Fixing and Drying PAGE Gels
1. Fixation
Gel MeOH EtOH AcCOOH Time
SDS-PAGE 40% 10% 30 minutes
Urea PAGE 20% 5% 30 minutes
Fix until gel floats off glass plate. Gently push
waves of fixative over gel to help remove urea.
2. Post-Fixation
Gel Comments
SDS-PAGE Soak twice in ddH2O for 15 minutes.
Soak SDS-PAGE gel in 1-2% glycerol (prevents cracking).
Urea PAGE Do not soak in water or glycerol.
3. Check trap and pump: If trap is full, gel will dry slowly or not at all. Clean if necessary.
4. Dry PAGE gel
SDS-PAGE Gel Urea PAGE Gel
Place plastic wrap on bench. Place 3MM paper over gel in fixative.
Place gel face down on plastic wrap. Lift out glass plate, gel, paper, and drain.
Place a piece of wet 3MM paper on gel. Place on metal screen and invert quickly.
Place white porous sheet on top. Place on absorbant pad. Cut away excess.
