    Cellular oxygen tension is sensed by a family of prolyl hydroxylases (PHD1-3) that regulate the degradation of hypoxia-inducible factors (HIF-1{alpha} and -2{alpha}). The PHD2 isoform is considered as the main downregulator of HIF in normoxia. Our previous results have shown that nuclear translocation of PHD2 associates with poorly differentiated tumor phenotype implying that nuclear PHD2 expression is advantageous for tumor growth. Here we show that a pool of PHD2 is shuttled between the nucleus and the cytoplasm. In line with this, accumulation of wild type PHD2 in the nucleus was detected in human colon adenocarcinomas and in cultured carcinoma cells. The PHD2 isoforms showing high nuclear expression increased anchorage-independent carcinoma cell growth. However, retention of PHD2 in the cytoplasm inhibited the anchorage-independent cell growth. A region that inhibits the nuclear localization of PHD2 was identified and the deletion of the region promoted anchorage-independent growth of carcinoma cells. Finally, the cytoplasmic PHD2, as compared with the nuclear PHD2, less efficiently downregulated HIF expression. Forced HIF-1{alpha} or -2{alpha} expression decreased and attenuation of HIF expression increased the anchorage-independent cell growth. However, hydroxylase-inactivating mutations in PHD2 had no effect on cell growth. The data imply that nuclear PHD2 localization promotes malignant cancer phenotype.

  3. Revamping AK-Ashland gas cleaning system

    AK Steel`s (formerly Armco) BOF shop was using a static precipitator for the primary collection. The system was designed for full combustion in the gas collecting hoods. No secondary dust collection was in place. A detailed study on alternative solutions led to a completely different system in 1990, and an order was awarded to Mannesmann Demag Corp. (MDC) in Dec. 1990. The new gas collection system is using suppressed combustion with the capability to collect Co at a later stage. The gas cleaning uses the Mannesmann Demag Baumco scrubber with a venturi throat for gas flow control. All auxiliary components, water treatment plant, electric substations and sludge handling were designed and supplied by MDC. The secondary dust collection covers the hot metal and scrap charging into the BOF`s, reladling, desulfurization and deslagging by a pulse jet baghouse. All emission limits set by the EPA and guaranteed by MDC have been met by the systems installed.

    The Anchorage, Alaska Roundtable on Tribal Energy Policy convened at 10:00 a.m., Thursday April 15th, at the downtown Anchorage Hilton. The meeting was held by the Department of Energy (DOE) Office of Indian Energy Policy and Programs (Office of Indian Energy). Tracey LeBeau, Director of the Office of Indian Energy, and Pilar Thomas, Deputy Director-Policy of the Office of Indian Energy, represented DOE. Approximately twenty-seven people attended the meeting, including representatives of three native Alaskan villages, four Alaskan tribal corporations representing more than 40 tribal governments, as well as representatives from tribal associations and conferences. Interested state, federal, and non-profit representatives also were present. A full list of attendees is at the end of this summary. The meeting was facilitated by the Udall Foundation’s U.S. Institute for Environmental Conflict Resolution (U.S. Institute).

  7. The anchorage function of CipA (CelL), a scaffolding protein of the Clostridium thermocellum cellulosome

    Enzymatic cellulose degradation is a heterogeneous reaction requiring binding of soluble cellulase molecules to the solid substrate. Based on our studies of the cellulase complex of Clostridium thermocellum (the cellulosome), we have previously proposed that such binding can be brought about by a special {open_quotes}anchorage subunit.{close_quotes} In this {open_quotes}anchor-enzyme{close_quotes} model, CipA (a major subunit of the cellulosome) enhances the activity of CelS (the most abundant catalytic subunit of the cellulosome) by anchoring it to the cellulose surface. We have subsequently reported that CelS contains a conserved duplicated sequence at its C terminus and the CipA contains nine repeated sequences with a cellulose binding domain (CBD) in between the second and third repeats. In this work, we reexamined the anchor-enzyme mechanism by using recombinant CelS (rCelS) and various CipA domains, CBD, R3 (the repeat next to CBD), and CBD/R3, expressed in Escherichia coli. As analyzed by non-denaturing gel electrophoresis, rCelS, through its conserved duplicated sequence, formed a stable complex with R3 or CBD/R3 but not with CBD. Although R3 or CBD alone did not affect the binding of rCelS to cellulose, such binding was dependent on CBD/R3, indicating the anchorage role of CBD/R3. Such anchorage apparently increased the rCelS activity toward crystalline cellulose. These results substantiate the proposed anchor-enzyme model and the expected roles of individual CipA domains and the conserved duplicated sequence of CelS.

    In 2008, Air Products and Chemicals, Inc. (Air Products) began development of a project to beneficially utilize waste blast furnace topgas generated in the course of the iron-making process at AK Steel Corporations Middletown, Ohio works. In early 2010, Air Products was awarded DOE Assistance Agreement DE-EE002736 to further develop and build the combined-cycle power generation facility. In June 2012, Air Products and AK Steel Corporation terminated work when it was determined that the project would not be economically viable at that time nor in the foreseeable future. The project would have achieved the FOA-0000044 Statement of Project Objectives by demonstrating, at a commercial scale, the technology to capture, treat, and convert blast furnace topgas into electric power and thermal energy.

    This document is a compilation of the characterization data for the TRISO-coated surrogate particle batch designated ZrO2-500-AK2 that was produced at Oak Ridge National Laboratory (ORNL) as part of the Advanced Gas Reactor Fuel Development and Qualification (AGR) program. The ZrO2-500-AK2 material contains nominally 500 {micro}m kernels of yttria-stabilized zirconia (YSZ) coated with all TRISO layers (buffer, inner pyrocarbon, silicon carbide, and outer pyrocarbon). The ZrO2-500-AK2 material was created for: (1) irradiation testing in the High Flux Isotope Reactor (HFIR) and (2) limited dissemination to laboratories as deemed appropriate to the AGR program. This material was created midway into a TRISO fuel development program to accommodate a sudden opportunity to perform irradiation testing on surrogate material. While the layer deposition processes were chosen based on the best technical understanding at the time, technical progress at ORNL has led to an evolution in the perceived optimal deposition conditions since the createion of ZrO2-500-AK2. Thus, ZrO2-500-AK2 contains a reasonable TRISO microstructure, but does differ significanly from currently produced TRISO surrogates and fuel at ORNL.     The reaction of Ca with a CaCrO/sub 4/-(LiCl-KCl eutectic) solution at temperatures of 400/sup 0/C to 500/sup 0/C was studied to better understand the nature of the chemical reactions and electrochemical processes that occur in the Ca/CaCrO/sub 4/ thermal battery at the anode during activation and discharge. Limited tests also were conducted with a CaCrO/sub 4/-(CaCl/sub 2/-NaCl-KCl eutectic) solution at 550/sup 0/C. Ca/CaCrO/sub 4/ and CaLi/sub 2//CaCrO/sub 4/ single cells were tested to observe the relative performance differences of Ca and CaLi/sub 2/ anodes. The discharged cells were analyzed by optical microscopy, electron microprobe, Auger electron spectroscopy, and secondary-ion mass spectroscopy. These analytical data were used in conjunction with the results of chemical-reaction experiments to propose a discharge mechanism for the Ca/CaCrO/sub 4/ thermal battery, consistent with experimental observations.

    We analyze new, high quality multicolor light curves of four overcontact binaries: AK Her, HI Dra, V1128 Tau, and V2612 Oph, and determine their orbital and physical parameters using the modeling program of G. Djurasevic and recently published results of radial velocity studies. The achieved precision in absolute masses is between 10% and 20%, and the precision in absolute radii is between 5% and 10%. All four systems are W UMa-type binaries with bright or dark spots indicative of mass and energy transfer or surface activity. We estimate the distances and the ages of the systems using the luminosities computed through our analysis, and perform an O – C study for V1128 Tau, which reveals a complex period variation that can be interpreted in terms of mass loss/exchange and either the presence of the third body, or the magnetic activity on one of the components. We conclude that further observations of these systems are needed to deepen our understanding of their nature and variability.

    In order to study the crystal structure of {beta}-bar Ca{sub 2}SiO{sub 4}, time-of-flight neutron powder diffraction experiments were carried out at temperatures between room temperature (RT) and 600 deg. C. Rietveld refinement at RT has shown that {beta}-bar Ca{sub 2}SiO{sub 4} is monoclinic based on P2{sub 1}/n symmetry and two different types of Ca sites, Ca(1) and Ca(2). All interatomic distances within 3A were calculated, with the valences of Ca(1) with seven Ca-O bonds and Ca(2) with eight were estimated to be 1.87+ and 2+ by the Zachariasen-Brown-Altermatt formula (bond valence sum). Applying charge neutrality the two charge states of Ca in {beta}-bar Ca{sub 2}SiO{sub 4} are [Ca(1)SiO{sub 4}]{sup 2-} and Ca(2){sup 2+}, respectively. Furthermore, the [Ca(1)SiO{sub 4}]{sup 2-} unit has the shortest Ca-O distance, and its length kept constant at 2.23A at all temperatures. In the short-range structure analysis at RT, the shortest Ca-O bond was also observed in a radial distribution function. These results imply that the [Ca(1)SiO{sub 4}]{sup 2-} unit has covalency on the shortest Ca-O in addition to Si-O.

    Using thermal analysis techniques, we investigated the corrosion process resulting from the reaction of iron, nickel, and stainless steel (used as current collectors in Ca/CaCrO/sub 4/ thermal batteries) with CaCrO/sub 4/ dissolved in LiCl-KCl eutectic. The reaction product for iron was synthesized in bulk external to the battery and was characterized by chemical analysis, X-ray diffraction, Moessbauer spectroscopy, X-ray photoelectron spectroscopy, static magnetization, and electrical conductivity. This characterization provides a better understanding of the cathodic corrosion processes that occur in the Ca/CaCrO/sub 4/ thermal battery, and how the properties of the reaction layer at the catholyte-current collector interface influence battery performance.

    As part of our effort to develop a semicontinuous PuO/sub 2/ reduction process, we are investigating promising materials for containing a 900/sup 0/C molten CaCl/sub 2/ . CaO chlorination reaction. We want the material to contain this reaction and to be reusable. We tested candidate materials in a simulated salt (no plutonium) using anhydrous HCl as the chlorinating agent.
