    Discharge of oilfield brines into fresh and estuarine waters is a common disposal practice in Texas. Petroleum crude oil (PCO) extraction from underground stores includes the removal of a significant amount of water along with the oil. Several methods may be used to separate the oil and water fractions, including tank batteries, heat separation, and skimming ponds. Disposal of the resultant produced water (oilfield brine) may be accomplished by deep-well injection or discharge to surface waters. In Texas, an estimated 766,000 barrels of oilfield brine were discharged daily into tidal waters in 1979. The maximum concentration for oil and grease in these discharges permitted by the Texas Railroad Commission is 25 ppm. Several studies have shown that oilfield brines are toxic to a wide range of marine life, yet little is known about their effects on birds and mammals. Exposure to petroleum in oilfield wastes could evoke toxicological effects in some waterbird species. Avian responses to PCO exposure are highly variable, including cessation of growth, osmoregulatory impairment, endocrine dysfunction, hemolytic anemia, altered blood chemistry, cytochrome P450 induction, reduced reproductive success, and mortality. Oilfield brine discharges may soon be the largest and most pervasive source of contaminants entering Texas estuaries. Migratory and resident birds feeding in the vicinity of discharge sites may be ingesting food items contaminated with petroleum hydrocarbons, heavy metals and salts in sufficient quantities to evoke toxicity. The present study of wintering western sandpipers (Calidris mauri) that feed and roost near discharge sites sought to examine oilfield brine exposure and effects through quantification of contaminant burdens, morphological characteristics, and cytochrome P450-associated monooxygenase activities. 20 refs., 2 tabs.

    The Marburg virus (MARV), an African filovirus closely related to the Ebola virus, causes a deadly hemorrhagic fever in humans, with up to 90% mortality. Currently, treatment of disease is only supportive, and no vaccines are available to prevent spread of MARV infections. In order to address this need, we have developed and characterized a novel recombinant vaccine that utilizes a single complex adenovirus-vectored vaccine (cAdVax) to overexpress a MARV glycoprotein (GP) fusion protein derived from the Musoke and Ci67 strains of MARV. Vaccination with the cAdVaxM(fus) vaccine led to efficient production of MARV-specific antibodies in both mice and guinea pigs. Significantly, guinea pigs vaccinated with at least 5 x 10{sup 7} pfu of cAdVaxM(fus) vaccine were 100% protected against lethal challenges by the Musoke, Ci67 and Ravn strains of MARV, making it a vaccine with trivalent protective efficacy. Therefore, the cAdVaxM(fus) vaccine serves as a promising vaccine candidate to prevent and contain multi-strain infections by MARV.

    It was demonstrated previously that mast cells play an important role in citric acid (CA)-induced airway constriction. To investigate the role of mast cells in CA-induced cough, three experiments were carried out in this study. In the first experiment, 59 guinea pigs were employed and we used compound 48/80 to deplete mast cells, cromolyn sodium to stabilize mast cells, MK-886 to inhibit leukotriene synthesis, pyrilamine to antagonize histamine H{sub 1} receptor, methysergide to antagonize serotonin receptor, and indomethacin to inhibit cyclooxygenase. In the second experiment, 56 compound 48/80-pretreated animals were divided into two parts; the first one was used to test the role of exogenous leukotriene (LT) C{sub 4}, while the second one to test the role of exogenous histamine in CA-induced cough. Each animal with one of the above pretreatments was exposed sequentially to saline (baseline) and CA (0.6 M) aerosol, each for 3 min. Then, cough was recorded for 12 min using a barometric body plethysmograph. In the third experiment, the activation of mast cells upon CA inhalation was investigated by determining arterial plasma histamine concentration in 17 animals. Exposure to CA induced a marked increase in cough number. Compound 48/80, cromolyn sodium, MK-886 and pyrilamine, but not indomethacin or methysergide, significantly attenuated CA-induced cough. Injection of LTC{sub 4} or histamine caused a significant increase in CA-induced cough in compound 48/80-pretreated animals.     Alcohol consumption increases the small intestinal bacterial overgrowth (SIBO) and intestinal permeability of endotoxin. The endotoxin mediated inflammatory signaling plays a major role in alcoholic liver fibrosis. We evaluated the effect of ascorbic acid (AA), silymarin and alcohol abstention on the alcohol induced endotoxemia and NF-?B activation cascade pathway in guinea pigs (Cavia porcellus). Guinea pigs were administered ethanol at a daily dose of 4 g/kg b.wt for 90 days. After 90 days, ethanol administration was stopped. The ethanol treated animals were divided into abstention, silymarin (250 mg/kg b.wt) and AA (250 mg/kg b.wt) supplemented groups and maintained for 30 days. The SIBO, intestinal permeability and endotoxin were significantly increased in the ethanol group. The mRNA expressions of intestinal proteins claudin, occludin and zona occludens-1 were significantly decreased in ethanol group. The mRNA levels of inflammatory receptors, activity of IKK? and the protein expressions of phospho-I?B?, NF-?B, TNF-?, TGF-?{sub 1} and IL-6 were also altered in ethanol group. The expressions of fibrosis markers ?-SMA, ?{sub 1} (I) collagen and sirius red staining in the liver revealed the induction of fibrosis. But the supplementation of AA could induce greater reduction of ethanol induced SIBO, intestinal barrier defects, NF-?B activation and liver fibrosis than silymarin. The possible mechanism may be the inhibitory effect of AA on SIBO, intestinal barrier defect and IKK?, which decreased the activation of NF-?B and synthesis of cytokines. This might have led to suppression of HSCs activation and liver fibrosis. - Highlights: Alcohol increases intestinal bacterial overgrowth and permeability of endotoxin. Endotoxin mediated inflammation plays a major role in alcoholic liver fibrosis. Ascorbic acid reduces endotoxemia, NF-?B activation and proinflammatory cytokines. AA's action is by inhibition of SIBO, IKK? and alteration of intestinal permeability. This might have led to suppression of HSCs activation and liver fibrosis.

    Ebola virus (EBOV) protein VP35 is a double-stranded RNA (dsRNA) binding inhibitor of host interferon (IFN)-{alpha}/{beta} responses that also functions as a viral polymerase cofactor. Recent structural studies identified key features, including a central basic patch, required for VP35 dsRNA binding activity. To address the functional significance of these VP35 structural features for EBOV replication and pathogenesis, two point mutations, K319A/R322A, that abrogate VP35 dsRNA binding activity and severely impair its suppression of IFN-{alpha}/{beta} production were identified. Solution nuclear magnetic resonance (NMR) spectroscopy and X-ray crystallography reveal minimal structural perturbations in the K319A/R322A VP35 double mutant and suggest that loss of basic charge leads to altered function. Recombinant EBOVs encoding the mutant VP35 exhibit, relative to wild-type VP35 viruses, minimal growth attenuation in IFN-defective Vero cells but severe impairment in IFN-competent cells. In guinea pigs, the VP35 mutant virus revealed a complete loss of virulence. Strikingly, the VP35 mutant virus effectively immunized animals against subsequent wild-type EBOV challenge. These in vivo studies, using recombinant EBOV viruses, combined with the accompanying biochemical and structural analyses directly correlate VP35 dsRNA binding and IFN inhibition functions with viral pathogenesis. Moreover, these studies provide a framework for the development of antivirals targeting this critical EBOV virulence factor.

    Significant rightholding changes took place in central and southern Africa during 1987. Angola, Benin, Congo, Gabon, Ghana, Guinea, Guinea Bissau, Mauritania, Seychelles, Somali Republic, Tanzania, Zaire, and Zambia announced awards or acreage open for bidding. Decreases in exploratory rightholdings occurred in Cameroon, Congo, Cote d'Ivoire, Equatorial Guinea, Gabon, Kenya, Namibia, South Africa, and Tanzania. More wells and greater footage were drilled in 1987 than in 1986. Total wells increased by 18% as 254 wells were completed compared to 217 in 1986. Footage drilled during the year increased by 46% as about 1.9 million ft were drilled compared to about 1.3 million ft in 1986. The success rate for exploration wells in 1987 improved slightly to 36% compared to 34% in 1986. Significant discoveries were made in Nigeria, Angola, Congo, and Gabon. Seismic acquisition in 1987 was the major geophysical activity during the year. Total oil production in 1987 was 773 million bbl (about 2.1 million b/d), a decrease of 7%. The decrease is mostly due to a 14% drop in Nigerian production, which comprises 60% of total regional production. The production share of OPEC countries (Nigeria and Gabon) versus non-OPEC countries of 67% remained unchanged from 1986. 24 figs., 5 tabs.

    This document describes the contents of a digital database containing maximum potential aboveground biomass, land use, and estimated biomass and carbon data for 1980. The biomass data and carbon estimates are associated with woody vegetation in Tropical Africa. These data were collected to reduce the uncertainty associated with estimating historical releases of carbon from land use change. Tropical Africa is defined here as encompassing 22.7 x 10{sup 6} km{sup 2} of the earth's land surface and is comprised of countries that are located in tropical Africa (Angola, Botswana, Burundi, Cameroon, Cape Verde, Central African Republic, Chad, Congo, Benin, Equatorial Guinea, Ethiopia, Djibouti, Gabon, Gambia, Ghana, Guinea, Ivory Coast, Kenya, Liberia, Madagascar, Malawi, Mali, Mauritania, Mozambique, Namibia, Niger, Nigeria, Guinea-Bissau, Zimbabwe (Rhodesia), Rwanda, Senegal, Sierra Leone, Somalia, Sudan, Tanzania, Togo, Uganda, Burkina Faso (Upper Volta), Zaire, and Zambia). The database was developed using the GRID module in the ARC/INFO{trademark} geographic information system. Source data were obtained from the Food and Agriculture Organization (FAO), the U.S. National Geophysical Data Center, and a limited number of biomass-carbon density case studies. These data were used to derive the maximum potential and actual (ca. 1980) aboveground biomass values at regional and country levels. The land-use data provided were derived from a vegetation map originally produced for the FAO by the International Institute of Vegetation Mapping, Toulouse, France.

Another issue that may impact the plans is the environmental taxes that may reduce the call on OPEC oil by 5 million b/d in 2000 and about 16 million b/d in the year 2010. This concluding part of a two-part series discusses the expansion possibilities of non-Middle East OPEC members, OPEC's capital requirements, and environmental concerns.     The vast continent of Africa hosts over eight sedimentary basins, covering approximately half its total area. Of these basins, only 82% have entered a mature exploration phase, 9% have had little or no exploration at all. Since oil was first discovered in Africa during the mid-1950s, old play concepts continue to bear fruit, for example in Egypt and Nigeria, while new play concepts promise to become more important, such as in Algeria, Angola, Chad, Egypt, Gabon, and Sudan. The most exciting developments of recent years in African oil exploration are: (1) the Gamba/Dentale play, onshore Gabon; (2) the Pinda play, offshore Angola; (3) the Lucula/Toca play, offshore Cabinda; (4) the Metlaoui play, offshore Libya/Tunisia; (5) the mid-Cretaceous sand play, Chad/Sudan; and (6) the TAG-I/F6 play, onshore Algeria. Examples of these plays are illustrated along with some of the more traditional oil plays. Where are the future oil plays likely to develop No doubt, the Saharan basins of Algeria and Libya will feature strongly, also the presalt of Equatorial West Africa, the Central African Rift System and, more speculatively, offshore Ethiopia and Namibia, and onshore Madagascar, Mozambique, and Tanzania.

    U{sup 6+}-phases are common alteration products, under oxidizing conditions, of uraninite and the UO{sub 2} in spent nuclear fuel. These U{sup 6+}-phases are subjected to a radiation field caused by the {alpha}-decay of U, or in the case of spent nuclear fuel, incorporated actinides, such as {sup 239}Pu and {sup 237}Np. In order to evaluate the effects of {alpha}-decay events on the stability of the U{sup 6+}-phases, we report, for the first time, the results of ion beam irradiations (1.0 MeV Kr{sup 2+}) of U{sup 6+}-phases. The heavy-particle irradiations are used to simulate the ballistic interactions of the recoil-nucleus of an {alpha}-decay event with the surrounding structure. The Kr{sup 2+}-irradiation decomposed the U{sup 6+}-phases to UO{sub 2} nanocrystals at doses as low as 0.006 displacements per atom (dpa). U{sup 6+}-phases accumulate substantial radiation doses ({approx}1.0 displacement per atom) within 100,000 years if the concentration of incorporated {sup 239}Pu is as high as 1 wt%. Similar nanocrystals of UO{sub 2} were observed in samples from the natural fission reactors at Oklo, Gabon.     There is strong evidence that the lower atmosphere has been warming at an unprecedented rate during the last 50 years, and it is expected to further increase at least for the next 100 years. Warmer air mass implies a higher capacity to hold water vapor and an increased likelihood of an acceleration of the global water cycle. This acceleration is not validated and considerable new research has gone into understanding aspects of the water cycle (e.g. Miller et al. 2003). Several significant findings on the hydrologic response to climate change can be reported. It is well understood that the observed and expected warming is related to sea level rise. In a recent seminar at Lawrence Berkeley National Laboratory, James Hansen (Director of the Institute for Space Studies, National Aeronautics and Space Administration) stressed that a 1.25 Wm{sup -2} increase in radiative forcing will lead to an increase in the near surface air temperature by 1 C. This small increase in temperature from 2000 levels is enough to cause very significant impacts to coasts. Maury Roos (Chief Hydrologist, California Department of Water Resources) has shown that a 0.3 m rise in sea level shifts the San Francisco Bay 100-year storm surge flood event to a 10-year event. Related coastal protection costs for California based on sea level rise are shown. In addition to rising sea level, snowmelt-related streamflow represents a particular problem in California. Model studies have indicated that there will be approximately a 50% decrease in snow pack by 2100. This potential deficit must be fully recognized and plans need to be put in place well in advance. In addition, the warmer atmosphere can hold more water vapor and result in more intense warm winter-time precipitation events that result in flooding. During anticipated high flow, reservoirs need to release water to maintain their structural integrity. California is at risk of water shortages, floods, and related ecosystem stresses. More research needs to be done to further improve our ability to forecast weather events at longer time scales. Seasonal predictions have been statistical and only recently have studies begun to use ensemble simulations and historical observations to constrain such predictions. Understanding the mechanisms of large-scale atmospheric dynamics and its local impacts remain topics of intensive research. The ability to predict extreme events and provide policy makers with this information, along with climate change and hydrologic response information, will help to guide planning to form a more resilient infrastructure in the future.

    During irradiation of nuclear fuel in a reactor, the five metals, Mo, Pd, Rh, Ru, and Tc, migrate to the fuel grain boundaries and form small metal particles of an alloy known as epsilon metal ({var_epsilon}-metal). When the fuel is dissolved in a reprocessing plant, these metal particles remain behind with a residue - the undissolved solids (UDS). Some of these same metals that comprise this alloy that have not formed the alloy are dissolved into the aqueous stream. These metals limit the waste loading for a borosilicate glass that is being developed for the reprocessing wastes. Epsilon metal is being developed as a waste form for the noble metals from a number of waste streams in the aqueous reprocessing of used nuclear fuel (UNF) - (1) the {var_epsilon}-metal from the UDS, (2) soluble Tc (ion-exchanged), and (3) soluble noble metals (TRUEX raffinate). Separate immobilization of these metals has benefits other than allowing an increase in the glass waste loading. These materials are quite resistant to dissolution (corrosion) as evidenced by the fact that they survive the chemically aggressive conditions in the fuel dissolver. Remnants of {var_epsilon}-metal particles have survived in the geologically natural reactors found in Gabon, Africa, indicating that they have sufficient durability to survive for {approx} 2.5 billion years in a reducing geologic environment. Additionally, the {var_epsilon}-metal can be made without additives and incorporate sufficient foreign material (oxides) that are also present in the UDS. Although {var_epsilon}-metal is found in fuel and Gabon as small particles ({approx}10 {micro}m in diameter) and has survived intact, an ideal waste form is one in which the surface area is minimized. Therefore, the main effort in developing {var_epsilon}-metal as a waste form is to develop a process to consolidate the particles into a monolith. Individually, these metals have high melting points (2617 C for Mo to 1552 C for Pd) and the alloy is expected to have a high melting point as well, perhaps exceeding 1500 C. The purpose of the work reported here is to find a potential commercial process with which {var_epsilon}-metal plus other components of UDS can be consolidated into a solid with minimum surface area and high strength Here, we report the results from the preliminary evaluation of spark-plasma sintering (SPS), hot-isostatic pressing (HIP), and microwave sintering (MS). Since bulk {var_epsilon}-metal is not available and companies could not handle radioactive materials, we prepared mixtures of the five individual metal powders (Mo, Ru, Rh, Pd, and Re) and baddeleyite (ZrO{sub 2}) to send the vendors of SPS, HIP, and MS. The processed samples were then evaluated at the Pacific Northwest National Laboratory (PNNL) for bulk density and phase assemblage with X-ray diffraction (XRD) and phase composition with scanning electron microscopy (SEM). Physical strength was evaluated qualitatively. Results of these scoping tests showed that fully dense cermet (ceramic-metal composite) materials with up to 35 mass% of ZrO{sub 2} were produced with SPS and HIP. Bulk density of the SPS samples ranged from 87 to 98% of theoretical density, while HIP samples ranged from 96 to 100% of theoretical density. Microwave sintered samples containing ZrO{sub 2} had low densities of 55 to 60% of theoretical density. Structurally, the cermet samples showed that the individual metals alloyed in to {var_epsilon}-phase - hexagonal-close-packed (HCP) alloy (4-95 mass %), the {alpha}-phase - face-centered-cubic (FCC) alloy structure (3-86 mass %), while ZrO{sub 2} remained in the monoclinic structure of baddeleyite. Elementally, the samples appeared to have nearly uniform composition, but with some areas rich in Mo and Re, the two components with the highest melting points. The homogeneity in distribution of the elements in the alloy is significantly improved in the presence of ZrO{sub 2}. However, ZrO{sub 2} does not appear to react with the alloy, nor was Zr found in the alloy.

  16. LFSC - Linac Feedback Simulation Code

    The computer program LFSC () is a numerical tool for simulation beam based feedback in high performance linacs. The code LFSC is based on the earlier version developed by a collective of authors at SLAC (L.Hendrickson, R. McEwen, T. Himel, H. Shoaee, S. Shah, P. Emma, P. Schultz) during 1990-2005. That code was successively used in simulation of SLC, TESLA, CLIC and NLC projects. It can simulate as pulse-to-pulse feedback on timescale corresponding to 5-100 Hz, as slower feedbacks, operating in the 0.1-1 Hz range in the Main Linac and Beam Delivery System. The code LFSC is running under Matlab for MS Windows operating system. It contains about 30,000 lines of source code in more than 260 subroutines. The code uses the LIAR ('Linear Accelerator Research code') for particle tracking under ground motion and technical noise perturbations. It uses the Guinea Pig code to simulate the luminosity performance. A set of input files includes the lattice description (XSIF format), and plane text files with numerical parameters, wake fields, ground motion data etc. The Matlab environment provides a flexible system for graphical output.

    Expression of internal receptors for MSH is an important criterion for responsiveness to MSH by Cloudman melanoma cells. Here, we show that internal and external receptors for MSH are of identical molecular weights (50-53 kDa) and share common antigenic determinants, indicating a structural relationship between the 2 populations of molecules. The internal receptors co-purified with a sub-cellular fraction highly enriched for small vesicles, many of which were coated. Ultraviolet B light (UVB) acted synergistically with MSH to increase tyrosinase activity and melanin content of cultured Cloudman melanoma cells, consistent with previous findings in the skin of mice and guinea pigs. Preceding the rise in tyrosinase activity in cultured cells, UVB elicited a decrease in internal MSH binding sites and a concomitant increase in external sites. The time frame for the UVB effects on MSH receptors and melanogenesis, 48 hours, was similar to that for a response to solar radiation in humans. Together, the results indicate a key role for MSH receptors in the induction of melanogenesis by UVB and suggest a potential mechanism of action for UVB: redistribution of MSH receptors with a resultant increase in cellular responsiveness to MSH.

  18. Inhalation toxicology of red and violet mixtures. Chamber concentration and particle-size distribution report

    An inhalation exposure facility was developed at the U.S. EPA, RTP, NC to conduct inhalation exposures of rodents and guinea pigs to dye mixtures used by the U.S. Army in the manufacture of smoke munitions. Initially, an evaluation of the prototype chamber aerosol homogeneity was conducted to determine the uniformity and reproducibility of the concentration and particle size of dye aerosol throughout the breathing zone of the test animals. The three dyes, DR11, SR1, and DB3, were chemically analyzed for purity and optically examined for size and shape. All pure dyes appeared to be stable at room temperature except DB3, which decomposes if not stored at 4 C. The particle size ranges varied for each pure dye and structures were either amorphous (azo dye) or crystalline (anthraquinone dyes). The bulk red and violet dye mixtures were analyzed for composition. The chemical analysis of the relative composition of each dye mixture, collected by cascade impactor sampling, revealed fractionation of the mixtures into component dyes.

    The tropical western Pacific (TWP) is an important climatic region. Strong solar heating, warm sea surface temperatures and the annual progression of the Intertropical Convergence Zone (ITCZ) across this region generate abundant convective systems, which through their effects on the heat and water budgets have a profound impact on global climate and precipitation. To accurately represent tropical cloud systems in models, measurements of tropical clouds, the environment in which they reside, and their impact on the radiation and water budgets are needed. Because of the remote location, ground-based datasets of cloud, atmosphere, and radiation properties from the TWP region have traditionally come primarily from short-term field experiments. While providing extremely useful information on physical processes, these datasets are limited in statistical and climatological information because of their short duration. To provide long-term measurements of the surface radiation budget in the tropics, and the atmospheric properties that affect it, the Atmospheric Radiation Measurement program established a measurement site on Manus Island, Papua New Guinea in 1996 and on the island republic of Nauru in late 1998. These sites provide unique datasets available from more than 10 years of operation in the equatorial western Pacific on Manus and Nauru. We present examples of the scientific use of these datasets including characterization of cloud properties, analysis of cloud radiative forcing, model studies of tropical clouds and processes, and validation of satellite algorithms. We also note new instrumentation recently installed at the Manus site that will expand opportunities for tropical atmospheric science.

  1. The bovine tuberculosis burden in cattle herds in zones with low dose radiation pollution in the Ukraine

    The authors describe a study of the tuberculosis (TB) incidence in cattle exposed to low doses of radiation resulting from the Chernobyl (pronounced Chornobyl in Ukrainian) nuclear plant catastrophe in 1986. The purpose of the study was to determine if ionising radiation influences the number of outbreaks of bovine TB and their severity on farms in the Kyiv, Cherkasy and Chernigiv regions of the Ukraine. These farms are all located within a 200 km radius of Chernobyl and have had low?dose radiation pollution. Pathological and blood samples were taken from cattle in those regions that had positive TB skin tests. Mycobacterium spp. were isolated, differentiated by PCR, analysed and tested in guinea?pigs and rabbits. Species differentiation showed a significant percentage of atypical mycobacteria, which resulted in the allergic reactions to tuberculin antigen in the skin test. Mixed infection of M. bovis and M. avium subsp. hominissuis was found in three cases. The results concluded that low?dose radiation plays a major role in the occurrence of bovine TB in regions affected by the Chernobyl nuclear disaster.

  2. Atmospheric Radiation Measurement (ARM) Data from the Tropical Western Pacific (TWP) Site.

    ARM maintains four major, permanent sites for data collection and deploys the ARM Mobile Facility to other sites as determined. The Tropical Western Pacific (TWP) site is one of the four fixed sites. It consists of three climate research facilities; the Manus facility on Los Negros Island in Manus, Papua New Guinea (established in 1996); the Nauru facility on Nauru Island, Republic of Nauru (1998); and the Darwin facility in Darwin, Northern Territory, Australia (2002). The operations are supported by government agencies in each host country. Covering the area roughly between 10 degrees N and 10 degrees S of the equator and from 130 degrees E to 167 degrees E, the TWP locale includes a region that plays a large role in the interannual variability observed in the global climate system. More than 250,000 TWP data sets from 1996 to the present reside in the ARM Archive. Begin at the TWP information page for links or access data directly from the ARM Archive at http://www.archive.arm.gov/.     The Epsilon-metal ({var_epsilon}-metal) phase was selected in FY 2009 as a potential waste form to for immobilizing the noble metals found in the undissolved solids + aqueous stream, and the soluble Tc from ion-exchange process, each resulting from proposed aqueous reprocessing. {var_epsilon}-metal phase is observed in used nuclear fuel and the natural reactors of Oklobono in Gabon, where the long-term corrosion behavior was demonstrated. This makes {var_epsilon}-metal a very attractive waste form. Last fiscal year, {var_epsilon}-metal was successfully fabricated by combining the five-metals, Mo, Ru, Rh, Pd and Re (surrogate for Tc), into pellets followed by consolidation with an arc melter. The arc melter produced fully dense samples with the epsilon structure. However, some chemistry differences were observed in the microstructure that resulted in regions rich in Re and Mo, and others rich in Pd, while Ru and Rh remained fairly constant throughout. This year, thermal stability (air), and corrosion testing of the samples fabricated by arc melting were the main focus for experimental work. Thermal stability was measured with a differential scanning calorimeter - thermogravimetric analyzer, by both ramp heating as well as step heating. There is clear evidence during the ramp heating experiment of an exothermic event + a weight loss peak both beginning at {approx}700 C. Step heating showed an oxidation event at {approx}690 C with minimal weight gain that occurs just before the weight loss event at 700 C. The conclusion being that the e-metal begins to oxidize and then become volatile. These findings are useful for considering the effects of voloxidation process. Three different pellets were subjected to electrochemical testing to study the corrosion behavior of the epsilon-metal phase in various conditions, namely acidic, basic, saline, and inert. Test was done according to an interim procedure developed for the alloy metal waste form. First an open circuit potential was measured, followed by linear polarization sweeps. The linear polarization sweep range was the Tafel equation was fit to the linear polarization sweep data to determine the corrosion rate of each pellet in each test solution. The average calculated corrosion rates of the three pellets according to solution conditions were: -1.91 x 10{sup -4} mm/yr (0.001 M NaOH), -1.48 x 10{sup -3} mm/yr (0.01 M NaCl), -8.77 x 10{sup -4} mm/yr (0.001 M H{sub 2}SO{sub 4}), -2.09 x 10{sup -3} mm/yr (0.001 M NaOH + 0.01 M NaCl), and -1.54 x 10{sup -3} mm/yr (0.001 M H{sub 2}SO{sub 4} + 0.01 M NaCl). Three single-pass flow through (SPFT) test were conducted at a flow rate of 10 ml/day, at 90 C, and pH of 2.5, 7.0, and 9.0 for up to 322 days. Results of the tests indicate that dissolution rates were 5 x 10{sup -4} g m{sup 2} d{sup -1} at pH 9.0, 1.2 x 10{sup -4} g m{sup -2} d{sup -1} at pH 7.0, and 2 x 10{sup -4} g m{sup -2} d{sup -1} at pH 2.5. The sample used for the pH 7.0 SPFT test contains extra Re compared to samples used for the other two SPFT test, which came from a single pellet. The corrosion data measured this year indicate that the {var_epsilon}-metal phase is chemically durable. The two chemically different phases, but structurally the same, behave differently during dissolution according to the microstructure changes observed in both the electrochemical and in SPFT test. Characterization of the test specimens after testing suggests that the dissolution is complex and involves oxidative dissolution followed by precipitation of both oxide and metallic phases. These data suggest that the dissolution in the electrochemical and SPFT tests is different; a process that needs further investigation.

    The intracellular calcium concentration ([Ca{sup 2+}]{sub i}) of isolated ventricular cardiac myocytes of the guinea pig was measured during the application of pulsed high-frequency electromagnetic fields. The high-frequency fields were applied in a transverse electromagnetic cell designed to allow microscopic observation of the myocytes during the presence of the high-frequency fields. The [Ca{sup 2+}]{sub i} was measured as fura-2 fluorescence by means of digital image analysis. Both the carrier frequency and the square-wave pulse-modulation pattern were varied during the experiments (carrier frequencies: 900, 1,300, and 1,800 MHz pulse modulated at 217 Hz with 14% duty cycle; pulsation pattern at 900 MHz; continuous wave, 16 Hz,and 50 Hz modulation with 50% duty cycle and 30 kHz modulation with 80% duty cycle). The mean specific absorption rate (SAR) values in the solution were within one order of magnitude of 1 mW/kg. They varied depending on the applied carrier frequency and pulse pattern. The experiments were designed in three phases: 500 s of sham exposure, followed by 500 s of field exposure, then chemical stimulation without field. The chemical stimulation (K{sup +}-depolarization) indicated the viability of the cells. The K{sup +} depolarization yielded a significant increase in [Ca{sup 2+}]{sub i}. Significant differences between sham exposure and high-frequency field exposure were not found except when a very small but statistically significant difference was detected in the case of 900 MHz/50 Hz. However, this small difference was not regarded as a relevant effect of the exposure.

  5. The formation and maintenance of single-thread tie channels entering floodplain lakes: observations from three diverse river systems

    Tie channels connect rivers to floodplain lakes on many lowland rivers and thereby play a central role in floodplain sedimentology and ecology, yet they are generally unrecognized and little studied. here we report the results of field studies focused on tie channel origin and morphodynamics in three contrasting systems: the Middle Fly River, Papua New Guinea, the Lower Mississippi River, and Birch Creek in Alaska. Across these river systems, tie channels vary by an order of magnitude in size but exhibit the same characteristic morphology and appear to develop and evolve by a similar set of processes. In all three systems, the channels are characterized by a narrow, leveed single-thread morphology with maximum width approximately one tenth the width of the mainstem river. The channels typically have a V shaped cross-section, unlike most fluvial channels. These channels develop as lakes become isolated from the river by sedimentation. Narrowing of the connection between river and lake causes a sediment-laden jet to develop. Levees develop along the margins of the jet leading to channel emergence and eventual levee aggradation to the height of the mainstem levees. Bi-directional flow in these channels is common. Outflows from the lake scour sediment and prevent channel blockage. We propose that channel geometry and size are then controlled by a dynamic balance between channel narrowing by suspended sediment deposition and incision and widening by mass failure of banks during outflows. Tie channels are laterally stable and may convey flow for hundreds to a few thousand of years.

  6. Inter-annual Tropospheric Aerosol Variability in Late Twentieth Century and its Impact on Tropical Atlantic and West African Climate by Direct and Semi-direct Effects

    A new high-resolution (0.9$^{\\circ}$x1.25$^{\\circ}$ in the horizontal) global tropospheric aerosol dataset with monthly resolution is generated using the finite-volume configuration of Community Atmosphere Model (CAM4) coupled to a bulk aerosol model and forced with recent estimates of surface emissions for the latter part of twentieth century. The surface emissions dataset is constructed from Coupled Model Inter-comparison Project (CMIP5) decadal-resolution surface emissions dataset to include REanalysis of TROpospheric chemical composition (RETRO) wildfire monthly emissions dataset. Experiments forced with the new tropospheric aerosol dataset and conducted using the spectral configuration of CAM4 with a T85 truncation (1.4$^{\\circ}$x1.4$^{\\circ}$) with prescribed twentieth century observed sea surface temperature, sea-ice and greenhouse gases reveal that variations in tropospheric aerosol levels can induce significant regional climate variability on the inter-annual timescales. Regression analyses over tropical Atlantic and Africa reveal that increasing dust aerosols can cool the North African landmass and shift convection southwards from West Africa into the Gulf of Guinea in the spring season in the simulations. Further, we find that increasing carbonaceous aerosols emanating from the southwestern African savannas can cool the region significantly and increase the marine stratocumulus cloud cover over the southeast tropical Atlantic ocean by aerosol-induced diabatic heating of the free troposphere above the low clouds. Experiments conducted with CAM4 coupled to a slab ocean model suggest that present day aerosols can shift the ITCZ southwards over the tropical Atlantic and can reduce the ocean mixed layer temperature beneath the increased marine stratocumulus clouds in the southeastern tropical Atlantic.

  7. Chemical, biomedical and ecological studies of SRC-I materials from the Fort Lewis Pilot Plant: a status report

    This document discusses studies performed with solvent refined coal (SRC) materials obtained from the Fort Lewis Pilot Plant during operation in the SRC-I mode. The development of analytical methodology is presented as well as results obtained from the application of these methods to light oil (LO), wash solvent (WS) and process solvent (PS). Results of cellular and animal studies with LO, WS and PS are included, along with a description of methods for the generation and characterization of LO and PS aerosols, and for exposing rats, mice and guinea pigs to these aerosols. The effects of SRC-I product on seed germination and plant growth which have also been studied are discussed. The SRC-I product, feed coal and the mineral residue have been analyzed for organic and inorganic constituents. The higher-boiling-point material, PS, exhibited significant mutagenic activity in the Ames assay; LO and WS were inactive. Process solvent also caused transformation of cultured Syrian hamster embryo cells. Additional chemical fractionation studies suggest that primary aromatic amines are major determinants of the observed mutagenic activity. Skin-painting studies with SRC-II naphtha, heavy distillate, shale oil and petroleum crude indicate a good correlation between the results of the cellular assays and skin carcinogenesis in mice. Wash solvent was more toxic after oral administration to rats than was light oil or process solvent. The effects of LO, WS and PS on development were studied after administration to pregnant rats. The tissue distribution of a number of components of PS was studied after oral administration of PS to rats. The effect of SRC-I product on the germination and growth of barley was investigated by mixing or layering the product with soil and placing the mixture in a field lysimeter.

  8. Mechanisms of aldehyde-induced bronchial reactivity: role of airway epithelium

    To investigate the relative irritant potencies of inhaled aldehydes, guinea pigs were exposed to formaldehyde or acrolein and specific total pulmonary resistance and bronchial reactivity to intravenous acetylcholine were assessed. The mechanisms associated with these responses were investigated by analyzing morphologic and biochemical changes in airway epithelial cells after in vivo and in vitro exposures. Immediately after exposure to formaldehyde or acrolein, specific resistance increased transiently and returned to control values within 30 to 60 minutes. Bronchial hyperreactivity, assessed by the acetylcholine dose necessary to double resistance, increased and became maximal two to six hours after exposure to at least 9 parts per million2 (ppm) formaldehyde or at least 1 ppm acrolein for two hours. The effect of exposure to 3 ppm formaldehyde for two hours was less than the effect of exposure to 1 ppm formaldehyde for eight hours; thus, extended exposures produced a disproportionate heightening of bronchial reactivity. Bronchial hyperreactivity often persisted for longer than 24 hours. Increases in three bronchoconstrictive eicosanoids, prostaglandin F2 alpha, thromboxane B2, and leukotriene C4, occurred immediately after exposure, whereas an influx of neutrophils into lavage fluid occurred 24 hours later. Histological examination of the tracheal epithelium and lamina propria also demonstrated a lack of inflammatory cell infiltration. Treatment with leukotriene synthesis inhibitors and receptor antagonists inhibited acrolein-induced hyperreactivity, supporting a causal role for these compounds in this response. Acrolein also stimulated eicosanoid release from bovine epithelial cells in culture. However, the profile of metabolites formed differed from that found in lavage fluid after in vivo exposure.

  9. The evolution and hydrocarbon habitat of the Papuan fold belt, PNG

    After over 70 years of hydrocarbon exploration in the Papuan fold belt of PNG (Papua New Guinea) there have been a number of hydrocarbon discoveries over recent years that have confirmed its potential as a significant producing province. The Papuan basin developed during the early Mesozoic as part of the northeast corner of the Australian passive margin. The basin's tertiary evolution and the development of the Papuan fold belt within the Papuan basin has evolved in response to oblique convergence between the northerly moving Australian plate and westerly moving Pacific plate. Restacking of the Mesozoic passive margin sequence within the Papuan Basin was initiated in the early miocene by southward abduction of the Solomon Sea plate and the subsequent collision, in the late Miocene, of the Melanesian Island arc along the northeastern margin of PNG. This later collision provided the driving mechanism for the development of the papuan thrust belt. To date, all the significant hydrocarbon discoveries made within the Papuan fold belt have been located within the frontal zone of the fold belt, which is characterized by relatively simple ramp anticlines and thick-skinned inversion structures. The primary proven reservoir fairway is the Jurassic Toro formation, which is a sequence of stacked submarine bars prograding out across a shallow-marine low-gradient shelf. Geochemical analysis of produced hydrocarbons and samples collected from the many surface seeps found in the fold belt indicate two main families of oil. A model explains the distribution of hydrocarbons discovered to date, which involves Jurassic and Cretaceous source intervals and a complex history of secondary migration and entrapment. The unique technical problems associated with exploration of the Papuan fold belt leave many elements of the proven play systems uncertain, but in so doing, they present many challenges and opportunities for the future.

    The plant alkaloid ibogaine has promising anti-addictive properties. Albeit not licenced as a therapeutic drug, and despite hints that ibogaine may perturb the heart rhythm, this alkaloid is used to treat drug addicts. We have recently reported that ibogaine inhibits human ERG (hERG) potassium channels at concentrations similar to the drugs affinity for several of its known brain targets. Thereby the drug may disturb the heart's electrophysiology. Here, to assess the drug's cardiac ion channel profile in more detail, we studied the effects of ibogaine and its congener 18-Methoxycoronaridine (18-MC) on various cardiac voltage-gated ion channels. We confirmed that heterologously expressed hERG currents are reduced by ibogaine in low micromolar concentrations. Moreover, at higher concentrations, the drug also reduced human Na{sub v}1.5 sodium and Ca{sub v}1.2 calcium currents. Ion currents were as well reduced by 18-MC, yet with diminished potency. Unexpectedly, although blocking hERG channels, ibogaine did not prolong the action potential (AP) in guinea pig cardiomyocytes at low micromolar concentrations. Higher concentrations (? 10 ?M) even shortened the AP. These findings can be explained by the drug's calcium channel inhibition, which counteracts the AP-prolonging effect generated by hERG blockade. Implementation of ibogaine's inhibitory effects on human ion channels in a computer model of a ventricular cardiomyocyte, on the other hand, suggested that ibogaine does prolong the AP in the human heart. We conclude that therapeutic concentrations of ibogaine have the propensity to prolong the QT interval of the electrocardiogram in humans. In some cases this may lead to cardiac arrhythmias. - Highlights: We study effects of anti-addiction drug ibogaine on ionic currents in cardiomyocytes. We assess the cardiac ion channel profile of ibogaine. Ibogaine inhibits hERG potassium, sodium and calcium channels. Ibogaines effects on ion channels are a potential source of cardiac arrhythmias. 18-Methoxycoronaridine has a lower affinity for cardiac ion channels than ibogaine.

  11. Radiative Heating of the ISCCP Upper Level Cloud Regimes and its Impact on the Large-scale Tropical Circulation

    Radiative heating profiles of the International Satellite Cloud Climatology Project (ISCCP) cloud regimes (or weather states) were estimated by matching ISCCP observations with radiative properties derived from cloud radar and lidar measurements from the Department of Energy (DOE) Atmospheric Radiation Measurement (ARM) sites at Manus, Papua New Guinea, and Darwin, Australia. Focus was placed on the ISCCP cloud regimes containing the majority of upper level clouds in the tropics, i.e., mesoscale convective systems (MCSs), deep cumulonimbus with cirrus, mixed shallow and deep convection, and thin cirrus. At upper levels, these regimes have average maximum cloud occurrences ranging from 30% to 55% near 12 km with variations depending on the location and cloud regime. The resulting radiative heating profiles have maxima of approximately 1 K/day near 12 km, with equal heating contributions from the longwave and shortwave components. Upper level minima occur near 15 km, with the MCS regime showing the strongest cooling of 0.2 K/day and the thin cirrus showing no cooling. The gradient of upper level heating ranges from 0.2 to 0.4 K/(day?km), with the most convectively active regimes (i.e., MCSs and deep cumulonimbus with cirrus) having the largest gradient. When the above heating profiles were applied to the 25-year ISCCP data set, the tropics-wide average profile has a radiative heating maximum of 0.45Kday-1 near 250 hPa. Column-integrated radiative heating of upper level cloud accounts for about 20% of the latent heating estimated by the Tropical Rainfall Measuring Mission (TRMM) Precipitation Radar (PR). The ISCCP radiative heating of tropical upper level cloud only slightly modifies the response of an idealized primitive equation model forced with the tropics-wide TRMM PR latent heating, which suggests that the impact of upper level cloud is more important to large-scale tropical circulation variations because of convective feedbacks rather than direct forcing by the cloud radiative heating profiles. However, the height of the radiative heating maxima and gradient of the heating profiles are important to determine the sign and patterns of the horizontal circulation anomaly driven by radiative heating at upper levels.

  12. Parameterization and analysis of 3-D radiative transfer in clouds

    This report provides a summary of major accomplishments from the project. The project examines the impact of radiative interactions between neighboring atmospheric columns, for example clouds scattering extra sunlight toward nearby clear areas. While most current cloud models donâ??t consider these interactions and instead treat sunlight in each atmospheric column separately, the resulting uncertainties have remained unknown. This project has provided the first estimates on the way average solar heating is affected by interactions between nearby columns. These estimates have been obtained by combining several years of cloud observations at three DOE Atmospheric Radiation Measurement (ARM) Climate Research Facility sites (in Alaska, Oklahoma, and Papua New Guinea) with simulations of solar radiation around the observed clouds. The importance of radiative interactions between atmospheric columns was evaluated by contrasting simulations that included the interactions with those that did not. This study provides lower-bound estimates for radiative interactions: It cannot consider interactions in cross-wind direction, because it uses two-dimensional vertical cross-sections through clouds that were observed by instruments looking straight up as clouds drifted aloft. Data from new DOE scanning radars will allow future radiative studies to consider the full three-dimensional nature of radiative processes. The results reveal that two-dimensional radiative interactions increase overall day-and-night average solar heating by about 0.3, 1.2, and 4.1 Watts per meter square at the three sites, respectively. This increase grows further if one considers that most large-domain cloud simulations have resolutions that cannot specify small-scale cloud variability. For example, the increases in solar heating mentioned above roughly double for a fairly typical model resolution of 1 km. The study also examined the factors that shape radiative interactions between atmospheric columns and found that local effects were often much larger than the overall values mentioned above, and were especially large for high sun and near convective clouds such as cumulus. The study also found that statistical methods such as neural networks appear promising for enabling cloud models to consider radiative interactions between nearby atmospheric columns. Finally, through collaboration with German scientists, the project found that new methods (especially one called â??stepwise krigingâ?) show great promise in filling gaps between cloud radar scans. If applied to data from the new DOE scanning cloud radars, these methods can yield large, continuous three-dimensional cloud structures for future radiative simulations.

    The extremely high fatality rates of many filovirus (FILV) strains the recurrent but rarely identified origin of human epidemics, the only partly identified viral reservoirs and the continuing non-human primate epizootics in Africa make a broadly-protective filovirus vaccine highly desirable. Cytotoxic T-cells (CTL) have been shown to be protective in mice, guinea pigs and non-human primates. In murine models the cytotoxic T-cell epitopes that are protective against Ebola virus have been mapped and in non-human primates CTL-mediated protection between viral strains (John Dye: specify) has been demonstrated using two filoviral proteins, nucleoprotein (NP) and glycoprotein (GP). These immunological results suggest that the CTL avenue of immunity deserves consideration for a vaccine. The poorly-understood viral reservoirs means that it is difficult to predict what strains are likely to cause epidemics. Thus, there is a premium on developing a pan-filoviral vaccine. The genetic diversity of FILV is large, roughly the same scale as human immunodeficiency virus (HIV). This presents a serious challenge for the vaccine designer because a traditional vaccine aspiring to pan-filoviral coverage is likely to require the inclusion of many antigenic reagents. A recent method for optimizing cytotoxic T-cell lymphocyte epitope coverage with mosaic antigens was successful in improving potential CTL epitope coverage against HIV and may be useful in the context of very different viruses, such as the filoviruses discussed here. Mosaic proteins are recombinants composed of fragments of wild-type proteins joined at locations resulting in exclusively natural k-mers, 9 {le} k {le} 15, and having approximately the same length as the wild-type proteins. The use of mosaic antigens is motivated by three conjectures: (1) optimizing a mosaic protein to maximize coverage of k-mers found in a set of reference proteins will give better odds of including broadly-protective CTL epitopes in a vaccine than is possible with a wild-type protein, (2) reducing the number of low-prevalence k-mers minimizes the likelihood of undesirable immunodominance, and (3) excluding exogenous k-mers will result in mosaic proteins whose processing for presentation is close to what occurs with wild-type proteins. The first and second applications of the mosaic method were to HIV and Hepatitis C Virus (HCV). HIV is the virus with the largest number of known sequences, and consequently a plethora of information for the CTL vaccine designer to incorporate into their mosaics. Experience with HIV and HCV mosaics supports the validity of the three conjectures above. The available FILV sequences are probably closer to the minimum amount of information needed to make a meaningful mosaic vaccine candidate. There were 532 protein sequences in the National Institutes of Health GenPept database in November 2007 when our reference set was downloaded. These sequences come from both Ebola and Marburg viruses (EBOV and MARV), representing transcripts of all 7 genes. The coverage of viral diversity by the 7 genes is variable, with genes 1 (nucleoprotein, NP), 4 (glycoprotein, GP; soluble glycoprotein, sGP) and 7 (polymerase, L) giving the best coverage. Broadly-protective vaccine candidates for diverse viruses, such as HIV or Hepatitis C virus (HCV) have required pools of antigens. FILV is similar in this regard. While we have designed CTL mosaic proteins using all 7 types of filoviral proteins, only NP, GP and L proteins are reported here. If it were important to include other proteins in a mosaic CTL vaccine, additional sequences would be required to cover the space of known viral diversity.

  14. Evaluation of cloud fraction and its radiative effect simulated by IPCC AR4 global models against ARM surface observations

    Cloud Fraction (CF) is the dominant modulator of radiative fluxes. In this study, we evaluate CF simulations in the IPCC AR4 GCMs against ARM ground measurements, with a focus on the vertical structure, total amount of cloud and its effect on cloud shortwave transmissivity, for both inter-model deviation and model-measurement discrepancy. Our intercomparisons of three CF or sky-cover related dataset reveal that the relative differences are usually less than 10% (5%) for multi-year monthly (annual) mean values, while daily differences are quite significant. The results also show that the model-observation and the inter-model deviations have a similar magnitude for the total CF (TCF) and the normalized cloud effect, and they are twice as large as the surface downward solar radiation and cloud transmissivity. This implies that the other cloud properties, such as cloud optical depth and height, have a similar magnitude of disparity to TCF among the GCMs, and suggests that a better agreement among the GCMs in solar radiative fluxes could be the result of compensating errors in either cloud vertical structure, cloud optical depth or cloud fraction. Similar deviation pattern between inter-model and model-measurement suggests that the climate models tend to generate larger bias against observations for those variables with larger inter-model deviation. The simulated TCF from IPCC AR4 GCMs are very scattered through all seasons over three ARM sites: Southern Great Plains (SGP), Manus, Papua New Guinea and North Slope of Alaska (NSA). The GCMs perform better at SGP than at Manus and NSA in simulating the seasonal variation and probability distribution of TCF; however, the TCF in these models is remarkably underpredicted and cloud transmissivity is less susceptible to the change of TCF than the observed at SGP. Much larger inter-model deviation and model bias are found over NSA than the other sites in estimating the TCF, cloud transmissivity and cloud-radiation interaction, suggesting that the Arctic region continues to challenge cloud simulations in climate models. Most of the GCMs tend to underpredict CF and fail to capture the seasonal variation of CF at middle and low levels in the tropics. The high altitude CF is much larger in the GCMs than the observation and the inter-model variability of CF also reaches maximum at high levels in the tropics. Most of the GCMs tend to underpredict CF by 50-150% relative to the measurement average at low and middle levels over SGP. While the GCMs generally capture the maximum CF in the boundary layer and vertical variability, the inter-model deviation is largest near surface over the Arctic. The internal variability of CF simulated in ensemble runs with the same model is very minimal.
