    Parabens are p-hydroxybenzoic acid esters that have been used extensively as preservatives in foods, cosmetics, drugs and toiletries. These intact esters are commonly detected in human breast cancer tissues and other human samples, thus arousing concern about the involvement of parabens in human breast cancer. In this study, an in vitro nuclear receptor coactivator recruiting assay was developed and used to evaluate the binding activities of parabens, salicylates and benzoates via antagonist competitive binding on the human oestrogen-related receptor ? (ERR?), which is known as both a diagnostic biomarker and a treatment target of breast cancer. The results showed that all of the test parabens (methyl-, ethyl-, propyl-, butyl- and benzylparaben) possessed clear inverse antagonist activities on ERR?, with a lowest observed effect level (LOEL) of 10{sup ?7} M and the 50% relative effective concentrations (REC50) varying from 3.09 10{sup ?7} to 5.88 10{sup ?7} M, whereas the salicylates possessed much lower activities and the benzoates showed no obvious activity. In silico molecular docking analyses showed that parabens fitted well into the active site of ERR?, with hydrogen bonds forming between the p-hydroxyl group of parabens and the Glu275/Arg316 of ERR?. As the paraben levels reported in breast cancer tissues are commonly higher than the LOELs observed in this study, parabens may play some role via ERR? in the carcinogenesis of human breast cancer. In addition, parabens may have significant effects on breast cancer patients who are taking tamoxifen, as ERR? is regarded as a treatment target for tamoxifen. - Highlights: An oestrogen-related receptor ? coactivator recruiting assay was developed.     One of the Plutonium Finishing Plant's (PFP) current Authorization Basis (AB) documents is the Final Safety Analysis Report (FSAR). This FSAR (HNF-SD-CP-SAR-02 1) was prepared to the format and content guidance specified in U.S. Nuclear Regulatory Commission (NRC) Regulatory Guide 3.39, Standard Format and Content of License Applications for Plutonium Processing and Fuel Fabrication Plants (RG 3.39). In April 1992, the US Department of Energy (DOE) issued DOE Order 5480.23 which established the FSAR requirements for DOE nonreactor nuclear facilities. In 1994, DOE issued DOE-STD-3009-94, Preparation Guide for US. Department of Energy Nonreactor Nuclear Facility Safety Analysis Reports, which is a format and content guide addressing the preparation of FSARs in accordance with DOE Order 5480.23. During the initial preparation and issuance of the PFP FSAR the format and content guidance contained in NRC Regulatory Guide 3.39 was utilized, since it was the most applicable guidance at the time for the preparation of Safety Analysis Reports for plutonium processing plants. With the adoption of DOE Order 5480.23 and DOE-STD-3009-94, DOE required the preparation of SARs to meet the format and content of those DOE documents. The PFP was granted an exemption to continue with RG 3.39 format for future FSAR revisions. PFP modifications and additions have required PFP FSAR modifications that have typically been prepared to the same NRC Regulatory Guide 3.39 format and content, to provide consistency with the PFP FSAR. This document provides a table comparison between the 3009 and RG 3.39 formats to validate the extent of PFP FSAR compliance with the intent of DOE Order 5480.23 and DOE-STD-3009-94. This evaluation was initially performed on Revisions 1 and 1A of the PFP FSAR.     Two standards have been developed that pertain to the safety of fusion facilities. These are DOE- STD-6002-96, Safety of Magnetic Fusion Facilities: Requirements, and DOE-STD-6003-96, Safety of Magnetic Fusion Facilities: Guidance. The first of these standards identifies requirements that subscribers to that standard must meet to achieve safety in fusion facilities. The second standard contains guidance to assist in meeting the requirements identified in the first This handbook provides additional documentation on good operations and design practices as well as lessons learned from the experiences of designers and operators of previous fusion facilities and related systems. It is intended to capture the experience gained in the various fields and pass it on to designers of future fusion facilities as a means of enhancing success and safety. The sections of this document are presented according to the physical location of the major systems of a fusion facility, beginning with the vacuum vessel and proceeding to those systems and components outside the vacuum vessel (the "Ex-vessel Systems"). The last section describes administrative procedures that cannot be localized to specific components.     Although worldwide demand for new construction of nuclear power stations has been on a decline, the available opportunities for the design and construction of qualified electrical penetrations continues to offer challenges, requiring a highly versatile design. Versatility is necessary in order to meet unique customer requirements within the constraints of a design basis qualified to IEEE Std. 317-1983. This paper summarizes such a versatile electrical penetration designed, built and tested to IEEE Std. 317-1983. The principal features are described including major materials of construction. Some of the design constraints such as sealing requirements, and conductor density (including numerical example) are discussed. The requirements for qualification testing of the penetration assembly to IEEE Std. 317-1983 are delineated in a general sense, and some typical test ranges for preconditioning, radiation exposure, and LOCA are provided. The paper concludes by describing ways in which this versatile design has been adapted to meet unique customer requirements in a variety of nuclear power plants.

    The United States Department of Energy (DOE) has published a standard that specifies the criteria for preparation and packaging of plutonium metals and oxides for safe long-term storage (DOE-STD-3013-96). This standard is followed for the packaging of materials resulting from the disassembly of nuclear weapons at Los Alamos National Laboratory under the Advanced Retirement and Integrated Extraction System (ARIES) project. Declassified plutonium metal or oxide material from the ARES project is packaged into doubly contained and welded type 304L stainless steel containers that comply with the DOE standard. The 3013-96 standard describes requirements for maximum contamination limits on the outer surface of the sealed inner container. These limits are 500 dpm per 100 cm2 for direct measurements and 20 dpm per 100 cm2 for removable contamination. For containers filled, welded, and handled inside a highly contaminated glovebox line, these limits are difficult to obtain. Simple handling within the line is demonstrated to contaminate surfaces from 10,000 to 10,000,000 dpm alpha per 100 cm2. To routinely achieve contamination levels below the maximum contamination levels specified by the 3013-96 standard within a processing operation, a decontamination step must be included. In the ARIES line, this decontamination step is an electrolytic process that produces a controlled uniform etch of the container surfaces. Decontamination of the 3013-96 compliant ARIES inner container is well demonstrated. Within 30 to 50 minutes electrolysis time, tixed contamination is reduced to hundreds of dpm generally occurring only at electrode contact points and welds. Removable contamination is routinely brought to non-detectable levels. The total process time for the cycle (includes electrolysis, rinse, and dry stages) is on the order of 1.5 to 2 hours per container.     The Surveillance and Monitoring Program (SMP) is a joint LANL/SRS effort funded by DOE/EM to provide the technical basis for the safe, long-term storage (up to 50 years) of over 6 metric tons of plutonium stored in over 5000 DOE-STD-3013 containers at various facilities around the DOE complex. The majority of this material is plutonium that is surplus to the nuclear weapons program, and much of it is destined for conversion to mixed oxide fuel for use in US nuclear power plants. The form of the plutonium ranges from relatively pure metal and oxide to very impure oxide. The performance of the 3013 containers has been shown to depend on moisture content and on the levels, types and chemical forms of the impurities. The oxide materials that present the greatest challenge to the storage container are those that contain chloride salts. The chlorides (NaCl, KCl, CaCl{sub 2}, and MgCl{sub 2}) range from less than half of the impurities present to nearly all the impurities. Other common impurities include oxides and other compounds of calcium, magnesium, iron, and nickel. Over the past 15 years the program has collected a large body of experimental data on over 60 samples of plutonium chosen to represent the broader population of materials in storage. This paper will summarize the characterization data, including the origin and process history, particle size, surface area, density, calorimetry, chemical analysis, moisture analysis, prompt gamma, gas generation and corrosion behavior.

    The 9977 is a certified Type B Packaging authorized to ship uranium and plutonium in metal and oxide forms. Historically, the standard container for these materials has been the DOE-STD-3013 which was specifically designed for the long term storage of plutonium bearing materials. The Department of Energy has used the 9975 Packaging containing a single 3013 container for the transportation and storage of these materials. In order to reduce container, shipping, and storage costs, the 9977 Packaging is being certified for transportation and storage of two 3013 containers.     This paper describes the approach taken by two Fluor Hanford projects for implementing of the seismic design criteria from DOE-STD-1189-2008, Appendix A. The existing seismic design criteria and the new seismic design criteria is described, and an assessment of the primary differences provided. The gaps within the new system of seismic design criteria, which necessitate conduct of portions of work to the existing technical standards pending availability of applicable industry standards, is discussed. Two Hanford Site projects currently in the Control Decision (CD)-1 phase of design have developed an approach to implementation of the new criteria. Calculations have been performed to determine the seismic design category for one project, based on information available in early CD-1. The potential effects of DOE-STD-1189-2008, Appendix A seismic design criteria on the process of project alternatives analysis is discussed. Present of this work is expected to benefit others in the DOE Complex that may be implementing DOE-STD-1189-2008.

    Diets containing coho salmon (Oncorhynchus kisutch Walbaum) from the Pacific Ocean or from Lakes Erie, Michigan, and Ontario (containing a gradation from low to high of halogenated aromatic hydrocarbons, (HAHs)) were fed to C57B1/6 and DBA/2 mice. Following a 4-month dietary exposure to Lake Ontario salmon, both strains of mice demonstrated hepatomegaly. The ethoxyresorufin-O-deethylase (ERR) enzyme levels were elevated in livers of C57B1/6 mice fed diets of salmon from all of the Great Lakes studied, with exceptionally high levels detected in C57B1/6 mice fed Lake Ontario salmon. Induction of ERR enzyme levels was detected in DBA/2 mice only following dietary exposure to Lake Ontario salmon. Serum levels of L-thyroxine (T4) and triiodo-L-thryonine (T3) were suppressed in C57B1/6 mice following consumption of Lake Ontario coho salmon, but T3 and T4 levels remained unchanged in DBA/2 mice. In general, pathobiological effects correlated with both dietary HAH exposure level and Ah receptor status.

    Three years ago there were no standards or published guidelines for quality in research environments. Today, one standard has been published, and three guidelines documents are in final draft form and about to be published. In this paper, I describe the events that led to the writing of DOE 5700.6C, 10CFR830.120, and DOE-ER-STD-6001-92, focusing on the cultural barriers that arose (largely in the community of quality assurance professionals) during this process. I go on to describe why I believe that implementing DOE 5700.6C and 10CFR830.120 must be pushed even further toward an approach that embodies the Malcolm Baldrige National Quality Award and why even this is not far enough. The reason is because the most crucial aspect of successfully implementing a quality initiative is to base it on a cohesive, unified foundation of organizational and individual values and beliefs. Stephen Covey`s 7 Habits of Highly Effective People and Principle Centered Leadership provide such a foundations.

Suggesting CB2R as a target to treat diseases with lipid dysregulation. -- Abstract: Abnormal fatty acid oxidation has been associated with obesity and type 2 diabetes. At the transcriptional level, peroxisome proliferator-activated receptor-gamma coactivator 1? (PGC-1?) has been reported to strongly increase the ability of hormone nuclear receptors PPAR? and ERR? to drive transcription of fatty acid oxidation enzymes. In this study, we report that a specific agonist of the type 2 cannabinoid receptor (CB2R) can lead to fatty acid oxidation through the PGC-1? pathway. We have found that CB2R is expressed in differentiated C2C12 myotubes, and that use of the specific agonist trans-caryophyllene (TC) stimulates sirtuin 1 (SIRT1) deacetylase activity by increasing the phosphorylation of cAMP response element-binding protein (CREB), thus leading to increased levels of PGC-1? deacetylation. This use of TC treatment increases the expression of genes linked to the fatty acid oxidation pathway in a SIRT1/PGC-1?-dependent mechanism and also drastically accelerates the rate of complete fatty acid oxidation in C2C12 myotubes, neither of which occur when CB2R mRNA is knocked down using siRNA. These results reveal that activation of CB2R by a selective agonist promotes lipid oxidation through a signaling/transcriptional pathway. Our findings imply that pharmacological manipulation of CB2R may provide therapeutic possibilities to treat metabolic diseases associated with lipid dysregulation.

    Hypoxia-inducible factors (HIFs) are differentially regulated in tumor cells. While the current paradigm supports post-translational regulation of the HIF-α subunits, we recently showed that hypoxic HIF-2α is also transcriptionally regulated via insulin-like growth factor (IGF)-II in the childhood tumor neuroblastoma. Here, we demonstrate that transcriptional regulation of HIF-2α seems to be restricted to neural cell-derived tumors, while HIF-1α is canonically regulated at the post-translational level uniformly across different tumor forms. Enhanced expression of HIF2A mRNA at hypoxia is due to de novo transcription rather than increased mRNA stability, and chemical stabilization of the HIF-α proteins at oxygen-rich conditions unexpectedly leads to increased HIF2A transcription. The enhanced HIF2A levels do not seem to be dependent on active HIF-1. Using a transcriptome array approach, we identified members of the Peroxisome proliferator-activated receptor gamma coactivator (PGC)/Estrogen-related receptor (ERR) complex families as potential regulators of HIF2A. Knockdown or inhibition of one of the members, ERRα, leads to decreased expression of HIF2A, and high expression of the ERRα gene ESRRA correlates with poor overall and progression-free survival in a clinical neuroblastoma material consisting of 88 tumors. Thus, targeting of ERRα and pathways regulating transcriptional HIF-2α are promising therapeutic avenues in neuroblastoma. - Highlights: • Transcriptional control of HIF-2α is restricted to neural cell-derived tumors. • Enhanced transcription of HIF2A is not due to increased mRNA stability. • Chemical stabilization of the HIF-α subunits leads to increased HIF2A transcription. • ERRα regulates HIF2A mRNA expression in neuroblastoma. • High expression of ESRRA correlates to poor outcome in neuroblastoma.

    This report provides the final hazard categorization (FHC) for the remediation of six solid waste disposal sites (referred to as burial grounds) located in the 300-FF-2 Operable Unit (OU) on the Hanford Site. These six sites (618-1, 618-2, 618-3, 618-7, 618-8, and 618-13 Burial Grounds) were determined to have a total radionuclide inventory (WCH 2005a, WCH 2005d, WCH 2005e and WCH 2006b) that exceeds the DOE-STD-1027 Category 3 threshold quantity (DOE 1997) and are the subject of this analysis. This FHC document examines the hazards, identifies appropriate controls to manage the hazards, and documents the FHC and commitments for the 300-FF-2 Burial Grounds Remediation Project.

    A principal focus of Task 2 under this project was for ORNL to evaluate the basis for susceptibility testing against high-frequency conducted interference and to establish recommendations to resolve concerns about the severity of test limits for the conducted susceptibility (CS) test, CS114, from MIL-STD-461. The primary concern about the test limit has been characterized by the EPRI EMI Working Group in the following terms: Demonstrating compliance with the CS114 test limits recommended in TR-102323 has proven to be problematic, even for components that have been tested to commercial standards and demonstrated proper operation in industrial applications [6]. Specifically, EPRI notes that the CS114 limits approved in regulatory documents are significantly higher than those invoked by the US military and similar commercial standards in the frequency range below 200 kHz. For this task, ORNL evaluated the original approach to establishing the test limit, EPRI technical findings from a review of the limit, and the regulatory basis through which the currently approved limits were accepted. Based on this analysis, strategies have been developed regarding changes to the CS114 limit that can resolve the technical concerns raised by the industry. Guided by the principles that reasonable assurance of safety must not be compromised but excessive conservatism should be reduced, recommendations on a suitable basis for a revised limit have been developed and can be incorporated into the planned Revision 2 of RG 1.180.

  12. Modeling CO{sub 2}-Brine-Rock Interaction Including Mercury and H{sub 2}S Impurities in the Context of CO{sub 2} Geologic Storage

    This study uses modeling and simulation approaches to investigate the impacts on injectivity of trace amounts of mercury (Hg) in a carbon dioxide (CO{sub 2}) stream injected for geologic carbon sequestration in a sandstone reservoir at ~2.5 km depth. At the range of Hg concentrations expected (7-190 ppbV, or ~ 0.06-1.6 mg/std.m{sup 3}CO{sub 2}), the total volumetric plugging that could occur due to complete condensation of Hg, or due to complete precipitation of Hg as cinnabar, results in a very small porosity change. In addition, Hg concentration much higher than the concentrations considered here would be required for Hg condensation to even occur. Concentration of aqueous Hg by water evaporation into CO{sub 2} is also unlikely because the higher volatility of Hg relative to H{sub 2}O at reservoir conditions prevents the Hg concentration from increasing in groundwater as dry CO{sub 2} sweeps through, volatilizing both H{sub 2}O and Hg. Using a model-derived aqueous solution to represent the formation water, batch reactive geochemical modeling show that the reaction of the formation water with the CO{sub 2}-Hg mixture causes the pH to drop to about 4.7 and then become buffered near 5.2 upon reaction with the sediments, with a negligible net volume change from mineral dissolution and precipitation. Cinnabar (HgS(s)) is found to be thermodynamically stable as soon as the Hg-bearing CO{sub 2} reacts with the formation water which contains small amounts of dissolved sulfide. Liquid mercury (Hg(l)) is not found to be thermodynamically stable at any point during the simulation. Two-dimensional radial reactive transport simulations of CO{sub 2} injection at a rate of 14.8 kg/s into a 400 m-thick formation at isothermal conditions of 106°C and average pressure near 215 bar, with varying amounts of Hg and H{sub 2}S trace gases, show generally that porosity changes only by about ±0.05% (absolute, i.e., new porosity = initial porosity ±0.0005) with Hg predicted to readily
