    DegS is a periplasmic Escherichia coli protease, which functions as a trimer to catalyze the initial rate-limiting step in a proteolytic cascade that ultimately activates transcription of stress response genes in the cytoplasm. Each DegS subunit consists of a protease domain and a PDZ domain. During protein folding stress, DegS is allosterically activated by peptides exposed in misfolded outer membrane porins, which bind to the PDZ domain and stabilize the active protease. It is not known whether allostery is conferred by the PDZ domains or is an intrinsic feature of the trimeric protease domain. Here, we demonstrate that free DegS{sup {Delta}PDZ} equilibrates between active and inactive trimers with the latter species predominating. Substrate binding stabilizes active DegS{sup {Delta}PDZ} in a positively cooperative fashion. Mutations can also stabilize active DegS{sup {Delta}PDZ} and produce an enzyme that displays hyperbolic kinetics and degrades substrate with a maximal velocity within error of that for fully activated, intact DegS. Crystal structures of multiple DegS{sup {Delta}PDZ} variants, in functional and non-functional conformations, support a two-state model in which allosteric switching is mediated by changes in specific elements of tertiary structure in the context of an invariant trimeric base. Overall, our results indicate that protein substrates must bind sufficiently tightly and specifically to the functional conformation of DegS{sup {Delta}PDZ} to assist their own degradation. Thus, substrate binding alone may have regulated the activities of ancestral DegS trimers with subsequent fusion of the protease domain to a PDZ domain, resulting in ligand-mediated regulation.

    The self consistent solution of Schrodinger and Poisson equations is used along with the total charge depletion model and applied with a novel approach of composite AlGaN barrier based HEMT heterostructure. The solution leaded to a completely new analytical model for Fermi energy level vs. 2DEG carrier concentration. This was eventually used to demonstrate a new analytical model for the temperature dependent 2DEG carrier concentration in AlGaN/GaN HEMT.

    A physics-based model based on the variational method for analyzing the two dimensional electron gas (2DEG) characteristics of polar AlGaN/GaN heterojunctions is developed. The 2DEG carrier concentration, the first and second energy subbands, and the position of the Fermi energy level are calculated for various barrier thicknesses, Al mole fractions, background dopant concentrations, and gate voltages for gated AlGaN/GaN heterojunctions. The results are in good agreement with the data reported based on self-consistent method. Whereas the aforementioned report has dealt with specific values of Al mole fraction, barrier thickness, and unintentional doping level, the present work provides a basis for calculating the 2DEG characteristics for the full range of these parameters. Furthermore, according to the proposed model, the applicability of the triangular approximation of the quantum well in AlGaN/GaN heterojunctions is evaluated.

    A numerical study was conducted to determine the effects of turning vanes on the flow characteristics, the pressure drop and the heat transfer distribution in the turn region of two-legged, coolant passages for application to cooling gas turbine blades. The channels consisted of two straight, square-sectioned legs connected by a 180-deg. turn, with an inner radius of 0.15 hydraulic diameters. The ribs were square-sectioned with a height of 0.1 D{sub h}, staggered between leading and trailing side and at an angle of 45-deg. to the flow. Three different configurations of the coolant channel were investigated: with smooth walls, with ribs on two walls and no vane and with ribs on two walls and a 180-deg guide vane in the turn. For all calculations, the Reynolds number at the inlet was Re{sub D} = 100,000, the fluid was water and buoyancy effects were neglected. The simulations were made for rotation numbers, Ro = 0.0 and 0.15. The flow and heat transfer simulations were made with the FLUENT structured code. Based on the results of several previous studies, the differential-Reynolds-Stress turbulence model with wall functions was used. The highest heat transfer in the turn was obtained for both the rotating and non-rotating cases with the ribbed channel without the guide vane. The pressure loss was also higher without the vane. Due to the staggering of the ribs, secondary flow and heat transfer in the turn was higher on the side of the channel where the last rib at the entrance reaches further into the turn. With increasing rotation number, the secondary flow and heat transfer increased on the leading side of the turn. For the configuration with a 180-deg. vane, the mass flux at the outer side of the vane increased with increasing rotation number.

    The effect of isothermal treatment (at 675, 750 and 900 deg. C) on HIC (hydrogen induced cracking) in sour environments containing hydrogen sulphide of a 2205 duplex stainless steel has been investigated. The performance and microstructure of failed material were characterized by optical microscopy, scanning electron microscopy and also X-ray diffraction. Two kinds of Cr-, Mo-enriched intermetallic phases, {sigma} and {chi}, were found to precipitate preferentially at {alpha}/{gamma} interfaces and within {alpha} grains after different times of aging in the temperature range of 650-900 deg. C. After performing tests according to the NACE Standard TM 0284 (1987) the specimens were investigated by using quantitative metallography methods. The volume fraction of {sigma} phase was changed with the time of aging and {sigma} phase developed into coarse particles due to the high diffusibility of solute atoms at high temperatures. The variation of size and shape of {sigma} phase particles was obtained by applying different heat treatment conditions to 2205 steel specimens. The results showed that 2205 duplex stainless steel containing nearly 12 vol.% of {sigma} phase in dispersed conditions was resistant to step cracking in wet environments containing hydrogen sulphide. It was highly possible that a crack would propagate faster along the embrittled {sigma} phase. However, very small cracks were found at austenite-ferrite boundaries where o phase particles were also present.

    The detailed mass transfer distributions around the sharp 180 deg turns in a two-pass, square, smooth channel and in an identical channel with two rib-roughened opposite walls were determined via the napthalene sublimation technique. The top, bottom, inner (divider), and outer walls of the test channel were napthalene-coated surfaces. For the ribbed channel tests, square, transverse, brass ribs were attached to the top and bottom walls of the channel in alignment. The rib height-to-hydraulic diameter ratios (e/D) were 0.063 and 0.094; the rib pitch-to-height ratios (P/e) were 10 and 20. Experiments were conducted for three Reynolds numbers of 15,000, 30,000 and 60,000. Results show that the Sherwood numbers on the top, outer, and inner walls around the turn in the rib-roughened channel are higher than the corresponding Sherwood numbers around the turn in the smooth channel. For both the smooth and the ribbed channels, the Sherwood numbers after the sharp turn are higher than those before the turn. The regional averages of the local Sherwood numbers are correlated and compared with published heat transfer data.

    The influence of uneven wall temperature on the local heat transfer coefficient in a rotating, two-pass, square channel with 60 deg ribs on the leading and trailing walls was investigated for Reynolds number from 2,500 to 25,000 and rotation numbers from 0 to 0.352. Each pass, composed of six isolated copper sections, had a length-to-hydraulic diameter ratio of 12. The mean rotating radius-to-hydraulic diameter ratio was 30. Three thermal boundary condition cases were studied: (A) all four walls at the same temperature, (B) all four walls at the same heat flux, and (C) trailing wall hotter than leading with side walls unheated and insulated. Results indicate that rotating ribbed wall heat transfer coefficients increase by a factor of 2 to 3 over the rotating smooth wall data and at reduced coefficient variation from inlet to exit. As rotation number (or buoyancy parameter) increases, the first pass (outflow) trailing heat transfer coefficients increase and the first pass leading heat transfer coefficients decrease, whereas the reverse is true for the second pass (inflow). The direction of the Coriolis force reverse from the outflow trailing wall to the inflow leading wall. Differences between the first pass leading and trailing heat transfer coefficients increase with rotation number. A similar behavior is seen for the second pass leading and trailing heat transfer coefficients, but the differences are reduced due to buoyancy changing from aiding to opposing the inertia force. The results suggest that uneven wall temperature has a significant impact on the local heat transfer coefficients. The heat transfer coefficients on the first pass leading wall for cases B and C are up to 70--100% higher than that for case A, while the heat transfer coefficients on the second pass trailing wall for cases B and C are up to 20--50% higher.

    We report the discovery of the optical afterglow of the ?-ray burst (GRB) 130702A, identified upon searching 71 deg{sup 2} surrounding the Fermi Gamma-ray Burst Monitor (GBM) localization. Discovered and characterized by the intermediate Palomar Transient Factory, iPTF13bxl is the first afterglow discovered solely based on a GBM localization. Real-time image subtraction, machine learning, human vetting, and rapid response multi-wavelength follow-up enabled us to quickly narrow a list of 27,004 optical transient candidates to a single afterglow-like source. Detection of a new, fading X-ray source by Swift and a radio counterpart by CARMA and the Very Large Array confirmed the association between iPTF13bxl and GRB 130702A. Spectroscopy with the Magellan and Palomar 200 inch telescopes showed the afterglow to be at a redshift of z = 0.145, placing GRB 130702A among the lowest redshift GRBs detected to date. The prompt ?-ray energy release and afterglow luminosity are intermediate between typical cosmological GRBs and nearby sub-luminous events such as GRB 980425 and GRB 060218. The bright afterglow and emerging supernova offer an opportunity for extensive panchromatic follow-up. Our discovery of iPTF13bxl demonstrates the first observational proof-of-principle for ?10 Fermi-iPTF localizations annually. Furthermore, it represents an important step toward overcoming the challenges inherent in uncovering faint optical counterparts to comparably localized gravitational wave events in the Advanced LIGO and Virgo era.

    We present results from the largest contiguous narrow-band survey in the near-infrared. We have used WIRCam/CFHT and the lowOH2 filter (1.187$\\pm$0.005 \\mu m) to survey ~10 deg$^2$ of contiguous extragalactic sky in the SA22 field. A total of ~6000 candidate emission-line galaxies are found. We use deep CFHTLS $ugriz$ and UKIDSS DXS $J$ and $K$ data to obtain robust photometric redshifts. We combine our data with HiZELS (COSMOS+UDS) and explore VVDS, VIPERS, KMOS and obtain our own spectroscopic follow-up with FMOS and MOSFIRE to derive large samples of high-redshift emission-line selected galaxies: 3471 H\\alpha\\ emitters at z=0.8, 1343 [OIII]+H\\beta\\ emitters at z=1.4 and 572 [OII] emitters at z=2.2. We probe co-moving volumes of >10$^6$ Mpc$^3$ and find significant over-densities, including an 8.5\\sigma\\ (spectroscopically confirmed) over-density of H\\alpha\\ emitters at z=0.81.     The Quantum Hall Effect (QHE) and Shubnikov-de Haas effect are investigated experimentally using n type modulation-doped GaAs/GaAlAs quantum wells (QWs) additionally doped in the well with beryllium acceptor atoms. It is presently shown that the localized magneto-acceptor (MA) states which possess discrete energies above the corresponding Landau levels (LLs) lead to two observable effects in magneto-transport: magnetic thaw-down and magnetic boil-off of 2D electrons. Both effects are related to the fact that electrons occupying the localized MA states cannot conduct. Thus in the thaw-down effect the electrons fall down from the MA states to the free Landau states. This leads to a shift of the Hall plateau towards higher magnetic fields as a consequence of an increase of the 2D electron density N{sub S}. In the boil-off effect the electrons are pushed from the free Landau states to the empty MA states under high enough Hall electric field. This process has an avalanche character leading to a dramatic increase of magneto-resistance, consequence of a decrease of N{sub S}.

A collaboration with that company was started in 2009. The system, based on a cluster of Joule-Thomson refrigerators running a special blend of mixed refrigerants is described. Both the advantages and problems of applying this technology to telescope camera refrigeration are discussed. Test results from a prototype refrigerator running in a realistic telescope configuration are reported. Current and future stages of the development program are described. (auth)

    Recent measurements of solar wind turbulence report the presence of intermittent, exponentially distributed angular discontinuities in the magnetic field. In this Letter, we study whether such discontinuities can be produced by magnetohydrodynamic (MHD) turbulence. We detect the discontinuities by measuring the fluctuations of the magnetic field direction, Delta theta, across fixed spatial increments Delta x in direct numerical simulations of MHD turbulence with an imposed uniform guide field B_0. A large region of the probability density function (pdf) for Delta theta is found to follow an exponential decay, proportional to exp(-Delta theta/theta_*), with characteristic angle theta_* ~ (14 deg) (b_rms/B_0)^0.65 for a broad range of guide-field strengths. We find that discontinuities observed in the solar wind can be reproduced by MHD turbulence with reasonable ratios of b_rms/B_0. We also observe an excess of small angular discontinuities when Delta x becomes small, possibly indicating an increasing statistical significance of dissipation-scale structures. The structure of the pdf in this case closely resembles the two-population pdf seen in the solar wind. We thus propose that strong discontinuities are associated with inertial-range MHD turbulence, while weak discontinuities emerge from near-dissipation-range turbulence. In addition, we find that the structure functions of the magnetic field direction exhibit anomalous scaling exponents, which indicates the existence of intermittent structures.

    The fast solar wind is generated by extended perpendicular ion heating in coronal holes, but the kinetic mechanism responsible for this heating has not been determined. One long-standing possibility is the resonant-cyclotron dissipation of ion-cyclotron waves, replenished from a turbulent cascade of interacting counter-propagating Alfven waves. We present results of a kinetic model for proton heating by the quasilinear resonant-cyclotron wave-particle interaction in a coronal hole. The resonant wave spectrum is taken as a power law in wavenumber, uniformly distributed in propagation direction between 0 deg. and 60 deg. with respect to the large-scale radial magnetic field. We obtain the steady-state solution of the kinetic guiding-center equation for the proton distribution in an expanding coronal hole, including the effects of large-scale forces of gravity, charge-separation electric field, Alfven wave ponderomotive force, and mirror force, along with the small-scale scattering from the wave dissipation. We find that plausible wave intensities can yield reasonable flow speeds and temperatures in the heliocentric radial range between 2 and 6 solar radii. We address the claim in earlier work that dissipation of parallel-propagating ion-cyclotron waves cannot provide enough acceleration and show that claim to be incorrect. We find that the combined action of the large-scale forces and the resonant-cyclotron scattering produces proton distribution functions with a characteristic structure: compressed in the sunward half of velocity space with a high-density shell separate from the origin, and relatively expanded in the anti-sunward half of velocity space. We suggest that qualitatively similar proton distributions would result from the kinetic evolution of any sufficiently effective perpendicular heating mechanism operating in an expanding coronal hole.

  15. Segmented Ultralight Pre-Aligned Rotor for Extreme-Scale Wind Turbines

    To alleviate the mass-scaling issues associated with conventional upwind rotors of extreme-scale turbines, a downwind rotor concept is proposed which employs fixed blade curvature based on force alignment at rated conditions. For a given peak stress constraint, the reduction in downwind cantilever loads allows reduced shell and spar thickness, and thus a reduced blade mass as compared with a conventional upwind rotor, especially as rotor sizes approach extreme-scales. To quantify this mass reduction, a Finite Element Analysis was conducted for a 10 MW rated rotor based on the NREL offshore 5 MW baseline wind turbine. The results show that this 'pre-alignment' yields a net downstream deflection of 32 deg, a downward hub-pitch angle of 6 deg, a 20% increase in blade length (to maintain the same radius as the conventional blade), and a net mass savings of about 50% through decreased shell and spar thicknesses. The pre-alignment may also allow a more straightforward and efficient segmentation of the blade since shear stresses near joints are substantially reduced. Segmenting, in turn, can dramatically reduce costs associated with fabrication, transport and assembly for extreme-scale off-shore systems. The pre-aligned geometric curvature can also help alleviate tower wake effects on the blades since blade tips (where shadow effects can be most problematic) are shifted downstream where the tower wake is weaker. In addition, the portion of the tower that is upstream of the blade tips can be faired with an externally-rotating aerodynamic shroud. Furthermore, the downwind rotor can allow a floating off-shore tri-pod platform to reduce tower weight and yaw-control requirements. A simple economic analysis of the segmented ultralight pre-aligned rotor (SUPAR) concept suggests that the overall system cost savings can be as much as 25%, indicating that more detailed (numerical and experimental) investigations are warranted.

    Measurements of small-scale turbulence made over the complex-terrain atmospheric boundary layer during the MATERHORN Program are used to describe the structure of turbulence in katabatic flows. Turbulent and mean meteorological data were continuously measured at multiple levels at four towers deployed along the East lower slope (2-4 deg) of Granite Mountain. The multi-level observations made during a 30-day long MATERHORN-Fall field campaign in September-October 2012 allowed studying of temporal and spatial structure of katabatic flows in detail, and herein we report turbulence and their variations in katabatic winds. Observed vertical profiles show steep gradients near the surface, but in the layer above the slope jet the vertical variability is smaller.     A wind turbine of the type having an airfoil blade (15) mounted on a flexible beam (20) and a pitch governor (55) which selectively, torsionally twists the flexible beam in response to wind turbine speed thereby setting blade pitch, is provided with a limiter (85) which restricts unwanted pitch change at operating speeds due to torsional creep of the flexible beam. The limiter allows twisting of the beam by the governor under excessive wind velocity conditions to orient the blades in stall pitch positions, thereby preventing overspeed operation of the turbine. In the preferred embodiment, the pitch governor comprises a pendulum (65,70) which responds to changing rotor speed by pivotal movement, the limiter comprising a resilient member (90) which engages an end of the pendulum to restrict further movement thereof, and in turn restrict beam creep and unwanted blade pitch misadjustment.

    We present the CFHTLS-Archive-Research Survey (CARS). It is a virtual multi-colour survey based on public archive images from the CFHT-Legacy-Survey. Our main scientific interests in CARS are optical searches for galaxy clusters from low to high redshift and their subsequent study with photometric and weak-gravitational lensing techniques. As a first step of the project we present multi-colour catalogues from 37 sq. degrees of the CFHTLS-Wide component. Our aims are to create astrometrically and photometrically well calibrated co-added images. Second goal are five-band (u*, g', r', i', z') multi-band catalogues with an emphasis on reliable estimates for object colours. These are subsequently used for photometric redshift estimates. The article explains in detail data processing, multi-colour catalogue creation and photometric redshift estimation. Furthermore we apply a novel technique, based on studies of the angular galaxy cross-correlation function, to quantify the reliability of photo-z's. The accuracy of our high-confidence photo-z sample (10-15 galaxies per sq. arcmin) is estimated to $\\sigma_{\\Delta_z/(1+z)}\\approx 0.04-0.05$ up to i'<24 with typically only 1-3% outliers. Interested users can obtain access to our data by request to the authors.

The approach for aggregating low pulse number electrical output from multiple wind turbines relies upon a pad mounted transformer at each wind turbine that performs phase multiplication on the output of each wind turbine. Phase multiplication converts a modified square wave from the wind turbine into a 6 pulse output. Phase shifting of the 6 pulse output from each wind turbine allows the aggregated output of multiple wind turbines to be a 24 pulse approximation of a sine wave. Additional filtering and VAR control is embedded within the wind farm to take advantage of the wind farm's electrical impedence characteristics to further enhance power quality at the point of common coupling.

    As part of its support of the US Department of Energy's (DOE's) Federal Wind Energy Program, the Pacific Northwest Laboratory (PNL) has initiated an effort to work jointly with the wind energy community to characterize wind turbulence in a variety of complex terrains at existing or potential sites of wind turbine installation. Five turbulence characterization systems were assembled and installed at four sites in the Tehachapi Pass in California, and one in the Green Mountains near Manchester, Vermont. Data processing and analyses techniques were developed to allow observational analyses of the turbulent structure; this analysis complements the more traditional statistical and spectral analyses. Preliminary results of the observational analyses, in the rotating framework or a wind turbine blade, show that the turbulence at a site can have two major components: (1) engulfing eddies larger than the rotor, and (2) fluctuating shear due to eddies smaller than the rotor disk. Comparison of the time series depicting these quantities at two sites showed that the turbulence intensity (the commonly used descriptor of turbulence) did not adequately characterize the turbulence at these sites. 9 refs., 10 figs.,

In India the research is carried out on wind energy utilization on big ways.There are still many unsolved challenges in expanding wind power, and there are numerous problems of interest to systems and control researchers. In this paper we study the pitch control mechanism of wind turbine. The pitch control system is one of the most widely used control techniques to regulate the output power of a wind turbine generator. The pitch angle is controlled to keep the generator power at rated power by reducing the angle of the blades. By regulating, the angle of stalling, fast torque changes from the wind will be reutilized. It also describes the design of the pitch controller and discusses the response of the pitch-controlled system to wind velocity variations. The pitch control system is found to have a large output power variation and a large settling time.

    The Illinois Wind Working Group (IWWG) was founded in 2006 with about 15 members. It has grown to over 200 members today representing all aspects of the wind industry across the State of Illinois. In 2008, the IWWG developed a strategic plan to give direction to the group and its activities. The strategic plan identifies ways to address critical market barriers to the further penetration of wind. The key to addressing these market barriers is public education and outreach. Since Illinois has a restructured electricity market, utilities no longer have a strong control over the addition of new capacity within the state. Instead, market acceptance depends on willing landowners to lease land and willing county officials to site wind farms. Many times these groups are uninformed about the benefits of wind energy and unfamiliar with the process. Therefore, many of the project objectives focus on conferences, forum, databases and research that will allow these stakeholders to make well-educated decisions.

    This project evaluated the technical feasibility of converting energy from the wind with a novel “wind fin” approach. This patent-pending technology has three major components: (1) a mast, (2) a vertical, hinged wind structure or fin, and (3) a power takeoff system. The wing structure responds to the wind with an oscillating motion, generating power. The overall project goal was to determine the basic technical feasibility of the wind fin technology. Specific objectives were the following: (1) to determine the wind energy-conversion performance of the wind fin and the degree to which its performance could be enhanced through basic design improvements; (2) to determine how best to design the wind fin system to survive extreme winds; (3) to determine the cost-effectiveness of the best wind fin designs compared to state-of-the-art wind turbines; and (4) to develop conclusions about the overall technical feasibility of the wind fin system. Project work involved extensive computer modeling, wind-tunnel testing with small models, and testing of bench-scale models in a wind tunnel and outdoors in the wind. This project determined that the wind fin approach is technically feasible and likely to be commercially viable. Project results suggest that this new technology has the potential to harvest wind energy at approximately half the system cost of wind turbines in the 10kW range. Overall, the project demonstrated that the wind fin technology has the potential to increase the economic viability of small wind-power generation. In addition, it has the potential to eliminate lethality to birds and bats, overcome public objections to the aesthetics of wind-power machines, and significantly expand wind-power’s contribution to the national energy supply.

    Broad Funding Opportunity Announcement Project: Makani Power is developing an Airborne Wind Turbine (AWT) that eliminates 90% of the mass of a conventional wind turbine and accesses a stronger, more consistent wind at altitudes of near 1,000 feet. At these altitudes, 85% of the country can offer viable wind resources compared to only 15% accessible with current technology. Additionally, the Makani Power wing can be economically deployed in deep offshore waters, opening up a resource which is 4 times greater than the entire U.S. electrical generation capacity. Makani Power has demonstrated the core technology, including autonomous launch, land, and power generation with an 8 meter wingspan, 20 kW prototype. At commercial scale, Makani Power aims to develop a 600 kW, 28 meter wingspan product capable of delivering energy at an unsubsidized cost competitive with coal, the current benchmark for low-cost power.
