    The Energy Sciences Network (ESnet) is the primary provider of network connectivity for the US Department of Energy Office of Science, the single largest supporter of basic research in the physical sciences in the United States. In support of the Office of Science programs, ESnet regularly updates and refreshes its understanding of the networking requirements of the instruments, facilities, scientists, and science programs that it serves. This focus has helped ESnet to be a highly successful enabler of scientific discovery for over 20 years. In April 2010 ESnet and the Office of Biological and Environmental Research, of the DOE Office of Science, organized a workshop to characterize the networking requirements of the science programs funded by BER. The requirements identified at the workshop are summarized and described in more detail in the case studies and the Findings section. A number of common themes emerged from the case studies and workshop discussions. One is that BER science, like many other disciplines, is becoming more and more distributed and collaborative in nature. Another common theme is that data set sizes are exploding. Climate Science in particular is on the verge of needing to manage exabytes of data, and Genomics is on the verge of a huge paradigm shift in the number of sites with sequencers and the amount of sequencer data being generated.

    The Environmental Molecular Sciences Laboratory (EMSL), a scientific user facility managed by Pacific Northwest National Laboratory for the U.S. Department of Energy, Office of Biological and Environmental Research (BER), conducted a one-day workshop on August 26, 2014 on the topic of “Multiscale Computation: Needs and Opportunities for BER Science.” Twenty invited participants, from various computational disciplines within the BER program research areas, were charged with the following objectives; Identify BER-relevant models and their potential cross-scale linkages that could be exploited to better connect molecular-scale research to BER research at larger scales and; Identify critical science directions that will motivate EMSL decisions regarding future computational (hardware and software) architectures.

    The Energy Sciences Network (ESnet) is the primary provider of network connectivity for the US Department of Energy Office of Science, the single largest supporter of basic research in the physical sciences in the United States of America. In support of the Office of Science programs, ESnet regularly updates and refreshes its understanding of the networking requirements of the instruments, facilities, scientists, and science programs that it serves. This focus has helped ESnet to be a highly successful enabler of scientific discovery for over 20 years. In July 2007, ESnet and the Biological and Environmental Research (BER) Program Office of the DOE Office of Science organized a workshop to characterize the networking requirements of the science programs funded by the BER Program Office. These included several large programs and facilities, including Atmospheric Radiation Measurement (ARM) Program and the ARM Climate Research Facility (ACRF), Bioinformatics and Life Sciences Programs, Climate Sciences Programs, the Environmental Molecular Sciences Laboratory at PNNL, the Joint Genome Institute (JGI). National Center for Atmospheric Research (NCAR) also participated in the workshop and contributed a section to this report due to the fact that a large distributed data repository for climate data will be established at NERSC, ORNL and NCAR, and this will have an effect on ESnet. Workshop participants were asked to codify their requirements in a 'case study' format, which summarizes the instruments and facilities necessary for the science and the process by which the science is done, with emphasis on the network services needed and the way in which the network is used. Participants were asked to consider three time scales in their case studies--the near term (immediately and up to 12 months in the future), the medium term (3-5 years in the future), and the long term (greater than 5 years in the future).     Our previous studies demonstrated that polychlorinated biphenyl (PCB) quinone induced oxidative DNA damage in HepG2 cells. To promote genomic integrity, DNA damage response (DDR) coordinates cell-cycle transitions, DNA repair and apoptosis. PCB quinone-induced cell cycle arrest and apoptosis have been documented, however, whether PCB quinone insult induce DNA repair signaling is still unknown. In this study, we identified the activation of DDR and corresponding signaling events in HepG2 cells upon the exposure to a synthetic PCB quinone, PCB29-pQ. Our data illustrated that PCB29-pQ induces the phosphorylation of p53, which was mediated by ataxia telangiectasia mutated (ATM) protein kinase. The observed phosphorylated histone H2AX (γ-H2AX) foci and the elevation of 8-hydroxy-2′-deoxyguanosine (8-OHdG) indicated that DDR was stimulated by PCB29-pQ treatment. Additionally, we found PCB29-pQ activates non-homologous end joining (NHEJ), base excision repair (BER) and nucleotide excision repair (NER) signalings. However, these repair pathways are not error-free processes and aberrant repair of DNA damage may cause the potential risk of carcinogenesis and mutagenesis. - Highlights: • Polychlorinated biphenyl quinone induces oxidative DNA damage in HepG2 cells. • The elevation of γ-H2AX and 8-OHdG indicates the activation of DNA damage response. • ATM-p53 signaling acts as the DNA damage sensor and effector. • Polychlorinated biphenyl quinone activates NHEJ, BER and NER signalings.

    The MH Tool software is designed to evaluate existing and new manufactured homes for structural adequacy in high winds. Users define design elements of a manufactured home and then select the hazard(s) for analysis. MH Tool then calculates and reports structural analysis results for the specified design and hazard Method of Solution: Design engineers input information (geometries, materials, etc.) describing the structure of a manufactured home, from which the software automatically creates a mathematical model.more » Windows, doors, and interior walls can be added to the initial design. HUD Code loads (wind, snow loads, interior live loads, etc.) are automatically applied. A finite element analysis is automatically performed using a third party solver to find forces and stresses throughout the structure. The designer may then employ components of strength (and cost) most appropriate for the loads that must be carried at each location, and then re-run the analysis for verification. If forces and stresses are still within tolerable limits (such as the HUD requirements), construction costs would be reduced without sacrificing quality.« less

    The project yielded papers on fluxes (energy, water and carbon dioxide)between each ecosystem and the atmosphere, and explained the temporal dynamics of fluxes based on intrinsic (physiology, canopy leaf area and structure) and extrinsic (atmospheric and edaphic conditions). Comparisons between any two of the ecosystems, and among all three followed, attributing differences in behavior to different patterns of phenology and differential sensitivities to soil and atmospheric humidity. Finally, data from one-to-three of the ecosystems (incorporated into FluxNet data archive) were used in syntheses across AmeriFlux sites and even more broadly across FluxNet sites.

    This article describes construction of the first manufactured home ever produced from structural insulated panels. The home was built in July 2000 by Champion Enterprises at its Silverton, Oregon, plant. The house was completed on the assembly line in 9 days including a 300-mile road test. The paper examines the design and approval process leading to the project, the manufacturing process and its adjustment to SIPs, and the transportation and energy performance of the house after it was built. PNNL coordinated this project and conducted long-term monitoring on the house. The WSU Energy Program conducted building diagnostics testing once the house was occupied. PNNL and WSU involvement was funded by the U.S. DOE Building America Program. The Oregon Office of Energy conducted blower door and duct blaster tests. The completed home was estimated to reduce energy consumption by 50% and to have twice the structural strength required by HUD code for manufactured homes. The demonstration proved that the manufactured home production line could support SIPs production simultaneously with traditional construction and without major modifications, the line work in parallel with SIPs and traditional materials. The project revealed severl possibilities for further improving cost and time savings with SIPs construction, that might translate into increased capacity.

It requires a tailored, easy-to-understand marketing campaign, and sometimes a little flair. This case study highlights the successful marketing approach of Tommy Williams Homes, which devotes resources to advertising, targeted social media outlets and blogs, realtor education seminars, and groundbreaking and open house celebrations. As a result, in one community, 2013 property sales records show that TWH outsells the only other builder in the development at a higher price, with fewer days on the market.
