ON OXIDATIVE METABOLISM IN STAPHYLOCOCCUS AUREUS AND A MUTANT OBTAINED BY ULTRAVIOLET IRRADIATION
The oxidative metabolism of Staphylococcus aureus-209 (St-209) and its mutant Staphylococcus aureus-UV-3 (StUV-3) was studied. The St-209 cells exhibited a drastic increase of oxygen uptake in the presence of glucose and oxidized also pyruvate, lactate, acetaldehyde, formalde hyde, ethanol, formic acid, oxalacetic acid, and malate. Under anaerobic conditions St-209 caused intensive decarboxylation of pyruvic and oxalacetic acid. Low concentrations of monoiodoacetate (0.2 mmol) completely inhibited glucose consumption and lactate accumulation by St-209. St-UV-3 did not show any increase in oxygen consumption in the presence of glucose but oxidized pyruvate, acetate, acetaldehyde, ethanol, oxalacetic acid, cis -aconitate, f umarate, succinate, and glyoxylate. Irregular malate and alpha -ketoglutarate oxidation was also noted. Under anaerobic conditions St-UV-3 only slightly decarboxylated pyruvic and oxalacetic acid. The Q/sub CO/sub 2//Qc/sub O/sub 2/ coefficient was more than 2 for pyrvate oxidation by St-209 while with St-UV-3 it approached 1. The effect was followed up of arsenite, malonate, cyanlde, and inorganic phosphate upon oxidation of some substrates. A discussion is presented of the pathways of oxidative transformations of respiratory substrates in St-209 and St-UV-3 as well as of the equivalency between the mutant and tumor tissue metabolism and of the possible mechanism of the effect of cancerostatic compounds upon St-UV-3 cells.
