    The Midlatitude Continental Convective Clouds Experiment (MC3E) will take place in central Oklahoma during the April–May 2011 period. The experiment is a collaborative effort between the U.S. Department of Energy (DOE) Atmospheric Radiation Measurement (ARM) Climate Research Facility and the National Aeronautics and Space Administration’s (NASA) Global Precipitation Measurement (GPM) mission Ground Validation (GV) program. The field campaign leverages the unprecedented observing infrastructure currently available in the central United States, combined with an extensive sounding array, remote sensing and in situ aircraft observations, NASA GPM ground validation remote sensors, and new ARM instrumentation purchased with American Recovery and Reinvestment Act funding. The overarching goal is to provide the most complete characterization of convective cloud systems, precipitation, and the environment that has ever been obtained, providing constraints for model cumulus parameterizations and space-based rainfall retrieval algorithms over land that have never before been available.

    Convective processes play a critical role in the Earth’s energy balance through the redistribution of heat and moisture in the atmosphere and subsequent impacts on the hydrologic cycle. Global observation and accurate representation of these processes in numerical models is vital to improving our current understanding and future simulations of Earth’s climate system. Despite improvements in computing power, current operational weather and global climate models are unable to resolve the natural temporal and spatial scales that are associated with convective and stratiform precipitation processes; therefore, they must turn to parameterization schemes to represent these processes. In turn, the physical basis for these parameterization schemes needs to be evaluated for general application under a variety of atmospheric conditions. Analogously, space-based remote sensing algorithms designed to retrieve related cloud and precipitation information for use in hydrological, climate, and numerical weather prediction applications often rely on physical “parameterizations” that reliably translate indirectly related instrument measurements to the physical quantity of interest (e.g., precipitation rate). Importantly, both spaceborne retrieval algorithms and model convective parameterization schemes traditionally rely on field campaign data sets as a basis for evaluating and improving the physics of their respective approaches. MC3E science objectives were motivated by the need to improve understanding of midlatitude continental convective cloud system lifecycles, microphysics, and GPM precipitation retrieval algorithms. To achieve these objectives a multi-scale surface- and aircraft-based in situ and remote sensing observing strategy was employed. A variety of cloud and precipitation events were sampled during the MC3E, of which results from three deepmore » convective events are highlighted. Vertical structure, air motions, precipitation drop-size distributions and ice properties were retrieved from multi-wavelength radar, profiler, and aircraft observations for an MCS on 11 May. Aircraft observations for another MCS observed on 20 May were used to test agreement between observed radar reflectivities and those calculated with forward-modeled reflectivity and microwave brightness temperatures using in situ particle size distributions and ice water content. Multi-platform observations of a supercell that occurred on 23 May allowed for an integrated analysis of kinematic and microphysical interactions. A core updraft of 25 ms-1 supported growth of hail and large rain drops. As a result, data collected during the MC3E campaign is being used in a number of current and ongoing research projects and is available through the DOE ARM and NASA data archives.« less

    This study attempts to understand interactions between midlatitude convective systems and their environments through a heat and moisture budget analysis using the sounding data collected from the Midlatitude Continental Convective Clouds Experiment (MC3E) in central Oklahoma. Distinct large-scale structures and diabatic heating and drying profiles are presented for cases of weaker and elevated thunderstorms as well as intense squall line and supercell thunderstorm events during the campaign. The elevated cell events were nocturnal convective systems occurring in an environment having low convective available potential energy (CAPE) and a very dry boundary layer. In contrast, deeper convective events happened during themore » morning into early afternoon within an environment associated with large CAPE and a near-saturated boundary layer. As the systems reached maturity, the diagnosed diabatic heating in the latter deep convective cases was much stronger and of greater vertical extent than the former. Both groups showed considerable diabatic cooling in the lower troposphere, associated with the evaporation of precipitation and low-level clouds. The horizontal advection of moisture also played a dominant role in moistening the lower troposphere, particularly for the deeper convective events, wherein the near surface southeasterly flow allows persistent low-level moisture return from the Gulf of Mexico to support convection. The moisture convergence often was present before these systems develop, suggesting a strong correlation between the large-scale moisture convergence and convection. As a result, sensitivity tests indicated that the uncertainty in the surface precipitation and the size of analysis domain mainly affected the magnitude of these analyzed fields rather than their vertical structures.« less

In particular, there has been recent focus on improving representations of ice collection processes (e.g., aggregation, riming), as these influence precipitation rate, heating profiles, and ultimately cloud life cycles. However, distinguishing these processes using conventional polarimetric radar observations is difficult, as they produce similar fingerprints. This necessitates improved analysis techniques and integration of complementary data sources. Furthermore, the Midlatitude Continental Convective Clouds Experiment (MC3E) provided such an opportunity.

    Recently the Radar Meteorology Group at Colorado State University has completed major case studies of some top cases from MC3E including 25 April, 20 May and 23 May 2011. A discussion on the analysis methods as well as radar quality control methods is included. For each case, a brief overview is first provided. Then, multiple Doppler (using available X-SAPR and C-SAPR data) analyses are presented including statistics on vertical air motions, sub-divided by convective and stratiform precipitation. Mean profiles and CFAD's of vertical motion are included to facilitate comparison with ASR model simulations. Retrieved vertical motion has also been verified with vertically pointing profiler data. Finally for each case, hydrometeor types are included derived from polarimetric radar observations. The latter can be used to provide comparisons to model-generated hydrometeor fields.     Despite the widespread importance of RING/U-box E3 ubiquitin ligases in ubiquitin (Ub) signaling, the mechanismby which this class of enzymes facilitates Ub transfer remains enigmatic. Here, we present a structural model for a RING/U-box E3:E2~Ub complex poised for Ub transfer. The model and additional analyses reveal that E3 binding biases dynamic E2~Ub ensembles toward closed conformations with enhanced reactivity for substrate lysines. We identify a key hydrogen bond between a highly conserved E3 side chain and an E2 backbone carbonyl, observed in all structures of active RING/ U-Box E3/E2 pairs, as the linchpin for allosteric activation of E2~Ub. The conformational biasing mechanism is generalizable across diverse E2s and RING/U-box E3s, but is not shared by HECT-type E3s. The results provide a structural model for a RING/ U-box E3:E2~Ub ligase complex and identify the long sought-after source of allostery for RING/UBox activation of E2~Ub conjugates.

    Blood coagulation factor VIII is a glycoprotein cofactor that is essential for the intrinsic pathway of the blood coagulation cascade. Inhibitory antibodies arise either spontaneously or in response to therapeutic infusion of functional factor VIII into hemophilia A patients, many of which are specific to the factor VIII C2 domain. The immune response is largely parsed into “classical” and “non-classical” inhibitory antibodies, which bind to opposing faces cooperatively. In this study, the 2.61 Å resolution structure of the C2 domain in complex with the antigen-binding fragment of the 3E6 classical inhibitory antibody is reported. The binding interface is largely conserved when aligned with the previously determined structure of the C2 domain in complex with two antibodies simultaneously. Further inspection of the B factors for the C2 domain in various X-ray crystal structures indicates that 3E6 antibody binding decreases the thermal motion behavior of surface loops in the C2 domain on the opposing face, thereby suggesting that cooperative antibody binding is a dynamic effect. Furthermore, understanding the structural nature of the immune response to factor VIII following hemophilia A treatment will help lead to the development of better therapeutic reagents.

    Bone morphogenetic proteins (BMPs) play important roles in osteoblast differentiation and maturation. In mammals, the BMP-induced receptor-regulated Smads form complexes with Smad4. These complexes translocate and accumulate in the nucleus, where they regulate the transcription of various target genes. However, the function of Smad4 remains unclear. We performed a yeast two-hybrid screen using Smad4 as bait and a cDNA library derived from bone marrow, to indentify the proteins interacting with Smad4. cDNA clones for Tumor necrosis factor (TNF) receptor-associated factor 2 (Traf2) were identified, and the interaction between the endogenous proteins was confirmed in the mouse osteoblast cell line MC3T3-E1. To investigate the function of Traf2, we silenced it with siRNA. The level of BMP-2 protein in the medium, the expression levels of the Bmp2 gene and BMP-induced transcription factor genes, including Runx2, Dlx5, Msx2, and Sp7, and the phosphorylated-Smad1 protein level were increased in cells transfected with Traf2 siRNA. The nuclear accumulation of Smad1 increased with TNF-{alpha} stimulation for 30 min at Traf2 silencing. These results suggest that the TNF-{alpha}-stimulated nuclear accumulation of Smad1 may be dependent on Traf2. Thus, the interaction between Traf2 and Smad4 may play a role in the cross-talk between TNF-{alpha} and BMP signaling pathways.

    The physics suite of the Community Atmosphere Model version 5 (CAM5) has recently been implemented in the Weather Research and Forecasting (WRF) model to explore the behavior of the parameterization suite at high resolution and in the more controlled setting of a limited area model. The initial paper documenting this capability characterized the behavior for northern high latitude conditions. This present paper characterizes the precipitation characteristics for continental, mid-latitude, springtime conditions during the Midlatitude Continental Convective Clouds Experiment (MC3E) over the central United States. This period exhibited a range of convective conditions from those driven strongly by large-scale synoptic regimes to more locally driven convection. The study focuses on the precipitation behavior at 32 km grid spacing to better anticipate how the physics will behave in the global model when used at similar grid spacing in the coming years. Importantly, one change to the Zhang-McFarlane deep convective parameterization when implemented in WRF was to make the convective timescale parameter an explicit function of grid spacing. This study examines the sensitivity of the precipitation to the default value of the convective timescale in WRF, which is 600 seconds for 32 km grid spacing, to the value of 3600 seconds used for 2 degree grid spacing in CAM5. For comparison, an infinite convective timescale is also used. The results show that the 600 second timescale gives the most accurate precipitation over the central United States in terms of rain amount. However, this setting has the worst precipitation diurnal cycle, with the convection too tightly linked to the daytime surface heating. Longer timescales greatly improve the diurnal cycle but result in less precipitation and produce a low bias. An analysis of rain rates shows the accurate precipitation amount with the shorter timescale is assembled from an over abundance of drizzle combined with too little heavy

    Sustainability is a critical national security issue for the U.S. and other nations. Sandia National Laboratories (SNL) is already a global leader in sustainability science and technology (SS&T) as documented in this report. This report documents the ongoing work conducted this year as part of the Sustainability Innovation Foundry (SIF). The efforts of the SIF support Sandia's national and international security missions related to sustainability and resilience revolving around energy use, water use, and materials, both on site at Sandia and externally. The SIF leverages existing Sandia research and development (R&D) in sustainability science and technology to support new solutions to complex problems. The SIF also builds on existing Sandia initiatives to support transformation of Sandia into a fully sustainable entity in terms of materials, energy, and water use. In the long term, the SIF will demonstrate the efficacy of sustainability technology developed at Sandia through prototyping and test bed approaches and will provide a common platform for support of solutions to the complex problems surrounding sustainability. Highlights from this year include the Sustainability Idea Challenge, improvements in facilities energy use, lectures and presentations from relevant experts in sustainability [Dr. Barry Hughes, University of Denver], and significant development of the Institutional Transformation (IX) modeling tools to support evaluation of proposed modifications to the SNL infrastructure to realize energy savings.

    We have measured the {sup 147}Sm(n,{alpha}) cross section from 3 eV to 500 keV. These data were used to test nuclear statistical models which must be relied on to calculate the rates for as yet unmeasurable reactions occurring in explosive nucleosynthesis scenarios. It was found that our data are in reasonably good agreement with the reaction rate predicted by an older model but that the rates predicted by two very recent models are roughly a factor of 3 different from the data (in opposite directions). A detailed analysis indicates the strong dependence on the employed optical {alpha} potentials. These results, together with counting rate estimates for future experiments indicate that (n,{alpha}) measurements will be useful for improving reaction rate predictions across the global range of masses needed for explosive nucleosynthesis calculations.

    Protein synthesis, or translation, consumes a sizable fraction of the cell’s energy budget, estimated at 5% and up to 50% in differentiated and growing cells, respectively. Plants also invest significant energy and biomass to construct and maintain the translation apparatus. Translation is regulated by a variety of external stimuli. Compared to transcriptional control, attributes of translational control include reduced sensitivity to stochastic fluctuation, a finer gauge of control, and more rapid responsiveness to environmental stimuli. Yet, our murky understanding of translational control allows few generalizations. Consequently, translational regulation is underutilized in the context of transgene regulation, although synthetic biologists are now beginning to appropriate RNA-level gene regulation into their regulatory circuits. We also know little about how translational control contributes to the diversity of plant form and function. This project explored how an emerging regulatory mRNA sequence element, upstream open reading frames (uORFs), is integrated with the general translation initiation machinery to permit translational regulation on specific mRNAs.

    The genes for aldolase-B (ALDOB), the [alpha]-microglobulin/bikunin precursor (AMBP), the paired box gene PAX5, and the anonymous DNA marker D9S3 map to human chromosome 9 (HSA9). The authors have set out to map the mouse homologues of each of these genes. The mouse genes for Pax-5 and Ambp previously been shown to map to MMU4. They have used an interspecific backcross to confirm these localizations and to map the mouse homologues of ALDOB (Aldo-2) and D9S3 (D4H9S3E) to the same chromosome. These genes were mapped with respect to the four anchor loci for MMU4. In addition, the panel of backcross DNAs had previously been typed for [delta]-amino levulinate dehydratase (Lv), orosomucoid-1 (Orm-1), and hexabranchion (Hxb), the human homologues of which map to HSA9q. The recombination distances [+-] the standard error between each pair of loci are D4Nds4-1.6[+-]1.1-D4H9S3E-4.0[+-]1.7 (Galt) 0.8[+-]0.8-Pax-5-4.8[+-]1.9-Aldo-2-6.3[+-]2.2-(Lv, Orm-1, Ambp)-1.6[+-]1.1-Hxb-4.0[+-]1.7-Tyrp-1-4.8[+-]1.9-Ifa. The data from this study have extended the known region of conserved synteny between human chromosome 9 and mouse chromosome 4. 17 refs., 2 figs.
