Ford Plug-In Project: Bringing PHEVs to Market Demonstration and Validation Project

SciTech Connect (OSTI)

This project is in support of our national goal to reduce our dependence on fossil fuels. By supporting efforts that contribute toward the successful mass production of plug-in hybrid electric vehicles, our nations transportation-related fuel consumption can be offset with energy from the grid. Over four and a half years ago, when this project was originally initiated, plug-in electric vehicles were not readily available in the mass marketplace. Through the creation of a 21 unit plug-in hybrid vehicle fleet, this program was designed to demonstrate the feasibility of the technology and to help build cross-industry familiarity with the technology and interface of this technology with the grid. Ford Escape PHEV Demonstration Fleet 3 March 26, 2014 Since then, however, plug-in vehicles have become increasingly more commonplace in the market. Ford, itself, now offers an all-electric vehicle and two plug-in hybrid vehicles in North America and has announced a third plug-in vehicle offering for Europe. Lessons learned from this project have helped in these production vehicle launches and are mentioned throughout this report. While the technology of plugging in a vehicle to charge a high voltage battery with energy from the grid is now in production, the ability for vehicle-to-grid or bi-directional energy flow was farther away than originally expected. Several technical, regulatory and potential safety issues prevented progressing the vehicle-to-grid energy flow (V2G) demonstration and, after a review with the DOE, V2G was removed from this demonstration project. Also proving challenging were communications between a plug-in vehicle and the grid or smart meter. While this project successfully demonstrated the vehicle to smart meter interface, cross-industry and regulatory work is still needed to define the vehicle-to-grid communication interface.

Cowlitz Falls Fish Passage.

SciTech Connect (OSTI)

The upper Cowlitz was once home to native salmon and steelhead. But the combined impacts of overharvest, farming, logging and road building hammered fish runs. And in the 1960s, a pair of hydroelectric dams blocked the migration path of ocean-returning and ocean-going fish. The lower Cowlitz still supports hatchery runs of chinook, coho and steelhead. But some 200 river miles in the upper river basin--much of it prime spawning and rearing habitat--have been virtually cut off from the ocean for over 26 years. Now the idea is to trap-and-haul salmon and steelhead both ways and bypass previously impassable obstacles in the path of anadromous fish. The plan can be summarized, for the sake of explanation, in three steps: (1) trap and haul adult fish--collect ocean-returning adult fish at the lowermost Cowlitz dam, and truck them upstream; (2) reseed--release the ripe adults above the uppermost dam, and let them spawn naturally, at the same time, supplement these runs with hatchery born fry that are reared and imprinted in ponds and net pens in the watershed; (3) trap and haul smolts--collection the new generation of young fish as they arrive at the uppermost Cowlitz dam, truck them past the three dams, and release them to continue their downstream migration to the sea. The critical part of any fish-collection system is the method of fish attraction. Scientists have to find the best combination of attraction system and screens that will guide young fish to the right spot, away from the turbine intakes. In the spring of 1994 a test was made of a prototype system of baffles and slots on the upriver face of the Cowlitz Falls Dam. The prototype worked at 90% efficiency in early tests, and it worked without the kind of expensive screening devices that have been installed on other dams. Now that the success of the attraction system has been verified, Harza engineers and consultants will design and build the appropriate collection part of the system.

Indian Monsoon Depression: Climatology and Variability

SciTech Connect (OSTI)

The monsoon climate is traditionally characterized by large seasonal rainfall and reversal of wind direction (e.g., Krishnamurti 1979). Most importantly this rainfall is the major source of fresh water to various human activities such as agriculture. The Indian subcontinent resides at the core of the Southeast Asian summer monsoon system, with the monsoon trough extended from northern India across Indochina to the Western Tropical Pacific (WTP). Large fraction of annual rainfall occurs during the summer monsoon season, i.e., June - August with two distinct maxima. One is located over the Bay of Bengal with rainfall extending northwestward into eastern and central India, and the other along the west coast of India where the lower level moist wind meets the Western Ghat Mountains (Saha and Bavardeckar 1976). The rest of the Indian subcontinent receives relatively less rainfall. Various weather systems such as tropical cyclones and weak disturbances contribute to monsoon rainfall (Ramage 1971). Among these systems, the most efficient rain-producing system is known as the Indian monsoon depression (hereafter MD). This MD is critical for monsoon rainfall because: (i) it occurs about six times during each summer monsoon season, (ii) it propagates deeply into the continent and produces large amounts of rainfall along its track, and (iii) about half of the monsoon rainfall is contributed to by the MDs (e.g., Krishnamurti 1979). Therefore, understanding various properties of the MD is a key towards comprehending the veracity of the Indian summer monsoon and especially its hydrological process.

Research results from the Ashland Exploration, Inc. Ford Motor Company 78 (ed) well, Pike County, Kentucky. Topical report, April 1992-December 1993

SciTech Connect (OSTI)

This report summarizes the work performed on the Ashland Exploration, Inc. Ford Motor Company 78 (Experimental Development (ED)) Well, in Pike County, KY. The ED well was the third well drilled in a research project conducted by GRI in eastern Kentucky targeting both the Devonian Shales and Berea Sandstone. Both the Shales and Berea were completed and tested in the ED well. The primary objective of the ED well was to apply what was learned from studying the Shalers in COOP 1 (first well drilled) and the Berea in COOP 2 (second well drilled) to both the Shales and the Berea in the ED well. Additionally, the ED well was used to evaluate the impact of different stimulation treatments on Shales production. Research in the ED well brings to a close GRI`s extensive field-based research program in the Appalachian Basin over the last ten years.

A fully relativistic radial fall

E-Print Network [OSTI]

Radial fall has historically played a momentous role. It is one of the most classical problems, the solutions of which represent the level of understanding of gravitation in a given epoch. A {\\it gedankenexperiment} in a modern frame is given by a small body, like a compact star or a solar mass black hole, captured by a supermassive black hole. The mass of the small body itself and the emission of gravitational radiation cause the departure from the geodesic path due to the back-action, that is the self-force. For radial fall, as any other non-adiabatic motion, the instantaneous identity of the radiated energy and the loss of orbital energy cannot be imposed and provide the perturbed trajectory. In the first part of this letter, we present the effects due to the self-force computed on the geodesic trajectory in the background field. Compared to the latter trajectory, in the Regge-Wheeler, harmonic and all others smoothly related gauges, a far observer concludes that the self-force pushes inward (not outward) the falling body, with a strength proportional to the mass of the small body for a given large mass; further, the same observer notes an higher value of the maximal coordinate velocity, this value being reached earlier on during infall. In the second part of this letter, we implement a self-consistent approach for which the trajectory is iteratively corrected by the self-force, this time computed on osculating geodesics. Finally, we compare the motion driven by the self-force without and with self-consistent orbital evolution. Subtle differences are noticeable, even if self-force effects have hardly the time to accumulate in such a short orbit.

U.S. Department of Energy FreedomCAR & Vehicle Technologies Program Advanced Vehicle Testing Activity, Hydrogen/CNG Blended Fuels Performance Testing in a Ford F-150

SciTech Connect (OSTI)

Federal regulation requires energy companies and government entities to utilize alternative fuels in their vehicle fleets. To meet this need, several automobile manufacturers are producing compressed natural gas (CNG)-fueled vehicles. In addition, several converters are modifying gasoline-fueled vehicles to operate on both gasoline and CNG (Bifuel). Because of the availability of CNG vehicles, many energy company and government fleets have adopted CNG as their principle alternative fuel for transportation. Meanwhile, recent research has shown that blending hydrogen with CNG (HCNG) can reduce emissions from CNG vehicles. However, blending hydrogen with CNG (and performing no other vehicle modifications) reduces engine power output, due to the lower volumetric energy density of hydrogen in relation to CNG. Arizona Public Service (APS) and the U.S. Department of Energys Advanced Vehicle Testing Activity (DOE AVTA) identified the need to determine the magnitude of these effects and their impact on the viability of using HCNG in existing CNG vehicles. To quantify the effects of using various blended fuels, a work plan was designed to test the acceleration, range, and exhaust emissions of a Ford F-150 pickup truck operating on 100% CNG and blends of 15 and 30% HCNG. This report presents the results of this testing conducted during May and June 2003 by Electric Transportation Applications (Task 4.10, DOE AVTA Cooperative Agreement DEFC36- 00ID-13859).

Klamath Falls geothermal field, Oregon

SciTech Connect (OSTI)

Klamath Falls, Oregon, is located in a Known Geothermal Resource Area which has been used by residents, principally to obtain geothermal fluids for space heating, at least since the turn of the century. Over 500 shallow-depth wells ranging from 90 to 2,000 ft (27 to 610 m) in depth are used to heat (35 MWt) over 600 structures. This utilization includes the heating of homes, apartments, schools, commercial buildings, hospital, county jail, YMCA, and swimming pools by individual wells and three district heating systems. Geothermal well temperatures range from 100 to 230{degree}F (38 to 110{degree}C) and the most common practice is to use downhole heat exchangers with city water as the circulating fluid. Larger facilities and district heating systems use lineshaft vertical turbine pumps and plate heat exchangers. Well water chemistry indicates approximately 800 ppM dissolved solids, with sodium sulfate having the highest concentration. Some scaling and corrosion does occur on the downhole heat exchangers (black iron pipe) and on heating systems where the geo-fluid is used directly. 73 refs., 49 figs., 6 tabs.

American Indian reservations: A showplace for renewable energy

SciTech Connect (OSTI)

The Indian Energy Resource Development Program, authorized by Title XXVI of the 1992 Energy Policy Act, provides funding to American Indian tribes to develop Indian renewable energy and other energy resources. In fiscal years 1994 and 1995, 35 grants totaling $6.5 million were awarded to 29 tribes and Alaskan native corporations in 13 states. The projects cover the development range from feasibility studies to purchase and installation of equipment for commercial projects. Technologies include photovoltaics, biomass, wind, building energy efficiency, hydroelectricity, integrated resource planning, coal-fired cogeneration, and multi-sector natural gas. The Title XXVI program provides an important opportunity for assessing the technical and economic feasibility of renewable energy on Indian lands, and also for demonstrating DOE-developed technologies in real-life settings.

Interior cavern conditions and salt fall potential

SciTech Connect (OSTI)

A relatively large number of salt caverns are used for fluid hydrocarbon storage, including an extensive set of facilities in the Gulf Coast salt domes for the Strategic Petroleum Reserve (SPR) Program. Attention is focused on the SPR caverns because of available histories that detail events involving loss and damage of the hanging string casing. The total number of events is limited, making the database statistically sparse. The occurrence of the events is not evenly distributed, with some facilities, and some caverns, more susceptible than others. While not all of these events could be attributed to impacts from salt falls, many did show the evidence of such impacts. As a result, a study has been completed to analyze the potential for salt falls in the SPR storage caverns. In this process, it was also possible to deduce some of the cavern interior conditions. Storage caverns are very large systems in which many factors could possibly play a part in casing damage. In this study, all of the potentially important factors such as salt dome geology, operational details, and material characteristics were considered, with all being logically evaluated and most being determined as secondary in nature. As a result of the study, it appears that a principal factor in determining a propensity for casing damage from salt falls is the creep and fracture characteristics of salt in individual caverns. In addition the fracture depends strongly upon the concentration of impurity particles in the salt. Although direct observation of cavern conditions is not possible, the average impurity concentration and the accumulation of salt fall material can be determined. When this is done, there is a reasonable correlation between the propensity for a cavern to show casing damage events and accumulation of salt fall material. The accumulation volumes of salt fall material can be extremely large, indicating that only a few of the salt falls are large enough to cause impact damage.

Electronic Out-fall Inspection Application - 12007

SciTech Connect (OSTI)

In early 2009 an exciting opportunity was presented to the Geographic Information Systems (GIS) team at the Savannah River Site (SRS). The SRS maintenance group was directed to maintain all Out-falls on Site, increasing their workload from 75 to 183 out-falls with no additional resources. The existing out-fall inspection system consisted of inspections performed manually and documented via paper trail. The inspections were closed out upon completion of activities and placed in file cabinets with no central location for tracking/trending maintenance activities. A platform for meeting new improvements required for documentation by the Department of Health and Environmental Control (DHEC) out-fall permits was needed to replace this current system that had been in place since the 1980's. This was accomplished by building a geographically aware electronic application that improved reliability of site out-fall maintenance and ensured consistent standards were maintained for environmental excellence and worker efficiency. Inspections are now performed via tablet and uploaded to a central point. Work orders are completed and closed either in the field using tablets (mobile application) or in their offices (via web portal) using PCs. And finally completed work orders are now stored in a central database allowing trending of maintenance activities. (authors)

Falling through the black hole horizon

E-Print Network [OSTI]

We consider the fate of a small classical object, a "stick", as it falls through the horizon of a large black hole (BH). Classically, the equivalence principle dictates that the stick is affected by small tidal forces, and Hawking's quantum-mechanical model of BH evaporation makes essentially the same prediction. If, on the other hand, the BH horizon is surrounded by a "firewall", the stick will be consumed as it falls through. We have recently extended Hawking's model by taking into account the quantum fluctuations of the geometry and the classical back-reaction of the emitted particles. Here, we calculate the strain exerted on the falling stick for our model. The strain depends on the near-horizon state of the Hawking pairs. We find that, after the Page time when the state of the pairs deviates significantly from maximal entanglement (as required by unitarity), the induced strain in our semiclassical model is still parametrically small. This is because the number of the disentangled pairs is parametrically ...

Quinault Indian Nation Renewable Energy Plan

SciTech Connect (OSTI)

The Quinault Indian Nation (Nation) initiated this study on conservation and production of renewable energy because this approach created the opportunity: To become self-sufficient and gain greater control over the energy the Nation uses; To generate jobs and businesses for its members; To better manage, sustain, and protect its resources; To express the cultural values of the Nation in an important new arena. The Nation has relatively small energy needs. These needs are concentrated at two separate points: the Quinault Beach Resort and Casino (QBRC) and Taholah on the Quinault Indian Reservation (QIR). Except for the town of Queets, energy needs are small and scattered. The needs vary greatly over the season. The small scale, widely dispersed, and variable nature of these needs presents a unique challenge to the Nation. Meeting these needs requires a resource and technology that is flexible, effective, and portable. Conservation is the most cost-effective way to meet any need. It is especially effective in a situation like this where production would leave a high per unit cost. This plan is based on first gaining energy savings through conservation. Major savings are possible through: 1. Upgrading home appliances on the QIR. 2. Weatherizing homes and facilities. 3. Changes in lighting/ventilation in the QBRC pool room. These elements of the plan are already being implemented and promise to save the Nation around a quarter of its present costs. Wood biomass is the best resource available to the QIN for energy production either on-site or for commercial development. It is abundant, flexible and portable. Its harvesting has high job potential and these jobs are a good fit for the present skill set of the QIN. This plan focuses on using wood biomass to produce energy and other value-added products. Our study considered various technologies and approaches to using wood for energy. We considered production for both on-site and commercial production. This plan calls for commercial-scale production only, with the QIN being the first customer for the product. This plan favors employing the pyrolysis technology to produce bio-oil, heat, and char. We favor this approach and technology because it is the most cost effective way to use the available resource. Its main product, bio-oil, has proven utility for the production of heat and electricity. It has promise for use as an alternative fuel, which is a much higher value than present uses of wood and it meets the QIN need for flexibility, scalability, and portability. Char, the secondary product from the pyrolysis process, has proven value-added uses. In addition to these direct benefits, the use of biomass in pyrolysis technology has significant indirect benefits. These benefits include the fact that the technology is a good fit with the Nations cultural values, i.e., environmental protection and the holistic use of a resource. It fits well with current QIN enterprises. For example, char could be processed into a charcoal co-product for QIN fish. Finally, the QIN can become a leader in developing and demonstrating this innovative approach to the use of wood. This plan proposes key organization steps to insure both excellent implementation of the plan and taking the best advantage of the processes and facilities put in place. This plan calls for two new QIN organizations: an energy production/distribution corporation and a community development corporation. The production/distribution corporation can be either a utility or a business enterprise that develops and markets renewable energy. The community development corporation can be a not-for-profit to support the QIN in taking best advantage of its energy opportunities. The production and distribution corporation is the subject of a further business planning effort now underway. This plan envisions a community development corporation that works directly with the Business Committee on research, education, and project development. Specifically, this corporation can seek grant funding to research energy m

Foreign direct investment in the electricity sector: the Indian perspective

SciTech Connect (OSTI)

So far, India is losing out in the competition against other emerging economies to attract more foreign direct investment to its electricity sector. This is in large part because the Indian approach towards power sector reforms is more haphazard than the more orderly and sensitive growth model of Singapore and Latin American economies. (author)

Final Report of a CRADA Between Pacific Northwest National Laboratory and the Ford Motor Company (CRADA No. PNNL/265): Deactivation Mechanisms of Base Metal/Zeolite Urea Selective Catalytic Reduction Materials, and Development of Zeolite-Based Hydrocarbon Adsorber Materials

SciTech Connect (OSTI)

Reducing NOx emissions and particulate matter (PM) are primary concerns for diesel vehicles required to meet current LEV II and future LEV III emission standards which require 90+% NOx conversion. Currently, urea SCR as the NOx reductant and a Catalyzed Diesel Particulate Filter (CDPF) are being used for emission control system components by Ford Motor Company for 2010 and beyond diesel vehicles. Because the use of this technology for vehicle applications is new, the relative lack of experience makes it especially challenging to satisfy durability requirements. Of particular concern is being able to realistically simulate actual field aging of the catalyst systems under laboratory conditions. This is necessary both as a rapid assessment tool for verifying improved performance and certifiability of new catalyst formulations, and to develop a good understanding of deactivation mechanisms that can be used to develop improved catalyst materials. In addition to NOx and PM, the hydrocarbon (HC) emission standards are expected to become much more stringent during the next few years. Meanwhile, the engine-out HC emissions are expected to increase and/or be more difficult to remove. Since HC can be removed only when the catalyst becomes warm enough for its oxidation, three-way catalyst (TWC) and diesel oxidation catalyst (DOC) formulations often contain proprietary zeolite materials to hold the HC produced during the cold start period until the catalyst reaches its operating temperature (e.g., >200C). Unfortunately, much of trapped HC tends to be released before the catalyst reaches the operating temperature. Among materials effective for trapping HC during the catalyst warm-up period, siliceous zeolites are commonly used because of their high surface area and high stability under typical operating conditions. However, there has been little research on the physical properties of these materials related to the adsorption and release of various hydrocarbon species found in the engine exhaust. For these reasons, automakers and engine manufacturers have difficulty improving their catalytic converters for meeting the stringent HC emission standards. In this collaborative program, scientists and engineers in the Institute for Integrated Catalysis at Pacific Northwest National Laboratory and at Ford Motor Company have investigated laboratory- and engine-aged SCR catalysts, containing mainly base metal zeolites. These studies are leading to a better understanding of various aging factors that impact the long-term performance of SCR catalysts and improve the correlation between laboratory and engine aging, saving experimental time and cost. We have also studied materials effective for the temporary storage of HC species during the cold-start period. In particular, we have examined the adsorption and desorption of various HC species produced during the combustion with different fuels (e.g., gasoline, E85, diesel) over potential HC adsorber materials, and measured the kinetic parameters to update Fords HC adsorption model. Since this CRADA has now been completed, in this final report we will provide brief summaries of most of the work carried out on this CRADA over the last several years.

Hawking radiation on a falling lattice

E-Print Network [OSTI]

Scalar field theory on a lattice falling freely into a 1+1 dimensional black hole is studied using both WKB and numerical approaches. The outgoing modes are shown to arise from incoming modes by a process analogous to a Bloch oscillation, with an admixture of negative frequency modes corresponding to the Hawking radiation. Numerical calculations show that the Hawking effect is reproduced to within 0.5% on a lattice whose proper spacing where the wavepacket turns around at the horizon is $\\sim0.08$ in units where the surface gravity is 1.

Market-Based Indian Grid Integration Study Options: Preprint

SciTech Connect (OSTI)

The Indian state of Gujarat is forecasting solar and wind generation expansion from 16% to 32% of installed generation capacity by 2015. Some states in India are already experiencing heavy wind power curtailment. Understanding how to integrate variable generation (VG) into the grid is of great interest to local transmission companies and India's Ministry of New and Renewable Energy. This paper describes the nature of a market-based integration study and how this approach, while new to Indian grid operation and planning, is necessary to understand how to operate and expand the grid to best accommodate the expansion of VG. Second, it discusses options in defining a study's scope, such as data granularity, generation modeling, and geographic scope. The paper also explores how Gujarat's method of grid operation and current system reliability will affect how an integration study can be performed.

Wind Resource Assessment Report: Mille Lacs Indian Reservation, Minnesota

SciTech Connect (OSTI)

The U.S. Environmental Protection Agency (EPA) launched the RE-Powering America's Land initiative to encourage development of renewable energy on potentially contaminated land and mine sites. EPA collaborated with the U.S. Department of Energy's (DOE's) National Renewable Energy Laboratory (NREL) and the Mille Lacs Band of Chippewa Indians to evaluate the wind resource and examine the feasibility of a wind project at a contaminated site located on the Mille Lacs Indian Reservation in Minnesota. The wind monitoring effort involved the installation of a 60-m met tower and the collection of 18 months of wind data at multiple heights above the ground. This report focuses on the wind resource assessment, the estimated energy production of wind turbines, and an assessment of the economic feasibility of a potential wind project sited this site.

Fisheries Enhancement on the Coeur d'Alene Indian Reservation; Hangman Creek, Annual Report 2001-2002.

SciTech Connect (OSTI)

Historically, Hangman Creek produced Chinook salmon (Oncorhynchus tshawytscha) and Steelhead trout (Oncorhynchus mykiss) for the Upper Columbia Basin Tribes. One weir, located at the mouth of Hangman Creek was reported to catch 1,000 salmon a day for a period of 30 days a year (Scholz et al. 1985). The current town of Tekoa, Washington, near the state border with Idaho, was the location of one of the principle anadromous fisheries for the Coeur d'Alene Tribe (Scholz et al. 1985). The construction, in 1909, of Little Falls Dam, which was not equipped with a fish passage system, blocked anadromous fish access to the Hangman Watershed. The fisheries were further removed with the construction of Chief Joseph and Grand Coulee Dams. As a result, the Coeur d'Alene Indian Tribe was forced to rely more heavily on native fish stocks such as Redband trout (Oncorhynchus mykiss gairdneri), Westslope Cutthroat trout (O. clarki lewisii), Bull trout (Salvelinus confluentus) and other terrestrial wildlife. Historically, Redband and Cutthroat trout comprised a great deal of the Coeur d'Alene Tribe's diet (Power 1997).

Development of energy-efficiency standards for Indian refrigerators

SciTech Connect (OSTI)

The application of advanced techniques in engineering simulation and economic analysis for the development of efficiency standards for Indian refrigerators is illustrated in this paper. A key feature of this methodology is refrigerator simulation to generate energy savings for a set of energy-efficient design options and life-cycle cost (LCC) analysis with these design options. The LCC of a refrigerator is analyzed as a function of five variables: nominal discount rate, fuel price, appliance lifetime, incremental price, and incremental energy savings. The frequency of occurrence of the LCC minimum at any design option indicates the optimum efficiency level or range. Studies carried out in the US and European Economic Community show that the location of the LCC minimum under different scenarios (e.g., variable fuel price, life-time, discount rate, and incremental price) is quite stable. Thus, an efficiency standard can be developed based on the efficiency value at the LCC minimum. This paper examines and uses this methodology in developing efficiency standards for Indian refrigerators. The potential efficiency standard value is indicated to be 0.65 kWh/day for a 165-liter, CFC-based, manual defrost, single-door refrigerator-freezer.

Nonlocal description of a falling body through the air

E-Print Network [OSTI]

In this present work we consider a falling body through the air under the influence of gravity. In particular, we consider the experimental data based upon the free fall of six men in the atmosphere of the earth. In order to describe this process we employ a nonlocal dissipative force. We show that our description, by using an exponential memory kernel, can fit the experimental data as well as that of a local dissipative force.

Life-Cycle Assessment of the Use of Jatropha Biodiesel in Indian Locomotives (Revised)

SciTech Connect (OSTI)

With India's transportation sector relying heavily on imported petroleum-based fuels, the Planning Commission of India and the Indian government recommended the increased use of blended biodiesel in transportation fleets, identifying Jatropha as a potentially important biomass feedstock. The Indian Oil Corporation and Indian Railways are collaborating to increase the use of biodiesel blends in Indian locomotives with blends of up to B20, aiming to reduce GHG emissions and decrease petroleum consumption. To help evaluate the potential for Jatropha-based biodiesel in achieving sustainability and energy security goals, this study examines the life cycle, net GHG emission, net energy ratio, and petroleum displacement impacts of integrating Jatropha-based biodiesel into locomotive operations in India. In addition, this study identifies the parameters that have the greatest impact on the sustainability of the system.

Complete genome sequencing and evolutionary analysis of Indian isolates of Dengue virus type 2

SciTech Connect (OSTI)

Highlights: Complete genome of Indian DENV-2 was deciphered for the first time in this study. The recent Indian DENV-2 revealed presence of many unique amino acid residues. Genotype shift (American to Cosmopolitan) characterizes evolution of DENV-2 in India. Circulation of a unique clade of DENV-2 in South Asia was identified. -- Abstract: Dengue is the most important arboviral infection of global public health significance. It is now endemic in most parts of the South East Asia including India. Though Dengue virus type 2 (DENV-2) is predominantly associated with major outbreaks in India, complete genome information of Indian DENV-2 is not available. In this study, the full-length genome of five DENV-2 isolates (four from 2001 to 2011 and one from 1960), from different parts of India was determined. The complete genome of the Indian DENV-2 was found to be 10,670 bases long with an open reading frame coding for 3391 amino acids. The recent Indian DENV-2 (20012011) revealed a nucleotide sequence identity of around 90% and 97% with an older Indian DENV-2 (1960) and closely related Sri Lankan and Chinese DENV-2 respectively. Presence of unique amino acid residues and non-conservative substitutions in critical amino acid residues of major structural and non-structural proteins was observed in recent Indian DENV-2. Selection pressure analysis revealed positive selection in few amino acid sites of the genes encoding for structural and non-structural proteins. The molecular phylogenetic analysis based on comparison of both complete coding region and envelope protein gene with globally diverse DENV-2 viruses classified the recent Indian isolates into a unique South Asian clade within Cosmopolitan genotype. A shift of genotype from American to Cosmopolitan in 1970s characterized the evolution of DENV-2 in India. Present study is the first report on complete genome characterization of emerging DENV-2 isolates from India and highlights the circulation of a unique clade in South Asia.

Albeni Falls Wildlife Mitigation Project, 2001 Annual Report.

SciTech Connect (OSTI)

The Albeni Falls Interagency Work Group was actively engaged in implementing wildlife mitigation activities in 2001. The Work Group met quarterly to discuss management and budget issues affecting the Albeni Falls Wildlife Mitigation Program. Work Group members protected 851 acres of wetland habitat in 2001. Wildlife habitat protected to date for the Albeni Falls project is approximately 5,248.31 acres ({approx}4,037.48 Habitat Units). Approximately 14% of the total wildlife habitat lost has been mitigated. Administrative activities increased as funding was more evenly distributed among Work Group members and protection opportunities became more time consuming. In 2001, Work Group members focused on development and implementation of the monitoring and evaluation program as well as completion of site-specific management plans. With the implementation of the monitoring and evaluation program, and as management plans are reviewed and executed, on the ground management activities are expected to increase in 2002.

Conceptual Model of the Klamath Falls, Oregon Geothermal Area

SciTech Connect (OSTI)

Over the last 50 years significant amounts of data have been obtained from the Klamath Falls geothermal resource. To date, the complexity of the system has stymied researchers, leading to the development of only very generalized hydrogeologic and geothermal models of the area. Recently, the large quantity of available temperature data have been re-evaluated, revealing new information on subsurface heat flow and locations of faults in the system. These inferences are supported by borehole, geochemical, geophysical, and hydrologic data. Based on re-evaluation of all available data, a detailed conceptual model for the Klamath Falls geothermal resource is proposed. 1 tab., 8 figs., 21 refs.

Long-term surveillance plan for the Falls City Disposal Site, Falls City, Texas. Revision 2

SciTech Connect (OSTI)

The need for ground water monitoring at the Falls City disposal site was evaluated in accordance with NRC regulations and guidelines established by the DOE in Guidance for Implementing the Long-term Surveillance Program for UMTRA Project Title 1 Disposal Sites (DOE, 1996). Based on evaluation of site characterization data, it has been determined that a program to monitor ground water for demonstration of disposal cell performance based on a set of concentration limits is not appropriate because ground water in the uppermost aquifer is of limited use, and a narrative supplemental standard has been applied to the site that does not include numerical concentration limits or a point of compliance. The limited use designation is based on the fact that ground water in the uppermost aquifer is not currently or potentially a source of drinking water in the area because it contains widespread ambient contamination that cannot be cleaned up using methods reasonably employed by public water supply systems. Background ground water quality varies by orders of magnitude since the aquifer is in an area of redistribution of uranium mineralization derived from ore bodies. The DOE plans to perform post-closure ground water monitoring in the uppermost aquifer as a best management practice (BMP) as requested by the state of Texas.
