jacob Fire site were completed. Measurements were made on burned ridge (upland) and drainage sites, and on burned and unburned sites beneath and between vegetation. A Portable In-Situ Wind Erosion Lab (PI-SWERL) was used to estimate emissions of suspended particles at different wind speeds. Context for these measurements was provided through a meteorological tower that was installed at the Jacob Fire site to obtain local, relevant environmental parameters. Filter samples, collected from the exhaust of the PI-SWERL during measurements, were analyzed for chemical composition. Runoff and water erosion were

International Offshore Chemicals (IOC; Aberdeen, UK), a project development and management company, has formed a consortium to seek permission to build a 2,500-m.t./day methanol plant near the St. Fergus terminal in northeast Scotland. Robert Spiers, technical adviser to IOC, tells CW that the $255-million plant, based on the ICI process, would begin production in second-quarter 1999. The key market would be Europe. Jacobs Engineering and Davy are negotiating for the construction contract. Spiers says Europe imports 3 million m.t./year of methanol, and-even with Statoil`s plant under construction in Norway-there is room for more. ICI is the UK`s only producer of methanol. Another company, reportedly Kemira, may build a 360,000-m.t./year ammonia plant nearby if the methanol scheme goes ahead.

Geologic ages and accumulation rates, estimated from regressions, were used to evaluate measured ages and interpreted stratigraphic and structural relations of basalt and sediment in the unsaturated zone and the Snake River Plain aquifer at the Idaho National Engineering Laboratory (INEL) in eastern Idaho. Geologic ages and accumulation rates were estimated from standard linear regressions of 21 mean potassium-argon (K-Ar) ages, selected mean paleomagnetic ages, and cumulative depths of a composite stratigraphic section composed of complete intervals of basalt and sediment that were deposited in areas of past maximum subsidence. Accumulation rates also were estimated from regressions of stratigraphic intervals in three wells in and adjacent to an area of interpreted uplift at and near the Idaho Chemical Processing Plant (ICPP) and the Test Reactor Area (TRA) to allow a comparison of rates in areas of past uplift and subsidence. Estimated geologic ages range from about 200 thousand to 1.8 million years before present and are reasonable approximations for the interval of basalt and sediment above the effective base of the aquifer, based on reported uncertainties of corresponding measured ages. Estimated ages between 200 and 800 thousand years are within the range of reported uncertainties for all 15 K-Ar ages used in regressions and two out of three -argon ({sup 40}Ar/{sup 39}Ar) ages of duplicate argon samples. Two sets of estimated ages between 800 thousand and 1.8 million years are within the range of reported uncertainties for all seven K-Ar ages used in regressions, which include one shared age of about 800 thousand years. Two sets of ages were estimated for this interval because K-Ar ages make up two populations that agree with previous and revised ages of three paleomagnetic subchrons. The youngest set of ages is consistent with a K-Ar age from the effective base of the aquifer that agrees with previous ages of the Olduvai Normal-Polarity Subchron.

The group I was responsible for studying has to remain confidential so as not to affect the current project. All dates, reference links and authors, and other distinguishing characteristics of the original group have been removed from this report. All references to the name of this group or the individual splinter groups has been changed to 'Group X'. I have been collecting texts from a variety of sources intended for the use of recruiting and radicalizing members for Group X splinter groups for the purpose of researching the motivation and intent of leaders of those groups and their influence over the likelihood of group radicalization. This work included visiting many Group X websites to find information on splinter group leaders and finding their statements to new and old members. This proved difficult because the splinter groups of Group X are united in beliefs, but differ in public opinion. They are eager to tear each other down, prove their superiority, and yet remain anonymous. After a few weeks of intense searching, a list of eight recruiting texts and eight radicalizing texts from a variety of Group X leaders were compiled.

    2015-02-01

Galaxy groups can be characterized by the radius of decoupling from cosmic expansion, the radius of the caustic of second turnaround, and the velocity dispersion of galaxies within this latter radius. These parameters can be a challenge to measure, especially for small groups with few members. In this study, results are gathered pertaining to particularly well-studied groups over four decades in group mass. Scaling relations anticipated from theory are demonstrated and coefficients of the relationships are specified. There is an update of the relationship between light and mass for groups, confirming that groups with mass of a few times 10{sup 12}M{sub ?} are the most lit up while groups with more and less mass are darker. It is demonstrated that there is an interesting one-to-one correlation between the number of dwarf satellites in a group and the group mass. There is the suggestion that small variations in the slope of the luminosity function in groups are caused by the degree of depletion of intermediate luminosity systems rather than variations in the number per unit mass of dwarfs. Finally, returning to the characteristic radii of groups, the ratio of first to second turnaround depends on the dark matter and dark energy content of the universe and a crude estimate can be made from the current observations of ?{sub matter}?0.15 in a flat topology, with a 68% probability of being less than 0.44.

    2010-01-14

Broad Funding Opportunity Announcement Project: MSU is developing a new engine for use in hybrid automobiles that could significantly reduce fuel waste and improve engine efficiency. In a traditional internal combustion engine, air and fuel are ignited, creating high-temperature and high-pressure gases which expand rapidly. This expansion of gases forces the engine’s pistons to pump and powers the car. MSU’s engine has no pistons. It uses the combustion of air and fuel to build up pressure within the engine, generating a shockwave that blasts hot gas exhaust into the blades of the engine’s rotors causing them to turn, which generates electricity. MSU’s redesigned engine would be the size of a cooking pot and contain fewer moving parts—reducing the weight of the engine by 30%. It would also enable a vehicle that could use 60% of its fuel for propulsion.

Thermoacoustic engines, or acoustic heat engines, are energy-conversion devices that achieve simplicity and concomitant reliability by use of acoustic technology. Their efficiency can be a substantial fraction of Carnot's efficiency. In thermoacoustic prime movers, heat flow from a high-temperature source to a low-temperature sink generates acoustic power (which may be converted to electric power using a transducer). In thermoacoustic heat pumps and refrigerators, acoustic power is used to pump heat from a low-temperature source to a high-temperature sink. This review teaches the fundamentals of thermoacoustic engines, by analysis, intuition, and example.

An engine based on a reciprocating piston engine that extracts work from pressurized working fluid. The engine includes a harmonic oscillator inlet valve capable of oscillating at a resonant frequency for controlling the flow of working fluid into of the engine. In particular, the inlet valve includes an inlet valve head and a spring arranged together as a harmonic oscillator so that the inlet valve head is moveable from an unbiased equilibrium position to a biased closed position occluding an inlet. Upon releasing the inlet valve the inlet valve head undergoes a single oscillation past the equilibrium positio to a maximum open position and returns to a biased return position close to the closed position to choke the flow and produce a pressure drop across the inlet valve causing the inlet valve to close. Protrusions carried either by the inlet valve head or piston head are used to bump open the inlet valve from the closed position and initiate the single oscillation of the inlet valve head, and protrusions carried either by the outlet valve head or piston head are used to close the outlet valve ahead of the bump opening of the inlet valve.

    2005-09-30

The goal of this project was to develop methods to optimize and control Homogeneous-Charge Compression Ignition (HCCI) engines, with emphasis on diesel-fueled engines. HCCI offers the potential of nearly eliminating IC engine NOx and particulate emissions at reduced cost over Compression Ignition Direct Injection engines (CIDI) by controlling pollutant emissions in-cylinder. The project was initiated in January, 2002, and the present report is the final report for work conducted on the project through December 31, 2004. Periodic progress has also been reported at bi-annual working group meetings held at USCAR, Detroit, MI, and at the Sandia National Laboratories. Copies of these presentation materials are available on CD-ROM, as distributed by the Sandia National Labs. In addition, progress has been documented in DOE Advanced Combustion Engine R&D Annual Progress Reports for FY 2002, 2003 and 2004. These reports are included as the Appendices in this Final report.

    SciTech Connect (OSTI)

    LOY,DOUGLAS A.; SHEA,KENNETH J.

    2000-05-09

Bridged polysilsesquioxanes are a class of hybrid organic-inorganic materials that permit molecular engineering of bulk properties including porosity. Prepared by sol-gel polymerization of monomers with two or more trialkoxysilyl groups, the materials are highly cross-linked amorphous polymers that are readily obtained as gels. The bridging configuration of the hydrocarbon group insures that network polymers are readily formed and that the organic functionality is homogeneously distributed throughout the polymeric scaffolding at the molecular level. This permits the bulk properties, including surface area, pore size, and dielectric constant to be engineered through the selection of the bridging organic group. Numerous bridging groups have been incorporated. This presentation will focus on the effects that the length, flexibility, and substitution geometry of the hydrocarbon bridging groups have on the properties of the resulting bridged polysilsesquioxanes. Details of the preparation, characterization, and some structure property relationships of these bridged polysilsesquioxanes will be given.

  14. Bellcrank mechanisms for Stirling engines

    SciTech Connect (OSTI)

    Senft, J.R.; Senft, V.J.

    1996-12-31

This paper describes a family of linkage drive systems for Stirling engines containing several new members. These mechanisms are adaptable to all three configurations of Stirling engine, impose minimal side loads on pistons and displacer rods, and include compact forms suitable for pressurized high performance engines. This group of drive systems is generated by a simple common scheme. Near sinusoidal motion is taken from a crankshaft carrying a single crankpin by two connecting rods each driving a bellcrank. The stationary pivots of the bellcranks are located so that their oscillatory motion has the phase angle separation required between the piston and displacer. The bellcranks are further configured to bring the third pin motion to a location suitable for coupling with the piston or displacer of the engine in a way which minimizes side loading. The paper presents a number of new linkage drives from the dual bellcrank family and indicates how they are embodied in beta and alpha type Stirling engines. The paper includes a design for a small multipurpose engine incorporating one of the subject mechanisms.

Ceramic and ceramic-coated components will be of increasing importance in the advanced engines now under development. Ceramics enable engines to run at much higher temperatures than the superalloys in more conventional engines can. The two options for noncontact high-temperature measurements of ceramic components are pyrometry and phosphor thermometry. This article describes how when properly applied as a thin coating, thermally sensitive phosphors can monitor the temperature of ceramic surfaces inside an engine.

    1993-11-09

The treatment of mixed waste remains one of this country`s most vexing environmental problems. Mixed waste is the combination of radioactive waste and hazardous waste, as defined by the Resource Conservation and Recovery Act (RCRA). The Department of Energy (DOE), as the country`s largest mixed waste generator, responsible for 95 percent of the Nation`s mixed waste volume, is now required to address a strict set of milestones under the Federal Facility Compliance Act of 1992. DOE`s earlier failure to adequately address the storage and treatment issues associated with mixed waste has led to a significant backlog of temporarily stored waste, significant quantities of buried waste, limited permanent disposal options, and inadequate treatment solutions. Between May and November of 1993, the Mixed Waste Working Group brought together stakeholders from around the Nation. Scientists, citizens, entrepreneurs, and bureaucrats convened in a series of forums to chart a course for accelerated testing of innovative mixed waste technologies. For the first time, a wide range of stakeholders were asked to examine new technologies that, if given the chance to be tested and evaluated, offer the prospect for better, safer, cheaper, and faster solutions to the mixed waste problem. In a matter of months, the Working Group has managed to bridge a gap between science and perception, engineer and citizen, and has developed a shared program for testing new technologies.

    2009-09-30

Energy independence and fuel savings are hallmarks of the nations energy strategy. The advancement of natural gas reciprocating engine power generation technology is critical to the nations future. A new engine platform that meets the efficiency, emissions, fuel flexibility, cost and reliability/maintainability targets will enable American manufacturers to have highly competitive products that provide substantial environmental and economic benefits in the US and in international markets. Along with Cummins and Waukesha, Caterpillar participated in a multiyear cooperative agreement with the Department of Energy to create a 50% efficiency natural gas powered reciprocating engine system with a 95% reduction in NOx emissions by the year 2013. This platform developed under this agreement will be a significant contributor to the US energy strategy and will enable gas engine technology to remain a highly competitive choice, meeting customer cost of electricity targets, and regulatory environmental standard. Engine development under the Advanced Reciprocating Engine System (ARES) program was divided into phases, with the ultimate goal being approached in a series of incremental steps. This incremental approach would promote the commercialization of ARES technologies as soon as they emerged from development and would provide a technical and commercial foundation of later-developing technologies. Demonstrations of the Phase I and Phase II technology were completed in 2004 and 2008, respectively. Program tasks in Phase III included component and system development and testing from 2009-2012. Two advanced ignition technology evaluations were investigated under the ARES program: laser ignition and distributed ignition (DIGN). In collaboration with Colorado State University (CSU), a laser ignition system was developed to provide ignition at lean burn and high boost conditions. Much work has been performed in Caterpillars DIGN program under the ARES program. This work has

  4. Perturbing engine performance measurements to determine optimal engine control settings

    DOE Patents [OSTI]

Jiang, Li; Lee, Donghoon; Yilmaz, Hakan; Stefanopoulou, Anna

    2014-12-30

Methods and systems for optimizing a performance of a vehicle engine are provided. The method includes determining an initial value for a first engine control parameter based on one or more detected operating conditions of the vehicle engine, determining a value of an engine performance variable, and artificially perturbing the determined value of the engine performance variable. The initial value for the first engine control parameter is then adjusted based on the perturbed engine performance variable causing the engine performance variable to approach a target engine performance variable. Operation of the vehicle engine is controlled based on the adjusted initial value for the first engine control parameter. These acts are repeated until the engine performance variable approaches the target engine performance variable.
