Use ACS grade chemicals. Use Sigma Trizma Tris Base and Tris-HCl for all buffers.
To check pH, dilute 10X stock solution to 1X. Use calibrated pH meter. pH of 1X phosphate buffers
should be 7.2-7.3. pH of 1X Tris buffers should be 7.5 or 8. The pH of 10X stocks will be different.
PBS: 1 L of 10X 1X Concentrations pH Adjustment
2.72 g KH2PO4 2 mM pH of 10X PBS will be ~6.6
11.36 g Na2HPO4 8 mM Adjust pH of 10X to 6.8
1.86 g KCl 2.5 mM (using KOH or NaOH)
81.8 g NaCl 140 mM pH of 1X PBS = 7.2-7.3
TBS: 1 L of 10X
19.6 g Tris-HCl 20 mM Tris
9.2 g Tris Base
81.8 g NaCl 140 mM
Final pH of 1X = ~8.0
IP buffer: 100 ml of 10 X
6.4 g Tris-HCl 50 mM Tris
1.2 g Tris base
8.8 g NaCl 150 mM
2 ml 0.5 M EDTA, pH 8.0 1 mM
