DNA adducts and genotypes of drug metabolizing enzymes in workers exposed to PAH and styrene

DNA adducts were measured by the postlabelling assay from lymphocytes or white blood cells of workers exposed to PAHs or styrene. The PAH-exposed groups included chimney sweeps, bus maintenance workers and truck terminal workers. Increased levels of adducts were noted in all the groups. In the sweeps the non-inducible CYP1A1 genotype, lacking a MSpI restriction site, and those lacking glutathine transferase mu had elevated levels of adducts. In bus maintenance and terminal workers no such relationship was noted. A postlabelling method was developed to measure specifically O{sup 6}-styrene oxide-guanine adducts. The levels in hand lamination workers around 5 adducts per 10{sup 8} normal nucleotides. In lymphocytes the adducts appeared to be very stable, as a summer vacation hardly affected the level of adducts in the workers.
