Identification of radioproducts resulting from the breakage of thymine moiety by gamma irradiation of E. coli DNA in an aerated aqueous solution
E. coli DNA labeled with /sup 14/C on thymine moiety was submitted to gamma rays in an aerated aqueous solution. The products resulting from the breakage of the thymidylic fragment and which were split off from the sugar phosphate backbone were characterized. Seven compounds were identified: thymine, pyruvamide, cis 5,6-dihydroxy-6,6-dihydro-thymine,. trans 5,6-dihydroxy- 5,6dihydro-thymine, 5-hydroxy-5-methyl hydantoin, 5-hydroxy- 5methyl-barbituric acid, N-formyl N'-pyruvyl urea. Modified nuclesosides did not appear in any significant amount, and thymidine was not obtained. All these components were characterized in two bidimensional chromatographic solvents systems on silicagel amd cellulose plates. Microreactions were performed in order to confirm the chromatographic results. The release of the radioproducts in the solution was measured versus the dose applied. The main component at the beginning of the irradiation was thymine and at the end was pyruvamide. (auth)
