TCR chain repertoire of peripheral T cells is generated through the stepwise assembly and subsequent selection of TCR
V region exons during thymocyte development. To evaluate the influence of a two-step recombination process on V rearrange-
ment and selection, we generated mice with a preassembled D 1J 1.1 complex on the J 1 allele, an endogenous TCR allele that
lacks the D 2-J 2 cluster, creating the J 1DJ
allele. As compared with J 1 /
