  1. Transport and intracellular accumulation of acetaldehyde in Saccharomyces cerevisiae

    The rate of acetaldehyde efflux from yeast cells and its intracellular concentration were studied in the light of recent suggestions that acetaldehyde inhibition may be an important factor in yeast ethanol fermentations. When the medium surrounding cells containing ethanol and acetaldehyde was suddenly diluted, the rate of efflux of acetaldehyde was slow relative to the rate of ethanol efflux, suggesting that acetaldehyde, unlike ethanol, may accumulate intracellularly. Intracellular acetaldehyde concentrations were measured during high cell density fermentations, using direct injection gas chromatography to avoid the need to concentrate or disrupt the cells. Intracellular acetaldehyde concentrations substantially exceeded the extracellular concentrations throughout fermentation and were generally much higher than the acetaldehyde concentrations normally recorded in the culture broth in ethanol fermentations. The technique used was sensitive to the time taken to cool and freeze the samples. Measured intracellular acetaldehyde concentrations fell rapidly as the time taken to freeze the suspensions was extended beyond 2 s. The results add weight to recent claims that acetaldehyde toxicity is responsible for some of the effects previously ascribed to ethanol in alcohol fermentations, especially Zymomonas fermentations. Further work is required to confirm the importance of acetaldehyde toxicity under other culture conditions.

    The lag phase of Saccharomyces cerevisiae subjected to a step increase in temperature or ethanol concentration was reduced by as much as 60% when acetaldehyde was added to the medium at concentrations less than 0.1 g/L. Maximum specific growth rates were also substantially increased. Even greater proportional reductions in lag time due to acetaldehyde addition were observed for ethanol-shocked cultures of Zymomonas mobilis. Acetaldehyde had no effect on S. cerevisiae cultures started from stationary phase inocula in the absence of environmental shock and its lag-reducing effects were greater in complex medium than in a defined synthetic medium. Acetaldehyde reacted strongly with the ingredients of complex culture media. It is proposed that the effect of added acetaldehyde may be to compensate for the inability of cells to maintain transmembrane acetaldehyde gradients following an environmental shock.

    Radon and its progeny are major contributors in the radiation dose received by general population of the world. Because radon is a noble gas, a large portion of it is free to migrate away from radium. The primary sources of radon in the houses are soils and rocks source emanations, emanation from building materials, and entry of radon into a structure from outdoor air. Keeping this in mind the study of radon exhalation rate from some soil samples of the Kharar, Punjab has been carried out using Can Technique. The equilibrium radon concentration in various soil samples of Kharar area of district Mohali varied from 12.7 Bqm{sup −3} to 82.9 Bqm{sup −3} with an average of 37.5 ± 27.0 Bqm{sup −3}. The radon mass exhalation rates from the soil samples varied from 0.45 to 2.9 mBq/kg/h with an average of 1.4 ± 0.9 mBq/kg/h and radon surface exhalation rates varied from 10.4 to 67.2 mBq/m{sup 2}/h with an average of 30.6 ± 21.8 mBq/m{sup 2}/h. The radon mass and surface exhalation rates of the soil samples of Kharar, Punjab were lower than that of the world wide average.

  8. The Impact of Soil Sampling Errors on Variable Rate Fertilization

    Variable rate fertilization of an agricultural field is done taking into account spatial variability in the soils characteristics. Most often, spatial variability in the soils fertility is the primary characteristic used to determine the differences in fertilizers applied from one point to the next. For several years the Idaho National Engineering and Environmental Laboratory (INEEL) has been developing a Decision Support System for Agriculture (DSS4Ag) to determine the economically optimum recipe of various fertilizers to apply at each site in a field, based on existing soil fertility at the site, predicted yield of the crop that would result (and a predicted harvest-time market price), and the current costs and compositions of the fertilizers to be applied. Typically, soil is sampled at selected points within a field, the soil samples are analyzed in a lab, and the lab-measured soil fertility of the point samples is used for spatial interpolation, in some statistical manner, to determine the soil fertility at all other points in the field. Then a decision tool determines the fertilizers to apply at each point. Our research was conducted to measure the impact on the variable rate fertilization recipe caused by variability in the measurement of the soils fertility at the sampling points. The variability could be laboratory analytical errors or errors from variation in the sample collection method. The results show that for many of the fertility parameters, laboratory measurement error variance exceeds the estimated variability of the fertility measure across grid locations. These errors resulted in DSS4Ag fertilizer recipe recommended application rates that differed by up to 138 pounds of urea per acre, with half the field differing by more than 57 pounds of urea per acre.     A novel theoretical framework for predicting the branching between roaming and bond fission channels in molecular dissociations is described and applied to the decomposition of acetaldehyde. This reduced dimensional trajectory (RDT) approach, which is motivated by the long-range nature of the roaming, bond fission, and abstraction dynamical bottlenecks, involves the propagation of rigid-body trajectories on an analytic potential energy surface. The analytic potential is obtained from fits to large-scale multireference ab initio electronic structure calculations. The final potential includes one-dimensional corrections from higher-level electronic structure calculations and for the effect of conserved mode variations along both the addition and abstraction paths. The corrections along the abstraction path play a significant role in the predicted branching. Master equation simulations are used to transform the microcanonical branching ratios obtained from the RDT simulations to the temperature- and pressure-dependent branching ratios observed in thermal decomposition experiments. For completeness, a transition-state theory treatment of the contributions of the tight transition states for the molecular channels is included in the theoretical analyses. The theoretically predicted branching between molecules and radicals in the thermal decomposition of acetaldehyde is in reasonable agreement with the corresponding shock tube measurement described in the companion paper. The prediction for the ratio of the tight to roaming contributions to the molecular channel also agrees well with results extracted from recent experimental and experimental/theoretical photodissociation studies.

  13. Measurements and calculations of oscillations and phase relations in the driven gas-phase combustion of acetaldehyde

    Oscillations in light emission and species concentrations, are measured as periodic perturbations are simultaneously applied to the input rates of acetaldehyde and oxygen in the gas-phase combustion of acetaldehyde in a continuous-flow stirred tank reactor for conditions where the autonomous reaction itself is oscillatory. The experimental results are compared with the predictions of a five-variable thermokinetic model. We measure periodic responses in the fundamental entrainment band (ratio of frequency of perturbation to frequency of response equal to unity) for four different values of phase shift between the acetaldehyde and oxygen perturbation wave forms as we vary the frequency and amplitude of the external periodic perturbations. Outside of the entrainment bands we find quasiperiodic response. We determine the phases of the light emission and six species concentrations, as measured with a mass spectrometer, with respect to the periodic perturbation, the variation of these phases across the fundamental entrainment band for different values of reactant phase shift and for different amplitudes of perturbation, and the effects of the phase shift between the two input perturbations on the light emission response of the system for different frequencies of perturbation. Both the experiments and calculations predict a widening of the entrainment band with an increase in perturbation amplitude, and the same variation in bandwidths for the four values of reactant phase shift studied. The experiments and calculations also predict the same general trends in light phase and species phases (difference between the light emission and species concentrations with respect to the perturbing wave form) as the band is traversed for different amplitudes of perturbation and for different values of reactant phase shift.

    This study reports and compares the adsorption and dissociation of acetaldehyde on oxidized and reduced CeOX(100) thin films. Acetaldehyde reacts and decomposes on fully oxidized CeO2(100) whereas it desorbs molecularly at low temperature on CeO2(111). The primary products are CO, CO2 and water along with trace amounts of crotonaldehyde and acetylene. The acetaldehyde adsorbs as the 2-acetaldehyde species, dioxyethylene. Decomposition proceeds by dehydrogenation through acetate and enolate intermediates. The reaction pathway is similar on the reduced CeO2-X(100) surface however the inability to react with surface O on the reduced surface results in H2 rather than H2O desorption and C is left on the surface rather than producing CO and CO2. C-O bond cleavage in the enolate intermediate followed by reaction with surface H results in ethylene desorption.

    Dissociation dynamics of the temporary negative ions of ethanol and acetaldehyde formed by the low-energy electron attachments is investigated by using the anion velocity map imaging technique and ab initio molecular dynamics simulations. The momentum images of the dominant fragments O{sup −}/OH{sup −} and CH{sub 3}{sup −} are recorded, indicating the low kinetic energies of O{sup −}/OH{sup −} for ethanol while the low and high kinetic energy distributions of O{sup −} ions for acetaldehyde. The CH{sub 3}{sup −} image for acetaldehyde also shows the low kinetic energy. With help of the dynamics simulations, the fragmentation processes are qualitatively clarified. A new cascade dissociation pathway to produce the slow O{sup −} ion via the dehydrogenated intermediate, CH{sub 3}CHO{sup −} (acetaldehyde anion), is proposed for the dissociative electron attachment to ethanol. After the electron attachment to acetaldehyde molecule, the slow CH{sub 3}{sup −} is produced quickly in the two-body dissociation with the internal energy redistributions in different aspects before bond cleavages.

By using static headspace-gas chromatography for liquid phase analysis, we identify acetaldehyde as a minor product and key intermediate in the electroreduction of CO to ethanol on OD-Cu electrodes. Acetaldehyde is produced with a Faradaic efficiency of ≈5 % at -0.33 V (vs. RHE).   18. False negative rate and other performance measures of a sponge-wipe surface sampling method for low contaminant concentrations.

    Recovery of spores from environmental surfaces is known to vary due to sampling methodology, techniques, spore size and characteristics, surface materials, and environmental conditions. A series of tests were performed to evaluate a new, validated sponge-wipe method. Specific factors evaluated were the effects of contaminant concentrations and surface materials on recovery efficiency (RE), false negative rate (FNR), limit of detection (LOD) - and the uncertainties of these quantities. Ceramic tile and stainless steel had the highest mean RE values (48.9 and 48.1%, respectively). Faux leather, vinyl tile, and painted wood had mean RE values of 30.3, 25.6, and 25.5, respectively, while plastic had the lowest mean RE (9.8%). Results show a roughly linear dependence of surface roughness on RE, where the smoothest surfaces have the highest mean RE values. REs were not influenced by the low spore concentrations tested (3 x 10{sup -3} to 1.86 CFU/cm{sup 2}). The FNR data were consistent with RE data, showing a trend of smoother surfaces resulting in higher REs and lower FNRs. Stainless steel generally had the lowest mean FNR (0.123) and plastic had the highest mean FNR (0.479). The LOD{sub 90} varied with surface material, from 0.015 CFU/cm{sup 2} on stainless steel up to 0.039 on plastic. Selecting sampling locations on the basis of surface roughness and using roughness to interpret spore recovery data can improve sampling. Further, FNR values, calculated as a function of concentration and surface material, can be used pre-sampling to calculate the numbers of samples for statistical sampling plans with desired performance, and post-sampling to calculate the confidence in characterization and clearance decisions.

    A Benthic Flux Sampling Device (BFSD) was demonstrated on site to determine the mobility of contaminants in sediments off the Puget Sound Naval Shipyard (PSNS) in Sinclair Inlet, WA. Quantification of toxicant flux from the sediments will support ongoing assessment studies and facilitate the design of appropriate remediation strategies, if required. In general, where release of contaminants was found, the measured rates do not represent a significant source relative to other major inputs such as sewer discharges, nonpoint source runoff, and marinas. They may, however, represent an exposure pathway for benthic biota with a subsequent potential for toxicological effects and/or bioaccumulation.     Acetaldehyde has been used as a fluorescent dopant for two-dimensional imaging of the flame front in an internal-combustion-engine simulator. The molecule was excited with a XeCl-laser-light sheet at 308 nm, and broadband fluorescence centered at 400 nm was detected. In this way, the flame front could be marked by mapping regions of unburned gas. Also, the intake process into the engine could be followed.

    Preliminary process economics data for the electrogenerative process to manufacture acetaldehyde from ethanol were generated based on patent information. The technology was assessed in four alternative processing options. The Electrohol process is viable in the US only if integrated to the production of 190 pf ethanol from corn in a large scale unit. To be competitive, the Electrohol process must show yields in excess of 93%. Its attractiveness depends on corn prices remaining under $2.90/bu and DDG selling for more than $132/T. A corn price of $2.00/bu is needed to make a farm-size corn-based processing alternative competitive. A plant based on the fermentation of molasses proved too expensive under the US economic assumptions. The Electrohol technology based on purchased ethanol cannot compete with the existing ethylene-based process under current conditions. To become attractive, the Electrohol process must have access to cheap ethanol ($1.43/gal). The zero electricity generation mode is the most attractive mode of operation for the Electrohol technology in the US. The penalty for low levels of generation (0.130 kwh/kg AcH) is, however, negligible. The optimum operating mode in W. Europe is the generation of 0.312 kwh/kg AcH. In Japan, the low generation level is perferred (0.130 kwh/kg AcH). In general, higher energy prices improve the competitiveness of the Electrohol processing alternatives.

  2. Insights into photodissociation dynamics of acetaldehyde from ab initio calculations and molecular dynamics simulations

    In the present paper we report a theoretical study on mechanistic photodissociation of acetaldehyde (CH{sub 3}CHO). Stationary structures for H{sub 2} and CO eliminations in the ground state (S{sub 0}) have been optimized with density functional theory method, which is followed by the intrinsic reaction coordinate and ab initio molecular dynamics calculations to confirm the elimination mechanism. Equilibrium geometries, transition states, and intersection structures for the C-C and C-H dissociations in excited states were determined by the complete-active-space self-consistent field (CASSCF) method. Based on the CASSCF optimized structures, the potential energy profiles for the dissociations were refined by performing the single-point calculations using the multireference configuration interaction method. Upon the low-energy irradiation of CH{sub 3}CHO (265 nm<{lambda}<318 nm), the T{sub 1} C-C bond fission following intersystem crossing from the S{sub 1} state is the predominant channel and the minor channel, the ground-state elimination to CH{sub 4}+CO after internal conversion (IC) from S{sub 1} to S{sub 0}, could not be excluded. With the photon energy increasing, another pathway of IC, achieved via an S{sub 1}/S{sub 0} intersection point resulting from the S{sub 1} C-C bond fission, becomes accessible and increases the yield of CH{sub 4}+CO.

    The objective of this work was to develop and evaluate a series of methods and validate their capability to measure differences in oxidized versus reduced saltstone. Validated methods were then applied to samples cured under field conditions to simulate Performance Assessment (PA) needs for the Saltstone Disposal Facility (SDF). Four analytical approaches were evaluated using laboratory-cured saltstone samples. These methods were X-ray absorption spectroscopy (XAS), diffuse reflectance spectroscopy (DRS), chemical redox indicators, and thin-section leaching methods. XAS and thin-section leaching methods were validated as viable methods for studying oxidation movement in saltstone. Each method used samples that were spiked with chromium (Cr) as a tracer for oxidation of the saltstone. The two methods were subsequently applied to field-cured samples containing chromium to characterize the oxidation state of chromium as a function of distance from the exposed air/cementitious material surface.

  4. Photochemical fabrication of size-controllable gold nanoparticles on chitosan and their application on catalytic decomposition of acetaldehyde

    In this work, we report a new pathway to prepare size-controllable gold nanoparticles (NPs) on chitosan (Ch) in aqueous solutions for improving catalytic decomposition of acetaldehyde by pure gold NPs at room temperature. First, Au substrates were cycled in deoxygenated aqueous solutions containing 0.1N NaCl and 1 g/L Ch from -0.28 to +1.22 V vs Ag/AgCl at 500 mV/s for 200 scans. Then the solutions were irradiated with UV lights of different wavelengths to prepare size-controllable Au NPs on Ch. Experimental results indicate that the particle sizes of prepared NPs are increased when UV lights with longer wavelengths were employed. The particle sizes of resulted Au NPs can be controlled from 10 to 50 nm.     These catalytic C–C coupling and deoxygenation reactions are essential for upgrading of biomass-derived oxygenates to fuel-range hydrocarbons. Detailed understanding of mechanistic and energetic aspects of these reactions is crucial to enabling and improving the catalytic upgrading of small oxygenates to useful chemicals and fuels. Using periodic density functional theory (DFT) calculations, we have investigated the reactions of furan and acetaldehyde in an HZSM-5 zeolite catalyst, a representative system associated with the catalytic upgrading of pyrolysis vapors. Comprehensive energy profiles were computed for self-reactions (i.e., acetaldehyde coupling and furan coupling) and cross-reactions (i.e., acetaldehyde + furan) of this representative mixture. Major products proposed from the computations are further confirmed using temperature controlled mass spectra measurements. Moreover, the computational results show that furan interacts with acetaldehyde in HZSM-5 via an alkylation mechanism, which is more favorable than the self-reactions, indicating that mixing furans with aldehydes could be a promising approach to maximize effective C–C coupling and dehydration while reducing the catalyst deactivation (e.g., coke formation) from aldehyde condensation.

  6. Recovery Efficiency, False Negative Rate, and Limit of Detection Performance of a Validated Macrofoam-Swab Sampling Method with Low Surface Concentrations of Two Bacillus anthracis Surrogates

    The performance of a macrofoam-swab sampling method was evaluated using Bacillus anthracis Sterne (BAS) and Bacillus atrophaeus Nakamura (BG) spores applied at nine low target amounts (2-500 spores) to positive-control plates and test coupons (2 in. × 2 in.) of four surface materials (glass, stainless steel, vinyl tile, and plastic). Test results from cultured samples were used to evaluate the effects of surrogate, surface concentration, and surface material on recovery efficiency (RE), false negative rate (FNR), and limit of detection. For RE, surrogate and surface material had statistically significant effects, but concentration did not. Mean REs were the lowest for vinyl tile (50.8% with BAS, 40.2% with BG) and the highest for glass (92.8% with BAS, 71.4% with BG). FNR values ranged from 0 to 0.833 for BAS and 0 to 0.806 for BG, with values increasing as concentration decreased in the range tested (0.078 to 19.375 CFU/cm2, where CFU denotes ‘colony forming units’). Surface material also had a statistically significant effect. A FNR-concentration curve was fit for each combination of surrogate and surface material. For both surrogates, the FNR curves tended to be the lowest for glass and highest for vinyl title. The FNR curves for BG tended to be higher than for BAS at lower concentrations, especially for glass. Results using a modified Rapid Viability-Polymerase Chain Reaction (mRV-PCR) analysis method were also obtained. The mRV-PCR results and comparisons to the culture results will be discussed in a subsequent report.

  7. False Negative Rates of a Macrofoam-Swab Sampling Method with Low Surface Concentrations of Two Bacillus anthracis Surrogates via Real-Time PCR

    Surface sampling for Bacillus anthracis spores has traditionally relied on detection via bacterial cultivation methods. Although effective, this approach does not provide the level of organism specificity that can be gained through molecular techniques. False negative rates (FNR) and limits of detection (LOD) were determined for two B. anthracis surrogates with modified rapid viability-polymerase chain reaction (mRV-PCR) following macrofoam-swab sampling. This study was conducted in parallel with a previously reported study that analyzed spores using a plate-culture method. B. anthracis Sterne (BAS) or B. atrophaeus Nakamura (BG) spores were deposited onto four surface materials (glass, stainless steel, vinyl tile, and plastic) at nine target concentrations (2 to 500 spores/coupon; 0.078 to 19.375 colony-forming units [CFU] per cm²). Mean FNR values for mRV-PCR analysis ranged from 0 to 0.917 for BAS and 0 to 0.875 for BG and increased as spore concentration decreased (over the concentrations investigated) for each surface material. FNRs based on mRV-PCR data were not statistically different for BAS and BG, but were significantly lower for glass than for vinyl tile. FNRs also tended to be lower for the mRV-PCR method compared to the culture method. The mRV-PCR LOD₉₅ was lowest for glass (0.429 CFU/cm² with BAS and 0.341 CFU/cm² with BG) and highest for vinyl tile (0.919 CFU/cm² with BAS and 0.917 CFU/cm² with BG). These mRV-PCR LOD₉₅ values were lower than the culture values (BAS: 0.678 to 1.023 CFU/cm² and BG: 0.820 to 1.489 CFU/cm²). The FNR and LOD₉₅ values reported in this work provide guidance for environmental sampling of Bacillus spores at low concentrations.

    Growing observational evidence indicates that nebular line emission has a significant impact on the rest-frame optical fluxes of z ∼ 5-7 galaxies. This line emission makes z ∼ 5-7 galaxies appear more massive, with lower specific star-formation rates (sSFRs). However, corrections for this line emission have been difficult to perform reliably because of huge uncertainties on the strength of such emission at z ≳ 5.5. In this paper, we present the most direct observational evidence thus far for ubiquitous high-equivalent-width (EW) [O III] + Hβ line emission in Lyman-break galaxies at z ∼ 7, and we present a strategy for an improved measurement of the sSFR at z ∼ 7. We accomplish this through the selection of bright galaxies in the narrow redshift window z ∼ 6.6-7.0 where the Spitzer/Infrared Array Camera (IRAC) 4.5 μm flux provides a clean measurement of the stellar continuum light, in contrast with the 3.6 μm flux, which is contaminated by the prominent [O III] + Hβ lines. To ensure a high signal-to-noise ratio for our IRAC flux measurements, we consider only the brightest (H {sub 160} < 26 mag) magnified galaxies we have identified behind galaxy clusters. It is remarkable that the mean rest-frame optical color for our bright seven-source sample is very blue, [3.6]-[4.5] = –0.9 ± 0.3. Such blue colors cannot be explained by the stellar continuum light and require that the rest-frame EW of [O III] + Hβ is greater than 637 Å for the average source. The four bluest sources from our seven-source sample require an even more extreme EW of 1582 Å. We can also set a robust lower limit of ≳ 4 Gyr{sup –1} on the sSFR of our sample based on the mean spectral energy distribution.

    This field study measured ventilation rates and indoor air quality parameters in 21 visits to retail stores in California. The data was collected to guide the development of new, science-based commercial building ventilation rate standards that balance the dual objectives of increasing energy efficiency and maintaining acceptable indoor air quality. Data collection occurred between September 2011 and March 2013. Three types of stores participated in this study: grocery stores, furniture/hardware stores, and apparel stores. Ventilation rates and indoor air contaminant concentrations were measured on a weekday, typically between 9 am and 6 pm. Ventilation rates measured using a tracer gas decay method exceeded the minimum requirement of California’s Title 24 Standard in all but one store. Even though there was adequate ventilation according to Title 24, concentrations of formaldehyde, acetaldehyde, and acrolein exceeded the most stringent chronic health guidelines. Other indoor air contaminants measured included carbon dioxide (CO{sub 2}), carbon monoxide (CO), ozone (O{sub 3}), and particulate matter (PM). Concentrations of CO{sub 2} were kept low by adequate ventilation, and were assumed low also because the sampling occurred on a weekday when retail stores were less busy. CO concentrations were also low. The indoor-outdoor ratios of O{sub 3} showed that the first-order loss rate may vary by store trade types and also by ventilation mode (mechanical versus natural). Analysis of fine and ultrafine PM measurements showed that a substantial portion of the particle mass in grocery stores with cooking-related emissions was in particles less than 0.3 μm. Stores without cooking as an indoor source had PM size distributions that were more similar indoors and outdoors. The whole-building emission rates of volatile organic compounds (VOCs) and PM were estimated from the measured ventilation rates and indoor and outdoor contaminant concentrations. A variable voltage-level shifter and a time-scale rampvoltage generator take the pulse height relative to the start of the waveform.     The laser induced {ital T}{sub 1}({ital n}{pi}{asterisk}){l_arrow}{ital S}{sub 0} phosphorescence excitation spectrum of jet-cooled acetaldehyde has been observed for the first time with a rotating slit nozzle excitation system. The vibronic origins were fitted to a set of levels that were obtained from a Hamiltonian that employed flexible torsion-wagging large amplitude coordinates. The potential surface extracted from the fitting procedure yielded barriers to torsion and inversion of 609.68 and 869.02 cm{sup {minus}1}, respectively. Minima in the potential hypersurface at {theta}=61.7{degree} and {alpha}=42.2{degree} defined the equilibrium positions for the torsion and wagging coordinates. A comparison to the corresponding {ital S}{sub 1}-state parameters showed that the torsion barrier (in cm{sup {minus}1}) does not greatly change, {ital S}{sub 1}/{ital T}{sub 1}=710.8/609.7, whereas the barrier height for the wagging-inversion barrier increases dramatically, 574.4/869.0. {copyright} {ital 1996 American Institute of Physics.}

    A system for sampling air and collecting particulate of a predetermined particle size range. A low pass section has an opening of a preselected size for gathering the air but excluding particles larger than the sample particles. An impactor section is connected to the low pass section and separates the air flow into a bypass air flow that does not contain the sample particles and a product air flow that does contain the sample particles. A wetted-wall cyclone collector, connected to the impactor section, receives the product air flow and traps the sample particles in a liquid.

    The current format of the OPCW proficiency tests has multiple sets of 2 samples sent to an analysis laboratory. In each sample set, one is identified as a sample, the other as a blank. This method of conducting proficiency tests differs from how an OPCW designated laboratory would receive authentic samples (a set of three containers, each not identified, consisting of the authentic sample, a control sample, and a blank sample). This exercise was designed to test the reporting if the proficiency tests were to be conducted. As such, this is not an official OPCW proficiency test, and the attached report is one method by which LLNL might report their analyses under a more realistic testing scheme. Therefore, the title on the report ''Report of the Umpteenth Official OPCW Proficiency Test'' is meaningless, and provides a bit of whimsy for the analyses and readers of the report.

Analyte/Combination (AnalCombo) A list of analytes and combinations of analytes that can be requested of the onsite and offsite labs. Whenever a specific combination of analytes or suite names appear on the same line as the code number, this indicates that one sample can be placed in one bottle to bemore » analyzed for these paremeters. Sampling Plans Database (SPDb) A database that contains all of the analytes and combinations of analytes along with the basic information required for preparing a sample plan. A biological sample collector may comprise a manifold plate for mounting a filter plate thereon, the filter plate having a plurality of filter wells therein; a hollow slider for engaging and positioning a tube that slides therethrough; and a slide case within which the hollow slider travels to allow the tube to be aligned with a selected filter well of the plurality of filter wells, wherein when the tube is aligned with the selected filter well, the tube is pushed through the hollow slider and into the selected filter well to sealingly engage the selected filter well and to allow the tube to deposit a biological sample onto a filter in the bottom of the selected filter well. The biological sample collector may be portable.

    A vertical core sample collection system for viscous sludge. A sample tube's upper end has a flange and is attached to a piston. The tube and piston are located in the upper end of a bore in a housing. The bore's lower end leads outside the housing and has an inwardly extending rim. Compressed gas, from a storage cylinder, is quickly introduced into the bore's upper end to rapidly accelerate the piston and tube down the bore. The lower end of the tube has a high sludge entering velocity to obtain a full-length sludge sample without disturbing strata detail. The tube's downward motion is stopped when its upper end flange impacts against the bore's lower end inwardly extending rim.

    The current focal point of the nuclear physics program at PNNL is the MAJORANA DEMONSTRATOR, and the follow-on Tonne-Scale experiment, a large array of ultra-low background high-purity germanium detectors, enriched in 76Ge, designed to search for zero-neutrino double-beta decay (0νββ). This experiment requires the use of germanium isotopically enriched in 76Ge. The MAJORANA DEMONSTRATOR is a DOE and NSF funded project with a major science impact. The DEMONSTRATOR will utilize 76Ge from Russia, but for the Tonne-Scale experiment it is hoped that an alternate technology, possibly one under development at Nonlinear Ion Dynamics (NID), will be a viable, US-based, lower-cost source of separated material. Samples of separated material from NID require analysis to determine the isotopic distribution and impurities. DOE is funding NID through an SBIR grant for development of their separation technology for application to the Tonne-Scale experiment. The Environmental Molecular Sciences facility (EMSL), a DOE user facility at PNNL, has the required mass spectroscopy instruments for making isotopic measurements that are essential to the quality assurance for the MAJORANA DEMONSTRATOR and for the development of the future separation technology required for the Tonne-Scale experiment. A sample of isotopically separated copper was provided by NID to PNNL in January 2011 for isotopic analysis as a test of the NID technology. The results of that analysis are reported here.     A fluid sampling tool is described for sampling fluid from a container. The tool has a fluid collecting portion which is drilled into the container wall, thereby affixing it to the wall. The tool may have a fluid extracting section which withdraws fluid collected by the fluid collecting section. The fluid collecting section has a fluted shank with an end configured to drill a hole into a container wall. The shank has a threaded portion for tapping the borehole. The shank is threadably engaged to a cylindrical housing having an inner axial passageway sealed at one end by a septum. A flexible member having a cylindrical portion and a bulbous portion is provided. The housing can be slid into an inner axial passageway in the cylindrical portion and sealed to the flexible member. The bulbous portion has an outer lip defining an opening. The housing is clamped into the chuck of a drill, the lip of the bulbous section is pressed against a container wall until the shank touches the wall, and the user operates the drill. Wall shavings (kerf) are confined in a chamber formed in the bulbous section as it folds when the shank advances inside the container. After sufficient advancement of the shank, an o-ring makes a seal with the container wall. 6 figs.

    The invention includes a rotatable tool for collecting fluid through the wall of a container. The tool includes a fluid collection section with a cylindrical shank having an end portion for drilling a hole in the container wall when the tool is rotated, and a threaded portion for tapping the hole in the container wall. A passageway in the shank in communication with at least one radial inlet hole in the drilling end and an opening at the end of the shank is adapted to receive fluid from the container. The tool also includes a cylindrical chamber affixed to the end of the shank opposite to the drilling portion thereof for receiving and storing fluid passing through the passageway. The tool also includes a flexible, deformable gasket that provides a fluid-tight chamber to confine kerf generated during the drilling and tapping of the hole. The invention also includes a fluid extractor section for extracting fluid samples from the fluid collecting section.

    We report deflagration rates on EDC-37 at high pressures. Experiments are conducted using the Lawrence Livermore National Laboratory High Pressure Strand Burner (HPSB) apparatus. The HPSB contains a deflagrating sample in a small volume, high pressure chamber. The sample consists of nine, 6.35 mm diameter, 6.35 mm length cylinders stacked on end, with burn wires placed between cylinders. Sample deflagration is limited to the cross-sectional surface of the cylinder by coating the cylindrical surface of the tower with Halthane 88-2 epoxy. Sample deflagration is initiated on one end of the tower by a B/KNO{sub 3} and HNS igniter train. Simultaneous temporal pressure history and burn front time of arrival measurements yield the laminar deflagration rate for a range of pressures and provide insight into deflagration uniformity. These measurements are one indicator of overall thermal explosion violence. Specific details of the experiment and the apparatus can be found in the literature.

The Specified Assurance Level (SAL) Sampling Procedure has been developed to permit the direct selection of attribute sampling plans which can meet commonly used assurance levels.     We present a Bayesian model for acceptance sampling where the population consists of two groups, each with different levels of risk of containing unacceptable items. Expert opinion, or judgment, may be required to distinguish between the high and low-risk groups. Hence, high-risk items are likely to be identifed (and sampled) using expert judgment, while the remaining low-risk items are sampled randomly. We focus on the situation where all observed samples must be acceptable. Consequently, the objective of the statistical inference is to quantify the probability that a large percentage of the unsampled items in the population are also acceptable. We demonstrate that traditional (frequentist) acceptance sampling and simpler Bayesian formulations of the problem are essentially special cases of the proposed model. We explore the properties of the model in detail, and discuss the conditions necessary to ensure that required samples sizes are non-decreasing function of the population size. The method is applicable to a variety of acceptance sampling problems, and, in particular, to environmental sampling where the objective is to demonstrate the safety of reoccupying a remediated facility that has been contaminated with a lethal agent.

    A compact pulse-rate divider circuit affording low impedance output and high input pulse repetition rates is described. The circuit features a single secondary emission tube having a capacitor interposed between its dynode and its control grid. An output pulse is produced at the anode of the tube each time an incoming pulse at the control grid drives the tube above cutoff and the duration of each output pulse corresponds to the charging time of the capacitor. Pulses incoming during the time the grid bias established by the discharging capacitor is sufficiently negative that the pulses are unable to drive the tube above cutoff do not produce output pulses at the anode; these pulses are lost and a dividing action is thus produced by the circuit.     Environmental surveillance of the Hanford Site and surrounding areas is conducted by the Pacific Northwest Laboratory (PNL) for the U.S. Department of Energy (DOE). This document contains the planned 1994 schedules for routine collection of samples for the Surface Environmental Surveillance Project (SESP), Drinking Water Project, and Ground-Water Surveillance Project. Samples are routinely collected for the SESP and analyzed to determine the quality of air, surface water, soil, sediment, wildlife, vegetation, foodstuffs, and farm products at Hanford Site and surrounding communities. The responsibility for monitoring onsite drinking water falls outside the scope of the SESP. PNL conducts the drinking water monitoring project concurrent with the SESP to promote efficiency and consistency, utilize expertise developed over the years, and reduce costs associated with management, procedure development, data management, quality control, and reporting. The ground-water sampling schedule identifies ground-water sampling .events used by PNL for environmental surveillance of the Hanford Site. Sampling is indicated as annual, semi-annual, quarterly, or monthly in the sampling schedule. Some samples are collected and analyzed as part of ground-water monitoring and characterization programs at Hanford (e.g. Resources Conservation and Recovery Act (RCRA), Comprehensive Environmental Response, Compensation, and Liability Act (CERCLA), or Operational). The number of samples planned by other programs are identified in the sampling schedule by a number in the analysis column and a project designation in the Cosample column. Well sampling events may be merged to avoid redundancy in cases where sampling is planned by both-environmental surveillance and another program.

    The Environmental Protection Agency (EPA) prescribed methodologies for sampling radionuclides in air effluents from stacks and ducts at US Department of Energy (DOE) facilities. Requirements include use of EPA Method 1 for the location of sampling sites and use of American National Standards Institute (ANSI) N13.1 for guidance in design of sampling probes and the number of probes at a given site. Application of ANSI N13.1 results in sampling being performed with multiprobe rakes that have as many as 20 probes. There can be substantial losses of aerosol particles in such sampling that will degrade the quality of emission estimates from a nuclear facility. Three alternate methods, technically justified herein, are proposed for effluent sampling. First, a shrouded aerosol sampling probe should replace the sharp-edged elbowed-nozzle recommended by ANSI. This would reduce the losses of aerosol particles in probes and result in the acquisition of more representative aerosol samples. Second, the rakes of multiple probes that are intended to acquire representative samples through spatial coverage should be replaced by a single probe located where contaminant mass and fluid momentum are both well mixed. A representative sample can be obtained from a well-mixed flow. Some effluent flows will need to be engineered to achieve acceptable mixing. Third, sample extraction should be performed at a constant flow rate through a suitable designed shrouded probe rather than at a variable flow rate through isokinetic probes. A shrouded probe is shown to have constant sampling characteristics over a broad range of stack velocities when operated at a fixed flow rate.

    This invention generally relates to the remote removal of pelletized samples from cylindrical containment capsules. V-blocks are used to receive the samples and provide guidance to push out rods. Stainless steel liners fit into the v-channels on the v-blocks which permits them to be remotely removed and replaced or cleaned to prevent cross contamination between capsules and samples. A capsule holder securely holds the capsule while allowing manual up/down and in/out movement to align each sample hole with the v-blocks. Both end sections contain identical v-blocks; one that guides the drive out screw and rods or manual push out rods and the other to receive the samples as they are driven out of the capsule.

    An improved apparatus is provided for sampling a gaseous mixture and for measuring mixture components. The apparatus includes two sampling containers connected in series serving as a duplex sampling apparatus. The apparatus is adapted to independently determine the amounts of condensable and noncondensable gases in admixture from a single sample. More specifically, a first container includes a first port capable of selectively connecting to and disconnecting from a sample source and a second port capable of selectively connecting to and disconnecting from a second container. A second container also includes a first port capable of selectively connecting to and disconnecting from the second port of the first container and a second port capable of either selectively connecting to and disconnecting from a differential pressure source. By cooling a mixture sample in the first container, the condensable vapors form a liquid, leaving noncondensable gases either as free gases or dissolved in the liquid. The condensed liquid is heated to drive out dissolved noncondensable gases, and all the noncondensable gases are transferred to the second container. Then the first and second containers are separated from one another in order to separately determine the amount of noncondensable gases and the amount of condensable gases in the sample.

Changes will be documented in the respective project files, but this plan will not be reissued.   14. Defining And Characterizing Sample Representativeness For DWPF Melter Feed Samples

    Representative sampling is important throughout the Defense Waste Processing Facility (DWPF) process, and the demonstrated success of the DWPF process to achieve glass product quality over the past two decades is a direct result of the quality of information obtained from the process. The objective of this report was to present sampling methods that the Savannah River Site (SRS) used to qualify waste being dispositioned at the DWPF. The goal was to emphasize the methodology, not a list of outcomes from those studies. This methodology includes proven methods for taking representative samples, the use of controlled analytical methods, and data interpretation and reporting that considers the uncertainty of all error sources. Numerous sampling studies were conducted during the development of the DWPF process and still continue to be performed in order to evaluate options for process improvement. Study designs were based on use of statistical tools applicable to the determination of uncertainties associated with the data needs. Successful designs are apt to be repeated, so this report chose only to include prototypic case studies that typify the characteristics of frequently used designs. Case studies have been presented for studying in-tank homogeneity, evaluating the suitability of sampler systems, determining factors that affect mixing and sampling, comparing the final waste glass product chemical composition and durability to that of the glass pour stream sample and other samples from process vessels, and assessing the uniformity of the chemical composition in the waste glass product. Many of these studies efficiently addressed more than one of these areas of concern associated with demonstrating sample representativeness and provide examples of statistical tools in use for DWPF. The time when many of these designs were implemented was in an age when the sampling ideas of Pierre Gy were not as widespread as they are today. Nonetheless, the engineers and

    Savannah River National Laboratory (SRNL) analyzed samples from Tank 48H in support of determining the cause for the unusually high dose rates at the sampling points for this tank. A set of two samples was taken from the quiescent tank, and two additional samples were taken after the contents of the tank were mixed. The results of the analyses of all the samples show that the contents of the tank have changed very little since the analysis of the previous sample in 2012. The solids are almost exclusively composed of tetraphenylborate (TPB) salts, and there is no indication of acceleration in the TPB decomposition. The filtrate composition shows a moderate increase in salt concentration and density, which is attributable to the addition of NaOH for the purposes of corrosion control. An older modeling simulation of the TPB degradation was updated, and the supernate results from a 2012 sample were run in the model. This result was compared to the results from the 2014 recent sample results reported in this document. The model indicates there is no change in the TPB degradation from 2012 to 2014. SRNL measured the buoyancy of the TPB solids in Tank 48H simulant solutions. It was determined that a solution of density 1.279 g/mL (~6.5M sodium) was capable of indefinitely suspending the TPB solids evenly throughout the solution. A solution of density 1.296 g/mL (~7M sodium) caused a significant fraction of the solids to float on the solution surface. As the experiments could not include the effect of additional buoyancy elements such as benzene or hydrogen generation, the buoyancy measurements provide an upper bound estimate of the density in Tank 48H required to float the solids.

The sampling fluid exhaust conduit has an inlet opening for receiving sampling fluid carried from the surface by the sampling sphere. A surface sampling probe and a method for sampling a surface are also disclosed.

    We present an analytical study of peak mode isotachophoresis (ITP), and provide closed form solutions for sample distribution and electric field, as well as for leading-, trailing-, and counter-ion concentration profiles. Importantly, the solution we present is valid not only for the case of fully ionized species, but also for systems of weak electrolytes which better represent real buffer systems and for multivalent analytes such as proteins and DNA. The model reveals two major scales which govern the electric field and buffer distributions, and an additional length scale governing analyte distribution. Using well-controlled experiments, and numerical simulations, we verify and validate the model and highlight its key merits as well as its limitations. We demonstrate the use of the model for determining the peak concentration of focused sample based on known buffer and analyte properties, and show it differs significantly from commonly used approximations based on the interface width alone. We further apply our model for studying reactions between multiple species having different effective mobilities yet co-focused at a single ITP interface. We find a closed form expression for an effective-on rate which depends on reactants distributions, and derive the conditions for optimizing such reactions. Interestingly, the model reveals that maximum reaction rate is not necessarily obtained when the concentration profiles of the reacting species perfectly overlap. In addition to the exact solutions, we derive throughout several closed form engineering approximations which are based on elementary functions and are simple to implement, yet maintain the interplay between the important scales. Both the exact and approximate solutions provide insight into sample focusing and can be used to design and optimize ITP-based assays.

    Tank Characterization Technical Sampling Basis (this document) is the first step of an in place working process to plan characterization activities in an optimal manner. This document will be used to develop the revision of the Waste Information Requirements Document (WIRD) (Winkelman et al. 1997) and ultimately, to create sampling schedules. The revised WIRD will define all Characterization Project activities over the course of subsequent fiscal years 1999 through 2002. This document establishes priorities for sampling and characterization activities conducted under the Tank Waste Remediation System (TWRS) Tank Waste Characterization Project. The Tank Waste Characterization Project is designed to provide all TWRS programs with information describing the physical, chemical, and radiological properties of the contents of waste storage tanks at the Hanford Site. These tanks contain radioactive waste generated from the production of nuclear weapons materials at the Hanford Site. The waste composition varies from tank to tank because of the large number of chemical processes that were used when producing nuclear weapons materials over the years and because the wastes were mixed during efforts to better use tank storage space. The Tank Waste Characterization Project mission is to provide information and waste sample material necessary for TWRS to define and maintain safe interim storage and to process waste fractions into stable forms for ultimate disposal. This document integrates the information needed to address safety issues, regulatory requirements, and retrieval, treatment, and immobilization requirements. Characterization sampling to support tank farm operational needs is also discussed.

  2. Sample Results from Routine Salt Batch 7 Samples

    Strip Effluent Hold Tank (SEHT) and Decontaminated Salt Solution Hold Tank (DSSHT) samples from several of the microbatches of Integrated Salt Disposition Project (ISDP) Salt Batch (Macrobatch) 7B have been analyzed for 238Pu, 90Sr, 137Cs, Inductively Coupled Plasma Emission Spectroscopy (ICPES), and Ion Chromatography Anions (IC-A). The results from the current microbatch samples are similar to those from earlier samples from this and previous macrobatches. The Actinide Removal Process (ARP) and the Modular Caustic-Side Solvent Extraction Unit (MCU) continue to show more than adequate Pu and Sr removal, and there is a distinct positive trend in Cs removal, due to the use of the Next Generation Solvent (NGS). The Savannah River National Laboratory (SRNL) notes that historically, most measured Concentration Factor (CF) values during salt processing have been in the 12-14 range. However, recent processing gives CF values closer to 11. This observation does not indicate that the solvent performance is suffering, as the Decontamination Factor (DF) has still maintained consistently high values. Nevertheless, SRNL will continue to monitor for indications of process upsets. The bulk chemistry of the DSSHT and SEHT samples do not show any signs of unusual behavior.

    When modeling solar energy technologies and other distributed energy systems, using high-quality expansive electricity rates is essential. The National Renewable Energy Laboratory (NREL) developed a utility rate platform for entering, storing, updating, and accessing a large collection of utility rates from around the United States. This utility rate platform lives on the Open Energy Information (OpenEI) website, OpenEI.org, allowing the data to be programmatically accessed from a web browser, using an application programming interface (API). The semantic-based utility rate platform currently has record of 1,885 utility rates and covers over 85% of the electricity consumption in the United States.

    The need for adaptive sampling arises in the context of high throughput data because the rates of data arrival are many orders of magnitude larger than the rates at which they can be analyzed. A very fast decision must therefore be made regarding the value of each incoming observation and its inclusion in the analysis. In this report we discuss one approach to adaptive sampling, based on the new data point’s similarity to the other data points being considered for inclusion. We present preliminary results for one real and one synthetic data set.

An understanding of the mechanisms of these processes is required for final resolution of the safety issue. Central to understanding is gathering information from such sources as historical records, tank sampling data, tank process data (temperatures, ventilation rates, etc.), and laboratory evaluations conducted on tank waste samples.

    A vertical core sample collection system for viscous sludge is disclosed. A sample tube's upper end has a flange and is attached to a piston. The tube and piston are located in the upper end of a bore in a housing. The bore's lower end leads outside the housing and has an inwardly extending rim. Compressed gas, from a storage cylinder, is quickly introduced into the bore's upper end to rapidly accelerate the piston and tube down the bore. The lower end of the tube has a high sludge entering velocity to obtain a full-length sludge sample without disturbing strata detail. The tube's downward motion is stopped when its upper end flange impacts against the bore's lower end inwardly extending rim.
