    2012-08-01

SL-01, an oral gemcitabine derivative, was synthesized by introducing the moiety of 3-(dodecyloxycarbonyl)pyrazine-2-carbonyl at the N4-position on the cytidine ring of gemcitabine. Our goal in this study was to evaluate the efficacy of SL-01 on the growth of human cancers with gemcitabine as control. Experiments were performed on human non-small cell lung cancer NCI-H460 and colon cancer HCT-116 both in vitro and in vivo. In vitro assays, SL-01 significantly inhibited the growth of cancer cells as determined by the 3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide (MTT) assay. Further studies indicated that SL-01 induced the cancer cells to apoptosis showing chromatin condensation and externalization of phosphatidylserine. In in vivo studies, we evaluated the efficacy of SL-01 in nude mice bearing human cancer xenografts. SL-01 effectively delayed the growth of NCI-H460 and HCT-116 without significant loss of body weight. Molecular analysis indicated that the high efficacy of SL-01 was associated with its ability to induce apoptosis as evidenced by increase of terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL) staining cells, activation of caspase-9, caspase-3 and cleaved poly ADP-ribose polymerase (PARP) in tumor tissues. SL-01 also increased Bax/Bcl-2 ratio in cancer cells. These biological activities of SL-01 were more potential than that of gemcitabine. Based on these in vitro and in vivo results, SL-01 is proposed as a potent oral anticancer agent that may supplant the use of gemcitabine in the clinic. -- Highlights: ► An oral gemcitabine derivative SL-01 was synthesized. ► The effects of SL-01 were evaluated and its efficacy was compared with gemcitabine. ► The biological activities of SL-01 were more potent than that of gemcitabine. ► SL-01 could replace gemcitabine for clinical use.

Highlights: •SL-01 is an oral derivative of gemcitabine. •SL-01 possessed activity against human breast cancer growth via apoptotic induction. •SL-01’s activity was more potently than that of gemcitabine. •SL-01 inhibited cancer growth without toxicity to mice. -- Abstract: SL-01 is an oral derivative of gemcitabine that was synthesized by introducing the moiety of 3-(dodecyloxycarbonyl) pyrazine-2-carbonyl at N4-position on cytidine ring of gemcitabine. We aimed to evaluate the efficacy of SL-01 on human breast cancer growth. SL-01 significantly inhibited MCF-7 proliferation as estimated by colorimetric assay. Flow cytometry assay indicated the apoptotic induction and cell cycle arrest in G1 phase. SL-01 modulated the expressions of p-ATM, p53 and p21 and decrease of cyclin D1 in MCF-7 cells. Further experiments were performed in a MCF-7 xenografts mouse model. SL-01 by oral administration strongly inhibited MCF-7 xenografts growth. This effect of SL-01 might arise from its roles in the induction of apoptosis. Immunohistochemistry assay showed the increase of TUNEL staining cells. Western blotting indicated the modulation of apoptotic proteins in SL-01-treated xenografts. During the course of study, there was no evidence of toxicity to mice. In contrast, the decrease of neutrophil cells in peripheral and increase of AST and ALT levels in serum were observed in the gemcitabine-treated mice. Conclusion: SL-01 possessed similar activity against human breast cancer growth with gemcitabine, whereas, with lower toxicity to gemcitabine. SL-01 is a potent oral agent that may supplant the use of gemcitabine.

For the SL(2,R) duality-invariant generalization of Maxwell electrodynamics in the presence of both electric and magnetic sources, we formulate a local, manifestly duality-symmetric, Zwanziger-type action by introducing a pair of four-potentials A{sup μ} and B{sup μ} in a judicious way. On the two potentials A{sup μ} and B{sup μ} the SL(2,R) duality transformation acts in a simple linear manner. In quantum theory including charged source fields, this action can be recast as a SL(2,Z)-invariant action. Also given is a Zwanziger-type action for SL(2,R) duality-invariant Born–Infeld electrodynamics which can be important for D-brane dynamics in string theory. -- Highlights: •We formulate a local, manifestly duality-symmetric, Zwanziger-type action. •Maxwell electrodynamics is generalized to include dilaton and axion fields. •SL(2,R) symmetry is manifest. •We formulate a local, manifestly duality-symmetric, nonlinear Born–Infeld action with SL(2,R) symmetry.

    2007-11-08

On January 3, 1961, at the National Reactor Testing Station, in Idaho Falls, Idaho, the Stationary Low Power Reactor No. 1 (SL-1) experienced a major nuclear excursion, killing three people, and destroying the reactor core. The SL-1 reactor, a 3 MW{sub t} boiling water reactor, was shut down and undergoing routine maintenance work at the time. This paper presents an analysis of the SL-1 reactor excursion using the RELAP5-3D thermal-hydraulic and nuclear analysis code, with the intent of simulating the accident from the point of reactivity insertion to destruction and vaporization of the fuel. Results are presented, along with a discussion of sensitivity to some reactor and transient parameters (many of the details are only known with a high level of uncertainty).

We construct the screening currents of the quantum superalgebra U{sub q}(sl-caret(N|1)) for an arbitrary level k{ne}-N+ 1. We show that these screening currents commute with the superalgebra modulo total difference. We propose bosonizations of the vertex operators by using the screening currents. We check that these vertex operators are the intertwiners among the Fock-Wakimoto representation and the typical representation for rank N Less-Than-Or-Slanted-Equal-To 4.

We explicitly determine all Rota-Baxter operators (of weight zero) on sl (2,C) under the Cartan-Weyl basis. For the skew-symmetric operators, we give the corresponding skew-symmetric solutions of the classical Yang-Baxter equation in sl (2,C), confirming the related study by Semenov-Tian-Shansky. In general, these Rota-Baxter operators give a family of solutions of the classical Yang-Baxter equation in the six-dimensional Lie algebra sl (2,C)⋉{sub ad{sup *}} sl (2,C){sup *}. They also give rise to three-dimensional pre-Lie algebras which in turn yield solutions of the classical Yang-Baxter equation in other six-dimensional Lie algebras.

    2013-02-15

The algebra H of the dual -1 Hahn polynomials is derived and shown to arise in the Clebsch-Gordan problem of sl{sub -1}(2). The dual -1 Hahn polynomials are the bispectral polynomials of a discrete argument obtained from the q{yields}-1 limit of the dual q-Hahn polynomials. The Hopf algebra sl{sub -1}(2) has four generators including an involution, it is also a q{yields}-1 limit of the quantum algebra sl{sub q}(2) and furthermore, the dynamical algebra of the parabose oscillator. The algebra H, a two-parameter generalization of u(2) with an involution as additional generator, is first derived from the recurrence relation of the -1 Hahn polynomials. It is then shown that H can be realized in terms of the generators of two added sl{sub -1}(2) algebras, so that the Clebsch-Gordan coefficients of sl{sub -1}(2) are dual -1 Hahn polynomials. An irreducible representation of H involving five-diagonal matrices and connected to the difference equation of the dual -1 Hahn polynomials is constructed.

    2009-12-15

In this paper we construct closed expressions that correspond to the topological charges of the various 1/2-BPS states of the maximal 10- and 11-dimensional supergravity theories. These expressions are related to the structure of the supersymmetry algebras in curved spacetimes. We mainly focus on IIB supergravity and 11-dimensional supergravity in a double M9-brane background, with an emphasis on the SL(2,R) multiplet structure of the charges and how these map between theories. This includes the charges corresponding to the multiplets of 7- and 9-branes in IIB. We find that examining the possible multiplet structures of the charges provides another tool for exploring the spectrum of BPS states that appear in these theories. As a prerequisite to constructing the charges we determine the field equations and multiplet structure of the 11-dimensional gauge potentials, extending previous results on the subject. The massive gauge transformations of the fields are also discussed. We also demonstrate how these massive gauge transformations are compatible with the construction of an SL(2,R) covariant kinetic term in the 11-dimensional Kaluza-Klein monopole worldvolume action.

U.S. Atomic Energy Commission (Idaho Operations Office) briefing about the SL-1 Nuclear Reactor Meltdown. The SL-1, or Stationary Low-Power Reactor Number One, was a United States Army experimental nuclear power reactor which underwent a steam explosion and meltdown on January 3, 1961, killing its three operators. The direct cause was the improper withdrawal of the central control rod, responsible for absorbing neutrons in the reactor core. The event is the only known fatal reactor accident in the United States. The accident released about 80 curies (3.0 TBq) of Iodine-131, which was not considered significant due to its location in a remote desert of Idaho. About 1,100 curies (41 TBq) of fission products were released into the atmosphere. The facility, located at the National Reactor Testing Station approximately 40 miles (64 km) west of Idaho Falls, Idaho, was part of the Army Nuclear Power Program and was known as the Argonne Low Power Reactor (ALPR) during its design and build phase. It was intended to provide electrical power and heat for small, remote military facilities, such as radar sites near the Arctic Circle, and those in the DEW Line. The design power was 3 MW (thermal). Operating power was 200 kW electrical and 400 kW thermal for space heating. In the accident, the core power level reached nearly 20 GW in just four milliseconds, precipitating the reactor accident and steam explosion.

We derive the matrix elements of generators of unitary irreducible representations of SL(2,C) with respect to basis states arising from a decomposition into irreducible representations of SU(1,1). This is done with regard to a discrete basis diagonalized by J{sup 3} and a continuous basis diagonalized by K{sup 1}, and for both the discrete and continuous series of SU(1,1). For completeness, we also treat the more conventional SU(2) decomposition as a fifth case. The derivation proceeds in a functional/differential framework and exploits the fact that state functions and differential operators have a similar structure in all five cases. The states are defined explicitly and related to SU(1,1) and SU(2) matrix elements.

Four tensor products of evaluation modules of the quantum affine algebra U{sub q}{sup Prime }sl-caret(2) obtained from the negative and positive series, the complementary and the strange series representations are investigated. Linear operators R(z) satisfying the intertwining property on finite linear combinations of the canonical basis elements of the tensor products are described in terms of two sets of infinite sums {l_brace}{tau}{sup (r,t)}{r_brace}{sub r,t Element-Of Z{sub {>=}{sub 0}}} and {l_brace}{tau}{sup (r,t)}{r_brace}{sub r,t Element-Of Z{sub {>=}{sub 0}}} involving big q{sup 2}-Jacobi functions or related nonterminating basic hypergeometric series. Inhomogeneous recurrence relations can be derived for both sets. Evaluations of the simplest sums provide the corresponding initial conditions. For the first set of sums the relations entail a big q{sup 2}-Jacobi function transform pair. An integral decomposition is obtained for the sum {tau}{sup (r,t)}. A partial description of the relation between the decompositions of the tensor products with respect to U{sub q}sl(2) or with respect to its complement in U{sub q}{sup Prime }sl-caret(2) can be formulated in terms of Askey-Wilson function transforms. For a particular combination of two tensor products, the occurrence of proper U{sub q}{sup Prime }sl-caret(2)-submodules is discussed.

Chemical reactions in starless molecular clouds are heavily dependent on interactions between gas phase material and solid phase dust and ices. We have observed the abundance and distribution of molecular gases in the cold, starless core DC 000.4-19.5 (SL42) in Corona Australis using data from the Swedish ESO Submillimeter Telescope. We present column density maps determined from measurements of C{sup 18}O (J = 2-1, 1-0) and N{sub 2}H{sup +} (J = 1-0) emission features. Herschel data of the same region allow a direct comparison to the dust component of the cloud core and provide evidence for gas phase depletion of CO at the highest extinctions. The dust color temperature in the core calculated from Herschel maps ranges from roughly 10.7 to 14.0 K. This range agrees with the previous determinations from Infrared Space Observatory and Planck observations. The column density profile of the core can be fitted with a Plummer-like density distribution approaching n(r) {approx} r {sup -2} at large distances. The core structure deviates clearly from a critical Bonnor-Ebert sphere. Instead, the core appears to be gravitationally bound and to lack thermal and turbulent support against the pressure of the surrounding low-density material: it may therefore be in the process of slow contraction. We test two chemical models and find that a steady-state depletion model agrees with the observed C{sup 18}O column density profile and the observed N(C{sup 18}O) versus A{sub V} relationship.

  5. Evidence of Progress - Measurement of Impacts of Australia's S&L Program from 1990-2010

    SciTech Connect (OSTI)

Lowenthal-Savy, Danielle; McNeil, Michael; Harrington, Lloyd

    2013-09-11

Australia first put categorical energy efficiency labels on residential appliances in the mid-1980s, and the first Minimum Energy Performance Standards (MEPS) for refrigerators was implemented in 1999. Updated in 2005, these MEPS were aligned with US 2001 levels. Considered together, these actions set Australia apart as having one of the most aggressive appliance efficiency programs in the world. For these reasons, together with good data on product sales over time, Australia represents a potentially fruitful case study for understanding the dynamics energy efficiency standards and labeling (EES&L) programs impacts on appliance markets. This analysis attempts to distinguish between the impacts of labeling alone as opposed to MEPS, and to probe the time-dependency of such impacts. Fortunately, in the Australian case, detailed market sales data and a comprehensive registration system provides a solid basis for the empirical evaluation of these questions. This paper analyzes Australian refrigerator efficiency data covering the years 1993-2009. Sales data was purchased from a commercial market research organization (in this case, the GfK Group) and includes sales and average price in each year for each appliance model; this can be used to understand broader trends by product class and star rating category, even where data is aggregated. Statistical regression analysis is used to model market introduction and adoption of high efficiency refrigerators according to logistic adoption model formalism, and parameterizes the way in which the Australian programs accelerated adoption of high-efficiency products and phased out others. Through this analysis, the paper presents a detailed, robust and quantitative picture of the impacts of EES&L in the Australian case, but also demonstrates a methodology of the evaluation of program impacts that could form the basis of an international evaluation framework for similar programs in other countries.

    1982-10-01

Fuel vaporization and combustion of the thereby achieved homogeneous mixtures improve the overall efficiency of SI engines in comparison to operation with liquid fuels. The improvements result from a recovery of waste heat and the thus achieved greater usable energy of the fuel, which is increased by the heat of vaporization over the lower calorific value of the liquid fuel, and from the fact that very lean mixtures can be burnt without misfiring. The favorable fuel economy of the air/fuel-vapor mixture-aspirating engine is explained with the aid of engine cycle computation which also enables comparison of different combustion processes. Consideration of common substances shows that methanol is the fuel best suited for this type of SI engine.

We report that Thermoanaerobacter saccharolyticum is a thermophilic microorganism that has been engineered to produce ethanol at high titer (30–70 g/L) and greater than 90 % theoretical yield. However, few genes involved in pyruvate to ethanol production pathway have been unambiguously identified. In T. saccharolyticum, the products of six putative pfor gene clusters and one pfl gene may be responsible for the conversion of pyruvate to acetyl-CoA. To gain insights into the physiological roles of PFOR and PFL, we studied the effect of deletions of several genes thought to encode these activities. We found that that pyruvate ferredoxin oxidoreductase enzymemore » (PFOR) is encoded by the pforA gene and plays a key role in pyruvate dissimilation. We further demonstrated that pyruvate formate-lyase activity (PFL) is encoded by the pfl gene. Although the pfl gene is normally expressed at low levels, it is crucial for biosynthesis in T. saccharolyticum. In pforA deletion strains, pfl expression increased and was able to partially compensate for the loss of PFOR activity. Deletion of both pforA and pfl resulted in a strain that required acetate and formate for growth and produced lactate as the primary fermentation product, achieving 88 % theoretical lactate yield. PFOR encoded by Tsac_0046 and PFL encoded by Tsac_0628 are only two routes for converting pyruvate to acetyl-CoA in T. saccharolyticum. The physiological role of PFOR is pyruvate dissimilation, whereas that of PFL is supplying C1 units for biosynthesis.« less

  5. Performance and emissions of non-petroleum fuels in a direct-injection stratified charge Sl engine

    SciTech Connect (OSTI)

    Freeman, L.E.; Chui, G.K.; Roby, R.J.

    1982-10-01

Seven fuels derived from coal and shale resources were evaluated using a direct-injection stratified charge engine. The fuels were refined to different degrees which ranged from those typical of gasoline blending components to those similar to current gasoline. Results showed that fuels refined to have properties similar to gasoline performed like gasoline. The less refined fuels were limited in performance. The total carbon monoxide and the hydrocarbon emissions varied with the volatility of the fuels. Most fuels with a higher overall distillation curve generally gave higher hydrocarbon and carbon monoxide emissions. The NOx emissions increased with the percent aromatics in the fuels. The hydrocarbon emissions were found to increase with fuel viscosity. Within the range of engine operation, nearly all the fuels evaluated gave satisfactory performance. With some modifications, even the less refined fuels can be potentially suitable for use in this engine.

    2011-01-01

The magnetic phase diagram of CuCrO2 is constructed as a function of magnetic field and anisotropy using a trial spin state built from harmonics of a fundamental ordering wavevector. Whereas the multiferroic phase of CuCrO2 is a modified spin spiral with a 3-sublattice (SL) period, the phase diagram also contains 1-SL, 2-SL, 4-SL, and 5-SL collinear states which may be accessi- ble in the nonstoichiometric compound CuCrO2+ . For small anisotropy, CuCrO2 is predicted to undergo a transition between two modified spiral states with an intervening 3-SL collinear phase.

/kWhe , which achieved the Phase 2 Go/No Go target of less than 0.12¢/kWhe. Abengoa Solar has high confidence that the primary risk areas have been addressed in the project and a commercial plant utilizing molten salt is economically and technically feasible. The strong results from the Phase 1 and 2 research, testing, and analyses, summarized in this report, led Abengoa Solar to recommend that the project proceed to Phase 3. However, a commercially viable collector interconnection was not fully validated by the end of Phase 2, combined with the uncertainty in the federal budget, forced the DOE and Abengoa Solar to close the project. Thus the resources required to construct and operate a molten salt pilot plant will be solely supplied by Abengoa Solar.

  17. Critical Anisotropies of a Geometrically-Frustrated Triangular-Lattice

    SciTech Connect (OSTI)

Swanson, Mason R; Haraldsen, Jason T; Fishman, Randy Scott

    2009-01-01

This work examines the critical anisotropy required for the local stability of the collinear ground states of a geometrically-frustrated triangular-lattice antiferromagnet (TLA). Using a Holstein-Primakoff expansion, we calculate the spin-wave frequencies for the 1, 2, 3, 4, and 8-sublattice (SL) ground states of a TLA with up to third neighbor interactions. Local stability requires that all spin-wave frequencies are real and positive. The 2, 4, and 8-SL phases break up into several regions where the critical anisotropy is a different function of the exchange parameters. We find that the critical anisotropy is a continuous function everywhere except across the 2-SL/3-SL and 3-SL/4-SL phase boundaries, where the 3-SL phase has the higher critical anisotropy.

  18. Conversion Tower for Dispatchable Solar Power: High-Efficiency Solar-Electric Conversion Power Tower

    SciTech Connect (OSTI)

    2012-01-11

HEATS Project: Abengoa Solar is developing a high-efficiency solar-electric conversion tower to enable low-cost, fully dispatchable solar energy generation. Abengoa’s conversion tower utilizes new system architecture and a two-phase thermal energy storage media with an efficient supercritical carbon dioxide (CO2) power cycle. The company is using a high-temperature heat-transfer fluid with a phase change in between its hot and cold operating temperature. The fluid serves as a heat storage material and is cheaper and more efficient than conventional heat-storage materials, like molten salt. It also allows the use of a high heat flux solar receiver, advanced high thermal energy density storage, and more efficient power cycles.

    2011-08-15

Flavivirus gene expression is modulated by RNA secondary structure elements at the terminal ends of the viral RNA molecule. For tick-borne encephalitis virus (TBEV), four stem-loop (SL) elements have been predicted in the first 180 nucleotides of the viral genome: 5'-SL1, 5'-SL2, 5'-SL3 and 5'-SL4. The last three of these appear to be unique to tick-borne flaviviruses. Here, we report their characterization by mutagenesis in a TBEV luciferase reporter system. By manipulating their thermodynamic properties, we found that an optimal stability of the 5'-SL2 is required for efficient RNA replication. 5'-SL3 formation is also important for viral RNA replication, but although it contains the viral start codon, its formation is dispensable for RNA translation. 5'-SL4 appears to facilitate both RNA translation and replication. Our data suggest that maintenance of the balanced thermodynamic stability of these SL elements is important for temporal regulation of its different functions.
