  18. Clock signal requirement for high-frequency, high dynamic range acquisition systems

    SciTech Connect (OSTI)

Viscor, Ivo; Halamek, Josef; Villa, Marco

    2005-11-15

Analog-to-digital converters (ADC's) are increasingly replacing mixers in frequency conversion schemes. To achieve superior performances, in terms of bandwidth and dynamic range, a nearly ideal ADC clock is needed, with a spectral purity higher than the reference signal of the classical mixing scheme. These requirements of spectral purity for the ADC clock are discussed by analyzing in detail the nonuniform sampling process and by characterizing an actual acquisition system. The effect of clock phase imperfections on the output is proportional to the input frequency over sampling frequency ratio. Moreover, at the output we may have a multiple folding of the phase jitter spectrum. These effects are illustrated by three sets of measurements performed using our system: transfer of spurious clock components, aliasing of these components, and transfer of clock phase noise.

    2013-07-21

This paper presents new methodologies developed at Pacific Northwest National Laboratory (PNNL) to estimate regulation capacity requirements in the California ISO control area. Two approaches have been developed: (1) an approach based on statistical analysis of actual historical area control error (ACE) and regulation data, and (2) an approach based on balancing authority ACE limit control performance standard. The approaches predict regulation reserve requirements on a day-ahead basis including upward and downward requirements, for each operating hour of a day. California ISO data has been used to test the performance of the proposed algorithms. Results show that software tool allows saving up to 30% on the regulation procurements cost .

We describe the versatile features of the attosecond beamline recently installed at CEA-Saclay on the PLFA kHz laser. It combines a fine and very complete set of diagnostics enabling high harmonic spectroscopy (HHS) through the advanced characterization of the amplitude, phase, and polarization of the harmonic emission. It also allows a variety of photo-ionization experiments using magnetic bottle and COLTRIMS (COLd Target Recoil Ion Momentum Microscopy) electron spectrometers that may be used simultaneously, thanks to a two-foci configuration. Using both passive and active stabilization, special care was paid to the long term stability of the system to allow, using both experimental approaches, time resolved studies with attosecond precision, typically over several hours of acquisition times. As an illustration, applications to multi-orbital HHS and electron-ion coincidence time resolved spectroscopy are presented.

  11. Extracellular Matrix-Regulated Gene Expression RequiresCooperation of SWI/SNF and Transcription Factors

    SciTech Connect (OSTI)

Xu, Ren; Spencer, Virginia A.; Bissell, Mina J.

    2006-05-25

Extracellular cues play crucial roles in the transcriptional regulation of tissue-specific genes, but whether and how these signals lead to chromatin remodeling is not understood and subject to debate. Using chromatin immunoprecipitation (ChIP) assays and mammary-specific genes as models, we show here that extracellular matrix (ECM) molecules and prolactin cooperate to induce histone acetylation and binding of transcription factors and the SWI/SNF complex to the {beta}- and ?-casein promoters. Introduction of a dominant negative Brg1, an ATPase subunit of SWI/SNF complex, significantly reduced both {beta}- and ?-casein expression, suggesting that SWI/SNF-dependent chromatin remodeling is required for transcription of mammary-specific genes. ChIP analyses demonstrated that the ATPase activity of SWI/SNF is necessary for recruitment of RNA transcriptional machinery, but not for binding of transcription factors or for histone acetylation. Coimmunoprecipitation analyses showed that the SWI/SNF complex is associated with STAT5, C/EBP{beta}, and glucocorticoid receptor (GR). Thus, ECM- and prolactin-regulated transcription of the mammary-specific casein genes requires the concerted action of chromatin remodeling enzymes and transcription factors.

  13. Identification of genes that regulate phosphate acquisition and plant performance during arbuscular my corrhizal symbiosis in medicago truncatula and brachypodium distachyon

    SciTech Connect (OSTI)

Harrison, Maria J; Hudson, Matthew E

    2015-11-24

Most vascular flowering plants have the ability to form symbiotic associations with arbuscular mycorrhizal (AM) fungi. The symbiosis develops in the roots and can have a profound effect on plant productivity, largely through improvements in plant mineral nutrition. Within the root cortical cells, the plant and fungus create novel interfaces specialized for nutrient transfer, while the fungus also develops a network of hyphae in the rhizosphere. Through this hyphal network, the fungus acquires and delivers phosphate and nitrogen to the root. In return, the plant provides the fungus with carbon. In addition, to enhancing plant mineral nutrition, the AM symbiosis has an important role in the carbon cycle, and positive effects on soil health. Here we identified and characterized plant genes involved in the regulation and functioning of the AM symbiosis in Medicago truncatula and Brachypodium distachyon. This included the identification and and characterization of a M. truncatula transcription factors that are required for symbiosis. Additionally, we investigated the molecular basis of functional diversity among AM symbioses in B. distachyon and analysed the transcriptome of Brachypodium distachyon during symbiosis.

  14. Integrating environmental considerations in the defense acquisition process

    SciTech Connect (OSTI)

    Cubbage, C.H.; Loeher, C.F. III; Bird, J.R.

    1995-12-01

The Federal Facilities compliance Act of 1992 directs all federal facilities including those under the auspices of the Department of Defense (DoD) to adhere to the ever growing number of environmental statutes. With a large percentage of the 1994 DoD budget dedicated to major acquisitions, it became apparent that an intensive study of the acquisition process was needed to identify milestone areas in which environmental protection requirements could be integrated. This paper provides a synopsis of the study which was undertaken to assist the DoD in understanding environmental considerations and complying with environmental legislation. The study utilized a three phased methodology which provided legislation analysis, process application, and guidance development. In phase one, over 25 Federal and 14 state environmental statutes and regulations, the tri-service regulations, and the DoD and EPA policy and procedures were analyzed. Phase two applied the environmental considerations and legislative analysis to the defense acquisition process, while phase three developed specific guidance to assist government personnel in their roles and responsibilities. The study resulted in the development of an expandable PC-based support system that integrated environmental protection considerations in the defense acquisition process and provided guidance to the responsible government official(s).

Whitney, John C.; Robinson, Howard; Whitfield, Gregory B.; Marmont, Lindsey S.; Yip, Patrick; Neculai, A. Mirela; Lobsanov, Yuri D.; Ohman, Dennis E.; Howell, P. Lynne

    2015-05-15

Pseudomonas aeruginosa is an opportunistic human pathogen that secretes the exopolysaccharide alginate during infection of the respiratory tract of individuals afflicted with cystic fibrosis and chronic obstructive pulmonary disease. Among the proteins required for alginate production, Alg44 has been identified as an inner membrane protein whose bis-(3',5')-cyclic dimeric guanosine monophosphate (c-di-GMP) binding activity post-translationally regulates alginate secretion. In this study, we report the 1.8 Å crystal structure of the cytoplasmic region of Alg44 in complex with dimeric self-intercalated c-di-GMP and characterize its dinucleotide-binding site using mutational analysis. The structure shows that the c-di-GMP binding region of Alg44 adopts a PilZmore » domain fold with a dimerization mode not previously observed for this family of proteins. Moreover, calorimetric binding analysis of residues in the c-di-GMP binding site demonstrate that mutation of Arg-17 and Arg-95 alters the binding stoichiometry between c-di-GMP and Alg44 from 2:1 to 1:1. Introduction of these mutant alleles on the P. aeruginosa chromosome show that the residues required for binding of dimeric c-di-GMP in vitro are also required for efficient alginate production in vivo. Our results suggest that the dimeric form of c-di-GMP represents the biologically active signaling molecule needed for the secretion of an important virulence factor produced by P. aeruginosa.« less

    SciTech Connect (OSTI)

    Tyacke, M.; Schmitt, R.

    1993-07-01

The purpose of this report is to identify the regulations and requirements for transporting greater-than-Class C (GTCC) low-level radioactive waste (LLW) and to identify planning activities that need to be accomplished in preparation for transporting GTCC LLW. The regulations and requirements for transporting hazardous materials, of which GTCC LLW is included, are complex and include several Federal agencies, state and local governments, and Indian tribes. This report is divided into five sections and three appendices. Section 1 introduces the report. Section 2 identifies and discusses the transportation regulations and requirements. The regulations and requirements are divided into Federal, state, local government, and Indian tribes subsections. This report does not identify the regulations or requirements of specific state, local government, and Indian tribes, since the storage, treatment, and disposal facility locations and transportation routes have not been specifically identified. Section 3 identifies the planning needed to ensure that all transportation activities are in compliance with the regulations and requirements. It is divided into (a) transportation packaging; (b) transportation operations; (c) system safety and risk analysis, (d) route selection; (e) emergency preparedness and response; and (f) safeguards and security. This section does not provide actual planning since the details of the Department of Energy (DOE) GTCC LLW Program have not been finalized, e.g., waste characterization and quantity, storage, treatment and disposal facility locations, and acceptance criteria. Sections 4 and 5 provide conclusions and referenced documents, respectively.

    1996-09-01

Although truly new technology is not required for successful acquisition of seismic data in deep Gulf of Mexico waters, it is helpful to review some basic aspects of these seismic surveys. Additionally, such surveys are likely to see early use of some emerging new technology which can improve data quality. Because such items as depth imaging, borehole seismic, 4-D and marine 3-component recording were mentioned in the May 1996 issue of World Oil, they are not discussed again here. However, these technologies will also play some role in the deepwater seismic activities. What is covered in this paper are some new considerations for: (1) longer data records needed in deeper water, (2) some pros and cons of very long steamer use, and (3) two new commercial systems for quantifying data quality.

  6. Requirements for Defining Utility Drive Cycles: An Exploratory Analysis of Grid Frequency Regulation Data for Establishing Battery Performance Testing Standards

    SciTech Connect (OSTI)

Hafen, Ryan P.; Vishwanathan, Vilanyur V.; Subbarao, Krishnappa; Kintner-Meyer, Michael CW

    2011-10-19

Battery testing procedures are important for understanding battery performance, including degradation over the life of the battery. Standards are important to provide clear rules and uniformity to an industry. The work described in this report addresses the need for standard battery testing procedures that reflect real-world applications of energy storage systems to provide regulation services to grid operators. This work was motivated by the need to develop Vehicle-to-Grid (V2G) testing procedures, or V2G drive cycles. Likewise, the stationary energy storage community is equally interested in standardized testing protocols that reflect real-world grid applications for providing regulation services. As the first of several steps toward standardizing battery testing cycles, this work focused on a statistical analysis of frequency regulation signals from the Pennsylvania-New Jersey-Maryland Interconnect with the goal to identify patterns in the regulation signal that would be representative of the entire signal as a typical regulation data set. Results from an extensive time-series analysis are discussed, and the results are explained from both the statistical and the battery-testing perspectives. The results then are interpreted in the context of defining a small set of V2G drive cycles for standardization, offering some recommendations for the next steps toward standardizing testing protocols.

An SSC data acquisition architecture applicable to high-p{sub T} detectors is described. The architecture is based upon a small set of design principles that were chosen to simplify communication between data acquisition elements while providing the required level of flexibility and performance. The architecture features an integrated system for data collection, event building, and communication with a large processing farm. The interface to the front end electronics system is also discussed. A set of design parameters is given for a data acquisition system that should meet the needs of high-p{sub T} detectors at the SSC.

    DOE Patents [OSTI]

Shapiro, Stephen L.; Mani, Sudhindra; Atlas, Eugene L.; Cords, Dieter H. W.; Holbrook, Britt

    1997-01-01

A data acquisition circuit for a particle detection system that allows for time tagging of particles detected by the system. The particle detection system screens out background noise and discriminate between hits from scattered and unscattered particles. The detection system can also be adapted to detect a wide variety of particle types. The detection system utilizes a particle detection pixel array, each pixel containing a back-biased PIN diode, and a data acquisition pixel array. Each pixel in the particle detection pixel array is in electrical contact with a pixel in the data acquisition pixel array. In response to a particle hit, the affected PIN diodes generate a current, which is detected by the corresponding data acquisition pixels. This current is integrated to produce a voltage across a capacitor, the voltage being related to the amount of energy deposited in the pixel by the particle. The current is also used to trigger a read of the pixel hit by the particle.

DOE Public Access Gateway for Energy & Science Beta (PAGES Beta)

    Thomas, John

    2016-04-05

A major driving force for change in light-duty vehicle design and technology is the National Highway Traffic Safety Administration (NHTSA) and the U.S. Environmental Protection Agency (EPA) joint final rules concerning Corporate Average Fuel Economy (CAFE) and greenhouse gas (GHG) emissions for model years (MY) 2016 through 2025 passenger cars and light trucks. The chief goal of this current study is to compare the already rapid pace of fuel economy improvement and technological change over the previous decade to the needed rate of change to meet regulations over the next decade. EPA and NHTSA comparisons of the MY 2004 USmore » light-duty vehicle fleet to the MY 2014 fleet shows improved fuel economy (FE) of approximately 28% using the same FE estimating method mandated for CAFE regulations. Future predictions by EPA and NHTSA concerning ensemble fleet fuel economy are examined as an indicator of needed vehicle rate-of-change. A set of 40 same-model vehicle pairs for MY 2005 and MY 2015 is compared to examine changes in energy use and related technological change over the 10 year period. Powertrain improvements measured as increased vehicle efficiency, and vehicle mass-glider improvements measured as decreased tractive work requirements are quantified. The focus is first on conventional gasoline powertrain vehicles which currently dominate the market, with hybrids also examined due to their high potential importance for CAFE compliance. Most hybrid vehicles with significant sales in 2014 were represented in the study. Results show 10 years of progress for the studied vehicle set includes lowered tractive effort of about 5.6% and improved powertrain efficiency of about 16.5%. Further analysis shows that this high rate of past progress must increase by about 50% in order to meet the 2025 CAFE standards. Examination of where certain MY 2015 vehicle compare to CAFE regulations is offered as well as some simple conjecture on what is needed to meet regulations under

    DOE Patents [OSTI]

    Phillips, David T.

    1979-01-01

A data acquisition system capable of resolving transient pulses in the subnanosecond range. A pulse in an information carrying medium such as light is transmitted through means which disperse the pulse, such as a fiber optic light guide which time-stretches optical pulses by chromatic dispersion. This time-stretched pulse is used as a sampling pulse and is modulated by the signal to be recorded. The modulated pulse may be further time-stretched prior to being recorded. The recorded modulated pulse is unfolded to derive the transient signal by utilizing the relationship of the time-stretching that occurred in the original pulse.

    2012-08-01

DAQMAN is a flexible configurable interface that allows the user to build and operate a VME-based data acquisition system on a Linux workstation. It consists of two parts: a Java-based Graphical User Interface to configure the system, and a C-based utility that reads out the data and creates the output ASCII data file, with two levels of diagnostic tools. The data acquisition system requires a CAEN CONET-VME Bridge to communicate between the hardware in themore » VME crate and the Linux workstation. Data acquisition modules, such as ADCs, TDC, Scalers, can be loaded into the system, or removed easily. The GUI allows users to activate modules, and channels within modules by clicking on icons. Running configurations are stored; data are collected and can be viewed either as raw numbers, or by charts and histograms that update as the data are accumulated. Data files are written to disk in ASCII format, with a date and time stamp.« less

    2011-03-01

Federal fleet managers face unique challenges in accomplishing their mission - meeting agency transportation needs while complying with Federal goals and mandates. Included in these challenges are a variety of statutory requirements, executive orders, and internal goals and objectives that typically focus on petroleum consumption and greenhouse gas (GHG) emissions reductions, alternative fuel vehicle (AFV) acquisitions, and alternative fuel use increases. Given the large number of mandates affecting Federal fleets and the challenges faced by all fleet managers in executing day-to-day operations, a primary challenge for agencies and other organizations is ensuring that they are as efficient as possible in using constrained fleet budgets. An NREL Optimal Vehicle Acquisition (NOVA) analysis makes use of a mathematical model with a variety of fleet-related data to create an optimal vehicle acquisition strategy for a given goal, such as petroleum or GHG reduction. The analysis can helps fleets develop a vehicle acquisition strategy that maximizes petroleum and greenhouse gas reductions.

    2006-05-10

The CRD addresses the requirements of Department of Energy (DOE) Order 413.3-Change 1, ''Program and Project Management for the Acquisition of Capital Assets'', by providing the Secretarial Acquisition Executive (Level 0) scope baseline and the Program-level (Level 1) technical baseline. The Secretarial Acquisition Executive approves the Office of Civilian Radioactive Waste Management's (OCRWM) critical decisions and changes against the Level 0 baseline; and in turn, the OCRWM Director approves all changes against the Level 1 baseline. This baseline establishes the top-level technical scope of the CRMWS and its three system elements, as described in section 1.3.2. The organizations responsible for design, development, and operation of system elements described in this document must therefore prepare subordinate project-level documents that are consistent with the CRD. Changes to requirements will be managed in accordance with established change and configuration control procedures. The CRD establishes requirements for the design, development, and operation of the CRWMS. It specifically addresses the top-level governing laws and regulations (e.g., ''Nuclear Waste Policy Act'' (NWPA), 10 Code of Federal Regulations (CFR) Part 63, 10 CFR Part 71, etc.) along with specific policy, performance requirements, interface requirements, and system architecture. The CRD shall be used as a vehicle to incorporate specific changes in technical scope or performance requirements that may have significant program implications. Such may include changes to the program mission, changes to operational capability, and high visibility stakeholder issues. The CRD uses a systems approach to: (1) identify key functions that the CRWMS must perform, (2) allocate top-level requirements derived from statutory, regulatory, and programmatic sources, and (3) define the basic elements of the system architecture and operational concept. Project-level documents address CRD requirements by further

David Petti; Tim Abram; Richard Hobbins; Jim Kendall

    2010-12-01

A Next Generation Nuclear Plant (NGNP) fuel acquisition strategy was first established in 2007. In that report, a detailed technical assessment of potential fuel vendors for the first core of NGNP was conducted by an independent group of international experts based on input from the three major reactor vendor teams. Part of the assessment included an evaluation of the credibility of each option, along with a cost and schedule to implement each strategy compared with the schedule and throughput needs of the NGNP project. While credible options were identified based on the conditions in place at the time, many changes in the assumptions underlying the strategy and in externalities that have occurred in the interim requiring that the options be re-evaluated. This document presents an update to that strategy based on current capabilities for fuel fabrication as well as fuel performance and qualification testing worldwide. In light of the recent Pebble Bed Modular Reactor (PBMR) project closure, the Advanced Gas Reactor (AGR) fuel development and qualification program needs to support both pebble and prismatic options under the NGNP project. A number of assumptions were established that formed a context for the evaluation. Of these, the most important are: • Based on logistics associated with the on-going engineering design activities, vendor teams would start preliminary design in October 2012 and complete in May 2014. A decision on reactor type will be made following preliminary design, with the decision process assumed to be completed in January 2015. Thus, no fuel decision (pebble or prismatic) will be made in the near term. • Activities necessary for both pebble and prismatic fuel qualification will be conducted in parallel until a fuel form selection is made. As such, process development, fuel fabrication, irradiation, and testing for pebble and prismatic options should not negatively influence each other during the period prior to a decision on reactor type

This paper is the first of a series which describes the Enrichment Diagnostic System (EDS) developed for the MARS project at Lawrence Livermore National Laboratory. Although EDS was developed for use on AVLIS, the functional requirements, overall design, and specific techniques are applicable to any experimental data acquisition system involving large quantities of transient data. In particular this paper will discuss the techniques and equipment used to do the data acquisition. Included are what types of hardware are used and how that hardware (CAMAC, digital oscilloscopes) is interfaced to the HP computers. In this discussion the author will address the problems encountered and the solutions used, as well as the performance of the instrument/computer interfaces. The second topic the author will discuss is how the acquired data is associated to graphics and analysis portions of EDS through efficient real time data bases. This discussion will include how the acquired data is folded into the overall structure of EDS providing the user immediate access to raw and analyzed data. By example you will see how easily a new diagnostic can be added to the EDS structure without modifying the other parts of the system. 8 figs.

    2006-05-01

Jefferson Lab's Continuous Electron Beam Accelerator Facility (CEBAF) occasionally experiences fast intermittent beam instabilities that are difficult to isolate and result in downtime. The Distributed Data Acquisition (Dist DAQ) system is being developed to detect and quickly locate such instabilities. It will consist of multiple Ethernet based data acquisition chassis distributed throughout the seven-eights of a mile CEBAF site. Each chassis will monitor various control system signals that are only available locally and/or monitored by systems with small bandwidths that cannot identify fast transients. The chassis will collect data at rates up to 40 Msps in circular buffers that can be frozen and unrolled after an event trigger. These triggers will be derived from signals such as periodic timers or accelerator faults and be distributed via a custom fiber optic event trigger network. This triggering scheme will allow all the data acquisition chassis to be triggered simultaneously and provide a snapshot of relevant CEBAF control signals. The data will then be automatically analyzed for frequency content and transients to determine if and where instabilities exist.

  11. CONTRACTOR LEGAL MANAGEMENT REQUIREMENTS: FINAL RULE

    Broader source: Energy.gov [DOE]

The Department has issued a Final Rule revising the existing contractor regulations covering contractor legal management requirements at 10 C.F.R. 719 (Federal Register May 3, 2013; http://www.gpo.gov/fdsys/pkg/FR-2013-05-03/pdf/2013-10485.pdf). One unusual aspect of the Final Rule is that it requires Contracting Officers to attempt to execute bilateral modifications to existing affected contracts to apply the Final Rule's revisions to those contracts. We will issue an Acquisition Letter shortly with suggested contract modification language. (The Final Rule's revisions "… are designed to clarify and streamline existing requirements, improve efficiency of contractor legal management, and facilitate oversight over the expenditure of taxpayer dollars." The contract modifications will state the revisions will only apply to costs incurred from date the bilateral modifications are executed forward. The revisions are administrative in nature. Regarding costs contractors have not yet incurred, the revisions could have been communicated to contractors without using a Final Rule to incorporate them into current contracts. Consequently, the modifications do not require equitable adjustments.)

    SciTech Connect (OSTI)

    Graniere, R.J.; Burns, R.E.

    1996-11-01

This is the first of a series of reports on utility mergers and acquisitions published by NRRI; because it was completed Nov. 1996, it does not cover FERC Order 592 (policy statement on merger policy under Federal Power Act). Since cost-benefit analyses are expensive, state regulators need guidelines that efficiently streamline the review process for mergers and acquisitions. Purpose of this paper is to suggest such guidelines; they are applicable only to mergers and acquisitions of vertically integrated electric utilities or combination electric/gas utilities.

Attached is new Acquisition Guide Chapter 9.4, Contractor Responsibility Determinations. This Chapter is being issued to address recommendations from a recent Inspector General (IG) report that found DOE Contracting Officers (CO) were not always conducting responsibility determinations properly. In particular, the IG found that for sole source awards, including 8(a) awards, the CO was not documenting reviews of past performance. The IG recommended that COs consistently follow existing FAR requirements for completing responsibility determinations. This Chapter reiterates and summarizes current FAR requirements for doing so.

    SciTech Connect (OSTI)

Wheat, Jr., Robert Mitchell; Dalmas, Dale A.; Dale, Gregory E.

    2015-08-11

Data acquisition from multiple beam current monitors is required for electron accelerator production of Mo-99. A two channel system capable of recording data from two beam current monitors has been developed, is currently in use, and is discussed below. The development of a cost-effective method of extending this system to more than two channels and integrating of these measurements into an accelerator control system is the main focus of this report. Data from these current monitors is digitized, processed, and stored by a digital data acquisition system. Limitations and drawbacks with the currently deployed digital data acquisition system have been identified as have been potential solutions, or at least improvements, to these problems. This report will discuss and document the efforts we've made in improving the flexibility and lowering the cost of the data acquisition system while maintaining the minimum requirements.

A multiple channel data acquisition system for the transfer of large amounts of data from a multiplicity of data channels has a plurality of modules which operate in parallel to convert analog signals to digital data and transfer that data to a communications host via a FASTBUS. Each module has a plurality of submodules which include a front end buffer (FEB) connected to input circuitry having an analog to digital converter with cache memory for each of a plurality of channels. The submodules are interfaced with the FASTBUS via a FASTBUS coupler which controls a module bus and a module memory. The system is triggered to effect rapid parallel data samplings which are stored to the cache memories. The cache memories are uploaded to the FEBs during which zero suppression occurs. The data in the FEBs is reformatted and compressed by a local processor during transfer to the module memory. The FASTBUS coupler is used by the communications host to upload the compressed and formatted data from the module memory. The local processor executes programs which are downloaded to the module memory through the FASTBUS coupler.

The proposed new data acquisition system architecture takes event fragments off a detector over fiber optics and to a parallel event building switch. The parallel event building switch concept, taken from the telephone communications industry, along with expected technology improvements in fiber-optic data transmission speeds over the next few years, should allow data acquisition system rates to increase dramatically and exceed those rates needed for the SSC. This report briefly describes the switch architecture and fiber optics for a SSC data acquisition system.

    1990-10-01

MDS-Plus, a model driven data acquisition system being jointly developed at Istituto Gas Ionizzati, MIT Plasma Fusion Center, and Los Alamos National Laboratory, is based on the concept of an experiment model. The model contains descriptions of experiment data, devices, and actions to be performed. The data acquisition engine, i.e., the part of the system which actually executes the acquisition process, is driven by the contents of the model itself and implemented as a set of independent processes. A scheduler keeps the data acquisition engine in step with a state machine reflecting the operation of the actual experiment; a dispatcher takes care of the appropriate sequencing of the operations associated with each state of the experiment; various servers actually execute actions on behalf of the dispatcher.

  11. Applying automated data acquisition and management technology to bioremediation

    SciTech Connect (OSTI)

    Widing, M.A.; Leser, C.

    1995-06-01

Operating a bioremediation process requires timely and accurate analysis of physical and chemical parameters that can affect the system. At a fuel oil spill site, the operation of an in-situ bioremediation system, consisting of fluid and nutrient injection, fluid withdrawal, and aeration cycles, is monitored by means of electronic downhole sensors and on-site chemical analysis. A data acquisition and management system was designed and implemented to rapidly analyze data for operational decision malting. A hardware suite, containing an electronic monitoring system data acquisition computer, and data analysis workstation, was also developed. Through the use of both commercial software products and custom software, suites of data management and analysis tools were provided. The data acquisition suite of software tools assisted in programming dataloggers, automatically recording monitored data, and integrating these data with manually sampled chemical data. The data analysis suite of software tools assisted in downloading data to remote workstations, sampling the database for trend analysis, and automating the interface to commercial analysis packages.

  20. Identification of a classical mutant in the industrial host Aspergillus niger by systems genetics: LaeA is required for citric acid production and regulates the formation of some secondary metabolites

    2015-11-13

The asexual filamentous fungus Aspergillus niger is an important industrial cell factory for citric acid production. In this study, we genetically characterized a UV-generated A. niger mutant that was originally isolated as a nonacidifying mutant, which is a desirable trait for industrial enzyme production. Physiological analysis showed that this mutant did not secrete large amounts of citric acid and oxalic acid, thus explaining the nonacidifying phenotype. As traditional complementation approaches to characterize the mutant genotype were unsuccessful, we used bulk segregant analysis in combination with high-throughput genome sequencing to identify the mutation responsible for the nonacidifying phenotype. Since A. nigermore » has no sexual cycle, parasexual genetics was used to generate haploid segregants derived from diploids by loss of whole chromosomes. We found that the nonacidifying phenotype was caused by a point mutation in the laeA gene. LaeA encodes a putative methyltransferase-domain protein, which we show here to be required for citric acid production in an A. niger lab strain (N402) and in other citric acid production strains. The unexpected link between LaeA and citric acid production could provide new insights into the transcriptional control mechanisms related to citric acid production in A. niger. Interestingly, the secondary metabolite profile of a ΔlaeA strain differed from the wild-type strain, showing both decreased and increased metabolite levels, indicating that LaeA is also involved in regulating the production of secondary metabolites. As a result, we show that our systems genetics approach is a powerful tool to identify trait mutations.« less

    Von Eschen, R.L.; Scheele, P.F.

    1962-04-24

A transistorized voltage regulator which provides very close voitage regulation up to about 180 deg F is described. A diode in the positive line provides a constant voltage drop from the input to a regulating transistor emitter. An amplifier is coupled to the positive line through a resistor and is connected between a difference circuit and the regulating transistor base which is negative due to the difference in voltage drop across thc diode and the resistor so that a change in the regulator output causes the amplifier to increase or decrease the base voltage and current and incrcase or decrease the transistor impedance to return the regulator output to normal. CH2M HILL Plateau Remediation Company (CH2M HILL) is the U.S. Department of Energy (DOE) prime contractor responsible for the environmental cleanup of the Hanford Site Central Plateau. As part of this responsibility, the CH2M HILL is faced with the task of complying with thousands of environmental requirements which originate from over 200 federal, state, and local laws and regulations, DOE Orders, waste management and effluent discharge permits, Comprehensive Environmental Response, Compensation, and Liability Act (CERCLA) response and Resource Conservation and Recovery Act (RCRA) corrective action documents, and official regulatory agency correspondence. The challenge is to manage this vast number of requirements to ensure they are appropriately and effectively integrated into CH2M HILL operations. Ensuring compliance with a large number of environmental requirements relies on an organization’s ability to identify, evaluate, communicate, and verify those requirements. To ensure that compliance is maintained, all changes need to be tracked. The CH2M HILL identified that the existing system used to manage environmental requirements was difficult to maintain and that improvements should be made to increase functionality. CH2M HILL established an environmental requirements management procedure and tools to assure that all environmental requirements are effectively and efficiently managed. Having a complete and accurate set of environmental requirements applicable to CH2M HILL operations will promote a more efficient approach to: • Communicating requirements • Planning work • Maintaining work controls • Maintaining compliance

    SciTech Connect (OSTI)

    Pierce, K.S.; Alton, C.C.; Linehan, A.O.

    1995-12-01

The Bonneville Power Administration (BPA) has a statutory obligation to supply cost-effective electric power to its customers. BPA must also be consistent with the Pacific Northwest Electric Power Planning and Conservation Act, which requires consideration of the Northwest Power Planning Council`s Conservation and Electric Power Plan and Fish and Wildlife Program. The challenge is to operate effectively within an increasingly competitive and dynamic electric utility market while continuing its commitment to the National Environmental Policy Act (NEPA). BPA developed a tiered approach to its environmental analyses associated with energy resource planning and acquisition. The Resource Programs Environmental Impact Statement (RPEIS) is a programmatic document designed to support energy resource decisions for several years. The RPEIS includes a description of the environmental impacts and potential mitigation measures for various energy resources-including conservation, efficiency improvements, renewable resources, cogeneration, combustion turbines, nuclear, and coal. Information was also provided on fuel switching, load management, and emerging technologies. After analyzing the environmental trade-offs among resources, the RPEIS focuses on the cumulative effects of adding these resources to the existing power system. The Record of Decision documents that BPA`s resource acquisitions will include all cost-effective conservation and efficiency improvements, supplemented by a mix of renewables and thermal resources. Subsequent site-specific documents have been prepared on individual resource acquisitions. By focusing environmental reviews on the actual issues ripe for decision and by providing timely environmental information to the public and to the decisionmakers, this tiered approach led to better decisionmaking. BPA was able to operate in a more business-like manner while assuring NEPA compliance.

  2. In situ acquisition of cathodic protection parameters

    SciTech Connect (OSTI)

    Baptista, W.; Costa, J.C.M. da

    1997-01-01

Present, offshore cathodic protection (CP) design in Brazil uses parameters found in the literature and in international norms, such as: initial and average current density, coating efficiency, leaked current at the buried part of the wet Xmas tree, etc. Five data acquisition systems (DAS) were launched at the Campos Basin water in depths of 102, 290, and 975 m. The DASs were recovered after 8, 11, and 14 months, respectively. Adequate methods of data processing guaranteed the acquisition of reliable parameters for use in CP projects in seawater: initial and average current densities, anode current capacity, and polarization curves at several time intervals. The potential of {minus}800 mV{sub Ag/AgCl} is generally accepted as norm for carbon steel protection in seawater. The cathodic current density necessary to obtain this potential is extremely dependent upon environmental conditions, and the geometry and superficial condition of the submerged steel structure.

    2011-01-01

The Data Acquisition System for the CDMS II dark matter experiment was designed and built when the experiment moved to its new underground installation at the Soudan Lab. The combination of remote operation and increased data load necessitated a completely new design. Elements of the original LabView system remained as stand-alone diagnostic programs, but the main data processing moved to a VME-based system with custom electronics for signal conditioning, trigger formation and buffering. The data rate was increased 100-fold and the automated cryogenic system was linked to the data acquisition. A modular server framework with associated user interfaces was implemented in Java to allow control and monitoring of the entire experiment remotely.

    2007-11-01

Numerous terrorist organizations have openly expressed interest in producing and deploying biological weapons. However, a limiting factor for many terrorists has been the acquisition of dangerous biological agents, as evidenced by the very few successful instances of biological weapons use compared to the number of documented hoaxes. Biological agents vary greatly in their ability to cause loss of life and economic damage. Some agents, if released properly, can kill many people and cause an extensive number of secondary infections; other agents will sicken only a small number of people for a short period of time. Consequently, several biological agents can potentially be used to perpetrate a bioterrorism attack but few are likely capable of causing a high consequence event. It is crucial, from a US national security perspective, to more deeply understand the likelihood that terrorist organizations can acquire the range of these agents. Few studies have attempted to comprehensively compile the technical information directly relevant to the acquisition of dangerous bacteria, viruses and toxins. In this report, technical fact sheets were assembled for 46 potentially dangerous biological agents. Much of the information was taken from various research sources which could ultimately and significantly expedite and improve bioterrorism threat assessments. By systematically examining a number of specific agent characteristics included in these fact sheets, it may be possible to detect, target, and implement measures to thwart future terrorist acquisition attempts. In addition, the information in these fact sheets may be used as a tool to help laboratories gain a rudimentary understanding of how attractive a method laboratory theft is relative to other potential acquisition modes.

    1998-08-27

The calorimeter Data Acquisition and Reporting Program performs the calculations necessary to calculate the calorimetric sample results in grams and provide a printable report for up to twelve Mound Calorimeters. To determine a standard''s wattage or sample gram fill, the reporting program retrieves the output voltage from the power supply at the calorimeter and a temperature resistant resistor via a voltmeter and digital input card in a Hewlett Packard Data Acquisition Unit (DAQ). From the retrieved voltage data, the reporting program can calculate a standard''s wattage output and sample gram fill. The reporting program also determines equilibrium (stability) by performing a stability algorithm bassed on user defined slope an/or sigma values for the previous forty values. Once the stability is determined, the reporting program will notify the user that the calorimeter has reached equilibrium. The Calorimeter Data Acquisition and Reporting Program operates continuously as described to monitor for calorimeter equilibrium and to generate a printable report with sample results.

This document describes the test procedure for the Project W-320 Tank C-106 Sluicing Data Acquisition System (W-320 DAS). The Software Test portion will test items identified in the WRSS DAS System Description (SD), HNF-2115. Traceability to HNF-2115 will be via a reference that follows in parenthesis, after the test section title. The Field Test portion will test sensor operability, analog to digital conversion, and alarm setpoints for field instrumentation. The W-320 DAS supplies data to assist thermal modeling of tanks 241-C-106 and 241-AY-102. It is designed to be a central repository for information from sources that would otherwise have to be read, recorded, and integrated manually. Thus, completion of the DAS requires communication with several different data collection devices and output to a usable PC data formats. This test procedure will demonstrate that the DAS functions as required by the project requirements stated in Section 3 of the W-320 DAS System Description, HNF-2115.
