    We previously characterized the G60A mutant of Ras and showed that the switch regions of the GTP-bound but not the GDP-bound form of this mutant adopt an 'open conformation' similar to that seen in nucleotide-free Ras. Here, we mutate Lys147 of the conserved {sup 145}SAK{sup 147} motif in the G60A background and characterize the resulting double mutant (DM). We show that RasDM is the first structure of a Ras protein with identical GDP- and GTP-bound structures. Both structures adopt the open conformation of the active form of RasG60A. The increase in the accessible surface area of the nucleotide is consistent with a 4-fold increase in its dissociation rate. Stopped-flow experiments show no major difference in the two-step kinetics of association of GDP or GTP with the wild type, G60A, or RasDM. Addition of Sos fails to accelerate nucleotide exchange. Overexpression of the G60A or double mutant of Ras in COS-1 cells fails to activate Erk and shows a strong dominant negative effect. Our data suggest that flexibility at position 60 is required for proper Sos-catalyzed nucleotide exchange and that structural information is somehow shared among the switch regions and the different nucleotide binding motifs.

    Mobile colloids can play an important role in contaminant transport in soils: many contaminants exist in colloidal form, and colloids can facilitate transport of otherwise immobile contaminants. In unsaturated soils, colloid transport is, among other factors, affected by water content and flow rate. Our objective was to determine whether water content or flow rate is more important for colloid transport. We passed negatively charged polystyrene colloids (220 nm diameter) through unsaturated sand-filled columns under steady-state flow at different water contents (effective water saturations Se ranging from 0.1 to 1.0, with Se = (? ?r)/(?s ?r)) and flow rates (pore water velocities v of 5 and 10 cm/min). Water content was the dominant factor in our experiments. Colloid transport decreased with decreasing water content, and below a critical water content (Se < 0.1), colloid transport was inhibited, and colloids were strained in water films. Pendular ring and water film thickness calculations indicated that colloids can move only when pendular rings are interconnected. The flow rate affected retention of colloids in the secondary energy minimum, with less colloids being trapped when the flow rate increased. These results confirm the importance of both water content and flow rate for colloid transport in unsaturated porous media and highlight the dominant role of water content.

    The crystal structures of the eubacterial translation initiation factor 2 in apo form and with bound GDP and GTP reveal conformational changes upon nucleotide binding and hydrolysis, notably of the catalytically important histidine in the switch II region. Translation initiation factor 2 (IF2) is involved in the early steps of bacterial protein synthesis. It promotes the stabilization of the initiator tRNA on the 30S initiation complex (IC) and triggers GTP hydrolysis upon ribosomal subunit joining. While the structure of an archaeal homologue (a/eIF5B) is known, there are significant sequence and functional differences in eubacterial IF2, while the trimeric eukaryotic IF2 is completely unrelated. Here, the crystal structure of the apo IF2 protein core from Thermus thermophilus has been determined by MAD phasing and the structures of GTP and GDP complexes were also obtained. The IF2GTP complex was trapped by soaking with GTP in the cryoprotectant. The structures revealed conformational changes of the protein upon nucleotide binding, in particular in the P-loop region, which extend to the functionally relevant switch II region. The latter carries a catalytically important and conserved histidine residue which is observed in different conformations in the GTP and GDP complexes. Overall, this work provides the first crystal structure of a eubacterial IF2 and suggests that activation of GTP hydrolysis may occur by a conformational repositioning of the histidine residue.

    Rat sarcoma (Ras) GTPases regulate cell proliferation and survival through effector pathways including Raf-MAPK, and are the most frequently mutated genes in human cancer. Although it is well established that Ras activity requires binding to both GTP and the membrane, details of how Ras operates on the cell membrane to activate its effectors remain elusive. Efforts to target mutant Ras in human cancers to therapeutic benefit have also been largely unsuccessful. Here we show that Ras-GTP forms dimers to activate MAPK. We used quantitative photoactivated localization microscopy (PALM) to analyze the nanoscale spatial organization of PAmCherry1-tagged KRas 4B (hereafter referred to KRas) on the cell membrane under various signaling conditions. We found that at endogenous expression levels KRas forms dimers, and KRasG12D, a mutant that constitutively binds GTP, activates MAPK. Overexpression of KRas leads to formation of higher order Ras nanoclusters. Conversely, at lower expression levels, KRasG12D is monomeric and activates MAPK only when artificially dimerized. Moreover, dimerization and signaling of KRas are both dependent on an intact CAAX (C, cysteine; A, aliphatic; X, any amino acid) motif that is also known to mediate membrane localization. These results reveal a new, dimerization-dependent signaling mechanism of Ras, and suggest Ras dimers as a potential therapeutic target in mutant Ras-driven tumors.

    Eukaryotic and archaeal translation initiation factors 2, heterotrimers that consist of {alpha}-, {beta}-, and {gamma}-subunits, deliver methionylated initiator tRNA to a small ribosomal subunit in a manner that depends on GTP. To evaluate correlation of the function and association of the subunits, we used isothermal titration calorimetry to analyze the thermodynamics of the interactions between the {alpha}- and {gamma}-subunits in the presence or absence of a nonhydrolyzable GTP analog or GDP. The {alpha}-subunits bound to the {gamma}-subunit with large heat capacity change ({delta}C{sub p}) values. The {delta}H and {delta}C{sub p} values for the interaction between the {alpha}- and {gamma}-subunits varied in the presence of the GTP analog but not in the presence of GDP. These results suggest that the binding of both the {alpha}-subunit and GTP changes the conformation of the switch region of the {gamma}-subunit and increases the affinity of the {gamma}-subunit for tRNA.

    Molecular traffic between the nucleus and the cytoplasm is regulated by the nuclear pore complex (NPC), which acts as a highly selective channel perforating the nuclear envelope in eukaryotic cells. The human immunodeficiency virus (HIV) exploits the nucleocytoplasmic pathway to export its RNA transcripts across the NPC to the cytoplasm. Despite extensive study on the HIV life cycle and the many drugs developed to target this cycle, no current drugs have been successful in targeting the critical process of viral nuclear export, even though HIV’s reliance on a single host protein, CRM1, to export its unspliced and partially spliced RNA transcripts makes it a tempting target. Due to recent findings implicating a DEAD-box helicase, DDX3, in HIV replication and a member of the export complex, it has become an appealing target for anti-HIV drug inhibition. In the present research, we have applied a hybrid computational protocol to analyze protein-protein interactions in the HIV mRNA export cycle. This method is based on molecular docking followed by molecular dynamics simulation and accompanied by approximate free energy calculation (MM/GBSA), computational alanine scanning, clustering, and evolutionary analysis.     Molecular traffic between the nucleus and the cytoplasm is regulated by the nuclear pore complex (NPC), which acts as a highly selective channel perforating the nuclear envelope in eukaryotic cells. The human immunodeficiency virus (HIV) exploits the nucleocytoplasmic pathway to export its RNA transcripts across the NPC to the cytoplasm. Despite extensive study on the HIV life cycle and the many drugs developed to target this cycle, no current drugs have been successful in targeting the critical process of viral nuclear export, even though HIV’s reliance on a single host protein, CRM1, to export its unspliced and partially spliced RNAmore » transcripts makes it a tempting target. Due to recent findings implicating a DEAD-box helicase, DDX3, in HIV replication and a member of the export complex, it has become an appealing target for anti-HIV drug inhibition. In the present research, we have applied a hybrid computational protocol to analyze protein-protein interactions in the HIV mRNA export cycle. This method is based on molecular docking followed by molecular dynamics simulation and accompanied by approximate free energy calculation (MM/GBSA), computational alanine scanning, clustering, and evolutionary analysis.     NREL conducted an annual program risk analysis on behalf of the U.S. Department of Energy Geothermal Technologies Program (GTP). NREL implemented a probabilistic risk analysis of GTP-sponsored research, development, and demonstration (RD&D) work, primarily for enhanced geothermal systems (EGS). The analysis examined estimates of improvement potential derived from program RD&D work for two types of technology performance metric (TPM): EGS-enabling technologies potential and EGS cost improvement potential. Four risk teams (exploration, wells/pumps/tools, reservoir engineering, and power conversion) comprised of industry experts, DOE laboratory researchers, academic researchers, and laboratory subcontractors estimated the RD&D impacts and TPM-improvement probability distributions. The assessment employed a risk analysis spreadsheet add-in that uses Monte Carlo simulation to drive the Geothermal Electric Technology Evaluation Model (GETEM). The GETEM-based risk analysis used baseline data from the experts' discussion of multiple reports and data sources. Risk results are expressed in terms of each metric's units and/or the program's top-level metric: levelized costs of electricity (LCOE). Results--both qualitative comments and quantitative improvement potential--are thorough and cohesive in three of the four expert groups. This conference paper summarizes the industry's current thinking on various metrics and potential for research improvement in geothermal technologies.

    The invention is an air flow control valve which is capable of maintaining a constant flow at the outlet despite changes in the inlet or outlet pressure. The device consists of a shell assembly with an inlet chamber and outlet chamber separated by a separation plate. The chambers are connected by an orifice. Also located within the inlet chamber is a port controller assembly. The port controller assembly consists of a differential pressure plate and port cap affixed thereon. The cap is able to slide in and out of the orifice separating the inlet and outlet chambers. When the pressure differential is sufficient, the differential pressure plate rises or falls to maintain a constant air flow. Movement of the port controller assembly does not require the use of seals, diaphragms, tight tolerances, bushings, bearings, hinges, guides, or lubricants.

    A linear, two-dimensional model of a transonic plasma flow in equilibrium is constructed and given an explicit solution in the form of a complex Laplace integral. The solution indicates that the transonic state can be solved as an elliptic boundary value problem, as is done in the numerical code FLOW [Guazzotto et al., Phys. Plasmas 11, 604 (2004)]. Moreover, the presence of a hyperbolic region does not necessarily imply the presence of a discontinuity or any other singularity of the solution.

    The Department of Energy (DOE) has developed this Standard to assist line managers in meeting their responsibilities for implementing occupational radiological control programs. DOE has established regulatory requirements for occupational radiation protection in Title 10 of the Code of Federal Regulations, Part 835 (10 CFR 835), ``Occupational Radiation Protection``. Failure to comply with these requirements may lead to appropriate enforcement actions as authorized under the Price Anderson Act Amendments (PAAA). While this Standard does not establish requirements, it does restate, paraphrase, or cite many (but not all) of the requirements of 10 CFR 835 and related documents (e.g., occupational safety and health, hazardous materials transportation, and environmental protection standards). Because of the wide range of activities undertaken by DOE and the varying requirements affecting these activities, DOE does not believe that it would be practical or useful to identify and reproduce the entire range of health and safety requirements in this Standard and therefore has not done so. In all cases, DOE cautions the user to review any underlying regulatory and contractual requirements and the primary guidance documents in their original context to ensure that the site program is adequate to ensure continuing compliance with the applicable requirements. To assist its operating entities in achieving and maintaining compliance with the requirements of 10 CFR 835, DOE has established its primary regulatory guidance in the DOE G 441.1 series of Guides. This Standard supplements the DOE G 441.1 series of Guides and serves as a secondary source of guidance for achieving compliance with 10 CFR 835.

    The low flow monitor provides a means for determining if a fluid flow meets a minimum threshold level of flow. The low flow monitor operates with a minimum of intrusion by the flow detection device into the flow. The electrical portion of the monitor is externally located with respect to the fluid stream which allows for repairs to the monitor without disrupting the flow. The electronics provide for the adjustment of the threshold level to meet the required conditions. The apparatus can be modified to provide an upper limit to the flow monitor by providing for a parallel electronic circuit which provides for a bracketing of the desired flow rate.

    At CW press time the house was considering a 10% cut in Department of Energy (DOE) conservation programs and a 14% cut in DOE funding for fossil fuel R&D. The cuts show that, while easing off this year on efforts to pare back EPA, House Republicans have not given up the fight over federal technology programs. House Republicans consider federal involvement in R&D {open_quotes}corporate welfare,{close_quotes} and, while the Commerce Department`s Advanced Technology Program (ATP) has been the lightning rod for their criticism, DOE programs have also been targeted for reduction. The $499-million conservation funding level contemplated by the House is 33% below the President`s request and follows an equally large cut last year.

    Flow induced migration, whereby polymer melts are fractionated by molecular weight across a flow field, represents a significant complication in the processing of polymer melts. Despite its long history, such phenomena remain relatively poorly understood. Here a simple analytical theory is presented which predicts the phenomena based on well-established principles of non-equilibrium thermodynamics. It is unambiguously shown that for purely viscous materials, a gradient in shear rate is needed to drive migration; for purely viscometric flows no migration is expected. Molecular scale simulations of flow migration effects in dense polymer melts are also presented. In shear flow the melts exhibit similar behavior as the quiescent case; a constant shear rate across the gap does not induce chain length based migration. In comparison, parabolic flow causes profound migration for both unentangled and entangled melts. These findings are consistent with the analytical theory. The picture that emerges is consistent with flow induced migration mechanisms predominating over competing chain degradation mechanisms.

    A system for determining the density, flow velocity, and mass flow of a fluid comprising at least one sing-around circuit that determines the velocity of a signal in the fluid and that is correlatable to a database for the fluid. A system for determining flow velocity uses two of the inventive circuits with directional transmitters and receivers, one of which is set at an angle to the direction of flow that is different from the others.

    The Price-Anderson Act provides indemnification to DOE contractors who manage and conduct nuclear activities in the DOE complex. The government acts as an insurer for these contractors against any findings of liability from the nuclear activities of the contractor within the scope of its contract. 10 CFR Part 820 establishes the legal framework for implementing DOE`s Nuclear Safety Enforcement Program. Integration with other DOE organizations and programs would assure that the enforcement process properly considers the actual or potential safety significance of a violation when determining an appropriate enforcement sanction. Achieving a proactive contractor compliance assurance rather than a heavy enforcement hand, will require a foundation of cooperation and teamwork across DOE organizations. This handbook identifies the areas of interface for the DOE Enforcement Program and provides guidance on roles and responsibilities for the key DOE organizational areas. It complements DOE-HDBK-1087-95 and 1089-95.

Saliva and/or buccal cells are collected from an individual using the sample collection unit. The sample collection unit is immersed in the reagent reservoir. The tip of the lateral flow strip is immersed in the reservoir and the reagent/sample mixture wicks up into the lateral flow strip to perform the assay.

    A fume hood is provided having an adequate level of safety while reducing the amount of air exhausted from the hood. A displacement flow fume hood works on the principal of a displacement flow which displaces the volume currently present in the hood using a push-pull system. The displacement flow includes a plurality of air supplies which provide fresh air, preferably having laminar flow, to the fume hood. The displacement flow fume hood also includes an air exhaust which pulls air from the work chamber in a minimally turbulent manner. As the displacement flow produces a substantially consistent and minimally turbulent flow in the hood, inconsistent flow patterns associated with contaminant escape from the hood are minimized. The displacement flow fume hood largely reduces the need to exhaust large amounts of air from the hood. It has been shown that exhaust air flow reductions of up to 70% are possible without a decrease in the hood's containment performance. The fume hood also includes a number of structural adaptations which facilitate consistent and minimally turbulent flow within a fume hood.

    The U.S. Department of Energy (DOE) Office of Analysis within the Office of Health, Safety and Security (HSS) publishes the annual DOE Occupational Radiation Exposure Report to provide an overview of the status of radiation protection practices at DOE.* The DOE 2010 Occupational Radiation Exposure Report provides an evaluation of DOE-wide performance regarding compliance with DOE Part 835 dose limits and as low as reasonably achievable (ALARA) process requirements. In addition, the report provides data to DOE organizations responsible for developing policies for protection of individuals from the effects of radiation. The report provides a summary and an analysis of occupational radiation exposure information from the monitoring of individuals involved in DOE activities. The occupational radiation exposure information is analyzed in terms of aggregate data, dose to individuals, and dose by site over the past 5 years.

The DOE 2011 Occupational Radiation Exposure Report provides an evaluation of DOE-wide performance regarding compliance with Title 10, Code of Federal Regulations (C.F.R.), Part 835, Occupational Radiation Protection dose limits and as low as reasonably achievable (ALARA) process requirements. In addition, the report provides data to DOE organizations responsible for developing policies for protection of individuals from the adverse health effects of radiation. The report provides a summary and an analysis of occupational radiation exposure information from the monitoring of individuals involved in DOE activities. The occupational radiation exposure information is analyzed in terms of aggregate data, dose to individuals, and dose by site over the past five years.

    A microelectromechanical (MEM) flow control apparatus is disclosed which includes a fluid channel formed on a substrate from a first layer of a nonconducting material (e.g. silicon nitride). A first electrode is provided on the first layer of the nonconducting material outside the flow channel; and a second electrode is located on a second layer of the nonconducting material above the first layer. A voltage applied between the first and second electrodes deforms the fluid channel to increase its cross-sectional size and thereby increase a flow of a fluid through the channel. In certain embodiments of the present invention, the fluid flow can be decreased or stopped by applying a voltage between the first electrode and the substrate. A peristaltic pumping of the fluid through the channel is also possible when the voltage is applied in turn between a plurality of first electrodes and the substrate. A MEM flow control assembly can also be formed by providing one or more MEM flow control devices on a common substrate together with a submicron filter. The MEM flow control assembly can optionally include a plurality of pressure sensors for monitoring fluid pressure and determining flow rates through the assembly.

    A turbine assembly includes a plurality of rotor blades comprising a root portion, an airfoil having a pressure sidewall and a suction sidewall, and a top portion having a cap. An outer shroud is concentrically disposed about said rotor blades, said shroud in combination with said tip portions defining a clearance gap. At least one circumferential shroud leakage discourager is disposed within the shroud. The leakage discourager(s) increase the flow resistance and thus reduce the flow of hot gas flow leakage for a given pressure differential across the clearance gap to improve overall turbine efficiency.

    An apparatus for noninvasively monitoring the flow and/or the composition of a flowing liquid using ultrasound is described. The position of the resonance peaks for a fluid excited by a swept-frequency ultrasonic signal have been found to change frequency both in response to a change in composition and in response to a change in the flow velocity thereof. Additionally, the distance between successive resonance peaks does not change as a function of flow, but rather in response to a change in composition. Thus, a measurement of both parameters (resonance position and resonance spacing), once calibrated, permits the simultaneous determination of flow rate and composition using the apparatus and method of the present invention.
