    Test Area North (TAN) was a site of the Aircraft Nuclear Propulsion (ANP) Project of the U.S. Air Force and the Atomic Energy Commission. Its Cold War mission was to develop a turbojet bomber propelled by nuclear power. The project was part of an arms race. Test activities took place in five areas at TAN. The Assembly & Maintenance area was a shop and hot cell complex. Nuclear tests ran at the Initial Engine Test area. Low-power test reactors operated at a third cluster. The fourth area was for Administration. A Flight Engine Test facility (hangar) was built to house the anticipated nuclear-powered aircraft. Experiments between 1955-1961 proved that a nuclear reactor could power a jet engine, but President John F. Kennedy canceled the project in March 1961. ANP facilities were adapted for new reactor projects, the most important of which were Loss of Fluid Tests (LOFT), part of an international safety program for commercial power reactors. Other projects included NASA's Systems for Nuclear Auxiliary Power and storage of Three Mile Island meltdown debris. National missions for TAN in reactor research and safety research have expired; demolition of historic TAN buildings is underway.

    This research centered on finding and perfecting methods of collection and disposal of refuse in Ibadan North Local Government Areas. The methodology used included questionnaire administration, personal interviews, field reconnaissance, and biochemical tests of water samples, all aimed at providing useful data for the design of effective methods of collecting and disposing refuse. The local government area was divided into three classes based on resident income: a high-income area (Bodija Avenue, etc.), a medium-income area (Sanngo, Oluyole, etc.), and a low-income area (Beere, Adeoyo, etc.). The research outcomes revealed that the waste generation rate for the local government ranged from 0.2 to 0.33 kg/cap/day and waste density ranged from 172.41 to 217.61 kg/m{sup 3}. Water analyses showed that the chloride, manganese, lead, and cadmium levels in water from low-income areas were above the WHO standard. The refuse generated in high and medium-income areas was collected and transported to the disposal site properly while only 54.5% of wastes were handled properly in low-income areas. Also, in order to make low-income areas free from wastes daily, an additional 15 metal skips and 9 refuse vehicles would be needed.

    We study a mechanism for reliable switching in biomolecular signal-transduction cascades. Steady bistable states are created by system-size cooperative effects in populations of proteins, in spite of the fact that the phosphorylation-state transitions of any molecule, by means of which the switch is implemented, are highly stochastic. The emergence of switching is a nonequilibrium phase transition in an energetically driven, dissipative system described by a master equation. We use operator and functional integral methods from reaction-diffusion theory to solve for the phase structure, noise spectrum, and escape trajectories and first-passage times of a class of minimal models of switches, showing how all critical properties for switch behavior can be computed within a unified framework.

    A novel charged-particle sensitive, pixel based detector array is described and its usage is demonstrated for a variety of applications, from detection of elemental particles (electrons) to hyper-thermal large biomolecular positive and negative ions including keV light atomic and molecular ions. The array detector is a modified light-sensitive charged coupled device (CCD). The IonCCDTM was engineered for direct charged particle detection by replacing the semi-conductor part of the CCD pixel by a conductor1. In contrast with the CCD, where the semi-conductive pixel is responsible for electron-hole pair formation upon photon bombardment, the IonCCD uses a capacitor coupled to the conductive electrode for direct charge integration. The detector can be operated from atmospheric pressure to high vacuum since no high voltages are needed. The IonCCD, presented in this work is an array of 2126 active pixels with 21 um pixel width and 3 um pixel gap. The detection area is 1.5x51mm2 where 1.5 mm and 51 mm are pixel and detector array length, respectively. The result is a one-dimensional position-sensitive detector with 24 um spatial resolution and 88 % pixel area ratio (PAR). In this work we demonstrate the capabilities and the performance of the detector. For the first time we show the direct detection of 250 eV electrons providing linearity response and detection efficiency of the IonCCD as function of electron beam current. Using positive ions from and electron impact source (E-I), we demonstrate that the detection efficiency of the IonCCD is virtually independent of particle energy [250 eV, 1250 eV], particle impact angle [45o, 90o] and particle flux. By combining the IonCCD with a double focusing sector field of Mattauch-Herzog geometry (M-H), we demonstrate fast acquisition of mass spectra in direct air sniffing mode. A first step towards fast in vivo breath analysis is presented. Detection of hyper-thermal biomolecular ions produced using an electrospray ionization source (ESI) is presented. The IonCCD was used as beam profiler to characterize the beam shape and intensity of 15 eV protonated and deprotonated biomolecular ions at the exit of an RF only collisional quadrupole. We present simultaneous detection of 140 eV doubly protonated biomolecular ions when the IonCCD is combined with the M-H analyzer. The latter, demonstrates the possibility of simultaneous separation and micro-array deposition of biological material using a miniature sector field.

    The accuracy of biomolecular simulations depends to some degree on the accuracy of the water model used to solvate the biomolecules. Because many biomolecules such as proteins are electrostatically rather inhomogeneous, containing apolar, polar, and charged moieties or side chains, a water model should be able to represent the polarisation response to a local electrostatic field, while being compatible with the force field used to model the biomolecules or protein. The two polarisable water models, COS/G2 and COS/D, that are compatible with the GROMOS biomolecular force fields leave room for improvement. The COS/G2 model has a slightly too large dielectric permittivity and the COS/D model displays a much too slow dynamics. The proposed COS/D2 model has four interaction sites: only one Lennard-Jones interaction site, the oxygen atom, and three permanent charge sites, the two hydrogens, and one massless off-atom site that also serves as charge-on-spring (COS) polarisable site with a damped or sub-linear dependence of the induced dipole on the electric field strength for large values of the latter. These properties make it a cheap and yet realistic water model for biomolecular solvation.

    Electromechanical coupling is ubiquitous in biological systems with examples ranging from simple piezoelectricity in calcified and connective tissues to voltage-gated ion channels, energy storage in mitochondria, and electromechanical activity in cardiac myocytes and outer hair cell stereocilia. Piezoresponse force microscopy (PFM) has originally emerged as a technique to study electromechanical phenomena in ferroelectric materials, and in recent years, has been employed to study a broad range of non-ferroelectric polar materials, including piezoelectric biomaterials. At the same time, the technique has been extended from ambient to liquid imaging on model ferroelectric systems. Here, we present results on local electromechanical probing of several model cellular and biomolecular systems, including insulin and lysozyme amyloid fibrils, breast adenocarcinoma cells, and bacteriorhodopsin in a liquid environment. The specific features of SPM operation in liquid are delineated and bottlenecks on the route towards nanometer-resolution electromechanical imaging of biological systems are identified.

    We evaluate the accuracy of density functional theory quantum calculations of biomolecular subsystems using a simple electrostatic embedding scheme. Our scheme is based on dividing the system of interest into a primary and secondary subsystem. A finite difference discretization of the Kohn-Sham equations is used for the primary subsystem, while its electrostatic environment is modeled with a simple one-electron potential. Force-field atomic partial charges are used to generate smeared Gaussian charge densities and to model the secondary subsystem. We illustrate the utility of this approach with calculations of truncated dipeptide chains. We analyze quantitatively the accuracy of this approach by calculating atomic forces and comparing results with fullQMcalculations. The impact of the choice made in terminating dangling bonds at the frontier of the QM region is also investigated.

    We present a hybrid method based on a combination of quantum/classical molecular dynamics (MD) simulations and a mod el Hamiltonian approach to describe charge transport through bio-molecular wires with variable lengths in presence o f a solvent. The core of our approach consists in a mapping of the bio-molecular electronic structure, as obtained f rom density-functional based tight-binding calculations of molecular structures along MD trajectories, onto a low di mensional model Hamiltonian including the coupling to a dissipative bosonic environment. The latter encodes fluctuat ion effects arising from the solvent and from the molecular conformational dynamics. We apply this approach to the c ase of pG-pC and pA-pT DNA oligomers as paradigmatic cases and show that the DNA conformational fluctuations are essential in determining and supporting charge transport.

    Often gaining insight into the functioning of biomolecular systems requires to follow their dynamics along a microscopic reaction coordinate (RC) on a macroscopic time scale, which is beyond the reach of current all atom molecular dynamics (MD) simulations. A practical approach to this inherently multiscale problem is to model the system as a fictitious overdamped Brownian particle that diffuses along the RC in the presence of an effective potential of mean force (PMF) due to the rest of the system. By employing the recently proposed FR method [I. Kosztin et al., J. of Chem. Phys. 124, 064106 (2006)], which requires only a small number of fast nonequilibrium MD simulations of the system in both forward and time reversed directions along the RC, we reconstruct the PMF: (1) of deca-alanine as a function of its end-to-end distance, and (2) that guides the motion of potassium ions through the gramicidin A channel.     The interactions of bio-molecules constitute the key steps of cellular functions. However, in vivo binding properties differ significantly from their in vitro measurements due to the heterogeneity of cellular environments. Here we introduce a coarse-grained model based on rigid-body representation to study how factors such as cellular crowding and membrane confinement affect molecular binding. The macroscopic parameters such as the equilibrium constant and the kinetic rate constant are calibrated by adjusting the microscopic coefficients used in the numerical simulations. By changing these model parameters that are experimentally approachable, we are able to study the kinetic and thermodynamic properties of molecular binding, as well as the effects caused by specific cellular environments. We investigate the volumetric effects of crowded intracellular space on bio-molecular diffusion and diffusion-limited reactions. Furthermore, the binding constants of membrane proteins are currently difficult to measure. We provide quantitative estimations about how the binding of membrane proteins deviates from soluble proteins under different degrees of membrane confinements. The simulation results provide biological insights to the functions of membrane receptors on cell surfaces. Overall, our studies establish a connection between the details of molecular interactions and the heterogeneity of cellular environments.

    In July 2005 the North American Solar Challenge (NASC) featured university built solar powered cars ran across the United States into Canada. The competition began in Austin, Texas with stops in Weatherford, Texas; Broken Arrow, Oklahoma; Topeka, Kansas; Omaha, Nebraska; Sioux Falls, South Dakota, Fargo, North Dakota; Winnipeg, Manitoba; Brandon, Manitoba; Regina, Saskatchewan; Medicine Hat, Alberta; mainly following U.S. Highway 75 and Canadian Highway 1 to the finish line in Calgary, Alberta, Canada, for a total distance of 2,500 miles. NASC major sponsors include the U.S. Department of Energy (DOE), Natural Resources Canada and DOEs National Renewable Energy Laboratory. The event is designed to inspire young people to pursue careers in science and engineering. NASCs predecessors, the American Solar Challenge and Sunrayce, generally have been held every two years since 1990. With each race, the solar cars travel faster and further with greater reliability. The NASC promotes: -Renewable energy technologies (specifically photovoltaic or solar cells) -Educational excellence in science, engineering and mathematics -Creative integration of technical and scientific expertise across a wide-range of disciplines -Hands-on experience for students and engineers to develop and demonstrate their technical and creative abilities. Safety is the first priority for the NASC. Each team put its car through grueling qualifying and technical inspections. Teams that failed to meet the requirements were not allowed participate. During the race, each team was escorted by lead and chase vehicles sporting rooftop hazard flashers. An official observer accompanied each solar car team to keep it alert to any safety issues.

    Many of the early North Sea platforms are reaching their end-of-field life. Most are still equipped with their original drilling package. In a few cases the package has either been removed or decommissioned. The early installations were designed for much simpler and less demanding wells than the horizontal, extended-reach or designer wells common today. Extended-reach wells now can be drilled realistically from ageing platforms, without incurring massive capital expenditure. This can be achieved using the existing drilling package to the limit of its capabilities and supplementing where necessary with relatively minor upgrades or the use of temporary equipment. Drilling even a few more wells from existing platforms not only prolongs field life, it enables any surplus processing capacity to be made available to develop near-field potential with extended-reach drilling (ERD) or by tying back subsea satellite wells, or for processing third-party fluids. The paper describes well design, surface equipment, mud pumps, shakers and solids control equipment, drill cuttings disposal systems, derrick and hoisting system, top drive and drillstring, downhole equipment, well planning, casing wear, logistics, rig preparations, and ERD vs. subsea tie-backs.

    Thermoacoustic engines, or acoustic heat engines, are energy-conversion devices that achieve simplicity and concomitant reliability by use of acoustic technology. Their efficiency can be a substantial fraction of Carnot's efficiency. In thermoacoustic prime movers, heat flow from a high-temperature source to a low-temperature sink generates acoustic power (which may be converted to electric power using a transducer). In thermoacoustic heat pumps and refrigerators, acoustic power is used to pump heat from a low-temperature source to a high-temperature sink. This review teaches the fundamentals of thermoacoustic engines, by analysis, intuition, and example.

    An engine based on a reciprocating piston engine that extracts work from pressurized working fluid. The engine includes a harmonic oscillator inlet valve capable of oscillating at a resonant frequency for controlling the flow of working fluid into of the engine. In particular, the inlet valve includes an inlet valve head and a spring arranged together as a harmonic oscillator so that the inlet valve head is moveable from an unbiased equilibrium position to a biased closed position occluding an inlet. Upon releasing the inlet valve the inlet valve head undergoes a single oscillation past the equilibrium positio to a maximum open position and returns to a biased return position close to the closed position to choke the flow and produce a pressure drop across the inlet valve causing the inlet valve to close. Protrusions carried either by the inlet valve head or piston head are used to bump open the inlet valve from the closed position and initiate the single oscillation of the inlet valve head, and protrusions carried either by the outlet valve head or piston head are used to close the outlet valve ahead of the bump opening of the inlet valve.

    The rotary engine has a circumferential main chamber and at least one smaller combustion chamber spaced from the main chamber. The rotor includes a plurality of radially-projecting sealing members in spaced relationship thereabout for maintaining a fluid-sealed condition along a single fixed transverse strip area on the interior surface of the main chamber. A single radially-oriented axially-parallel piston vane is also carried by the rotor and moves through the fixed strip area of the main chamber at each revolution of the rotor. Plural passages for intake, compression, expansion, and exhaust are ported into the main chamber at locations proximate to the fixed strip area. Valve means in the passages selectively open and close the same for a cycle of engine operation involving intake, compression, burning, and exhaust.

    This report summarizes die research by an Energy Modeling Forum working group on the evolution of the North American natural gas markets between now and 2010. The group's findings are based partly on the results of a set of economic models of the natural gas industry that were run for four scenarios representing significantly different conditions: two oil price scenarios (upper and lower), a smaller total US resource base (low US resource case), and increased potential gas demand for electric generation (high US demand case). Several issues, such as the direction of regulatory policy and the size of the gas resource base, were analyzed separately without the use of models.

    An engine has at least two piston-cylinder assemblies each comprising a cylinder formed in an engine block with a cylinder head and a piston therein in sliding relationship toward and away from the head, a piston rod operatively connected to the piston and to a crankshaft, motion producing member of shape-memory material, e.g. Nitinol, having a transformation temperature range, secured to the cylinder head and the side of the piston opposite from the connecting rod, the motion producing member having a heat treated high temperature extended shape memory position and a low temperature low energy compressed position, the Nitinol member being of hollow tubular form and having pressure and return hoses connected thereto for supplying and removing cooling fluid into and from the Nitinol member, an electrical heating device connected to the Nitinol member, whereby the Nitinol member is easily compressed with relatively little force from the extended shape memory position to the compressed position when cooling fluid is supplied thereto to reduce the temperature of the Nitinol member to or below the lower limit of the transformation temperature range and the Nitinol member is automatically extended with relatively great force from the compressed position to the shape memory position when heated by the heating device to or above the upper limit of the transformation temperature range.

    The major components of this rotary engine are two equal sized rotary units, the housing containing them along with associated ignition and cooling systems. Each of the rotary units consists of a shaft, gear, two outer compressor wheels, and one center power wheel which has twice the axial thickness as the compressor wheel. All the wheels are cylindrical in shape with a lobe section comprising a 180/sup 0/ arc on the periphery of each wheel which forms an expanding and contracting volumetric chamber by means of leading and trailing lips. The lobes of the first rotary unit are situated 180/sup 0/ opposite the lobes of the second adjacent mating rotary unit, thus lobes can intermesh with its corresponding wheel.

    According to a 2008 report issued by the United States Geological Survey, North Dakota and Montana have an estimated 3.0 to 4.3 billion barrels of undiscovered, technically recoverable oil in an area known as the Bakken Formation. With the size and remoteness of the discovery, the question became 'can a business case be made for increasing refining capacity in North Dakota?' And, if so what is the impact to existing players in the region. To answer the question, a study committee comprised of leaders in the region's petroleum industry were brought together to define the scope of the study, hire a consulting firm and oversee the study. The study committee met frequently to provide input on the findings and modify the course of the study, as needed. The study concluded that the Petroleum Area Defense District II (PADD II) has an oversupply of gasoline. With that in mind, a niche market, naphtha, was identified. Naphtha is used as a diluent used for pipelining the bitumen (heavy crude) from Canada to crude markets. The study predicted there will continue to be an increase in the demand for naphtha through 2030. The study estimated the optimal configuration for the refinery at 34,000 barrels per day (BPD) producing 15,000 BPD of naphtha and a 52 percent refinery charge for jet and diesel yield. The financial modeling assumed the sponsor of a refinery would invest its own capital to pay for construction costs. With this assumption, the internal rate of return is 9.2 percent which is not sufficient to attract traditional investment given the risk factor of the project. With that in mind, those interested in pursuing this niche market will need to identify incentives to improve the rate of return.

    Over the past ten years, methods have been developed to construct discrete nanostructures using nanocrystals and biomolecules. While these frequently consist of gold nanocrystals and DNA, semiconductor nanocrystals as well as antibodies and enzymes have also been used. One example of discrete nanostructures is dimers of gold nanocrystals linked together with complementary DNA. This type of nanostructure is also known as a nanocrystal molecule. Discrete nanostructures of this kind have a number of potential applications, from highly parallel self-assembly of electronics components and rapid read-out of DNA computations to biological imaging and a variety of bioassays. My research focused in three main areas. The first area, the refinement of electrophoresis as a purification and characterization method, included application of agarose gel electrophoresis to the purification of discrete gold nanocrystal/DNA conjugates and nanocrystal molecules, as well as development of a more detailed understanding of the hydrodynamic behavior of these materials in gels. The second area, the development of methods for quantitative analysis of transmission electron microscope data, used computer programs written to find pair correlations as well as higher order correlations. With these programs, it is possible to reliably locate and measure nanocrystal molecules in TEM images. The final area of research explored the use of DNA ligase in the formation of nanocrystal molecules. Synthesis of dimers of gold particles linked with a single strand of DNA possible through the use of DNA ligase opens the possibility for amplification of nanostructures in a manner similar to polymerase chain reaction. These three areas are discussed in the context of the work in the Alivisatos group, as well as the field as a whole.

    Work is reported in these areas: Large-scale & reconfigurable 3D structures of precise nanoparticle assemblies in self-assembled collagen peptide grids; Binary QD-Au NP 3D superlattices assembled with collagen-like peptides and energy transfer between QD and Au NP in 3D peptide frameworks; Catalytic peptides discovered by new hydrogel-based combinatorial phage display approach and their enzyme-mimicking 2D assembly; New autonomous motors of metal-organic frameworks (MOFs) powered by reorganization of self-assembled peptides at interfaces; Biomimetic assembly of proteins into microcapsules on oil-in-water droplets with structural reinforcement via biomolecular recognition-based cross-linking of surface peptides; and Biomimetic fabrication of strong freestanding genetically-engineered collagen peptide films reinforced by quantum dot joints. We gained the broad knowledge about biomimetic material assembly from nanoscale to microscale ranges by coassembling peptides and NPs via biomolecular recognition. We discovered: • Genetically-engineered collagen-like peptides can be self-assembled with Au NPs to generate 3D superlattices in large volumes (> ?m{sup 3}). • The assembly of the 3D peptide-Au NP superstructures is dynamic and the interparticle distance changes with assembly time as the reconfiguration of structure is triggered by pH change; • QDs/NPs can be assembled with the peptide frameworks to generate 3D superlattices and these QDs/NPs can be electronically coupled for the efficient energy transfer; • The controlled assembly of catalytic peptides mimicking the catalytic pocket of enzymes can catalyze chemical reactions with high selectivity; • For the bacteria-mimicking swimmer fabrication, peptide-MOF superlattices can power translational and propellant motions by the reconfiguration of peptide assembly at the MOF-liquid interface.

    The objective of the Cummins ARES program, in partnership with the US Department of Energy (DOE), is to develop advanced natural gas engine technologies that increase engine system efficiency at lower emissions levels while attaining lower cost of ownership. The goals of the project are to demonstrate engine system achieving 50% Brake Thermal Efficiency (BTE) in three phases, 44%, 47% and 50% (starting baseline efficiency at 36% BTE) and 0.1 g/bhp-hr NOx system out emissions (starting baseline NOx emissions at 2 – 4 g/bhp-hr NOx). Primary path towards above goals include high Brake Mean Effective Pressure (BMEP), improved closed cycle efficiency, increased air handling efficiency and optimized engine subsystems. Cummins has successfully demonstrated each of the phases of this program. All targets have been achieved through application of a combined set of advanced base engine technologies and Waste Heat Recovery from Charge Air and Exhaust streams, optimized and validated on the demonstration engine and other large engines. The following architectures were selected for each Phase: Phase 1: Lean Burn Spark Ignited (SI) Key Technologies: High Efficiency Turbocharging, Higher Efficiency Combustion System. In production on the 60/91L engines. Over 500MW of ARES Phase 1 technology has been sold. Phase 2: Lean Burn Technology with Exhaust Waste Heat Recovery (WHR) System Key Technologies: Advanced Ignition System, Combustion Improvement, Integrated Waste Heat Recovery System. Base engine technologies intended for production within 2 to 3 years Phase 3: Lean Burn Technology with Exhaust and Charge Air Waste Heat Recovery System Key Technologies: Lower Friction, New Cylinder Head Designs, Improved Integrated Waste Heat Recovery System. Intended for production within 5 to 6 years Cummins is committed to the launch of next generation of large advanced NG engines based on ARES technology to be commercialized worldwide.
