    The crystal structures of the eubacterial translation initiation factor 2 in apo form and with bound GDP and GTP reveal conformational changes upon nucleotide binding and hydrolysis, notably of the catalytically important histidine in the switch II region. Translation initiation factor 2 (IF2) is involved in the early steps of bacterial protein synthesis. It promotes the stabilization of the initiator tRNA on the 30S initiation complex (IC) and triggers GTP hydrolysis upon ribosomal subunit joining. While the structure of an archaeal homologue (a/eIF5B) is known, there are significant sequence and functional differences in eubacterial IF2, while the trimeric eukaryotic IF2 is completely unrelated. Here, the crystal structure of the apo IF2 protein core from Thermus thermophilus has been determined by MAD phasing and the structures of GTP and GDP complexes were also obtained. The IF2–GTP complex was trapped by soaking with GTP in the cryoprotectant. The structures revealed conformational changes of the protein upon nucleotide binding, in particular in the P-loop region, which extend to the functionally relevant switch II region. The latter carries a catalytically important and conserved histidine residue which is observed in different conformations in the GTP and GDP complexes. Overall, this work provides the first crystal structure of a eubacterial IF2 and suggests that activation of GTP hydrolysis may occur by a conformational repositioning of the histidine residue.

  5. The structure of YqeH: An AtNOS1/AtNOA1 ortholog that couples GTP hydrolysis to molecular recognition

    AtNOS1/AtNOA1 was identified as a nitric oxide-generating enzyme in plants, but that function has recently been questioned. To resolve issues surrounding AtNOA1 activity, we report the biochemical properties and a 2.36 {angstrom} resolution crystal structure of a bacterial AtNOA1 ortholog (YqeH). Geobacillus YqeH fused to a putative AtNOA1 leader peptide complements growth and morphological defects of Atnoa1 mutant plants. YqeH does not synthesize nitric oxide from L-arginine but rather hydrolyzes GTP. The YqeH structure reveals a circularly permuted GTPase domain and an unusual C-terminal {beta}-domain. A small N-terminal domain, disordered in the structure, binds zinc. Structural homology among the C-terminal domain, the RNA-binding regulator TRAP, and the hypoxia factor pVHL define a recognition module for peptides and nucleic acids. TRAP residues important for RNA binding are conserved by the YqeH C-terminal domain, whose positioning is coupled to GTP hydrolysis. YqeH and AtNOA1 probably act as G-proteins that regulate nucleic acid recognition and not as nitric-oxide synthases.

  6. Characterization of a Ras Mutant with Identical GDP- and GTP-Bound Structures

    We previously characterized the G60A mutant of Ras and showed that the switch regions of the GTP-bound but not the GDP-bound form of this mutant adopt an 'open conformation' similar to that seen in nucleotide-free Ras. Here, we mutate Lys147 of the conserved {sup 145}SAK{sup 147} motif in the G60A background and characterize the resulting double mutant (DM). We show that RasDM is the first structure of a Ras protein with identical GDP- and GTP-bound structures. Both structures adopt the open conformation of the active form of RasG60A. The increase in the accessible surface area of the nucleotide is consistent with a 4-fold increase in its dissociation rate. Stopped-flow experiments show no major difference in the two-step kinetics of association of GDP or GTP with the wild type, G60A, or RasDM. Addition of Sos fails to accelerate nucleotide exchange. Overexpression of the G60A or double mutant of Ras in COS-1 cells fails to activate Erk and shows a strong dominant negative effect. Our data suggest that flexibility at position 60 is required for proper Sos-catalyzed nucleotide exchange and that structural information is somehow shared among the switch regions and the different nucleotide binding motifs.

    Rat sarcoma (Ras) GTPases regulate cell proliferation and survival through effector pathways including Raf-MAPK, and are the most frequently mutated genes in human cancer. Although it is well established that Ras activity requires binding to both GTP and the membrane, details of how Ras operates on the cell membrane to activate its effectors remain elusive. Efforts to target mutant Ras in human cancers to therapeutic benefit have also been largely unsuccessful. Here we show that Ras-GTP forms dimers to activate MAPK. We used quantitative photoactivated localization microscopy (PALM) to analyze the nanoscale spatial organization of PAmCherry1-tagged KRas 4B (hereafter referred to KRas) on the cell membrane under various signaling conditions. We found that at endogenous expression levels KRas forms dimers, and KRasG12D, a mutant that constitutively binds GTP, activates MAPK. Overexpression of KRas leads to formation of higher order Ras nanoclusters. Conversely, at lower expression levels, KRasG12D is monomeric and activates MAPK only when artificially dimerized. Moreover, dimerization and signaling of KRas are both dependent on an intact CAAX (C, cysteine; A, aliphatic; X, any amino acid) motif that is also known to mediate membrane localization. These results reveal a new, dimerization-dependent signaling mechanism of Ras, and suggest Ras dimers as a potential therapeutic target in mutant Ras-driven tumors.

    Insulin increases glucose transport in fat and muscle cells by stimulating the exocytosis of specialized vesicles containing the glucose transporter GLUT4. This process, which is referred to as GLUT4 translocation, increases the amount of GLUT4 at the cell surface. Previous studies have provided evidence that insulin signaling increases the amount of Rab10-GTP in the GLUT4 vesicles and that GLUT4 translocation requires the exocyst, a complex that functions in the tethering of vesicles to the plasma membrane, leading to exocytosis. In the present study we show that Rab10 in its GTP form binds to Exoc6 and Exoc6b, which are the two highly homologous isotypes of an exocyst subunit, that both isotypes are found in 3T3-L1 adipocytes, and that knockdown of Exoc6, Exoc6b, or both inhibits GLUT4 translocation in 3T3-L1 adipocytes. These results suggest that the association of Rab10-GTP with Exoc6/6b is a molecular link between insulin signaling and the exocytic machinery in GLUT4 translocation. - Highlights: • Insulin stimulates the fusion of vesicles containing GLUT4 with the plasma membrane. • This requires vesicular Rab10-GTP and the exocyst plasma membrane tethering complex. • We find that Rab10-GTP associates with the Exoc6 subunit of the exocyst. • We find that knockdown of Exoc6 inhibits fusion of GLUT4 vesicles with the membrane. • The interaction of Rab10-GTP with Exoc6 potentially links signaling to exocytosis.

    Eukaryotic and archaeal translation initiation factors 2, heterotrimers that consist of {alpha}-, {beta}-, and {gamma}-subunits, deliver methionylated initiator tRNA to a small ribosomal subunit in a manner that depends on GTP. To evaluate correlation of the function and association of the subunits, we used isothermal titration calorimetry to analyze the thermodynamics of the interactions between the {alpha}- and {gamma}-subunits in the presence or absence of a nonhydrolyzable GTP analog or GDP. The {alpha}-subunits bound to the {gamma}-subunit with large heat capacity change ({delta}C{sub p}) values. The {delta}H and {delta}C{sub p} values for the interaction between the {alpha}- and {gamma}-subunits varied in the presence of the GTP analog but not in the presence of GDP. These results suggest that the binding of both the {alpha}-subunit and GTP changes the conformation of the switch region of the {gamma}-subunit and increases the affinity of the {gamma}-subunit for tRNA.

    Molecular traffic between the nucleus and the cytoplasm is regulated by the nuclear pore complex (NPC), which acts as a highly selective channel perforating the nuclear envelope in eukaryotic cells. The human immunodeficiency virus (HIV) exploits the nucleocytoplasmic pathway to export its RNA transcripts across the NPC to the cytoplasm. Despite extensive study on the HIV life cycle and the many drugs developed to target this cycle, no current drugs have been successful in targeting the critical process of viral nuclear export, even though HIV’s reliance on a single host protein, CRM1, to export its unspliced and partially spliced RNAmore » transcripts makes it a tempting target. Due to recent findings implicating a DEAD-box helicase, DDX3, in HIV replication and a member of the export complex, it has become an appealing target for anti-HIV drug inhibition. In the present research, we have applied a hybrid computational protocol to analyze protein-protein interactions in the HIV mRNA export cycle. This method is based on molecular docking followed by molecular dynamics simulation and accompanied by approximate free energy calculation (MM/GBSA), computational alanine scanning, clustering, and evolutionary analysis. We highlight here some of the most likely binding modes and interfacial residues between DDX3 and CRM1 both in the absence and presence of RanGTP. This work shows that although DDX3 can bind to free CRM1, addition of RanGTP leads to more concentrated distribution of binding modes and stronger binding between CRM1 and RanGTP.« less

    This study used DOE ARM data and facilities to: 1) study macroscopic properties of continental stratus clouds at SGP and the factors controlling these properties, 2) develop a scientific basis for understanding the pocesses responsible for the formation of boundary layer clouds using ARM observations in conjunction with simple parametric models and LES, and 3) evaluate cumulus cloud characteristics retrieved retrieved from the MMCR operating at TWP-Nauru. In addition we have used high resolution 94 GHz observations of boundary layer clouds and precipitation to: 1)develop techniques for using high temporal resolution Doppler velocities to study large-eddy circulations and turbulence in boundary layer clouds and estimate the limitations of using current and past MMCR data for boundary layer cloud studies, 2) evaluate the capability and limitation of the current MMCR data for estimating reflectivity, vertical velocities, and spectral under low-signal-to-noise conditions associated with weak non-precipitating clouds, 3) develop possible sampling modes for the new MMCR processors to allow for adequate sampling of boundary layer clouds, and 4) retrieve updraft and downdraft structures under precipitating conditions.

The complete method comprises the steps of locating the ground with reference to the mildpoint of the parallel branches by connecting a potentiometer across the terminals of the circuit and connecting the slider of the potentiometer to ground through a current indicating instrument, adjusting the slider to right or left of the mildpoint so as to cause the instrument to indicate zero, connecting the terminal of the network which is farthest from the ground as thus indicated by the potentiometer to ground through a condenser, impressing a ripple voltage on the circuit, and then measuring the ripple voltage at the midpoint of each parallel branch to find the branch in which is the lowest value of ripple voltage, and then measuring the distribution of the ripple voltage along this branch to determine the point at which the ripple voltage drops off to zero or substantially zero due to the existence of a ground. The invention has particular application where a circuit ground is present which will disappear if the normal circuit voltage is removed.

Ground-water investigations are carried out to fulfill the requirements for the US Department of Energy (DOE) to meet the requirements of DOE Orders. Investigations are also performed for various clients to meet the requirements of the Resource Conservation and Recovery Act of 1976 (RCRA) and the Comprehensive Environmental Response, Compensation and Liability Act of 1980 (CERCLA). National standards including procedures published by the American Society for Testing and Materials (ASTM) and the US Geological Survey were utilized in developing the procedures contained in this manual.

    This document is a users manual and applications guide for the software package SGA. This package comprises four computer programs, namely SOMIP, SMECC, SGSYS, and TGRND. The first three programs are analysis models which are to be used in the design process of substation grounding systems. The fourth program, TGRND, is an analysis program for determining the transient response of a grounding system. This report, Volume 5, is an applications guide of the three computer programs. SOMIP, SMECC, and SGSYS, for the purpose of designing a safe substation grounding system. The applications guide utilizes four example substation grounding systems for the purpose of illustrating the application of the programs, SOMIP, SMECC, and SGSYS. The examples are based on data provided by four contributing utilities, namely, Houston Lighting and Power Company, Southern Company Services, Puget Sound Power and Light Company, and Arizona Public Service Company. For the purpose of illustrating specific capabilities of the computer programs, the data have been modified. As a result, the final designs of the four systems do not necessarily represent actual grounding system designs by these utilities. The example system 1 is a 138 kV/35 kV distribution substation. The example system 2 is a medium size 230 kV/115 kV transmission substation. The third example system is a generation substation while the last is a large 525 kV/345 kV/230 kV transmission substation. The four examples cover most of the practical problems that a user may encounter in the design of substation grounding systems.

    In the last few years the public and governments of many nations have become increasingly aware of the need for improving oil tanker safety. The requirements for double hull tankers are an attempt to address this need through legislation. Even though a number of investigations on the mechanics of collisions have been done in the past, until recently very little research supported the development of structural improvements to reduce oil tanker damage during grounding and stranding accidents. An aggressive evaluation of double hull tanker crashworthiness in stranding and grounding accidents is underway at CD/NSWC (formerly the David Taylor Research Center). The ability to predict damage from grounding accidents accurately is not currently available. The objective of this paper is to present qualitatively the structural failure mechanisms associated with stranding and grounding events for candidate double hull tanker structures and to present some simple methods for comparing damage scenarios. A comparison of the structural performance of key features in several very different designs will provide useful information toward this understanding.

    NREL conducted an annual program risk analysis on behalf of the U.S. Department of Energy Geothermal Technologies Program (GTP). NREL implemented a probabilistic risk analysis of GTP-sponsored research, development, and demonstration (RD&D) work, primarily for enhanced geothermal systems (EGS). The analysis examined estimates of improvement potential derived from program RD&D work for two types of technology performance metric (TPM): EGS-enabling technologies potential and EGS cost improvement potential. Four risk teams (exploration, wells/pumps/tools, reservoir engineering, and power conversion) comprised of industry experts, DOE laboratory researchers, academic researchers, and laboratory subcontractors estimated the RD&D impacts and TPM-improvement probability distributions. The assessment employed a risk analysis spreadsheet add-in that uses Monte Carlo simulation to drive the Geothermal Electric Technology Evaluation Model (GETEM). The GETEM-based risk analysis used baseline data from the experts' discussion of multiple reports and data sources. Risk results are expressed in terms of each metric's units and/or the program's top-level metric: levelized costs of electricity (LCOE). Results--both qualitative comments and quantitative improvement potential--are thorough and cohesive in three of the four expert groups. This conference paper summarizes the industry's current thinking on various metrics and potential for research improvement in geothermal technologies.

The voltage of the first electrode and second electrode is attenuated by an attenuation factor creating an attenuated voltage. The true RMS voltage of the attenuated voltage is determined creating an attenuated true RMS voltage. The attenuated true RMS voltage is then multiplied by the attenuation factor creating a calculated true RMS voltage. If the calculated true RMS voltage is greater than a first predetermined voltage threshold, a first alarm is enabled at a local location. If user input is received at a remote location acknowledging the first alarm, a first alarm acknowledgment signal is transmitted. The first alarm acknowledgment signal is then received at which time the first alarm is disabled.

    A critical U.S. Department of Energy (DOE) mission is to plan, implement, and complete DOE Environmental Restoration (ER) programs at facilities that were operated by or in support of the former Atomic Energy Commission (AEC). These facilities include the 24 inactive processing sites the Uranium Mill Tailings Radiation Control Act (UMTRCA) (42 USC Section 7901 et seq.) identified as Title I sites, which had operated from the late 1940s through the 1970s. In UMTRCA, Congress acknowledged the potentially harmful health effects associated with uranium mill tailings and directed the DOE to stabilize, dispose of, and control the tailings in a safe and environmentally sound manner. The UMTRA Surface Project deals with buildings, tailings, and contaminated soils at the processing sites and any associated vicinity properties (VP). Surface remediation at the processing sites will be completed in 1997 when the Naturita, Colorado, site is scheduled to be finished. The UMTRA Ground Water Project was authorized in an amendment to the UMTRCA (42 USC Section 7922(a)), when Congress directed DOE to comply with U.S. Environmental Protection Agency (EPA) ground water standards. The UMTRA Ground Water Project addresses any contamination derived from the milling operation that is determined to be present at levels above the EPA standards.

  15. Regional analysis of ground and above-ground climate

    The regional suitability of underground construction as a climate control technique is discussed with reference to (1) a bioclimatic analysis of long-term weather data for 29 locations in the United States to determine appropriate above ground climate control techniques, (2) a data base of synthesized ground temperatures for the coterminous United States, and (3) monthly dew point ground temperature comparisons for identifying the relative likelihood of condensation from one region to another. It is concluded that the suitability of earth tempering as a practice and of specific earth-sheltered design stereotypes varies geographically; while the subsurface almost always provides a thermal advantage on its own terms when compared to above ground climatic data, it can, nonetheless, compromise the effectiveness of other, regionally more important climate control techniques. Also contained in the report are reviews of above and below ground climate mapping schemes related to human comfort and architectural design, and detailed description of a theoretical model of ground temperature, heat flow, and heat storage in the ground. Strategies of passive climate control are presented in a discussion of the building bioclimatic analysis procedure which has been applied in a computer analysis of 30 years of weather data for each of 29 locations in the United States.

In over 20 years in some aspect of this profession, the author has never faced that particular challenge. By far the majority of information management opportunities involve taking over from someone else. Sometimes, a predecessor has gone on to better things on his/her initiative; that is not always the case. Sometimes the group is one you were a part of yesterday. If the function functions, time moves on and changes may be needed to accommodate new technology, additional and/or changed tasks, and alterations in corporate missions. If the function does not, it is a good bet that you were hired or promoted as an agent of change. Each of these situations poses challenges. This presentation is about that first few months and first year in a new assignment. In other words, you have the job, now what?

    The freezing of ground for the containment of subsurface hazardous waste is a promising method that is environmentally friendly and offers a safe alternative to other methods of waste retention in many cases. The frozen soil method offers two concepts for retaining waste. One concept is to freeze the entire waste area into a solid block of frozen soil thus locking the waste in situ. For small areas where the contaminated soil does not include vessels that would rupture from frost action, this concept may be simpler to install. A second concept, of course, is to create a frozen soil barrier to confine the waste within prescribed unfrozen soil boundaries; initial research in this area was funded by EPA, Cincinnati, OH, and the Army Corps of Engineers. The paper discusses advantages and limitations, a case study from Oak Ridge, TN, and a mesh generation program that simulates the cryogenic technology.

    Amount 61% of the water used by Texans is ground-water. Some areas, both municipal and rural, depend entirely on ground-water. In many areas long term withdrawal is lowering the water levels, causing surface land subsidence, salt-water encroachment, and reducing future reservoir availability. The increasing probability of seepage from radioactive and toxic wastes, herbicide residues, septic systems, and oilfield brines is threatening dangerous contamination of fresh ground-water reservoirs. The Texas Department of Water Resources, the Texas Department of Health, State and private colleges and universities, the US Geological Survey, the Environmental Protection Agency, various underground water districts, among others, are cooperating with concerned hydrologists in a concentrated program to increase the efficiency of ground-water use and development, preserve the aquifer reservoirs, and decrease the pollution potential. 88 references.

    The ground water project evaluates the nature and extent of ground water contamination resulting from the uranium ore processing activities. This report is a site specific document that will be used to evaluate current and future impacts to the public and the environment from exposure to contaminated ground water. Currently, no one is using the ground water and therefore, no one is at risk. However, the land will probably be developed in the future and so the possibility of people using the ground water does exist. This report examines the future possibility of health hazards resulting from the ingestion of contaminated drinking water, skin contact, fish ingestion, or contact with surface waters and sediments.

    Perhaps the greatest risk to both equipment and personnel associated with highwall mining is from ground control. The two most significant ground control hazards are rock falls from highwall and equipment entrapment underground. In the central Appalachians, where the majority of highwall mining occurs in the USA, hillseams (or mountain cracks) are the most prominent structure that affects highwall stability. The article discusses measures to minimise the risk of failure associated with hillstreams. A 'stuck' or trapped highwall miner, and the ensuring retrieval or recovery operation, can be extremely disruptive to the highwall mining process. Most entrapment, are due to roof falls in the hole. The options for recovery are surface retrieval, surface excavation or underground recovery. Proper pillar design is essential to maintain highwall stability and prevent entrapments. NIOSH has developed the Analysis of Retreat Mining Pillar stability-Highwall Mining (ARMPS-HWM) computer program to help mine planners with this process. 10 figs.

    Vertically oriented radiation detectors may not provide sufficient screening in rail or aviation applications. Railcars can be heavily shielded on the sides, reducing the sensitivity of vertically mounted monitors. For aviation, the distance required for wingspan clearance reduces a vertical detector’s coverage of the fuselage. To surmount these, and other, challenging operational and sensitivity issues, we have investigated the use of in-ground radiation detectors. (PIET-43741-TM-605).

    This GNDD Technology Roadmap is intended to provide guidance to potential researchers and help management define research priorities to achieve technology advancements for ground-based nuclear explosion monitoring science being pursued by the Ground-based Nuclear Detonation Detection (GNDD) Team within the Office of Nuclear Detonation Detection in the National Nuclear Security Administration (NNSA) of the U.S. Department of Energy (DOE). Four science-based elements were selected to encompass the entire scope of nuclear monitoring research and development (R&D) necessary to facilitate breakthrough scientific results, as well as deliver impactful products. Promising future R&D is delineated including dual use associated with the Comprehensive Nuclear-Test-Ban Treaty (CTBT). Important research themes as well as associated metrics are identified along with a progression of accomplishments, represented by a selected bibliography, that are precursors to major improvements to nuclear explosion monitoring.

    The Department of Energy (DOE) has developed this Standard to assist line managers in meeting their responsibilities for implementing occupational radiological control programs. DOE has established regulatory requirements for occupational radiation protection in Title 10 of the Code of Federal Regulations, Part 835 (10 CFR 835), ``Occupational Radiation Protection``. Failure to comply with these requirements may lead to appropriate enforcement actions as authorized under the Price Anderson Act Amendments (PAAA). While this Standard does not establish requirements, it does restate, paraphrase, or cite many (but not all) of the requirements of 10 CFR 835 and related documents (e.g., occupational safety and health, hazardous materials transportation, and environmental protection standards). Because of the wide range of activities undertaken by DOE and the varying requirements affecting these activities, DOE does not believe that it would be practical or useful to identify and reproduce the entire range of health and safety requirements in this Standard and therefore has not done so. In all cases, DOE cautions the user to review any underlying regulatory and contractual requirements and the primary guidance documents in their original context to ensure that the site program is adequate to ensure continuing compliance with the applicable requirements. To assist its operating entities in achieving and maintaining compliance with the requirements of 10 CFR 835, DOE has established its primary regulatory guidance in the DOE G 441.1 series of Guides. This Standard supplements the DOE G 441.1 series of Guides and serves as a secondary source of guidance for achieving compliance with 10 CFR 835.

    This document reports the findings of a performance assessment (PA) analysis for the disposal of solid low-level radioactive waste (LLW) in the 200 West Area Low-Level Waste Burial Grounds (LLBG) in the northwest corner of the 200 West Area of the Hanford Site. This PA analysis is required by US Department of Energy (DOE) Order 5820.2A (DOE 1988a) to demonstrate that a given disposal practice is in compliance with a set of performance objectives quantified in the order. These performance objectives are applicable to the disposal of DOE-generated LLW at any DOE-operated site after the finalization of the order in September 1988.     This baseline risk assessment at the former uranium mill tailings site near Shiprock, New Mexico, evaluates the potential impact to public health or the environment resulting from ground water contamination at the former uranium mill processing site. The tailings and other contaminated material at this site were placed in an on-site disposal cell in 1986 through the US Department of Energy (DOE) Uranium Mill Tailings Remedial Action (UMTRA) Project. Currently, the UMTRA Project is evaluating ground water contamination. This risk assessment is the first document specific to this site for the Ground Water Project. There are no domestic or drinking water wells in the contaminated ground water of the two distinct ground water units: the contaminated ground water in the San Juan River floodplain alluvium below the site and the contaminated ground water in the terrace alluvium area where the disposal cell is located. Because no one is drinking the affected ground water, there are currently no health or environmental risks directly associated with the contaminated ground water. However, there is a potential for humans, domestic animals, and wildlife to the exposed to surface expressions of ground water in the seeps and pools in the area of the San Juan River floodplain below the site. For these reasons, this risk assessment evaluates potential exposure to contaminated surface water and seeps as well as potential future use of contaminated ground water.

Surface cleanup at this site is under way and is scheduled for completion in 1996. The tailings are being stabilized in-place at this site. The disposal area has been withdrawn from public use by the DOE and is referred to as the permanent withdrawal area. The Ground Water Project evaluates the nature and extent of ground water contamination resulting from past uranium ore processing activities. The Ground Water Project at this site is in its beginning stages. This report is a site-specific document that will be used to evaluate current and future potential impacts to the public and the environment from exposure to contaminated ground water. The results of this evaluation and further site characterization will determine whether any action is needed to protect human health or the environment. Currently, no points of exposure (e.g. a drinking water well); and no receptors of contaminated ground water have been identified at the Maybell site. Therefore, there are no current human health and ecological risks associated with exposure to contaminated ground water. Furthermore, if current site conditions and land- and water-use patterns do not change, it is unlikely that contaminated ground water would reach people or the ecological communities in the future.

This Environmental Assessment (EA) discusses two alternatives and the effects associated with each. The two alternatives are (1) natural flushing coupled with institutional controls and continued monitoring and (2) no action. The compliance strategy must meet U.S. Environmental Protection Agency (EPA) ground water standards defined in Title 40 ''Code of Federal Regulations'' Part 192, Subpart B, in areas where ground water beneath and around the site is contaminated as a result of past milling operations.     This paper will briefly outline major activities in Department of Energy (DOE) Laboratories focused on mobile platforms, both Unmanned Ground Vehicles (UGV’s) as well as Unmanned Air Vehicles (UAV’s). The activities will be discussed in the context of the science and technology construct used by the DOE Technology Roadmap for Robotics and Intelligent Machines (RIM)1 published in 1998; namely, Perception, Reasoning, Action, and Integration. The activities to be discussed span from research and development to deployment in field operations. The activities support customers in other agencies. The discussion of "perception" will include hyperspectral sensors, complex patterns discrimination, multisensor fusion and advances in LADAR technologies, including real-world perception. "Reasoning" activities to be covered include cooperative controls, distributed systems, ad-hoc networks, platform-centric intelligence, and adaptable communications. The paper will discuss "action" activities such as advanced mobility and various air and ground platforms. In the RIM construct, "integration" includes the Human-Machine Integration. Accordingly the paper will discuss adjustable autonomy and the collaboration of operator(s) with distributed UGV’s and UAV’s. Integration also refers to the applications of these technologies into systems to perform operations such as perimeter surveillance, large-area monitoring and reconnaissance. Unique facilities and test beds for advanced mobile systems will be described. Given that this paper is an overview, rather than delve into specific detail in these activities, other more exhaustive references and sources will be cited extensively.

  2. Final Harvest of Above-Ground Biomass and Allometric Analysis of the Aspen FACE Experiment

    The Aspen FACE experiment, located at the US Forest Service Harshaw Research Facility in Oneida County, Wisconsin, exposes the intact canopies of model trembling aspen forests to increased concentrations of atmospheric CO2 and O3. The first full year of treatments was 1998 and final year of elevated CO2 and O3 treatments is scheduled for 2009. This proposal is to conduct an intensive, analytical harvest of the above-ground parts of 24 trees from each of the 12, 30 m diameter treatment plots (total of 288 trees) during June, July & August 2009. This above-ground harvest will be carefully coordinated with the below-ground harvest proposed by D.F. Karnosky et al. (2008 proposal to DOE). We propose to dissect harvested trees according to annual height growth increment and organ (main stem, branch orders, and leaves) for calculation of above-ground biomass production and allometric comparisons among aspen clones, species, and treatments. Additionally, we will collect fine root samples for DNA fingerprinting to quantify biomass production of individual aspen clones. This work will produce a thorough characterization of above-ground tree and stand growth and allocation above ground, and, in conjunction with the below ground harvest, total tree and stand biomass production, allocation, and allometry.

    Ground source heat pumps (GSHP) use the constant temperature of the Earth as the heat exchange medium instead of the outside air temperature. During the winter, a GSHP uses the ground as a heat source to provide heating, and during the summer, a GSHP uses the ground as a heat sink to provide cooling.     Proper substation grounding grid design requires good, accurate soil resistivity measurements. This data is essential to model the substation ground grid to design a safe ground grid with a satisfactory ground grid resistance at minimum cost. For substations with several decades of service, there is some concern that a grid may have deteriorated, been damaged during equipment installation or excavation, or that initial soil resistivity measurements were lost or may not have been correctly performed. Ground grid conductors change the substation surface voltage distribution. Any voltage measurements taken at the complete substation will also vary from the tests made without conductors present. During testing, current was injected in the soil by probes placed near the ground grid. The current tends to follow the ground grid conductors since copper is a far better conductor than the soil it is placed in. Resistance readings near grids will be lower than readings in undisturbed soil. Since computer models were unavailable for many years, analyzing the effect of the grid conductors on soil resistivity measurements was very difficult. As a result, soil resistivity measurements made close to substations were of little use to the engineer unless some means of correcting the measured values could be developed. This paper will present results of soil resistivity measurements near a substation ground grid before and after a ground grid has been installed and describes a means of calculating the undisturbed soil model.

    Optimal performance of closed-loop, ground-source heat pumps (ground-coupled heat pumps) is dependent upon the thermal properties of the backfill in the annual region between the ground heat exchanger (GHEX) tubes and the outer bore wall. Equally important is the protection of groundwater aquifers from contaminants that may flow from the surface of other aquifers through poorly sealed boreholes. Conventional cement and bentonite-based grouts have relatively low thermal conductivities. Loop requirements often increase beyond the allotted budget in applications where regulatory bodies require the entire heat exchanger length to be grouted. This paper reports on the results of four mixes of thermally enhanced cementitious grouts. Four grouts were evaluated in a test stand to minimize the impact of external factors typically present in field tests. The test stand accepts up to 6 in. (15 cm) ground heat exchangers in a 10 ft (3 m) test section. Controlled testing is performed in either the cooling mode (loop above 85 F [29 C]) or heating mode (loop at 32 F [0 C]), and the temperature of the outer bore wall is held constant with a groundwater source. Results indicate cement grouts that are enhanced with low-cost additives have thermal conductivities three to four times as large as conventional high-solids bentonite grouts. This would result in reduced heat exchanger lengths compared to those grouted with bentonite. There appears to be no measurable increase in overall borehole resistance due to separation of the colder tubes from the grout in the heating mode. This discussion does not include pumpability, permeability, and material handling issues, which must be thoroughly investigated before any grout can be recommended for use.

    A stepped frequency ground penetrating radar system is described comprising an RF signal generating section capable of producing stepped frequency signals in spaced and equal increments of time and frequency over a preselected bandwidth which serves as a common RF signal source for both a transmit portion and a receive portion of the system. In the transmit portion of the system the signal is processed into in-phase and quadrature signals which are then amplified and then transmitted toward a target. The reflected signals from the target are then received by a receive antenna and mixed with a reference signal from the common RF signal source in a mixer whose output is then fed through a low pass filter. The DC output, after amplification and demodulation, is digitized and converted into a frequency domain signal by a Fast Fourier Transform. A plot of the frequency domain signals from all of the stepped frequencies broadcast toward and received from the target yields information concerning the range (distance) and cross section (size) of the target.

    An improved ground flare is provided comprising a stack, two or more burner assemblies, and a servicing port so that some of the burner assemblies can be serviced while others remain in operation. The burner assemblies comprise a burner conduit and nozzles which are individually fitted to the stack's burner chamber and are each removably supported in the chamber. Each burner conduit is sealed to and sandwiched between a waste gas inlet port and a matching a closure port on the other side of the stack. The closure port can be opened for physically releasing the burner conduit and supplying sufficient axial movement room for extracting the conduit from the socket, thereby releasing the conduit for hand removal through a servicing port. Preferably, the lower end of the stack is formed of one or more axially displaced lower tubular shells which are concentrically spaced for forming annular inlets for admitting combustion air. An upper tubular exhaust stack, similarly formed, admits additional combustion air for increasing the efficiency of combustion, increasing the flow of exhausted for improved atmospheric dispersion and for cooling the upper stack.

    The Price-Anderson Act provides indemnification to DOE contractors who manage and conduct nuclear activities in the DOE complex. The government acts as an insurer for these contractors against any findings of liability from the nuclear activities of the contractor within the scope of its contract. 10 CFR Part 820 establishes the legal framework for implementing DOE`s Nuclear Safety Enforcement Program. Integration with other DOE organizations and programs would assure that the enforcement process properly considers the actual or potential safety significance of a violation when determining an appropriate enforcement sanction. Achieving a proactive contractor compliance assurance rather than a heavy enforcement hand, will require a foundation of cooperation and teamwork across DOE organizations. This handbook identifies the areas of interface for the DOE Enforcement Program and provides guidance on roles and responsibilities for the key DOE organizational areas.     The US Department of Energy (DOE) proposes to modify an existing reactor containment building (decommissioned Plutonium Recycle Test Reactor (PRTR) 309 Building) to provide ground test capability for the prototype SP-100 reactor. The 309 Building (Figure 1.1) is located in the 300 Area on the Hanford Site in Washington State. The National Environmental Policy Act (NEPA) requires that Federal agencies assess the potential impacts that their actions may have on the environment. This Environmental Assessment describes the consideration given to environmental impacts during reactor concept and test site selection, examines the environmental effects of the DOE proposal to ground test the nuclear subsystem, describes alternatives to the proposed action, and examines radiological risks of potential SP-100 use in space. 73 refs., 19 figs., 7 tabs.
