Crystal structure of the anti-viral APOBEC3G catalytic domain and functional implications

The APOBEC family members are involved in diverse biological functions. APOBEC3G restricts the replication of human immunodeficiency virus (HIV), hepatitis B virus and retroelements by cytidine deamination on single-stranded DNA or by RNA binding. Here we report the high-resolution crystal structure of the carboxy-terminal deaminase domain of APOBEC3G (APOBEC3G-CD2) purified from Escherichia coli. The APOBEC3G-CD2 structure has a five-stranded {beta}-sheet core that is common to all known deaminase structures and closely resembles the structure of another APOBEC protein, APOBEC2. A comparison of APOBEC3G-CD2 with other deaminase structures shows a structural conservation of the active-site loops that are directly involved in substrate binding. In the X-ray structure, these APOBEC3G active-site loops form a continuous 'substrate groove' around the active centre. The orientation of this putative substrate groove differs markedly (by 90 degrees) from the groove predicted by the NMR structure. We have introduced mutations around the groove, and have identified residues involved in substrate specificity, single-stranded DNA binding and deaminase activity. These results provide a basis for understanding the underlying mechanisms of substrate specificity for the APOBEC family.

The advent of the cooperating IP-enabled heterogeneous radio networks will offer, under the ďBeyond 3G Ē vision, ubiquitous services. In this new challenging context, a novel architecture has been designed within the control plane to avoid modification of existing networks. Therefore, a new entity in the network, a complementary middleware in the terminal, and enhanced signaling protocols were introduced. Robinson Unit 2 Pressure Vessel Benchmark is analyzed with a suite of Westinghouse-developed codes and data libraries. The radiation transport from the reactor core to the surveillance capsule and ex-vessel locations is performed by RAPTOR-M3G, a parallel deterministic radiation transport code that calculates high-resolution neutron flux information in three dimensions. The cross-section library used in this analysis is the ALPAN library, an Evaluated Nuclear Data File (ENDF)/B-VII.0-based library designed for reactor dosimetry and fluence analysis applications. Dosimetry is evaluated with the industry-standard SNLRML reactor dosimetry cross-section data library. (authors)

AbstractóWe proposed a new class of asymmetric turbo encoder for 3G systems that performs well in both ďwater fall Ē and ďerror floor Ē regions in [7]. In this paper, a modified (optimal) power allocation scheme for the different bits of new class of asymmetric turbo encoder has been investigated to enhance the performance. The simulation results and performance bound for proposed asymmetric turbo code with modified Unequal Power Allocation (UPA) scheme for the frame length, N=400, code rate, r=1/3 with Log-MAP decoder over Additive White Gaussian Noise (AWGN) channel are obtained and compared with the system with typical UPA and without UPA. The performance tests are extended over AWGN channel for different frame size to verify the possibility of implementation of the modified UPA scheme for the proposed asymmetric turbo code. From the performance results, it is observed that the proposed asymmetric turbo code with modified UPA performs better than the system without UPA and with typical UPA and it provides a coding gain of 0.4 to 0.52dB. An Analysis of the Broadband (22-3900 MHz) Radio Spectrum of HB3 (G132.7+1.3): The Detection of Thermal Radio Emission from an Evolved Supernova Remnant?

We present an analysis of the broadband radio spectrum (from 22 to 3900 MHz) of the Galactic supernova remnant (SNR) HB3 (G132.7+1.3). Published observations have revealed that a curvature is present in the radio spectrum of this SNR, indicating that a single synchrotron component appears is insufficient to adequately fit the spectrum. We present here a fit to this spectrum using a combination of a synchrotron component and a thermal bremsstrahlung component. We discuss properties of this latter component and estimate the ambient density implied by the presence of this component to be n \\~ 10 cm^-3. We have also analyzed extracted X-ray spectra from archived {\\it ASCA} GIS observations of different regions of HB3 to obtain independent estimates of the density of the surrounding interstellar medium (ISM). From this analysis, we have derived electron densities of 0.1-0.4 f^-1/2 cm^-3 for the ISM for the three different regions of the SNR, where f is the volume filling factor. By comparing these density estimates with the estimate derived from the thermal bremsstrahlung component, we argue that the radio thermal bremsstrahlung emission is emitted from a thin shell enclosing HB3. The presence of this thermal bremsstrahlung component in the radio spectrum of HB3 suggests that this SNR is in fact interacting with an adjacent molecular cloud associated with the HII region W3. By extension, we argue that the presence of thermal emission at radio wavelengths may be a useful tool for identifying interactions between SNRs and molecular clouds, and for estimating the ambient density near SNRs using radio continuum data.

Observers at rest in a stationary spacetime flat at infinity can measure small amounts of rest-mass+internal energies+kinetic energies+pressure energy in a small volume of fluid attached to a local inertial frame. The sum of these small amounts is the total "matter energy" for those observers. The total mass-energy minus the matter energy is the binding gravitational energy. Misner, Thorne and Wheeler evaluated the gravitational energy of a spherically symmetric static spacetime. Here we show how to calculate gravitational energy in any static and stationary spacetime for isolated sources with a set of observers at rest. The result of MTW is recovered and we find that electromagnetic and gravitational 3-covariant energy densities in conformastatic spacetimes are of opposite signs. Various examples suggest that gravitational energy is negative in spacetimes with special symmetries or when the energy-momentum tensor satisfies usual energy conditions.

Motivated and informed by perspectives on sustainability and design, this paper draws on a diverse body of scholarly works related to energy and materiality to articulate a perspective on energy-as-materiality and propose a design approach of materializing energy. Three critical themes are presented: the intangibility of energy, the undifferentiatedness of energy, and the availability of energy. Each theme is developed through combination of critical investigation and design exploration, including the development and deployment of several novel design artifacts: Energy Mementos and The Local Energy Lamp. A framework for interacting with energy-as-materiality is proposed involving collecting, keeping, sharing, and activating energy. A number of additional concepts are also introduced, such as energy attachment, energy engagement, energy attunement, local energy and energy meta-data. Our work contributes both a broader, more integrative design perspective on energy and materiality as well as a diversity of more specific concepts and artifacts that may be of service to designers and researchers of interactive systems concerned with sustainability and energy. This Transportation Energy Project is comprised of four unique tasks which work within the railroad industry to provide solutions in various areas of energy conservation. These tasks addressed: energy reducing yard related decision issues; alternate fuels; energy education, and energy storage for railroad applications. The NIU Engineering and Technology research team examined these areas and provided current solutions which can be used to both provide important reduction in energy usage and system efficiency in the given industry. This project also sought a mode in which rural and long-distance education could be provided. The information developed in each of the project tasks can be applied to all of the rail companies to assist in developing efficiencies.

Magma energy

The thermal energy contained in magmatic systems represents a huge potential resource. In the US, useful energy contained in molten and partially-molten magma within the upper 10 km of the crust has been estimated at 5 to 50 x 10/sup 22/ J (50,000 to 500,000 Quads). The objective of the Magma Energy Extraction Program is to determine the engineering feasibility of locating, accessing, and utilizing magma as a viable energy resource. This program follows the DOE/OBES-funded Magma Energy Research Project that concluded scientific feasibility of the magma energy concept. A primary long-range goal of this program is to conduct an energy extraction experiment directly in a molten, crustal magma body. Critical to determining engineering feasibility are several key technology tasks: (1) Geophysics - to obtain detailed definition of potential magma targets, (2) Geochemistry/Materials - to characterize the magma environment and select compatible engineering materials, (3) Drilling - to develop drilling and completion techniques for entry into a magma body, and (4) Energy Extraction - to develop heat extraction technology.
