Degradation of delighted sprucewood by purified mannanase, xylanase and cellulases
The actions of endo-beta-1,4-mannanase (I) of Aspergillus niger and endo-beta-1,4-xylanase (II), cellulose (III) and cellobiohydrolase (IV) from Trichoderma viride on sprucewood holocellulose were studied. Mannan (V) was approximately 1/4 hydrolyzed in 2-3 day by I without xylan (VI) or cellulose (VII) degradation, whereas approximately 1/2 VI was degraded by II with 10% V solubilization, and less than or equal to 45% VII and less than or equal to 20% VI were hydrolyzed by III and IV with 40-70% V solubilization. Combined I-II treatment degraded approximately 1/2 V and VI, and addition of I to III increased the degradation of V and VII. Micromorphological studies of variously treated specimens showed a loss of substances in the primary, secondary and tertiary wall regions.
