    The human homolog of the yeast cse1 gene (CSE1L) is over-expressed in ovarian cancer. CSE1L forms complex with Ran and importin-α and has roles in nucleocytoplasmic traffic and gene expression. CSE1L accumulated in the nucleus of ovarian cancer cell lines, while it was localized also in the cytoplasm of other cancer cell lines. Nuclear localization depended on AKT, which was constitutively active in ovarian cancer cells, as the CSE1L protein translocated to the cytoplasm when AKT was inactivated. Moreover, the expression of a constitutively active AKT forced the translocation of CSE1L from the cytoplasm to the nucleus in other cancer cells. Nuclear accrual of CSE1L was associated to the nuclear accumulation of the phosphorylated Ran Binding protein 3 (RanBP3), which depended on AKT as well. Also in samples of human ovarian cancer, AKT activation was associated to nuclear accumulation of CSE1L and phosphorylation of RanBP3. Expression profiling of ovarian cancer cells after CSE1L silencing showed that CSE1L was required for the expression of genes promoting invasion and metastasis. In agreement, CSE1L silencing impaired motility and invasiveness of ovarian cancer cells. Altogether these data show that in ovarian cancer cells activated AKT by affecting RanBP3 phosphorylation determines the nuclear accumulation of CSE1L and likely the nuclear concentration of transcription factors conveying pro-oncogenic signals. - highlights: • CSE1L is a key player in nucleocytoplasmic traffic by forming complex with Ran. • AKT phosphorylates RanBP3 that regulates the nucleocytoplasmic gradient of Ran. • The activated oncogenic AKT drives the nuclear accumulation of CSE1L. • CSE1L in the nucleus up-regulates genes conveying pro-oncogenic signals. • CSE1L might contribute to tumor progression driven by the activated oncogenic AKT.

    An ultraviolet (UV) in-line Fraunhofer holography diagnostic has been developed for making high-resolution spatial measurements of ejecta particles traveling at many mm/?sec. This report will discuss the development of the diagnostic, including the high-powered laser system and high-resolution optical relay system. In addition, we will also describe the system required to reconstruct the images from the hologram and the corresponding analysis of those images to extract particles. Finally, we will present results from six high-explosive (HE), shock-driven Sn-ejecta experiments. Particle-size distributions will be shown that cover most of the ejecta velocities for experiments conducted in a vacuum, and helium gas environments. In addition, a modification has been made to the laser system that produces two laser pulses separated by 6.8 ns. This double-pulsed capability allows a superposition of two holograms to be acquired at two different times, thus allowing ejecta velocities to be measured directly. Results from this double-pulsed experiment will be described.

    Abstract Information system security risk, defined as the product of the monetary losses associated with security incidents and the probability that they occur, is a suitable decision criterion when considering different information system architectures. Risk assessment is the widely accepted process used to understand, quantify, and document the effects of undesirable events on organizational objectives so that risk management, continuity of operations planning, and contingency planning can be performed. One technique, the Cyberspace Security Econometrics System (CSES), is a methodology for estimating security costs to stakeholders as a function of possible risk postures. In earlier works, we presented a computational infrastructure that allows an analyst to estimate the security of a system in terms of the loss that each stakeholder stands to sustain, as a result of security breakdowns. Additional work has applied CSES to specific business cases. The current state-of-the-art of CSES addresses independent events.     Double wall technologies utilizing wood framing have been well-known and used in North American buildings for decades. Most of double wall designs use only natural materials such as wood products, gypsum, and cellulose fiber insulation, being one of few building envelope technologies achieving high thermal performance without use of plastic foams or fiberglass. Today, after several material and structural design modifications, these technologies are considered as highly thermally efficient, sustainable option for new constructions and sometimes, for retrofit projects. Following earlier analysis performed for U.S. Department of Energy by Fraunhofer CSE, this paper discusses different ways to build double walls and to optimize their thermal performance to minimize the space conditioning energy consumption. Description of structural configuration alternatives and thermal performance analysis are presented as well. Laboratory tests to evaluate thermal properties of used insulation and whole wall system thermal performance are also discussed in this paper. Finally, the thermal loads generated in field conditions by double walls are discussed utilizing results from a joined project performed by Zero Energy Building Research Alliance and Oak Ridge National Laboratory (ORNL), which made possible evaluation of the market viability of low-energy homes built in the Tennessee Valley. Experimental data recorded in two of the test houses built during this field study is presented in this work.

    In this paper we present a system for visualizing volume data from remote supercomputers (PermWeb). We have developed both parallel volume rendering algorithms, and the World Wide Web software for accessing the data at the remote sites. The implementation uses Hypertext Markup Language (HTML), Java, and Common Gateway Interface (CGI) scripts to connect World Wide Web (WWW) servers/clients to our volume renderers. The front ends are interactive Java classes for specification of view, shading, and classification inputs. We present performance results, and implementation details for connections to our computing resources at the University of California Santa Cruz including a MasPar MP-2, SGI Reality Engine-RE2, and SGI Challenge machines. We apply the system to the task of visualizing trabecular bone from finite element simulations. Fast volume rendering on remote compute servers through a web interface allows us to increase the accessibility of the results to more users. User interface issues, overviews of parallel algorithm developments, and overall system interfaces and protocols are presented.     Anisodamine, an antagonist of muscarinic acetylcholine receptors (mAChRs), has been used therapeutically to improve smooth muscle function, including microvascular, intestinal and airway spasms. Our previous studies have revealed that airway hyper-reactivity could be prevented by anisodamine. However, whether anisodamine prevents smoking-induced airway smooth muscle (ASM) cell proliferation remained unclear. In this study, a primary culture of rat ASM cells was used to evaluate an ASM phenotype through the ability of the cells to proliferate and express contractile proteins in response to cigarette smoke extract (CSE) and intervention of anisodamine. Our results showed that CSE resulted in an increase in cyclin D1 expression concomitant with the G0/G1-to-S phase transition, and high expression of M2 and M3. Functional studies showed that tracheal hyper-contractility accompanied contractile marker α-SMA high-expression. These changes, which occur only after CSE stimulation, were prevented and reversed by anisodamine, and CSE-induced cyclin D1 expression was significantly inhibited by anisodamine and the specific inhibitor U0126, BAY11-7082 and LY294002. Thus, we concluded that the protective and reversal effects and mechanism of anisodamine on CSE-induced events might involve, at least partially, the ERK, Akt and NF-κB signaling pathways associated with cyclin D1 via mAChRs. Our study validated that anisodamine intervention on ASM cells may contribute to anti-remodeling properties other than bronchodilation. -- Highlights: ► CSE induces tracheal cell proliferation, hyper-contractility and α-SMA expression. ► Anisodamine reverses CSE-induced tracheal hyper-contractility and cell proliferation. ► ERK, PI3K, and NF-κB pathways and cyclin D1 contribute to the reversal effect.

    Software lifecycles are becoming an increasingly important issue for computational science and engineering (CSE) software. The process by which a piece of CSE software begins life as a set of research requirements and then matures into a trusted high-quality capability is both commonplace and extremely challenging. Although an implicit lifecycle is obviously being used in any effort, the challenges of this process - respecting the competing needs of research vs. production - cannot be overstated. Here we describe a proposal for a well-defined software lifecycle process based on modern Lean/Agile software engineering principles. What we propose is appropriate for many CSE software projects that are initially heavily focused on research but also are expected to eventually produce usable high-quality capabilities. The model is related to TriBITS, a build, integration and testing system, which serves as a strong foundation for this lifecycle model, and aspects of this lifecycle model are ingrained in the TriBITS system. Here, we advocate three to four phases or maturity levels that address the appropriate handling of many issues associated with the transition from research to production software. The goals of this lifecycle model are to better communicate maturity levels with customers and to help to identify and promote Software Engineering (SE) practices that will help to improve productivity and produce better software. An important collection of software in this domain is Trilinos, which is used as the motivation and the initial target for this lifecycle model. However, many other related and similar CSE (and non-CSE) software projects can also make good use of this lifecycle model, especially those that use the TriBITS system. Indeed this lifecycle process, if followed, will enable large-scale sustainable integration of many complex CSE software efforts across several institutions.

    The Second SIAM Conference on Computational Science and Engineering was held in San Diego from February 10-12, 2003. Total conference attendance was 553. The focus of this conference was to draw attention to the tremendous range of major computational efforts on large problems in science and engineering, to promote the interdisciplinary culture required to meet these large-scale challenges, and to encourage the training of the next generation of computational scientists. Computational Science & Engineering (CS&E) is now widely accepted, along with theory and experiment, as a crucial third mode of scientific investigation and engineering design. Aerospace, automotive, biological, chemical, semiconductor, and other industrial sectors now rely on simulation for technical decision support. For federal agencies also, CS&E has become an essential support for decisions on resources, transportation, and defense. CS&E is, by nature, interdisciplinary. It grows out of physical applications and it depends on computer architecture, but at its heart are powerful numerical algorithms and sophisticated computer science techniques. From an applied mathematics perspective, much of CS&E has involved analysis, but the future surely includes optimization and design, especially in the presence of uncertainty. Another mathematical frontier is the assimilation of very large data sets through such techniques as adaptive multi-resolution, automated feature search, and low-dimensional parameterization.     Prior research suggests that poor programmable thermostats usability may prevent their effective use to save energy. The Fraunhofer team hypothesized that home occupants with high-usability thermostats would be more likely to use them to save energy than people with a basic thermostats. In this report, the team discusses results of a project in which the team monitored and compared programmable thermostats with basic thermostats in an affordable housing apartment complex.

    To comply with anticipated OSHA regulatory requirements concerning Permit-Required Confined Spaces, Westinghouse Hanford Company (WHC) created a Confined Spaces Task Team. The primary focus of the task team was to prepare a formal Confined Space Entry (CSE) Program that would ensure full compliance with the anticipated OSHA requirements. A comprehensive training plan was also prepared and submitted for approval as soon as the new CSE Program was approved and released for implementation. On January 14, 1993, OSHA released their final ruling which contained several further changes, requiring the WHC Confined Space Entry Program and Training Plan to be revised. The revised training manual and lessons learned in establishing a Confined Space Entry Program are presented.

    We report on a pilot imaging line survey (36.0-37.0 GHz, with {approx}1 km s{sup -1} spectral channels) with the Expanded Very Large Array for two asymptotic giant branch stars, RW LMi (= CIT6, which has a carbon-rich circumstellar envelope, CSE) and IK Tau (=NML Tau, with an oxygen-rich CSE). Radio continuum emission consistent with photospheric emission was detected from both stars. From RW LMi we imaged the HC{sub 3}N (J = 4{yields}3) emission. The images show several partial rings of emission; these multiple shells trace the evolution of the CSE from 400 to 1200 years. SiS (J = 2{yields}1) emission was detected from both RW LMi and IK Tau. For both stars the SiS emission is centrally condensed with the peak line emission coincident with the stellar radio continuum emission. In addition, we have detected weak HC{sub 7}N (J = 32{yields}31) emission from RW LMi.

    End-use energy efficiency is increasingly being relied upon as a resource for meeting electricity and natural gas utility system needs within the United States. There is a direct connection between the maturation of energy efficiency as a resource and the need for consistent, high-quality data and reporting of efficiency program costs and impacts. To support this effort, LBNL initiated the Cost of Saved Energy Project (CSE Project) and created a Demand-Side Management (DSM) Program Impacts Database to provide a resource for policy makers, regulators, and the efficiency industry as a whole. This study is the first technical report of the LBNL CSE Project and provides an overview of the project scope, approach, and initial findings, including: • Providing a proof of concept that the program-level cost and savings data can be collected, organized, and analyzed in a systematic fashion; • Presenting initial program, sector, and portfolio level results for the program administrator CSE for a recent time period (2009-2011); and • Encouraging state and regional entities to establish common reporting definitions and formats that would make the collection and comparison of CSE data more reliable. The LBNL DSM Program Impacts Database includes the program results reported to state regulators by more than 100 program administrators in 31 states, primarily for the years 2009–2011. In total, we have compiled cost and energy savings data on more than 1,700 programs over one or more program-years for a total of more than 4,000 program-years’ worth of data, providing a rich dataset for analyses. We use the information to report costs-per-unit of electricity and natural gas savings for utility customer-funded, end-use energy efficiency programs. The program administrator CSE values are presented at national, state, and regional levels by market sector (e.g., commercial, industrial, residential) and by program type (e.g., residential whole home programs, commercial new

    This document serves as the final report documenting work completed by Oak Ridge National Laboratory (ORNL) and the Fraunhofer Institute in Building Physics (Holzkirchen, Germany) under an international CRADA No. 0575 with Fraunhofer Institute of Bauphysics of the Federal Republic of Germany for Researching Complex Heat, Air and Moisture Interactions for a Wide Range of Building Envelope Systems and Environmental Loads. This CRADA required a multi-faceted approach to building envelope research that included a moisture engineering approach by blending extensive material property analysis, laboratory system and sub-system thermal and moisture testing, and advanced moisture analysis prediction performance. The Participant's Institute for Building physics (IBP) and the Contractor's Buildings Technology Center (BTC) identified potential research projects and activities capable of accelerating and advancing the development of innovative, low energy and durable building envelope systems in diverse climates. This allowed a major leverage of the limited resources available to ORNL to execute the required Department of Energy (DOE) directives in the area of moisture engineering. A joint working group (ORNL and Fraunhofer IBP) was assembled and a research plan was executed from May 2000 to May 2005. A number of key deliverables were produced such as adoption of North American loading into the WUFI-software. in addition the ORNL Weather File Analyzer was created and this has been used to address environmental loading for a variety of US climates. At least 4 papers have been co-written with the CRADA partners, and a chapter in the ASTM Manual 40 on Moisture Analysis and Condensation Control. All deliverables and goals were met and exceeded making this collaboration a success to all parties involves.
