A multi-dimensional user oriented synthetic environment system allows application programs to be programmed and accessed with input/output device independent, generic functional commands which are a distillation of the actual functions performed by any application program. A shared memory structure allows the translation of device specific commands to device independent, generic functional commands. Complete flexibility of the mapping of synthetic environment data to the user is thereby allowed. Accordingly, synthetic environment data may be provided to the user on parallel user information processing channels allowing the subcognitive mind to act as a filter, eliminating irrelevant information and allowing the processing of increase amounts of data by the user. The user is further provided with a craft surrounding the user within the synthetic environment, which craft, imparts important visual referential an motion parallax cues, enabling the user to better appreciate distances and directions within the synthetic environment. Display of this craft in close proximity to the user's point of perspective may be accomplished without substantially degrading the image resolution of the displayed portions of the synthetic environment.

OAK-B135 The homeotic gene AGAMOUS (AG) has dual roles in specifying floral organ fate and limiting stem cell proliferation in Arabidopsis flowers. We show here that the floral identity protein LEAFY (LFY), a transcription factor expressed throughout the flower, acts together with the homeodomain protein WUSCHEL (WUS) to activate AG in the center of flowers. WUS was previously identified because of its role in maintaining a stem cell population in the center of both shoot and floral meristems. The unsuspected additional role of WUS in regulating floral homeotic gene expression supports the hypothesis that floral patterning makes use of a general meristem patterning system that was present before flowers evolved. We also show that AG represses WUS at later stages of floral development, thus creating a negative feedback loop that is required for the determinate growth of floral meristems.

The synthesis and characterization of some linear acene quinones of up to nine fused rings prepared by the Friedel-Craft's reaction of hydroquinone (HQ) and 1,4,9,10 tetrahydroxy anthracene (THA) with pyromellitic dianhydride (PMDA), and fused AlCl/sub 3/, was carried out. The intermediate product of the reaction of THA and PMDA, 1,4 dihydroxy anthraquinone, 6,7 dicarboxylic acid (DADCA) was also isolated and its synthesis optimized. This material was reduced to 1,4,9,10-tetrahydroxy anthracene 6,7-dicarboxylic acid (TADCA) and further dehydrated to its anhydride (TADCAmh). These compounds contain the necessary chemical functionalities which may lead to the facile synthesis of higher molecular weight quinones. These acene quinones show electronic spectral absorptions extending far into the NIR region, an indication of their long conjunction length.

  7. Interaction of LEAFY, AGAMOUS, and TERMINAL FLOWER1 in maintaining floral identity in Arabidopsis

    SciTech Connect (OSTI)

    Parcy, F., Bomblies, K., and Weigel, D.

    2002-02-22T23:59:59.000Z

OAK-B135 The Arabidopsis transcription factor LEAFY (LFY) acts upstream of homeotic genes such as AGAMOUS (AG) to confer floral identity on meristems that arise after the transition to reproductive development. Compared to the genetic circuitry regulating the establishment of floral meristem identity, little is known about its maintenance. Previous experiments with fly heterozygous plants and ag mutants grown in conditions that reduce the floral inductive stimulus have shown that both genes are required to prevent reversion of floral to inflorescence meristems. Here, we present evidence that LFY maintains floral meristem identity independently of AG, and that the primary role of LFY is either direct repression of shoot identity genes or repression of an intermediate factor that activates shoot identity genes. The latter conclusions were deduced from the phenotypes conferred by a gain-of-function transgene, LFY:VP16, that appears to act as a dominant negative, or antimorphic, allele during maintenance of floral meristem identity. These observations contrast with previous findings that LFY acts as a direct activator of floral homeotic genes, supporting the hypothesis that the transcriptional activity of LFY is dependent on specific co-regulators.

Highlights: ? Effective management of temple floral offerings using E. fetida. ? Physico-chemical properties in TW VC were better especially EC, C/N, C/P and TK. ? TW VC as plant growth promoter at much lower application rates than KW and FYW VC. - Abstract: Recycling of temple waste (TW) mainly comprising of floral offerings was done through vermitechnology using Eisenia fetida and its impact on seed germination and plant growth parameters was studied by comparing with kitchen waste (KW) and farmyard waste (FYW) vermicompost (VC). The worm biomass was found to be maximum in TW VC compared to KW and FYW VCs at both 40 and 120 days old VCs. Physico-chemical analysis of worm-worked substrates showed better results in TW VC especially in terms of electrical conductivity, C/N, C/P and TK. 10% TW VCwater extract (VCE) showed stimulatory effect on germination percentage of chickpea seeds while KW and FYW VCE proved effective at higher concentration. Variation in growth parameters was also observed with change in the VCsoil ratio and TW VC showed enhanced shoot length, root length, number of secondary roots and total biomass at 12.5% VC compared to KW and FYW VC.

In a continuing effort to facilitate communications and resource exchange among the DOE contractors, the TRADE (Training Resources and Data Exchange) Committee composed of representatives from 10 DOE contractors, has compiled the Training Programs Inventories. Taken together there are over 2700 courses that reflect the range and focus of in-house training programs being offered throughour the DOE GOCO system by approximately 49 contractors. The Technical Training Programs Inventories include those programs or courses designed specifically for in-house employees and which provide development and practical information about job-related subjects not included in the nontechnical training area. The focus of the Inventories has been on targeted audiences, resources, and course design. The contact person has been listed with each course to encourage the use of the inventories and facilitate the excchange of information between GOCOs through a peer-to-peer exchange between training personnel of various contractors. This publication contains 140 courses focusing on crafts programs and miscellanous programs at all levels. Information of the inventories is available on the DOE Integrated Technical Information System (ITIS) as the TRD file.

OAK-B135 In Arabidopsis thaliana, cis-regulatory sequences of the floral homeotic gene AGAMOUS (AG) are located in the second intron. This 3 kb intron contains binding sites for two direct activators of AG, LEAFY (LFY) and WUSCHEL (WUS), along with other putative regulatory elements. We have used phylogenetic footprinting and the related technique of phylogenetic shadowing to identify putative cis-regulatory elements in this intron. Among 29 Brassicaceae, several other motifs, but not the LFY and WUS binding sites previously identified, are largely invariant. Using reporter gene analyses, we tested six of these motifs and found that they are all functionally important for activity of AG regulatory sequences in A. thaliana. Although there is little obvious sequence similarity outside the Brassicaceae, the intron from cucumber AG has at least partial activity in A. thaliana. Our studies underscore the value of the comparative approach as a tool that complements gene-by-gene promoter dissection, but also highlight that sequence-based studies alone are insufficient for a complete identification of cis-regulatory sites.

Under the Clean Energy Works (CEW) program, Craft3 developed a loan product that widened access to financing for homeowners, offered long term funding, and collected repayments through the customer?s utility bill. The program?s success led Craft3 to pursue the sale of the loan portfolio to both mitigate its own risks and replenish funds for lending. This sale breaks new ground for energy efficiency finance and is notable as it was completed even with many novel program design elements. It replenished Craft3?s program capital and uncovered some valuable lessons that may facilitate future transactions. However, the lack of data history and the unproven nature of the loan portfolio meant that Craft3 had to limit the risk of losses to Self-Help, the purchaser of the portfolio. It remains to be seen whether this experience will pave the way for more sales of on-bill energy efficiency loan portfolios. This case study illustrates how certain program design decisions can sometimes both facilitate programmatic objectives and possibly present challenges for the sale of a portfolio of energy efficiency loans.

At the request of the US Department of Energy (DOE), a team from Oak Ridge National Laboratory conducted a radiological survey at the former Alba Craft Laboratory Site Properties, Oxford, Ohio. The survey was performed in July and September of 1992. The purpose of the survey was to determine whether the property was contaminated with radioactive residues, primarily [sup 238]U, from uranium machining operations conducted for National Lead of Ohio, a prime Atomic Energy Commission contractor. The survey included scan measurement of direct radiation levels inside and outside the former laboratory, outdoors on eight properties adjoining the former laboratory, and the city right-of-way adjacent to the surveyed properties. Radionuclide concentrations were determined in outdoor surface and subsurface soil samples taken from each property and the exterior of the laboratory. Fixed surface residual radioactivity was measured inside the laboratory and outside the building. Air samples were collected, direct exposure was measured, and samples were collected to measure transferable radioactivity inside the building. Results of the survey indicate areas where surface and soil contamination level s are above the DOE guidelines for uncontrolled areas.

The Naval Postgraduate School has been conducting Tactical Network Topology (TNT) Maritime Interdiction Operations (MIO) experiments with Lawrence Livermore National Laboratory (LLNL) since early in 2005. In this work, we are investigating cutting edge technology to evaluate use of networks, advanced sensors and collaborative technology for globally-supported maritime interdiction operations. Some examples of our research include communications in harsh environments, between moving ships at sea; small boat drive-by radiation detection; network-centric collaboration with global partners; situational awareness; prototype sensors & biometric instruments. Since 2006, we have studied the concept of using a small vessel with fixed radiation sensors to do initial searches for illicit radioactive materials. In our work, we continue to evaluate concepts of operation for small boat monitoring. For example, in San Francisco Bay we established a simulated choke point using two RHIBs. Each RHIB had a large sodium iodide radiation sensor on board, mounted on the side nearest to the passing potential target boats. Once detections were made, notification over the network prompted a chase RHIB also equipped with a radiation sensor to further investigate the potential target. We have also used an unmanned surface vessel (USV) carrying a radiation sensor to perform the initial discovery. The USV was controlled remotely and to drive by boats in different configurations. The potential target vessels were arranged in a line, as a choke point and randomly spaced in the water. Search plans were problematic when weather, waves and drift complicated the ability to stay in one place. A further challenge is to both detect and identify the radioactive materials during the drive-by. Our radiation detection system, ARAM, Adaptable Radiation Area Monitor, is able to detect, alarm and quickly identify plausible radionuclides in real time. We have performed a number of experiments to better understand parameters of vessel speed, time, shielding, and distance in this complex three-dimensional space. At the NMIOTC in September 2009, we employed a dual detector portal followed by a chase. In this event, the challenge was to maintain communications after a lapse. When the chase went past the line-of sight reach of the Tactical Operational Center's (TOC) antenna, with interference from a fortress island in Suda Bay, Wave Relay extended the network for continued observation. Sodium iodide radiation detectors were mounted on two Hellenic Navy SEAL fast boats. After making the detection one of the portal boats maintained line-of sight while the other pursued the target vessel. Network access via Wave Relay antennas was maintained until the conclusion of the chase scenario. Progress has been made in the detection of radioactive materials in the maritime environment. The progression of the TNT MIO experiments has demonstrated the potential of the hardware to solve the problems encountered in this physically challenging environment. There continue to be interesting opportunities for research and development. These experiments provide a variety of platforms and motivated participants to perform real-world testing as solutions are made available.

    2003-09-01T23:59:59.000Z

We theoretically demonstrate that electromagnetic energy can be obtained by direct, lossless, conversion from gravitational and kinetic energies. For this purpose we discuss the properties of an electromechanical system which consists of a superconducting coil submitted to a constant external force and to magnetic fields. The coil oscillates and has induced in it a rectified electrical current whose magnitude may reach hundreds of Ampere. There is no need for an external electrical power source for the system to start out and it can be kept working continuously if linked to large capacitors. We extensively discuss the issue of energy dissipation in superconductors and show that the losses for such a system can be made extremely small for certain operational conditions, so that by reaching and keeping resonance the system main application should be in magnetic energy storage and transmission.

  18. Progress Report for DOE DE-FG03-98ER20317 ''Regulation of the floral homeotic gene AGAMOUS'' Current and Final Funding Period: September 1, 2002, to December 31, 2002

    SciTech Connect (OSTI)

    Weigel, D.

    2003-03-11T23:59:59.000Z

OAK-B135 Results obtained during this funding period: (1) Phylogenetic footprinting of AG regulatory sequences Sequences necessary and sufficient for AGAMOUS (AG) expression in the center of Arabidopsis flowers are located in the second intron, which is about 3 kb in size. This intron contains binding sites for two transcription factors, LEAFY (LFY) and WUSCHEL (WUS), which are direct activators of AG. We used the new method of phylogenetic shadowing to identify new regulatory elements. Among 29 Brassicaceae, several other motifs, but not the LFY and WUS binding sites previously identified, are largely invariant. Using reporter gene analyses, we tested six of these motifs and found that they are all functionally important for activity of AG regulatory sequences in A. thaliana. (2) Repression of AG by MADS box genes A candidate for repressing AG in the shoot apical meristem has been the MADS box gene FUL, since it is expressed in the shoot apical meristem and since an activated version (FUL:VP16) leads to ectopic AG expression in the shoot apical meristem. However, there is no ectopic AG expression in full single mutants. We therefore started to generate VP16 fusions of several other MADS box genes expressed in the shoot apical meristem, to determine which of these might be candidates for FUL redundant genes. We found that AGL6:VP16 has a similar phenotype as FUL:VP16, suggesting that AGL6 and FUL interact. We are now testing this hypothesis. (3) Two candidate AG regulators, WOW and ULA Because the phylogenetic footprinting project has identified several new candidate regulatory motifs, of which at least one (the CCAATCA motif) has rather strong effects, we had decided to put the analysis of WOW and ULA on hold, and to focus on using the newly identified motifs as tools. We conduct ed yeast one-hybrid screen with two of the conserved motifs, and identified several classes of transcription factors that can interact with them. One of these is encoded by the PAN gene, previously known to be expressed in a domain that overlaps the AG domain, but not known before to regulate AG. (4) New genetic modifiers of AG This part of the project was concluded in the previous funding period.

The present invention provides a method of human-computer interfacing. Force feedback allows intuitive navigation and control near a boundary between regions in a computer-represented space. For example, the method allows a user to interact with a virtual craft, then push through the windshield of the craft to interact with the virtual world surrounding the craft. As another example, the method allows a user to feel transitions between different control domains of a computer representation of a space. The method can provide for force feedback that increases as a user's locus of interaction moves near a boundary, then perceptibly changes (e.g., abruptly drops or changes direction) when the boundary is traversed.
