    IL-15 interacts with a heterotrimeric receptor that consists of the {beta} and {gamma} subunits of the IL-2 receptor (IL-2R) as well as a specific, high-affinity IL-15-binding subunit, which is designated IL-15R{alpha}. Since both the {beta} and the {gamma} subunits of the IL-2R are required for signaling by either IL-2 or IL-15, it is not surprising that these cytokines share many activities in vitro. However, the differential expression of these cytokines and the {alpha} chains of their receptors within various tissues and cell types suggests that IL-2 and IL-15 may perform at least partially distinct physiological functions. The production of IL-15 by macrophages, and possibly other cell types, in response to environmental stimuli and infectious agents suggests that IL-15 may play a role in protective immune responses, allograft rejection, and the pathogenesis of autoimmune diseases. 56 refs.

    IL-7 and IL-7R{alpha} bind the {gamma}{sub c} receptor, forming a complex crucial to several signaling cascades leading to the development and homeostasis of T and B cells. We report that the IL-7R{alpha} ectodomain uses glycosylation to modulate its binding constants to IL-7, unlike the other receptors in the {gamma}{sub c} family. IL-7 binds glycosylated IL-7R{alpha} 300-fold more tightly than unglycosylated IL-7R{alpha}, and the enhanced affinity is attributed primarily to an accelerated on rate. Structural comparison of IL-7 in complex to both forms of IL-7R{alpha} reveals that glycosylation does not participate directly in the binding interface. The SCID mutations of IL-7R{alpha} locate outside the binding interface with IL-7, suggesting that the expressed mutations cause protein folding defects in IL-7R{alpha}. The IL-7/IL-7R{alpha} structures provide a window into the molecular recognition events of the IL-7 signaling cascade and provide sites to target for designing new therapeutics to treat IL-7-related diseases.

    From October 2012 through September 2013, the second ARM Mobile Facility (AMF2) was deployed on the container ship Spirit, operated by Horizon Lines, for the Marine ARM GPCI* Investigation of Clouds (MAGIC) field campaign. During approximately 20 round trips between Los Angeles, California, and Honolulu, Hawaii, AMF2 obtained continuous on-board measurements of cloud and precipitation, aerosols, and atmospheric radiation; surface meteorological and oceanographic variables; and atmospheric profiles from weather balloons launched every six hours. During two two-week intensive observational periods in January and July 2013, additional instruments were deployed and balloon soundings were be increased to every three hours. These additional data provided a more detailed characterization of the state of the atmosphere and its daily cycle during two distinctly different seasons. The primary objective of MAGIC was to improve the representation of the stratocumulus-to-cumulus transition in climate models. AMF2 data documented the small-scale physical processes associated with turbulence, convection, and radiation in a variety of marine cloud types.

    Highlights: {yields} Curcumin inhibits CD4{sup +} T-lymphocyte proliferation. {yields} Curcumin inhibits interleukin-2 (IL-2) synthesis and CD25 expression by CD4{sup +} T-lymphocytes. {yields} Curcumin interferes with IL-2 receptor signaling by inhibiting JAK3 and STAT5 phosphorylation. {yields} IL-2-dependent regulatory T-lymphocyte function and Foxp3 expression is downregulated by curcumin. -- Abstract: Curcumin (diferulomethane) is the principal curcuminoid in the spice tumeric and a potent inhibitor of activation-induced T-lymphocyte proliferation; however, the molecular basis of this immunosuppressive effect has not been well studied. Here we show that micromolar concentrations of curcumin inhibited DNA synthesis by mouse CD4{sup +} T-lymphocytes, as well as interleukin-2 (IL-2) and CD25 ({alpha} chain of the high affinity IL-2 receptor) expression in response to antibody-mediated cross-linking of CD3 and CD28. Curcumin acted downstream of protein kinase C activation and intracellular Ca{sup 2+} release to inhibit I{kappa}B phosphorylation, which is required for nuclear translocation of the transcription factor NF{kappa}B. In addition, IL-2-dependent DNA synthesis by mouse CTLL-2 cells, but not constitutive CD25 expression, was impaired in the presence of curcumin, which demonstrated an inhibitory effect on IL-2 receptor (IL-2R) signaling. IL-2-induced phosphorylation of STAT5A and JAK3, but not JAK1, was diminished in the presence of curcumin, indicating inhibition of critical proximal events in IL-2R signaling. In line with the inhibitory action of curcumin on IL-2R signaling, pretreatment of CD4{sup +}CD25{sup +} regulatory T-cells with curcumin downregulated suppressor function, as well as forkhead box p3 (Foxp3) expression. We conclude that curcumin inhibits IL-2 signaling by reducing available IL-2 and high affinity IL-2R, as well as interfering with IL-2R signaling.

    Sublethally irradiated mice undergo thymic regeneration which follows a phenotypic pattern of events similar to that observed during normal fetal development. Thymic regeneration after irradiation is the product of a limited pool of intrathymic radioresistant stem cells undergoing simultaneous differentiation. We show that in this model of T cell development, thymic regeneration follows a pathway in which the IL-2R is transiently expressed on CD4-/CD8- cells. IL-2R expression occurred during the exponential growth period of thymic regeneration, and IL-2R blocking prevented this explosive growth. Flow cytometry analysis revealed that the IL-2R blockade affected primarily the development of the immature CD3-/CD4-/CD8- (triple negative) cells and their ability to generate CD3+/CD4+/CD8+ or CD3+/CD4+/CD8- and CD3+/CD4-/CD8+ thymocytes. Thus, our findings demonstrate that blocking of the IL-2R resulted in an arrest in proliferation and differentiation by intrathymic radioresistant stem cells, indicating that the IL-2/IL-2R pathway is necessary for the expansion of immature triple negative T cells.

    Human peripheral blood T cell expression of IL-2 receptors (IL-2R), detected by both immunocytofluorometry and /sup 125/I-IL-2 binding, was studied using lymphocytes stimulated with monoclonal anti-T3 antibodies (Leu-4, OKT3). Lymphocytes, isolated from healthy individuals, were prescreened and classified as Leu-4 responders or non-responders according to 72 h /sup 3/H-thymidine incorporation experiments. Leu-4 non-responder lymphocytes, though capable of normal IL-2R expression and IL-2 secretion when cultured with OKT3 (IgG2a), expressed little to no IL-2R nor secreted IL-2 when stimulated with Leu-4 (IgG1). In addition, the amount of IL-2 secreted by Leu-4 stimulated, Leu-4 responder cells, was one-third- to one-fifth of that detected when OKT3 was used as the stimulant. The addition of recombinant IL-2 (rIL-2) to a Leu-4 stimulated, Leu-4 non-responder lymphocyte culture, resulted in the expression of IL-2R and cellular proliferation, indicating that IL-2 upregulated its biologically relevant receptor. As expected, cyclosporin-A (CSA) inhibited the secretion of IL-2 and subsequent proliferation of Leu-4 stimulated, Leu-4 responder cells. Unexpectedly, however, the expression of IL-2R was also blocked. Exogenous rIL-2 partially reversed the effect of CSA on IL-2R expression and proliferation. The results indicate that IL-2 may provide an additional, required signal for optimal IL-2R expression.

    Although blood pH is maintained in a narrow range of around pH 7.4 in living organisms, inflammatory loci are characterized by acidic conditions. Mast cells tend to reside close to the surface of the body in areas such as the mucosa and skin where they may be exposed to exogenous acids, and they play an important role in immune responses. However, little is known about the effects of extracellular acidification on the functions of mast cell. Here, we found that extracellular acidification increased the dinitrophenyl-conjugated human serum albumin (DNP-HSA)-induced production of interleukin (IL)-6 and IL-13 in MC/9 cells or bone marrow-derived mouse mast cells sensitized with anti-DNP IgE. Extracellular acidification also inhibited migration of MC/9 cells toward DNP-HSA. In addition, acidic pH stimulated antigen-induced activation of p38 mitogen-activated protein kinase (MAPK) and protein kinase B (Akt). These findings suggest that extracellular acidification augmented antigen/IgE-induced and FcεRI-mediated production of IL-6 and IL-13 in mast cells, and that this was associated with the enhancement of p38 MAPK and Akt activation. - Highlights: • Antigen-induced IL-6 and IL-13 production was augmented by acidic pH in mast cells. • Acidic pH-induced actions were associated with activation of p38 MAPK and Akt. • Inhibition of p38 MAPK and Akt attenuated cytokine responses to acidic pH. • Acidic pH effects are not attributable to actions of known proton-sensing GPCRs.

    A case-control association study was conducted in Caucasian patients with schizophrenia (DSM-III-R, n = 42) and unaffected controls (n = 47) matched for ethnicity and area of residence. Serum interleukin-2 receptor (IL-2R) concentrations, as well as a dinucleotide repeat polymorphism in the IL-2RP chain gene, were examined in both groups. No significant differences in IL-2R concentrations or in the distribution of the polymorphism were noted.     Binding of interleukin-2 (IL-2) to its specific receptor induces activation of two members of Jak family protein tyrosine kinases, Jak1 and Jak3. An IL-2 receptor (IL-2R)-reconstituted NIH 3T3 fibroblast cell line proliferates in response to IL-2 only when hematopoietic lineage-specific Jak3 is ectopically expressed. However, the mechanism of Jak3-dependent proliferation in the fibroblast cell line is not known. Here, I showed that Jak3 expression is dispensable for IL-2-induced activation of Jak1 and Stat proteins and expression of nuclear proto-oncogenes in the IL-2R-reconstituted fibroblast cell line. Jak3 expression markedly enhanced these IL-2-induced signaling events. In contrast, Jak3 expression was essential for induction of cyclin genes involved in the G1-S transition. These data suggest a critical role of Jak3 in IL-2 signaling in the fibroblast cell line and may provide further insight into the cell type-specific mechanism of cytokine signaling.

We show here that IL-3 significantly inhibits receptor activator of NF-{kappa}B (RANK) ligand (RANKL)-induced activation of c-Jun N-terminal kinase (JNK). IL-3 down-regulates expression of c-Fos and nuclear factor of activated T cells (NFATc1) transcription factors. In addition, IL-3 down-regulates RANK expression posttranscriptionally in both purified osteoclast precursors and whole bone marrow cells. Furthermore, the inhibitory effect of IL-3 on RANK expression was irreversible. Interestingly, IL-3 inhibits in vivo RANK expression in mice. Thus, we provide the first evidence that IL-3 irreversibly inhibits RANK expression that results in inhibition of important signaling molecules induced by RANKL.

    Interleukin (IL)-33 is a recently discovered proinflammatory cytokine that belongs to the IL-1 family. Several studies have reported that IL-33 inhibits osteoclast differentiation. However, the mechanism of IL-33 regulation of osteoclastogenesis remains unclear. In the present study, we examined the effect of IL-33 on osteoclast formation in vitro. IL-33 suppressed osteoclast formation in both mouse bone marrow cells and monocyte/macrophage cell line RAW264.7 cells induced by receptor activator of NF-κB ligand (RANKL) and/or macrophage stimulating factor (M-CSF). IL-33 also inhibited the expression of RANKL-induced nuclear factor of activated T-cell cytoplasmic 1 (NFATc1), thereby decreasing the expression of osteoclastogenesis-related marker genes, including Cathepsin K, Osteoclast stimulatory transmembrane protein (Oc-stamp) and Tartrate-resistant acid phosphatase (Trap). Blockage of IL-33-ST2 binding suppressed the IL-33-mediated inhibition of NFATc1.     Nanoparticles are becoming promising carriers for gene delivery because of their high capacity in gene loading and low cell cytotoxicity. In this study, a chitosan-based nanoparticle encapsulated within a recombinant pcDNA3.1-dsNKG2D-IL-15 plasmid was generated. The fused dsNKG2D-IL-15 gene fragment consisted of double extracellular domains of NKG2D with IL-15 gene at downstream. The average diameter of the gene nanoparticles ranged from 200 nm to 400 nm, with mean zeta potential value of 53.8 ± 6.56 mV. The nanoparticles which were loaded with the dsNKG2D-IL-15 gene were uptaken by tumor cells with low cytotoxicity. Tumor cells pre-transfected by gene nanopartilces stimulated NK and T cells in vitro. Intramuscular injection of gene nanoparticles suppressed tumor growth and prolonged survival of tumor-bearing mice through activation of NK and CD8{sup +} T cells. Thus, chitosan-based nanoparticle delivery of dsNKG2D-IL-15 gene vaccine can be potentially used for tumor therapy. - Highlights: • Generation of a nanoparticle for delivery of dsNKG2D-IL-15 gene. • Characterization of the gene nanoparticle. • Antitumor activity mediated by the gene nanoparticle.

    Keratinocytes are the major cellular components of human epidermis and play a key role in the modulating cutaneous inflammation and toxic responses. In human chronic skin diseases, the common skin inflammatory phenotypes like skin barrier disruption and epidermal hyperplasia are manifested in epidermal keratinocytes by interactions with T helper (Th) cells. To find a common gene expression signature of human keratinocytes in chronic skin diseases, we performed a whole genome microarray analysis on normal human epidermal keratinocytes (NHKs) treated with IFNγ, IL-4, IL-17A or IL-22, major cytokines from Th1, Th2, Th17 or Th22 cells, respectively. The microarray results showed that the four genes, IL-24, PDZK1IP1, H19 and filaggrin, had common expression profiles in NHKs exposed to Th cell cytokines. In addition, the acute phase pro-inflammatory cytokines, IL-1β, IL-6 and TNFα, also change the gene transcriptional profile of IL-24, PDZK1IP1, H19, and filaggrin in NHKs as those of Th cytokines. Therefore, the signature gene set, consisting of IL-24, PDZK1IP1, H19, and filaggrin, provides essential insights for understanding the process of cutaneous inflammation and toxic responses. We demonstrate that environmental toxic stressors, such as chemical irritants and ultraviolet irradiation stimulate the production of IL-24 in NHKs. IL-24 stimulates the JAK1-STAT3 and MAPK pathways in NHKs, and promotes the secretion of pro-inflammatory mediators IL-8, PGE2, and MMP-1.     COX-2 is a major inflammatory mediator implicated in colorectal inflammation and cancer. However, the exact origin and role of COX-2 on colorectal inflammation and carcinogenesis are still not well defined. Recently, we reported that COX-2 and iNOS signalings interact in colonic CCD18Co fibroblasts. In this article, we investigated whether activation of COX-2 signaling by IL1{beta} in primary colonic fibroblasts obtained from normal and cancer patients play a critical role in regulation of proliferation and invasiveness of human colonic epithelial cancer cells. Our results demonstrated that COX-2 level was significantly higher in cancer associated fibroblasts than that in normal fibroblasts with or without stimulation of IL-1{beta}, a powerful stimulator of COX-2. Using in vitro assays for estimating proliferative and invasive potential, we discovered that the proliferation and invasiveness of the epithelial cancer cells were much greater when the cells were co-cultured with cancer associated fibroblasts than with normal fibroblasts, with or without stimulation of IL1{beta}. Further analysis indicated that the major COX-2 product, prostaglandin E{sub 2}, directly enhanced proliferation and invasiveness of the epithelial cancer cells in the absence of fibroblasts. Moreover, a selective COX-2 inhibitor, NS-398, blocked the proliferative and invasive effect of both normal and cancer associate fibroblasts on the epithelial cancer cells, with or without stimulation of IL-1{beta}. Those results indicate that activation of COX-2 signaling in the fibroblasts plays a major role in promoting proliferation and invasiveness of the epithelial cancer cells. In this process, PKC is involved in the activation of COX-2 signaling induced by IL-1{beta} in the fibroblasts.

    IL-2 receptors on T cells exist in at least three forms which differ in their ligand-binding affinity. The low-affinity IL-2 receptor (IL-2R) consists of the 55-kDa Tac protein (p55 alpha), the intermediate-affinity site corresponds to the 70-kDa molecule (p70 beta), and the high-affinity IL-2R consists of a noncovalent heterodimeric structure involving both p55 alpha and p70 beta. We studied 24 B cell lines (8 EBV-negative and 16 EBV-positive) for IL-2R expression in the presence or absence of the tumor promoter, teleocidin. 125I-IL-2 radioreceptor binding assays and crosslinking studies demonstrated the sole expression of p55 alpha in EBV-negative cell lines only, whereas p55 alpha present in EBV-positive cell lines was always associated with p70 beta to construct high-affinity IL-2R. p70 beta was not detected in any of the EBV-negative cell lines, but was expressed on most of the EBV-positive cell lines (13 of 16). Our data also indicate that the expression of p55 alpha and p70 beta by radiolabeling correlates with their expression in flow cytometry, and that a large excess of p55 alpha is required to construct high-affinity IL-2R. Coexpression of p55 alpha and p70 beta on human B cells contributed to constructing high-affinity IL-2R hybrid complex as shown by rapid association rate contributed by p55 alpha and slow dissociation rate by p70 beta; teleocidin's ability to induce p55 alpha on cell lines which express p70 beta only, resulting in appearance of high-affinity IL-2R; and blocking p55 alpha by anti-Tac mAb in cell lines which constitutively express high-affinity IL-2R eliminated both high- and low-affinity components. The existence of low, intermediate, and high IL-2R on human B cells bears important future implications for understanding the mechanism of IL-2 signaling and the role of IL-2 in B cell activation, proliferation, and differentiation.

    This work is aimed at giving proof that Eu(Tf2N)(3) (Tf2N = bis(trifluoromethanesulfonyl)amide) can act as both an optical and electrochemical probe for the determination of the Lewis acidity of an ionic liquid anion. For that reason the luminescence spectra and cyclic voltammograms of dilute solutions of Eu(Tf2N)(3) in various ionic liquids were investigated. The Eu2+/3+ redox potential in the investigated ILs can be related to the Lewis basicity of the IL anion. The IL cation had little influence. The lower the determined halfwave potential, the higher the IL anion basicity. The obtained ranking can be confirmed by luminescence spectroscopy where a bathochromic shift of the D-5(0) -> F-7(4) transition indicates a stronger Lewis basicity of the IL anion.     Crystal structure of the SEFIR domain from human IL-17 receptor A provides new insights into IL-17 signaling. Interleukin 17 (IL-17) cytokines play a crucial role in mediating inflammatory and autoimmune diseases. A unique intracellular signaling domain termed SEFIR is found within all IL-17 receptors (IL-17Rs) as well as the key adaptor protein Act1. SEFIR-mediated protein–protein interaction is a crucial step in IL-17 cytokine signaling. Here, the 2.3 Å resolution crystal structure of the SEFIR domain of IL-17RA, the most commonly shared receptor for IL-17 cytokine signaling, is reported. The structure includes the complete SEFIR domain and an additional α-helical C-terminal extension, which pack tightly together to form a compact unit. Structural comparison between the SEFIR domains of IL-17RA and IL-17RB reveals substantial differences in protein topology and folding. The uniquely long insertion between strand βC and helix αC in IL-17RA SEFIR is mostly well ordered, displaying a helix (αCC′{sub ins}) and a flexible loop (CC′). The DD′ loop in the IL-17RA SEFIR structure is much shorter; it rotates nearly 90° with respect to the counterpart in the IL-17RB SEFIR structure and shifts about 12 Å to accommodate the αCC′{sub ins} helix without forming any knots. Helix αC was identified as critical for its interaction with Act1 and IL-17-stimulated gene expression. The data suggest that the heterotypic SEFIR–SEFIR association via helix αC is a conserved and signature mechanism specific for IL-17 signaling. The structure also suggests that the downstream motif of IL-17RA SEFIR together with helix αC could provide a composite ligand-binding surface for recruiting Act1 during IL-17 signaling.

    We have revealed in a porcine skin injury model that eosinophil recruitment was dose-dependently enhanced by a single high-dose irradiation. In this study, we investigated the underlying mechanism of eosinophil-associated skin fibrosis and the effect of high-dose-per-fraction radiation. The dorsal skin of a mini-pig was divided into two sections containing 4-cm{sup 2} fields that were irradiated with 30 Gy in a single fraction or 5 fractions and biopsied regularly over 14 weeks. Eosinophil-related Th2 cytokines such as interleukin (IL)-4, IL-5, and C–C motif chemokine-11 (CCL11/eotaxin) were evaluated by quantitative real-time PCR. RNA-sequencing using 30 Gy-irradiated mouse skin and functional assays in a co-culture system of THP-1 and irradiated-human umbilical vein endothelial cells (HUVECs) were performed to investigate the mechanism of eosinophil-mediated radiation fibrosis. Single high-dose-per-fraction irradiation caused pronounced eosinophil accumulation, increased profibrotic factors collagen and transforming growth factor-β, enhanced production of eosinophil-related cytokines including IL-4, IL-5, CCL11, IL-13, and IL-33, and reduced vessels compared with 5-fraction irradiation. IL-33 notably increased in pig and mouse skin vessels after single high-dose irradiation of 30 Gy, as well as in irradiated HUVECs following 12 Gy. Blocking IL-33 suppressed the migration ability of THP-1 cells and cytokine secretion in a co-culture system of THP-1 cells and irradiated HUVECs.     In this study, rutile films consisting of rectangular nanorods were facilely deposited on glass substrates from strongly acid solution of TiCl{sub 4}. The highly ordered array of nanorods was realized in presence of ionic liquid (IL) of [Bmim]Br by following a hydrothermal process. In this process, Degussa P25 nanoparticles served as seeds that were pre-deposited on the substrates to facilitate the array of rutile nanorods. X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy (TEM), and Raman spectrum were used to characterize the obtained nanorod films. The measurements showed that the nanorods were rectangular with width of 100-200 nm and length of more than 1 {mu}m, and grew up typically along c-axis to form the arrays against the substrate. The presence of IL was found vital for the formation of rutile nanorods, and the suitable molar ratio of [Bmim]Br to TiCl{sub 4} ranged from 500:1 to 1500:1. The excessive [Bmim]Br may hinder the precipitation of rutile particles. - Graphical abstract: The rutile film consisting of rectangular nanorods is successfully deposited on glass substrate in presence of ionic liquid (IL) of [Bmim]Br.     X-linked severe combined immunodeficiency (SCID) is characterized by profound defects in cellular and humoral immunity and, in humans, is associated with mutations in the gene for the {gamma} chain of the IL-2 receptor (IL-2R{gamma}). We have examined this gene in a colony of dogs established from a single X-linked SCID carrier female. Affected dogs have a 4-bp deletion in the first exon of the IL-2R{gamma} gene, which precludes the production of a functional protein, demonstrating that the canine disease is a true homologue of human X-linked SCID. 37 refs., 3 figs.

    Expression of the IL-2R{alpha} gene is regulated by members of the c-Rel/NF-{kappa}B family of transcription factors binding to the {kappa}B site in the promoter. Previous work has not defined the role of individual members of the c-Rel family in the activation of the IL-2R{alpha} gene. Using the COS cell system, we were able to reconstitute the regulation of the IL-2R{alpha} promoter by expressing cloned Rel family members with serum response factor (SRF). We found that c-rel alone activated the IL-2R{alpha} promoter only weakly but worked with the p50 subunit of NF-{kappa}B (NFKB1) to give a higher level of expression. We showed that c-rel heterodimerizes with p50 and the amount of this heterodimer correlated with the level of IL-2R{alpha} gene expression. Our results provide evidence that c-rel/p50 heterodimers activate gene expression in the context of a cellular promoter. We show that c-rel or p65 can cooperate with SRF in the activation of this promoter and the transactivation by c-rel with SRF was enhanced by p50. Synergistic activation required both {kappa}B an CArG sites, and binding studies show that these that these adjacent sites can be occupied simultaneously. The transactivation observed with cloned transcription factors mimics the physiologic induction of the IL-2R{alpha} gene since multiple sequence elements cooperate to give gene activation. The data support the model that c-rel/p50 or p65 can cooperate with SRF to specifically target the expression of the IL-2R{alpha} gene in activated T cells.

    Recently, prolongation of cardiac allograft survival in mice was reported using a rat anti-IL-2R mAb (AMT-13). However, its immunosuppressive action in vivo, alone and in combination with other immunosuppressants, and its effect on other organ transplants has not been extensively studied. We grafted cultured fetal pancreas from CBA (H-2k) donors to Balb/c (H-2d) mice. Recipients were treated with 10 consecutive daily injections each of 20 micrograms AMT-13 only, or with an additional mild immunosuppression of 350 rads irradiation. Control groups received rat immunoglobulin or 350 rads irradiation. Graft survival and the phenotype of infiltrating cells were assessed histologically and immunocytochemically on days 12, 17, and 21, and soluble IL-2R levels were measured in the serum with a quantitative ELISA in all recipients. Two of five grafts in the AMT-13-treated group had islets on day 12 posttransplantation despite lymphocytic infiltration in all grafts, while at this time all grafts of rat Ig treated control mice were completely rejected with only scar tissue and a few lymphocytes remaining. Additional immunosuppression with 350 rads irradiation had a marked additive effect with AMT-13. Soluble IL-2R levels in the serum of untreated recipients were not elevated compared with normal serum levels, but recipients injected with AMT-13 had multifold increased soluble IL-2R levels. The percentage of IL-2R+ cells in the grafts of AMT-13-treated animals was either normal (less than 5%) or increased (20%) in the additionally irradiated mice, providing strong evidence that the immunosuppressive effect of AMT-13 is not due to a depletion of activated IL-2R+ lymphocytes.

    This report documents a field demonstration at the Rock Island Arsenal, IL, that validated the thermal spray vitrification (TSV) process as a safe and effective technique for removing lead-based paint from a steel bridge. Specially formulated glass was applied in a molten state to painted steel using a conventional thermal spray application system. The molten glass reacts with the paint, and encapsulates the lead. The cooled glass readily cracks and falls off, removing the paint. After onsite remelting of the glass waste to complete the encapsulation process, the final waste product is chemically inert and may be disposed of in a regular landfill. The Illinois Environmental Protection Agency, Division of Air Pollution Control determined that the glass remelt process could be considered a paint-removal operation for which no air quality permit was required.

    Carbon nanotubes (CNTs) have potential applications in biosensors, tissue engineering, and biomedical devices because of their unique physico-chemical, electronic and mechanical properties. However, there is limited literature data available concerning the biological properties and toxicity of CNTs. This study aimed to assess the toxicity exhibited by multi-walled CNTs (MWCNTs) and to elucidate possible molecular mechanisms underlying the biological effects of MWCNTs in A549 cells. Exposing A549 cells to MWCNTs led to cell death, changes in cell size and complexity, reactive oxygen species (ROS) production, interleukin-8 (IL-8) gene expression and nuclear factor (NF)-{kappa}B activation. Treatment of A549 cells with antioxidants prior to adding MWCNTs decreased ROS production and abrogated expression of IL-8 mRNA. Pretreatment of A549 cells with NF-{kappa}B inhibitors suppressed MWCNTs-induced IL-8 mRNA expression. These results indicate that MWCNTs are able to induce expression of IL-8 in A549 cells, at least in part, mediated by oxidative stress and NF-{kappa}B activation.

    The meeting began with presentations by the facilitator, Mr. Spiegel, and Dr. Lewis, the representative from DOE. The facilitator introduced those on the podium. He then described the general structure of the meeting and its purpose: to hear the issues and concerns of those present regarding the proposed Hawaiian Geothermal Project. He described his role as assuring the impartiality and fairness of the meeting. Dr. Lewis of DOE further defined the scope of the project, introduced members of the EIS team, and briefly described the EIS process. Ninety percent of those presenting expressed concern about the potential impacts of the HGP to the environment. They asked that the EIS investigate the impacts of 1.he HGP on quality of air, water and soils, flora and fauna, land, socioeconomics, religion and culture. Seventy percent of the commenters want the EIS to investigate the impacts of all components of the HGP (eg. generation facilities, pipes, roads, drill pads, transmission corridors, and cable) on terrestrial ecosystems (e.g. the rainforest), threatened and endangered species, species diversity, species used for ritual or medicinal purposes, and climate stability. The EIS should address the effects of segmentation of habitat, introduction of exotic species, acid rain, herbicides, and enhanced fire danger. It should consider the entire Kilauea East Rift Zone when studying the impacts of HGP on the ecology of the Puna District. Of importance is the interrelationship between the lava, the biota of the region, and the regeneration that occurs following an eruption. DOE should examine the impacts of the network of roads that would be constructed for the HGP. Roads may serve as barriers that segment habitats, prevent regeneration and natural spread of biota, or serve as conduits for introduction of exotic species or for mobilization of herbicides. They may enhance the potential for logging or collecting of endangered native species. The EIS should determine the impacts of the HGP on marine life, particularly threatened and endangered species such as the humpback whale. It should address the effects of emf. In particular, the EIS should establish whether the clearing of land for HGP increase the problems of silting in the near-shore ocean. Ninety percent of the presenters requested that the EIS address long- and short-term socioeconomic impacts of the HGP. Sixty percent want the EIS to provide a detailed economic analysis of the costs (to the Consumer, rate payer and non-user) of the HGP, including the cable, from inception (planning) through decommissioning, to determine both feasibility and impacts to economic systems. The EIS should investigate the effects of the presence of transmission lines making large regions of the State: less desirable for living in terms of property values, cost of living, etc . b.b. This impact would affect all residents of Hawai'i, not just those on Hawai'i, Maui, Moloka'i and Oahu. The EIS should analyze the economic impacts of failures once geothermal energy provides a significant proportion of Hawai'i's energy needs, including the costs of developing backup power supply on Oahu. One commenter asked who would be responsible for the consequences of lower property values or property condemnation associated with the HGP and suggested that the developer(s) should be bonded. Fifty percent want the EIS to identify what the benefits of HGP are and who would benefit from development of the HGP.

    Arsenite is an established human carcinogen, and arsenite-induced inflammation contributes to malignant transformation of cells, but the molecular mechanisms by which cancers are produced remain to be established. The present results showed that, evoked by arsenite, secretion of interleukin-6 (IL-6), a pro-inflammatory cytokine, led to the activation of STAT3, a transcription activator, and to increased levels of a microRNA, miR-21. Blocking IL-6 with anti-IL-6 antibody and inhibiting STAT3 activation reduced miR-21 expression. For human bronchial epithelial cells, cultured in the presence of anti-IL-6 antibody for 3 days, the arsenite-induced EMT and malignant transformation were reversed. Thus, IL-6, acting on STAT3 signaling, which up-regulates miR-21in an autocrine manner, contributes to the EMT induced by arsenite. These data define a link from inflammation to EMT in the arsenite-induced malignant transformation of HBE cells. This link, mediated through miRNAs, establishes a mechanism for arsenite-induced lung carcinogenesis. - Highlights: Arsenite evokes IL-6 secretion. IL-6 autocrine mediates STAT3 signaling and up-regulates miR-21expression. Inflammation is involved in arsenite-induced EMT.

    Endothelin (ET) signaling provokes nephrotoxicity induced by the immunosuppressant drug cyclosporine A (CSA). We tested the hypotheses that (i): celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, counterbalances renal derangements caused by CSA in rats and (ii) the COX-2/endothelin ET{sub B} receptor signaling mediates the CSA-celecoxib interaction. Ten-day treatment with CSA (20 mg/kg/day) significantly increased biochemical indices of renal function (serum urea, creatinine), inflammation (interleukin-2, IL-2) and fibrosis (transforming growth factor-?{sub 1}, TGF-?{sub 1}). Histologically, CSA caused renal tubular atrophy along with interstitial fibrosis. These detrimental renal effects of CSA were largely reduced in rats treated concurrently with celecoxib (10 mg/kg/day). We also report that cortical glomerular and medullary tubular protein expressions of COX-2 and ET{sub B} receptors were reduced by CSA and restored to near-control values in rats treated simultaneously with celecoxib. The importance of ET{sub B} receptors in renal control and in the CSA-celecoxib interaction was further verified by the findings (i) most of the adverse biochemical, inflammatory, and histopathological profiles of CSA were replicated in rats treated with the endothelin ET{sub B} receptor antagonist BQ788 (0.1 mg/kg/day, 10 days), and (ii) the BQ788 effects, like those of CSA, were alleviated in rats treated concurrently with celecoxib. Together, the data suggest that the facilitation of the interplay between the TGF-?1/IL-2/COX-2 pathway and the endothelin ET{sub B} receptors constitutes the cellular mechanism by which celecoxib ameliorates the nephrotoxic manifestations of CSA in rats. - Highlights: Celecoxib abolishes nephrotoxic manifestations of CSA in rats. In the present study, we investigated the signaling pathway involved in TGF-{beta}1-promoted MMP-2 secretion and invasion by human pancreatic cancer cells SW1990. Using specific inhibitors, we found that stimulation of these tumor cells with TGF-{beta}1 induced secretion and activation of the collagenase MMP-2, which was required for TGF-{beta}1-stimulated invasion. Our results also indicate that signaling events involved in TGF-{beta}1-enhanced SW1990 invasiveness comprehend activation of Rac1 followed by generation of reactive oxygen species through nicotinamide adenine dinucleotide phosphate-oxidase, activation of nuclear factor-kappa beta, release of interleukin-6, and secretion and activation of MMP-2.

    The present study has demonstrated the translocation of zinc ferrite nanoparticles (ZnFe{sub 2}O{sub 4}-NPs) into the cytoplasm of human amnion epithelial (WISH) cells, and the ensuing cytotoxicity and genetic damage. The results suggested that in situ NPs induced oxidative stress, alterations in cellular membrane and DNA strand breaks. The [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] (MTT) and neutral red uptake (NRU) cytotoxicity assays indicated 64.48 1.6% and 50.73 2.1% reduction in cell viability with 100 ?g/ml of ZnFe{sub 2}O{sub 4}-NPs exposure. The treated WISH cells exhibited 1.2-fold higher ROS level with 0.9-fold decline in membrane potential (??m) and 7.4-fold higher DNA damage after 48 h of ZnFe{sub 2}O{sub 4}-NPs treatment. Real-time PCR (qPCR) analysis of p53, CASP 3 (caspase-3), and bax genes revealed 5.3, 1.6, and 14.9-fold upregulation, and 0.18-fold down regulation of bcl 2 gene vis--vis untreated control. RT{sup 2} Profiler PCR array data elucidated differential up-regulation of mRNA transcripts of IL-1b, NFKB1, NOS2 and CCL21 genes in the range of 1.5 to 3.7-folds. The flow cytometry based cell cycle analysis suggested the transfer of 15.2 2.1% (p < 0.01) population of ZnFe{sub 2}O{sub 4}-NPs (100 ?g/ml) treated cells into apoptotic phase through intrinsic pathway. Over all, the data revealed the potential of ZnFe{sub 2}O{sub 4}-NPs to induce cellular and genetic toxicity in cells of placental origin. Thus, the significant ROS production, reduction in ??m, DNA damage, and activation of genes linked to inflammation, oxidative stress, proliferation, DNA damage and repair could serve as the predictive toxicity and stress markers for ecotoxicological assessment of ZnFe{sub 2}O{sub 4}-NPs induced cellular and genetic damage. - Highlights: First report on the molecular toxicity of ZnFe{sub 2}O{sub 4}-NPs in cells of placental origin WISH cells treated with ZnFe{sub 2}O{sub 4}-NPs exhibited cytoplasmic localization of NPs

    There is a clear evidence that environmental pollutants, such as benzo[a]pyrene (B[a]P), can have detrimental effects on the immune system, whereas the underlying mechanisms still remain elusive. Jurkat T cells share many properties with native T lymphocytes and therefore are an appropriate model to analyze the effects of environmental pollutants on T cells and their activation. Since environmental compounds frequently occur at low, not acute toxic concentrations, we analyzed the effects of two subtoxic concentrations, 50 nM and 5 μM, on non- and activated cells. B[a]P interferes directly with the stimulation process as proven by an altered IL-2 secretion. Furthermore, B[a]P exposure results in significant proteomic changes as shown by DIGE analysis. Pathway analysis revealed an involvement of the AhR independent Nrf2 pathway in the altered processes observed in unstimulated and stimulated cells. A participation of the Nrf2 pathway in the change of IL-2 secretion was confirmed by exposing cells to the Nrf2 activator tBHQ. tBHQ and 5 μM B[a]P caused similar alterations of IL-2 secretion and glutamine/glutamate metabolism. Moreover, the proteome changes in unstimulated cells point towards a modified regulation of the cytoskeleton and cellular stress response, which was proven by western blotting. Additionally, there is a strong evidence for alterations in metabolic pathways caused by B[a]P exposure in stimulated cells. Especially the glutamine/glutamate metabolism was indicated by proteome pathway analysis and validated by metabolite measurements. The detrimental effects were slightly enhanced in stimulated cells, suggesting that stimulated cells are more vulnerable to the environmental pollutant model compound B[a]P. - Highlights: • B[a]P affects the proteome of Jurkat T cells also at low concentrations. • Exposure to B[a]P (50 nM, 5 μM) did not change Jurkat T cell viability. • Both B[a]P concentrations altered the IL-2 secretion of stimulated cells.

    This is the first technical progress report of the remediation of two wells and a water injection well in Clay County, Illinois. The location is identified as the Routt lease and the wells will be identified as the Routt No.3 and Routt No.4 respectively throughout this report. The Clay County portion of this project has met all legal, financial, and environmental requirements to drill and /or pump oil at this lease. We have also obtained all available information about this site and have taken the necessary steps to improve access roads, dig the necessary pits and build the necessary firewalls. Both wells have been drilled to the Salem formation. Gas gun technology was used to stimulate the reservoir of the Routt No.3. This report will address the technical aspects of the remediation.

    Forty-two uranium anomalies meet the minimum statistical requirements as defined in Volume I. These anomalies are listed and are shown on the Uranium Anomaly Interpretation Map. Potassium (%K), equivalent Uranium (ppM eU), equivalent Thorium (ppM eTh), eU/eTh, eU/K, eTh/K, and magnetic pseudo-contour maps are presented in Appendix E. Stacked Profiles showing geologic strip maps along each flight-line, together with sensor data, and ancillary data are presented in Appendix F. All maps and profiles were prepared on a scale of 1:250,000, but have been reduced to 1:500,000 for presentation in Volume II. Anomalies No. 1, No. 4 to No. 17, No. 22 to No. 23, and No. 27 to No. 30 are over areas underlain by the Radnor Till member (Qti) of the Pleistocene Glasford formation. Anomalies No. 2 and No. 3 are over areas underlain by Pleistocene Cahokia Alluvium (Qc) and the Radnor Till member (Qti) of the Glasford formation.     An airborne combined radiometric and magnetic survey was performed for the Department of Energy (DOE) over the area covered by the Peoria, Decatur, and Belleville, 1:250,000 National Topographic Map Series (NTMS), quadrangle maps. The survey was part of DOE's National Uranium Resource Evaluation (NURE) program. Data were collected by a helicopter equipped with a gamma-ray spectrometer with a large crystal volume, and with a high sensitivity proton procession magnetometer. The radiometric system was calibrated at the Walker Field Calibration pads and the Lake Mead Dynamic Test Range. Data quality was ensured during the survey by daily test flights and equipment checks. Radiometric data were corrected for live time, aircraft and equipment background, cosmic background, atmospheric radon, Compton scatter, and altitude dependence. The corrected data were statistically evaluated, plotted, and contoured to produce anomaly maps based on the radiometric response of individual geological units. The anomalies were interpreted and an interpretation map produced. Volume I contains a description of the systems used in the survey, a discussion of the calibration of the systems, the data collection procedures, the data processing procedures, the data presentation, the interpretation rationale, and the interpretation methodology. A separate Volume II for each quadrangle contains the data displays and the interpretation results.

    This report summarizes the efforts completed in identifying candidate fluorescence compounds and radioisotopes for a developing tagging system. The tagging system is being developed as a deterrent to nuclear smuggling, by providing a means of: (1) tracing materials and pilferers to the facility of origin for any recovered special nuclear materials, (2) inventory control of long-term stored items containing special nuclear materials, and (3) tracking materials transferred between facilities. The system uses three types of materials to cover a range of applications intended to prevent the pilfering of special nuclear materials. One material, fluorescent compounds which are invisible without ultraviolet or near-infrared detection systems, is marked on controlled items with a tracking pattern that corresponds to a specified item in a specified location in the data control system. The tagging system uses an invisible, fluorescent dusting powder to mark equipment and personnel who inappropriately handle the tagged material. The tagging system also uses unique combinations of radionuclides to identify the facility of origin for any special nuclear material. This report also summarizes the efforts completed in identifying hardware that will be used for the tagging system. This hardware includes the devices for applying the tagging materials, the commercially available fluorescence detection systems, and gamma ray detection systems assembled from existing, commercially available technologies.

    Residential development within the watershed of a fen complex has resulted in encroachment of ground-water borne anthropogenic contaminants into 3 high quality fens. Groundwater samples were collected from peat and marl and analyzed for inorganic constituents. Height and density of Typha angustifolia L. and Scirpus acutus Muhl. were recorded in belt transects coincident with ground-water samples. Contaminant plumes from septic systems of houses near the fens had higher Na, Cl, NO[sub 3], and TDS levels than uncontaminated areas of the fens. Typha density increased significantly as Na, Cl, CL/SO[sub 4], and TDS levels increased; Scirpus occurred only where pH [ge] 7.1 (X[sub 2]=11.656, sl=.02). Scirpus density was generally independent of other water chemistry levels. In one fen Scirpus density declined significantly as Typha density increased (r[sub 2]=25.86). Contamination appears to favor Tvpha invasion, which subsequently leads to displacement of Scirpus.

    This report summarizes the efforts completed in identifying candidate fluorescence compounds and radioisotopes for a developing tagging system. The tagging system is being developed as a deterrent to nuclear smuggling, by providing a means of: (1) tracing materials and pilferers to the facility of origin for any recovered special nuclear materials; (2) inventory control of long-term stored items containing special nuclear materials; and (3) tracking materials transferred between facilities. The tagging system uses four types of tagging materials to cover a range of applications intended to prevent the pilfering of special nuclear materials. One material, fluorescent compounds which are invisible without ultraviolet or near-infrared detection systems, is marked on controlled items with a tracking pattern that corresponds to a specified item in a specified location in the data control system. The tagging system uses an invisible, fluorescent dusting powder to mark equipment and personnel who inappropriately handle the tagged material. The tagging system also uses unique combinations of radionuclides to identify the facility of origin for any special nuclear material. Currently, 18 long-lived radioisotopes, 38 short-live radioisotopes and 10 fluorescent compounds have been selected as candidate materials for the tagging system.

    The overall objective of the Workshop was to identify opportunities for accelerating the use of both natural gas (NG) and hydrogen (H{sub 2}) as motor fuels and in stationary power applications. Specific objectives of the Workshop were to: (1) Convene industry and other stakeholders to share current status/state-of-the-art of NG and H{sub 2} infrastructure; (2) Identify key challenges (including non-technical challenges, such as permitting, installation, codes, and standards) preventing or delaying the widespread deployment of NG and H{sub 2} infrastructure. Identify synergies between NG and H{sub 2} fuels; and (3) Identify and prioritize opportunities for addressing the challenges identified above, and determine roles and opportunities for both the government and industry stakeholders. Plenary speakers and panel discussions summarized the current status of the NG and H{sub 2} infrastructure, technology for their use in transportation and stationary applications, and some of the major challenges and opportunities to more widespread use of these fuels. Two break-out sessions of three groups each addressed focus questions on: (1) infrastructure development needs; (2) deployment synergies; (3) natural gas and fuel cell vehicles (NGVs, FCVs), specialty vehicles, and heavy-duty trucks; (4) CHP (combined heat and power), CHHP (combined hydrogen, heat, and power), and synergistic approaches; and (5) alternative uses of natural gas.

    Basic principles of anaerobic digestion are considered along with the status of the Imperial Valley Biogas Project, the Department of Energy program for the recovery of energy and materials from urban waste, the principles of alcohol production from wastes, the mechanical recovery of a refuse-derived cellulosic feedstock for ethanol production, and the production of ethanol from cellulosic biomass. Attention is given to on-farm alcohol fuel production, the current status and future role of gasohol production, methane generation from small scale farms, farmsite installations of energy harvester anaerobic digesters, biogas/composting and landfill recovery, farm-scale composting as an option to anaerobic digestion, designing a high-quality biogas system, and methane as fuel of the future. A description is presented of subjects which are related to landfill gas recovery, biogas purification with permselective membranes, and anaerobic digestion of marine biomass. Other topics studied include the application of biogas technology in India, biogas production in China, biogasification of organic wastes in the Republic of the Philippines, and economics and operational experience of full-scale anaerobic dairy manure digester.

This report summarizes the findings from the evaluations, including the best practices identified at each site and opportunities for improving energy efficiency and renewable energy. The findings from this evaluation are intended to inform energy efficient building design, energy efficiency technology, and management protocols for buildings in subtropical climates.

    The U.S. Bureau of Mines diskette contains analysis and related source data for 2,357 natural gas samples collected from miscellaneous states, which include the following states: Alabama, Arkansas (except Arkoma Basin), Florida, Georgia, Illinois, Indiana, Iowa, Kentucky, Louisiana, Maryland, Michigan, Mississippi, Missouri, New Jersey, New York, North Carolina, Ohio, Pennsylvania, Tennessee, Virginia, and West Virginia. All samples were obtained and analyzed as part of the Bureau's investigations of occurrences of helium in natural gases of countries with free market economies. The survey has been conducted since 1917. The analysis contained on the diskette contain the full range of component analysis data. Five files are on the diskette: READ.ME, MISC.TXT, MISC.DBF, USHEANAL.DBF, and BASINCDE.TXT.
