  16. WE-G-17A-07: Investigation of the Influence of the Electron Return Effect (ERE) On the Dose Distribution in Rectal Cancer Patients On a 1.5T MR-Linac

    SciTech Connect (OSTI)

    Uilkema, S; Heide, U; Nijkamp, J; Sonke, J; Moreau, M

    2014-06-15

Purpose: The purpose of this planning study is to investigate the influence of the ERE on the day-to-day dose distribution in rectal cancer patients, where changes in gas-pockets frequently occur. Methods: Daily CT scans of 5 patients treated neo-adjuvant with 5x5Gy for rectal cancer were used. We optimized two plans on the planning CT (Monaco, 1 mm3 dosegrid), a conventional 7-field 6MV IMRT plan (Dconv) and a plan in the presence of a 1.5T field (Dmrl). We recalculated the plans on all repeat-CT scans and evaluated under/over-dosage of the daily CTVs. Changes of more than 1% were considered significant. In the bowel area, we investigated the relative dose changes due to the ERE, where the contribution of the ERE was separated from other effects such as attenuation. Results: Both plans were comparable and compliant with ICRU 62 for all patients. For 2 fractions in one patient under-dosage in the CTV was significant, due to a disappearing gas-pocket. Here the V95 was 96.82 and 97.36% in in Dmrl compared to 98.85 and 98.66% in Dconv, respectively. For 3 fractions in another patient appearing gas-pockets resulted in significant over-dosage of the CTV. In these fractions the V107 was 1.882.68% in Dmrl compared to 0.331.27% in Dconv. In the bowel area the dose changes attributable to the ERE were approximately 5% in 1cc, at low dose levels. Conclusion: We were able to calculate acceptable treatment plans with and without a magnetic field. The ERE was present in the Dmrl, but the volumetric effect within the CTV was limited. Outside the CTV relative dose differences were similar, but on small volumes at lower, less relevant dose levels. This suggests that there is no clinical relevant ERE on dose distributions in rectal cancer patients on a 1.5T MR-Linac.

More than 80% of the microbial genomes in GenBank are of draft quality (12,553 draft vs. 2,679 finished, as of October, 2013). We examined the microbial DNA sequences available for complete, draft, and Sequence Read Archive genomes in GenBank as well as three other major public databases and assigned quality scores for more than 30,000 prokaryotic genome sequences. Scores were assigned using four categories: the completeness of the assembly, the presence of full-length rRNA genes, tRNA composition and the presence of a set of 120 conserved genes in prokaryotes. Most (~87.6%) of the genomes had quality scores of 0.8 or better and can be safely used for standard comparative genomics analysis, although only 6.1% of the genomes had a perfect quality score. We find that about 2.9% of the genomes had a score below 0.6 and probably have too low a quality to yield reliable analysis. This score corresponds to more than 1000 contigs. Comparison of the codon usage across 15,000 quality genomes found that anti-codons beginning with an A (codons ending with a U ) are almost non-existent, with the exception of one arginine codon (CGU).

Using a fluorescein di-{beta}-d-galactopyranoside (FDG) substrate we show that in live cells of an estrogen-sensitive yeast strain RMY/ER-ERE with human estrogen receptor (ER{alpha}) gene and the lacZ gene which encodes {beta}-galactosidase, the uptake of 17{beta}-estradiol (E2) and the subsequent production of {beta}-galactosidase enzyme occur quite rapidly, with maximal enzyme-catalyzed product formation evident after about 30 min of exposure to E2. This finding which agrees with the well-known rates of enzyme-catalyzed reactions could have implications for shortening the duration of environmental sample screening and monitoring regimes using yeast-based estrogen assays, and the development of biosensors for environmental estrogens to complement quantification methods.

    SciTech Connect (OSTI)

Wang, Na; Goel, Supriya; Gorrissen, Willy J.; Makhmalbaf, Atefe

    2013-06-24

The U.S. Department of Energy (DOE) is developing a national voluntary energy asset score system to help building owners to evaluate the as-built physical characteristics (including building envelope, the mechanical and electrical systems) and overall building energy efficiency, independent of occupancy and operational choices. The energy asset score breaks down building energy use information by simulating building performance under typical operating and occupancy conditions for a given use type. A web-based modeling tool, the energy asset score tool facilitates the implementation of the asset score system. The tool consists of a simplified user interface built on a centralized simulation engine (EnergyPlus). It is intended to reduce both the implementation cost for the users and increase modeling standardization compared with an approach that requires users to build their own energy models. A pilot project with forty-two buildings (consisting mostly offices and schools) was conducted in 2012. This paper reports the findings. Participants were asked to collect a minimum set of building data and enter it into the asset score tool. Participants also provided their utility bills, existing ENERGY STAR scores, and previous energy audit/modeling results if available. The results from the asset score tool were compared with the building energy use data provided by the pilot participants. Three comparisons were performed. First, the actual building energy use, either from the utility bills or via ENERGY STAR Portfolio Manager, was compared with the modeled energy use. It was intended to examine how well the energy asset score represents a buildings system efficiencies, and how well it is correlated to a buildings actual energy consumption. Second, calibrated building energy models (where they exist) were used to examine any discrepancies between the asset score model and the pilot participant buildings [known] energy use pattern. This comparison examined the end use breakdowns and more detailed time series data. Third, ASHRAE 90.1 prototype buildings were also used as an industry standard modeling approach to test the accuracy level of the asset score tool. Our analysis showed that the asset score tool, which uses simplified building simulation, could provide results comparable to a more detailed energy model. The buildings as-built efficiency can be reflected in the energy asset score. An analysis between the modeled energy use through the asset score tool and the actual energy use from the utility bills can further inform building owners about the effectiveness of their buildings operation and maintenance.

Purpose: To evaluate the prognostic utility of the cell cycle progression (CCP) score, a RNA signature based on the average expression level of 31 CCP genes, for predicting biochemical recurrence (BCR) in men with prostate cancer treated with external beam radiation therapy (EBRT) as their primary curative therapy. Methods and Materials: The CCP score was derived retrospectively from diagnostic biopsy specimens of men diagnosed with prostate cancer from 1991 to 2006 (n=141). All patients were treated with definitive EBRT; approximately half of the cohort was African American. Outcome was time from EBRT to BCR using the Phoenix definition. Median follow-up for patients without BCR was 4.8 years. Association with outcome was evaluated by Cox proportional hazards survival analysis and likelihood ratio tests. Results: Of 141 patients, 19 (13%) had BCR. The median CCP score for patient samples was 0.12. In univariable analysis, CCP score significantly predicted BCR (P=.0017). The hazard ratio for BCR was 2.55 for 1-unit increase in CCP score (equivalent to a doubling of gene expression). In a multivariable analysis that included Gleason score, prostate-specific antigen, percent positive cores, and androgen deprivation therapy, the hazard ratio for CCP changed only marginally and remained significant (P=.034), indicating that CCP provides prognostic information that is not provided by standard clinical parameters. With 10-year censoring, the CCP score was associated with prostate cancer-specific mortality (P=.013). There was no evidence for interaction between CCP and any clinical variable, including ethnicity. Conclusions: Among men treated with EBRT, the CCP score significantly predicted outcome and provided greater prognostic information than was available with clinical parameters. If validated in a larger cohort, CCP score could identify high-risk men undergoing EBRT who may need more aggressive therapy.

    2014-12-18

Background: Top-down mass spectrometry plays an important role in intact protein identification and characterization. Top-down mass spectra are more complex than bottom-up mass spectra because they often contain many isotopomer envelopes from highly charged ions, which may overlap with one another. As a result, spectral deconvolution, which converts a complex top-down mass spectrum into a monoisotopic mass list, is a key step in top-down spectral interpretation. Results: In this paper, we propose a new scoring function, L-score, for evaluating isotopomer envelopes. By combining L-score with MS-Deconv, a new software tool, MS-Deconv+, was developed for top-down spectral deconvolution. Experimental results showedmore » that MS-Deconv+ outperformed existing software tools in top-down spectral deconvolution. Conclusions: L-score shows high discriminative ability in identification of isotopomer envelopes. Using L-score, MS-Deconv+ reports many correct monoisotopic masses missed by other software tools, which are valuable for proteoform identification and characterization.« less

    1998-07-01

A technical maturity assessment method was performed by systems engineers in collaboration with an advisory panel composed of team members from different Department of Energy sites and from different engineering and science disciplines. Various stabilization technologies were assessed annually as to their relative maturity and availability for use in stabilizing nuclear materials. After three years of assessments, several of the technologies are now components of operational systems. A regression analysis of the historical assessments was performed, and it was concluded that the numerical technical maturity score produced by a team of experts can provide a powerful predictor of the time remaining until the operational application of technologies.

    2014-12-04

Colorectal cancer is the third and the second most common cancer worldwide in men and women respectively, and the second in Malaysia for both genders. Surgery, chemotherapy and radiotherapy are among the options available for treatment of patients with colorectal cancer. In clinical trials, the main purpose is often to compare efficacy between experimental and control treatments. Treatment comparisons often involve several responses or endpoints, and this situation complicates the analysis. In the case of colorectal cancer, sets of responses concerned with survival times include: times from tumor removal until the first, the second and the third tumor recurrences, and time to death. For a patient, the time to recurrence is correlated to the overall survival. In this study, global score test methodology is used in combining the univariate score statistics for comparing treatments with respect to each survival endpoint into a single statistic. The data of tumor recurrence and overall survival of colorectal cancer patients are taken from a Malaysian hospital. The results are found to be similar to those computed using the established Wei, Lin and Weissfeld method. Key factors such as ethnic, gender, age and stage at diagnose are also reported.

It’s becoming easier to demonstrate a home’s energy efficiency performance. This month, through its Better Buildings Colorado program, Colorado became the second state to offer the Home Energy Score statewide, which provides a home’s efficiency score and recommendations for efficiency improvements. Similar to a vehicle’s miles-per-gallon rating, the Home Energy Score is a free tool from the Energy Department that provides an energy efficiency score for a home and recommendations for energy efficiency improvements. The program is uniquely targeting real estate professionals to implement the benefits that Home Energy Score can bring to home buyers and home sellers.

  7. Preradiotherapy Calcium Scores of the Coronary Arteries in a Cohort of Women With Early-Stage Breast Cancer: A Comparison With a Cohort of Healthy Women

    SciTech Connect (OSTI)

    2012-07-01

Purpose: Breast cancer radiotherapy has been associated with an increased risk of cardiac toxicity. However, no data are available on the probability of developing coronary artery disease (CAD) in breast cancer patients when compared with healthy women. Therefore, baseline coronary artery calcium (CAC) scores, as an accepted tool to predict CAD, were determined and compared with the CAC scores of a healthy, asymptomatic cohort, the Multi-Ethnic Study of Atherosclerosis (MESA) cohort. Methods and Materials: Eighty consecutive patients with ductal carcinoma in situ or infiltrative breast cancer referred for radiotherapy after breast-conserving surgery were included in our study. Their cardiovascular risk profile was registered, and a 64 multislice CT scan was performed. The CAC scores of an unselected (Caucasian only) Radiotherapy Centre West (RCWEST) cohort, as well as of those of a selected (comorbidity and race adjusted) RCWEST cohort, were determined. The scores of both cohorts were compared with those of the female (Caucasian only) MESA cohort. Results: For the unselected RCWEST cohort (n = 62) we found significant (p < .01) higher scores for women in the 55-64 age category compared with those of the MESA cohort. In the selected cohort (n = 55) the CAC scores of the women in the age category 55-64 were significantly (p = .02) higher compared with the MESA cohort. No significant differences were noted in the other age categories. Conclusion: Both cohorts revealed that CAC scores in the 55-64 age category were significantly higher than the CAC scores in the asymptomatic (female) MESA population. These data suggest that breast cancer patients bear a higher risk of developing coronary heart disease before the start of radiotherapy. Therefore, measures to decrease cardiac dose further in breast cancer radiotherapy are even more important.

  11. Commercial Building Energy Asset Score System: Program Overview and Technical Protocol (Version 1.0)

    SciTech Connect (OSTI)

    Wang, Na; Gorrissen, Willy J.

    2013-01-11

The U.S. Department of Energy (DOE) is developing a national voluntary energy asset score system that includes an energy asset score tool to help building owners evaluate their buildings with respect to the score system. The goal of the energy asset score system is to facilitate cost-effective investment in energy efficiency improvements of commercial buildings. The system will allow building owners and managers to compare their building infrastructure against peers and track building upgrade progress over time. The system can also help other building stakeholders (e.g., building operators, tenants, financiers, and appraisers) understand the relative efficiency of different buildings in a way that is independent from their operations and occupancy. This report outlines the technical protocol used to generate the energy asset score, explains the scoring methodology, and provides additional details regarding the energy asset score tool. This report also describes alternative methods that were considered prior to developing the current approach. Finally, this report describes a few features of the program where alternative approaches are still under evaluation.

    Broader source: Energy.gov [DOE]

Connecticut recently launched a statewide residential energy labeling program that will make energy efficiency labels ubiquitous across the state. Using the Energy Department’s Home Energy Score, EnergizeCT’s Home Energy Solutions program will provide an energy efficiency score and recommend efficiency improvements to residents across the state. Similar to a vehicle’s miles-per-gallon rating, the Home Energy Score helps homeowners and homebuyers determine a home’s expected energy use. It also provides recommendations for improving energy efficiency.

  17. Methodology for Scaling Fusion Power Plant Availability

    SciTech Connect (OSTI)

    Lester M. Waganer

    2011-01-04

Normally in the U.S. fusion power plant conceptual design studies, the development of the plant availability and the plant capital and operating costs makes the implicit assumption that the plant is a 10th of a kind fusion power plant. This is in keeping with the DOE guidelines published in the 1970s, the PNL report1, "Fusion Reactor Design Studies - Standard Accounts for Cost Estimates. This assumption specifically defines the level of the industry and technology maturity and eliminates the need to define the necessary research and development efforts and costs to construct a one of a kind or the first of a kind power plant. It also assumes all the "teething" problems have been solved and the plant can operate in the manner intended. The plant availability analysis assumes all maintenance actions have been refined and optimized by the operation of the prior nine or so plants. The actions are defined to be as quick and efficient as possible. This study will present a methodology to enable estimation of the availability of the one of a kind (one OAK) plant or first of a kind (1st OAK) plant. To clarify, one of the OAK facilities might be the pilot plant or the demo plant that is prototypical of the next generation power plant, but it is not a full-scale fusion power plant with all fully validated "mature" subsystems. The first OAK facility is truly the first commercial plant of a common design that represents the next generation plant design. However, its subsystems, maintenance equipment and procedures will continue to be refined to achieve the goals for the 10th OAK power plant.

ATF2 is an accelerator test facility modeled after the final focus beamline envisioned for the ILC. By the end of 2008, KEK plans to commission the ATF2 [1]. SLAC and OCEM collaborated on the design of 38 power systems for beamline magnets. The systems range in output power from 1.5 kW to 6 kW. Since high availability is essential for the success of the ILC, Collaborators employed an N+1 modular approach, allowing for redundancy and the use of a single power module rating. This approach increases the availability of the power systems. Common power modules reduces inventory and eases maintenance. Current stability requirements are as tight as 10 ppm. A novel, SLAC designed 20-bit Ethernet Power Supply Controller provides the required precision current regulation. In this paper, Collaborators present the power system design, the expected reliability, fault immunity features, and the methods for satisfying the control and monitoring challenges. Presented are test results and the status of the power systems.

    2008-09-25

This paper reports the design and test results on novel topology, high-efficiency, and low operating temperature, 1,320-watt power modules for high availability power supplies. The modules permit parallel operation for N+1 redundancy with hot swap capability. An embedded DSP provides intelligent start-up and shutdown, output regulation, general control and fault detection. PWM modules in the DSP drive the FET switches at 20 to 100 kHz. The DSP also ensures current sharing between modules, synchronized switching, and soft start up for hot swapping. The module voltage and current have dedicated ADCs (>200 kS/sec) to provide pulse-by-pulse output control. A Dual CAN bus interface provides for low cost redundant control paths. Over-rated module components provide high reliability and high efficiency at full load. Low on-resistance FETs replace conventional diodes in the buck regulator. Saturable inductors limit the FET reverse diode current during switching. The modules operate in a two-quadrant mode, allowing bipolar output from complimentary module groups. Controllable, low resistance FETs at the input and output provide fault isolation and allow module hot swapping.

This dataset consists of measurements of plant available nutrients made using Plant Root Simulator probes (Western Ag Innovations Inc.) during 2012 and 2013. In 2012, Ca, Mg, K, P, Fe, Mn, Cu, Zn, B, S, Pb, Al, Cd, NO3-N and NH4-N were measured during spring, summer and winter in the centers, edges and troughs of four polygons in each of four areas of contrasting moisture regime and polygon type. In 2013, probes were installed in centers, edges and troughs of four polygons in each of two areas (high-centered and low-centered polygons) at two-week intervals and at 3 soil depths to capture fine-scale season dynamics of NO3-N and NH4-N. PRS probes are ion exchange resin membranes held in plastic supports that are inserted into soil to measure ion supply in situ. The anion and cation exchange with the membrane is intended to mimic plant uptake and thus provide a relevant measure of soil nutrient bioavailability. Measurements are made per area of probe membrane and cannot be converted to concentrations or related to soil volume.

  16. Optimizing the availability of a buffered industrial process

    DOE Patents [OSTI]

Martz, Jr., Harry F.; Hamada, Michael S.; Koehler, Arthur J.; Berg, Eric C.

    2004-08-24

A computer-implemented process determines optimum configuration parameters for a buffered industrial process. A population size is initialized by randomly selecting a first set of design and operation values associated with subsystems and buffers of the buffered industrial process to form a set of operating parameters for each member of the population. An availability discrete event simulation (ADES) is performed on each member of the population to determine the product-based availability of each member. A new population is formed having members with a second set of design and operation values related to the first set of design and operation values through a genetic algorithm and the product-based availability determined by the ADES. Subsequent population members are then determined by iterating the genetic algorithm with product-based availability determined by ADES to form improved design and operation values from which the configuration parameters are selected for the buffered industrial process.
