    HH 222 is a giant shocked region in the L1641 cloud, and is popularly known as the Orion Streamers or ''the waterfall'' on account of its unusual structure. At the center of these streamers are two infrared sources coincident with a nonthermal radio jet aligned along the principal streamer. The unique morphology of HH 222 has long been associated with this radio jet. However, new infrared images show that the two sources are distant elliptical galaxies, indicating that the radio jet is merely an improbable line-of-sight coincidence. Accurate proper motion measurements of HH 222 reveal that the shock structure is a giant bow shock moving directly away from the well-known, very young, Herbig Be star V380 Ori. The already known Herbig-Haro object HH 35 forms part of this flow. A new Herbig-Haro object, HH 1041, is found precisely in the opposite direction of HH 222 and is likely to form part of a counterflow. The total projected extent of this HH complex is 5.3 pc, making it among the largest HH flows known. A second outflow episode from V380 Ori is identified as a pair of HH objects, HH 1031 to the northwest and the already known HH 130 to the southeast, along an axis that deviates from that of HH 222/HH 1041 by only 3.7. V380 Ori is a hierarchical quadruple system, including a faint companion of spectral type M5 or M6, which at an age of ?1 Myr corresponds to an object straddling the stellar-to-brown dwarf boundary. We suggest that the HH 222 giant bow shock is a direct result of the dynamical interactions that led to the conversion from an initial non-hierarchical multiple system into a hierarchical configuration. This event occurred no more than 28,000 yr ago, as derived from the proper motions of the HH 222 giant bow shock.

    Photometric and polarimetric monitoring observations of UX Ori begun in 1986 in the Crimea and Bolivia have resulted in the observation of two deep minima of the brightness during which a growth of the linear polarization (to approx. =7%) was observed, together with a tendency for the circular polarization to increase (up to approx. =1%). Analysis of the observational data shows that the main source of the polarized radiation in the deep minima is the emission of the star scattered by grains of circumstellar dust. On the basis of Mie's theory for a polydisperse graphite-silicate mixtures of particles the optical properties of ellipsoidal dust envelopes have been calculated and a model of the Algol-like minimum constructed.

    The idea that non-steady accretion during the embedded phase of protostar evolution can produce the observed luminosity spread in the Herzsprung-Russell diagram (HRD) of young clusters has recently been called into question. Observations of FU Ori, for instance, suggest an expansion of the star during strong accretion events, whereas the luminosity spread implies a contraction of the accreting objects, decreasing their radiating surface. In this paper, we present a global scenario based on calculations coupling episodic accretion histories derived from numerical simulations of collapsing cloud prestellar cores of various masses and subsequent protostar evolution. Our calculations show that, assuming an initial protostar mass M{sub i} {approx} 1 M{sub Jup}, typical of the second Larson's core, both the luminosity spread in the HRD and the inferred properties of FU Ori events (mass, radius, accretion rate) can be explained by this scenario, providing two conditions. First, there must be some variation within the fraction of accretion energy absorbed by the protostar during the accretion process. Second, the range of this variation should increase with increasing accretion burst intensity and thus with the initial core mass and final star mass. The numerical hydrodynamics simulations of collapsing cloud prestellar cores indeed show that the intensity of the accretion bursts correlates with the mass and initial angular momentum of the prestellar core. Massive prestellar cores with high initial angular momentum are found to produce intense bursts characteristic of FU Ori-like events. Our results thus suggest a link between the burst intensities and the fraction of accretion energy absorbed by the protostar, with some threshold in the accretion rate, of the order of 10{sup -5} M{sub Sun} yr{sup -1}, delimitating the transition from 'cold' to 'hot' accretion.     The minimal replicator of the Epstein-Barr virus (EBV) latent cycle origin of DNA replication oriP is composed of two binding sites for the Epstein-Barr virus nuclear antigen-1 (EBNA-1) and flanking inverted repeats that bind the telomere repeat binding factor TRF2. Although not required for minimal replicator activity, additional binding sites for EBNA-1 and TRF2 and one or more auxiliary elements located to the right of the EBNA-1/TRF2 sites are required for the efficient replication of oriP plasmids. Another region of oriP that is predicted to be destabilized by DNA supercoiling is shown here to be an important functional component of oriP. The ability of DNA fragments of unrelated sequence and possessing supercoiled-induced DNA duplex destabilized (SIDD) structures, but not fragments characterized by helically stable DNA, to substitute for this component of oriP demonstrates a role for the SIDD region in the initiation of oriP-plasmid DNA replication.

    Quercetin is a bioflavonoid that exhibits several biological functions in vitro and in vivo. Quercetin 3-O-methyl ether (Q3) is a natural product reported to have pharmaceutical activities, including antioxidative and anticancer activities. However, little is known about the mechanism by which it protects cells from oxidative stress. This study was designed to investigate the mechanisms by which Q3 protects against Cu{sup 2+}-induced cytotoxicity. Exposure to Cu{sup 2+} resulted in the death of mouse liver FL83B cells, characterized by apparent apoptotic features, including DNA fragmentation and increased nuclear condensation. Q3 markedly suppressed Cu{sup 2+}-induced apoptosis and mitochondrial dysfunction, characterized by reduced mitochondrial membrane potential, caspase-3 activation, and PARP cleavage, in Cu{sup 2+}-exposed cells. The involvement of PI3K, Akt, Erk, FOXO3A, and Mn-superoxide dismutase (MnSOD) was shown to be critical to the survival of Q3-treated FL83B cells. The liver of both larval and adult zebrafish showed severe damage after exposure to Cu{sup 2+} at a concentration of 5 μM. Hepatic damage induced by Cu{sup 2+} was reduced by cotreatment with Q3. Survival of Cu{sup 2+}-exposed larval zebrafish was significantly increased by cotreatment with 15 μM Q3. Our results indicated that Cu{sup 2+}-induced apoptosis in FL83B cells occurred via the generation of ROS, upregulation and phosphorylation of Erk, overexpression of 14-3-3, inactivation of Akt, and the downregulation of FOXO3A and MnSOD. Hence, these results also demonstrated that Q3 plays a protective role against oxidative damage in zebrafish liver and remarked the potential of Q3 to be used as an antioxidant for hepatocytes.     The processes of defining managerial roles and providing for delegation of authority are essential to any enterprise. At most large organizations, these processes are defined in policy manuals and through sets of standard operating procedures for many, if not all, business and administrative functions. Many of these staff-initiated, administrative functions require the routing of documents for approval to one or more levels of management. These employee-oriented, back office types of workflows tend to require more flexibility in determining to whom these documents should go to, while, at the same time, providing the responsible parties with the flexibility to delegate their approval authority or allow others to review their work. Although this practice is commonplace in manual, paper-based processes that exist in many organizations, it is difficult to provide the same flexibility in the more structured, electronic-based, workflow systems. The purpose of this report is to present a framework or architecture for creating a R/DA system and provide some insights associated with its design and utilization. To improve understanding and clarify subsequent discussion, the goals and requirements for the major R/DA system components, namely, the database and interface modules, are initially presented along with the identification of important concepts and the definition of critical terms. Next high-level functions relating the types of inputs to the outputs of the R/DA interface module are introduced and discussed. Then the relationships between the major R/DA modules and the primary components associated with its creation and maintenance are presented and analyzed. Finally, some conclusions are drawn relative to the advantages associated with developing a R/DA system for use in implementing an enterprise-wide, work-facilitating information system.

    We present a spectroscopically identified catalog of 70 DA white dwarfs (WDs) from the LAMOST pilot survey. Thirty-five are found to be new identifications after cross-correlation with the Eisenstein et al. and Villanova catalogs. The effective temperature and gravity of these WDs are estimated by Balmer lines fitting. Most of them are hot WDs. The cooling times and masses of these WDs are estimated by interpolation in theoretical evolution tracks. The peak of the mass distribution is found to be {approx}0.6 M {sub Sun }, which is consistent with prior work in the literature. The distances of these WDs are estimated using the method of synthetic spectral distances. All of these WDs are found to be in the Galactic disk from our analysis of space motions. Our sample supports the expectation that WDs with high mass are concentrated near the plane of the Galactic disk.

    The atmospheres of very cool, hydrogen-rich white dwarfs (WDs) (T{sub eff} < 6000 K) are challenging to model because of the increased complexity of the equation of state, chemical equilibrium, and opacity sources in a low-temperature, weakly ionized dense gas. In particular, many models that assume relatively simple models for the broadening of atomic levels and mostly ideal gas physics overestimate the flux in the blue part of their spectra. A solution to this problem that has met with some success is that additional opacity at short wavelengths comes for the extreme broadening of the Lyman α line of atomic H by collisions primarily with H{sub 2}. For the purpose of validating this model more rigorously, we acquired Hubble Space Telescope STIS spectra of eight very cool WDs (five DA and three DC stars). Combined with their known parallaxes, BVRIJHK, and Spitzer IRAC photometry, we analyze their entire spectral energy distribution (from 0.24 to 9.3 μm) with a large grid of model atmospheres and synthetic spectra. We find that the red wing of the Lyman α line reproduces the rapidly decreasing near-UV flux of these very cool stars very well. We determine better constrained values of T{sub eff} and gravity as well as upper limits to the helium abundance in their atmospheres.

    Rozitis et al. recently reported that near-Earth asteroid (29075) 1950DA, whose bulk density ranges from 1.0 g cm{sup 3} to 2.4g cm{sup 3}, is a rubble pile and requires a cohesive strength of at least 44-76 Pa to keep from failing due to its fast spin period. Since their technique for giving failure conditions required the averaged stress over the whole volume, it discarded information about the asteroid's failure mode and internal stress condition. This paper develops a finite element model and revisits the stress and failure analysis of 1950DA. For the modeling, we do not consider material hardening and softening. Under the assumption of an associated flow rule and uniform material distribution, we identify the deformation process of 1950DA when its constant cohesion reaches the lowest value that keeps its current shape. The results show that to avoid structural failure the internal core requires a cohesive strength of at least 75-85 Pa. It suggests that for the failure mode of this body, the internal core first fails structurally, followed by the surface region. This implies that if cohesion is constant over the whole volume, the equatorial ridge of 1950DA results from a material flow going outward along the equatorial plane in the internal core, but not from a landslide as has been hypothesized. This has additional implications for the likely density of the interior of the body.

    The electron-positron collider DA{Phi}NE, the Italian {Phi} factory, has been recently upgraded in order to implement an innovative collision scheme based on large crossing angle, small beam sizes at the crossing point, and compensation of beam-beam interaction by means of sextupole pairs creating a ''crab-waist'' configuration in the interaction region. Experimental tests of the novel scheme exhibited an increase by a factor of 3 in the peak luminosity of the collider with respect to the performances reached before the upgrade. In this Letter we present the new collision scheme, discuss its advantages, describe the hardware modifications realized for the upgrade, and report the results of the experimental tests carried out during commissioning of the machine in the new configuration and standard operation for the users.

  2. NMR Analysis of Methyl Groups at 100-500 kDa: Model Systems and Arp2/3 Complex

    Large macromolecular machines are among the most important and challenging targets for structural and mechanistic analyses. Consequently, there is great interest in development of NMR methods for the study of multicomponent systems in the 50-500 kDa range. Biochemical methods also must be developed in concert to produce such systems in selectively labeled form. Here, we present 1H/13C-HSQC spectra of protonated methyl groups in a model system that mimics molecular weights up to ~560 kDa. Signals from side chain methyl groups of Ile, Leu, and Val residues are clearly detectable at correlation times up to ~330 ns. We have also developed a biochemical procedure to produce the 240 kDa, heteroheptameric Arp2/3 actin nucleation complex selectively labeled at one subunit and obtained 1H/13C-HSQC spectra of this assembly. Sensitivity in spectra of both the Arp2/3 complex and the model system indicate that methyl groups will be useful sources of information in nonsymmetric systems with molecular weights greater than 600 kDa at concentrations less than 100 μM. Methyl analyses will complement TROSY and CRINEPT analyses of amides in NMR studies of structure and molecular interactions of extremely large macromolecules and assemblies.

    The American Academy of Microbiology convened a colloquium November 6-7, 2003, in Key Largo, Florida, to discuss the application of genomic methods to the study of pathogenesis. Professionals in the fields of genomics, bacteriology, virology, eukaryotic microbiology, medicine, clinical diagnostics, bioinformatics, and forensics contributed their expertise to discussions on the recent advancements in the field and the outlook for future research. A number of recommendations were made for ensuring that progress in the field continues. The availability of genome sequences from pathogenic bacteria, viruses, and fungi and other eukaryotes has opened new horizons for the field of pathogenesis. The genomes of over 100 bacterial pathogens have been fully sequenced, and scientists are busy investigating the mechanisms that regulate the diversity of bacterial pathogens and their myriad abilities to evade host defenses. Close to 1,200 viral genomes have been sequenced, and virologists are now beginning to examine the genomes of those viruses that cause undetected, cryptic infections. These virus-host interactions can serve as a reservoir of viral genes that later emerge in novel pathogens with the potential to infect humans, economically important animals, or crops. A number of eukaryotic microbes, including several pathogenic fungi, have also been sequenced, revealing unimagined diversity among these important pathogens. Certain themes have emerged from analyses of pathogen genome sequences, and the possibility exists that a sequence-based common thread may be found linking pathogens from different taxa. The results of genome sequencing efforts have also informed the study of pathogenesis, helping to identify the ways in which pathogens bring about disease. The advances of the past several years have been great, and we are closer than ever to a comprehensive understanding of pathogenesis, but much work lies ahead. If the science is to move forward, the genome sequences of many more organisms are needed. The sequences of many hosts, pathogens, their nonpathogenic relatives, commensals, as well as a diverse array of microorganisms, are all needed to complete the picture of pathogenesis and provide a phylogenetic framework for understanding the phenomenon. Moreover, improvements are needed in the two most important tools of genomics: annotation methodologies and sequence databases.

The full-length 21.5-kDa MBP isoform has an additional 26 residues encoded by exon-II of the classic gene, which causes it to be trafficked to the nucleus of oligodendrocytes (OLGs). We have performed site-directed mutagenesis of selected residues within this segment in red fluorescent protein (RFP)-tagged constructs, which were then transfected into the immortalized N19-OLG cell line to view protein localization using epifluorescence microscopy. We found that 21.5-kDa MBP contains two non-traditional PY-nuclear-localization signals, and that arginine and lysine residues within these motifs were involved in subcellular trafficking of this protein to the nucleus, where it may have functional roles during myelinogenesis.

This document selects a new interim remedy to address an estimated 45,000 cubic yards of pentachlorophenol (PCP) and dioxin-contaminated source material (i.e., soil, sediment, and debris) and expands the scope of the groundwater remedy to permanently address PCP-, and potentially dioxin-, contaminated groundwater in the upper surficial aquifer.

    Crystals of the C-terminal 10 kDa lid subdomain from the C. elegans chaperone Hsp70 have been obtained that diffract X-rays to ?3.5 and belong to space group I2{sub 1}2{sub 1}2{sub 1}. Analysis of X-ray data and initial heavy-atom phasing reveals 24 monomers in the asymmetric unit related by 432 non-crystallographic symmetry. Hsp70 is an important molecular chaperone involved in the regulation of protein folding. Crystals of the C-terminal 10 kDa helical lid domain (residues 542640) from a Caenorhabditis elegans Hsp70 homologue have been produced that diffract X-rays to ?3.4 . Crystals belong to space group I2{sub 1}2{sub 1}2{sub 1}, with unit-cell parameters a = b = 197, c = 200 . The Matthews coefficient, self-rotation function and Patterson map indicate 24 monomers in the asymmetric unit, showing non-crystallographic 432 symmetry. Molecular-replacement studies using the corresponding domain from rat, the only eukaryotic homologue with a known structure, failed and a mercury derivative was obtained. Preliminary MAD phasing using SHELXD and SHARP for location and refinement of the heavy-atom substructure and SOLOMON for density modification produced interpretable maps with a clear proteinsolvent boundary. Further density-modification, model-building and refinement are currently under way.

    The 7-acre Whitehouse Waste Oil Pits site was used by Allied Petroleum Products (Allied) to dispose of acidic waste oil sludges from its oil reclamation process in Whitehouse, Duval County, Florida. A cypress swamp system and residential area are immediately adjacent to the site. The acid sludge produced in the first step and clay used to decolorize the oil were dumped into the unlined pits at the site. A 1985 ROD addressed source control as a containment remedy consisting of a slurry wall construction, soil cap, and a ground water recovery and treatment system; however, EPA has re-evaluated the 1985 ROD selection and determined that the containment remedy failed to meet the requirements of SARA. As a result, the ROD Amendment focuses on an alternative for treating Whitehouse wastes by eliminating direct contact risk associated with pit soil/sludge wastes and preventing contaminated ground water in the surficial aquifer from migrating laterally. The primary contaminants of concern that affect the soil, sediment, surface water, and ground water are VOCs, including benzene, toluene, and xylenes; organics, including PCBs and phenols; and metals, including arsenic, chromium, and lead. The amended remedial action for the site are included.

    The first component of the bacterial phosphotransferase system, enzyme I (EI), is a multidomain 128 kDa dimer that undergoes large rigid-body conformational transitions during the course of its catalytic cycle. Here we investigate the solution structure of a non-phosphorylatable active-site mutant in which the active-site histidine is substituted by glutamine. We show that perturbations in the relative orientations and positions of the domains and subdomains can be rapidly and reliably determined by conjoined rigid-body/torsion angle/Cartesian simulated annealing calculations driven by orientational restraints from residual dipolar couplings and shape and translation information afforded by small- and wide-angle X-ray scattering. Although histidine and glutamine are isosteric, the conformational space available to a Gln side chain is larger than that for the imidazole ring of His. An additional hydrogen bond between the side chain of Gln189 located on the EIN{sup {alpha}/{beta}} subdomain and an aspartate (Asp129) on the EIN{sup {alpha}} subdomain results in a small ({approx}9{sup o}) reorientation of the EIN{sup {alpha}} and EIN{sup {alpha}/{beta}} subdomains that is in turn propagated to a larger reorientation ({approx}26{sup o}) of the EIN domain relative to the EIC dimerization domain, illustrating the positional sensitivity of the EIN domain and its constituent subdomains to small structural perturbations.

    There are several definitions of symbiosis, but in this book it involves an association where one organism (the symbiont) lives within or on the body of another organism (the host), regardless of the actual effect on the host. Some symbioses are mutualistic, some parasitic, and some involve commensalism, in which one partner derives some benefit without either harming or benefiting the other. This is the second volume in this exciting and rapidly advancing topic by these editors.     The U.S. Bureau of Mines diskette contains analysis and related source data for 2,357 natural gas samples collected from miscellaneous states, which include the following states: Alabama, Arkansas (except Arkoma Basin), Florida, Georgia, Illinois, Indiana, Iowa, Kentucky, Louisiana, Maryland, Michigan, Mississippi, Missouri, New Jersey, New York, North Carolina, Ohio, Pennsylvania, Tennessee, Virginia, and West Virginia. All samples were obtained and analyzed as part of the Bureau's investigations of occurrences of helium in natural gases of countries with free market economies. The survey has been conducted since 1917. The analysis contained on the diskette contain the full range of component analysis data. Five files are on the diskette: READ.ME, MISC.TXT, MISC.DBF, USHEANAL.DBF, and BASINCDE.TXT.

    It is difficult to overestimate the importance that physical measurements performed with nondestructive assay instruments play throughout the nuclear fuel cycle. They underpin decision making in many areas and support: criticality safety, radiation protection, process control, safeguards, facility compliance, and waste measurements. No physical measurement is complete or indeed meaningful, without a defensible and appropriate accompanying statement of uncertainties and how they combine to define the confidence in the results. The uncertainty budget should also be broken down in sufficient detail suitable for subsequent uses to which the nondestructive assay (NDA) results will be applied. Creating an uncertainty budget and estimating the total measurement uncertainty can often be an involved process, especially for non routine situations. This is because data interpretation often involves complex algorithms and logic combined in a highly intertwined way. The methods often call on a multitude of input data subject to human oversight. These characteristics can be confusing and pose a barrier to developing and understanding between experts and data consumers. ASTM subcommittee C26-10 recognized this problem in the context of how to summarize and express precision and bias performance across the range of standards and guides it maintains. In order to create a unified approach consistent with modern practice and embracing the continuous improvement philosophy a consensus arose to prepare a procedure covering the estimation and reporting of uncertainties in non destructive assay of nuclear materials. This paper outlines the needs analysis, objectives and on-going development efforts. In addition to emphasizing some of the unique challenges and opportunities facing the NDA community we hope this article will encourage dialog and sharing of best practice and furthermore motivate developers to revisit the treatment of measurement uncertainty.
