1 Introduction
TheLAGLIDADGproteinfamilyincludesthefirstidentifiedandbiochemical-
ly characterizedintron-encodedproteins(Dujon 1980;Lazowskaet al. 1980;
|acquierand Dujon 1985),asdescribedin this volumeby Dujon.It hasbeen
variouslytermedthe'DOD', todecapeptide','dodecamer',and'decapeptide'
endonucleasefamily,basedontheconservationof aten-residuesequencemo-
tif(Dujon 1989;Dujonetal.1989;Belfortetal.1995;BelfortandRoberts1997;
Dalgaardetal.1997;chevalierandstoddard2001).TheLAGLIDADGendonu-
cleasesarethemostdiverseof thehomingendonucleasefamilies.Theirhost
rangeincludesthe genomesof plant and algalchloroplasts,fungaland pro-
tozoanmitochondria,bacteriaandArchaea(Dalgaardet al.1997).Onerea-
sonfor the wide phylogeneticdistributionof LAGLIDADGgenesappearsto
betheir remarkableabilityto invadeunrelatedtypesof interveningsequenc-
es,includinggroupI introns,archaealintronsand inteins(Belfortand Rob-
erts 1997;Chevalierand Stoddard2001).Descendentsof LAGLIDADGhom-
ing endonucleasesalsoincludetheyeastHO matingtype switchendonucle-
ase(Jinetal.1997),whichisencodedby anindependentreadingframerather
thanwithin an intron,but doescarry remnantsof an inactiveintein domain
